Semi-quantitative and visual assay of copper ions by fluorescent test paper constructed with dual-emission carbon dots.

ABSTRACT: A novel, simple and effective dual-emissive fluorescent probe for the sensitive and selective detection of Cu(ii) has been developed by mixing blue carbon dots and orange carbon dots, with a sensitive detection limit of 7.31 nM. The blue fluorescence can be selectively quenched by Cu(ii), while the orange fluorescence is a internal reference, resulting in a distinguishable fluorescence color change from blue to orange under a UV lamp. Meanwhile, its as-prepared text paper provides a convenient and simple approach for the visual detection of Cu(ii) and successfully applied in real water samples, with a dose-discerning ability as low as 50 nM. The methodology reported here opens a novel pathway toward the real applications of fluorescent test papers.

Project description:A simple and portable paper-based analytical device was developed for visual and semiquantitative detection of ferric ion in real samples using green emitting carbon dots (CDs), which were prepared via microwave method using sodium citrate, urea and sodium hydroxide as raw materials and then loaded on the surface of paper substrate. When Fe3+ exists, the green fluorescence of CDs was quenched and significant color change from green to dark blue were observed, resulting the visual detection of Fe3+ with a minimum distinguishable concentration of 100 μM. By analyzing the intensity changes of green channels of test paper with the help of smartphone, the semiquantitative detection was realized within the range of 100 μM to 1200 μM. The proposed paper-based analytical devices have great application prospects in on site detection of Fe3+ in real samples.

Quantitative Visual Detection of Mercury Ions With Ratiometric Fluorescent Test Paper Sensor.

Project description:A novel ratiometric fluorescence probe based on nitrogen-doped blue carbon dots (NCDs) and red gold nanoclusters (Au NCs) for mercuric ion (Hg2+) has been prepared and characterized. A user friendly fluorescent test paper based sensor combined with smartphone was fabricated for rapid visual and quantitative detection. Hg2+ can specifically bind to Au+ on the surface of Au NCs, leading to the quench of red fluorescence while the fluorescence intensity of the NCDs with blue fluorescence remained unchanged as a internal standard signal. The implement of paper-based sensor address some common drawback in analytical process such as the detection time, analysis cost. In a further demonstration, a homemade detection device with smartphone was used to qualify the Hg2+. After adding different concentration of Hg2+, red, purple, and blue colors were obtained on the detection zones of the fluorescent test paper. The Android App Color Grab was used to identify the red, green and blue (RGB) values of fluorescent color. The rapid visual and quantitative detection of Hg2+ was accomplished with the detection limit of 2.7 nM for fluorescence, 25 nM for smartphone and 32 nM for paper strip. The developed multi-mode detection platform was successfully applied to the detection of mercury ions in water samples with acceptable recoveries. The NCDs and Au NCs probe facilitate the one-site environmental monitoring for Hg2+ with "naked-eye" and smartphone.

Colorimetric and Fluorescent Sensing of Copper Ions in Water through o-Phenylenediamine-Derived Carbon Dots.

Project description:Fluorescent nitrogen and sulfur co-doped carbon dots (NSCDs) were synthesized using a simple one-step hydrothermal method starting from o-phenylenediamine (OPD) and ammonium sulfide. The prepared NSCDs presented a selective dual optical response to Cu(II) in water through the arising of an absorption band at 660 nm and simultaneous fluorescence enhancement at 564 nm. The first effect was attributed to formation of cuprammonium complexes through coordination with amino functional groups of NSCDs. Alternatively, fluorescence enhancement can be explained by the oxidation of residual OPD bound to NSCDs. Both absorbance and fluorescence showed a linear increase with an increase of Cu(II) concentration in the range 1-100 µM, with the lowest detection limit of 100 nM and 1 µM, respectively. NSCDs were successfully incorporated in a hydrogel agarose matrix for easier handling and application to sensing. The formation of cuprammonium complexes was strongly hampered in an agarose matrix while oxidation of OPD was still effective. As a result, color variations could be perceived both under white light and UV light for concentrations as low as 10 µM. Since these color changes were similarly perceived in tap and lake water samples, the present method could be a promising candidate for simple, cost-effective visual monitoring of copper onsite.

Ratiometric fluorescent test paper based on silicon nanocrystals and carbon dots for sensitive determination of mercuric ions.

Project description:Classical dye absorption-based pH test papers are widely used to measure the degree of acidity of media for quantification or semi-quantification by the observation of the naked eye on the variation of colour. However, it remains a challenging task to extend portable and cheap methods to a wide range of analytes for accurate quantification. Here, we report a dosage-sensitive test paper for the assay of mercury ions (Hg2+) with wide colour evolution. The ratiometric fluorescent probe was prepared by mixing blue-emission silicon nanocrystals (Si NCs) and red-emission carbon dots (r-CDs). The Si NCs serve as the reaction site and the r-CDs as the internal reference. Upon the addition of Hg2+, the blue fluorescence of the Si NCs was quenched, while the red fluorescence of the r-CDs remained constant, resulting in the consecutive fluorescence colour changes from blue to orange red. The probe demonstrates high selectivity and sensitivity of the visualization assay of Hg2+ with a detection limit of 7.63 nM. Moreover, we printed the ratiometric fluorescence probe result onto a piece of filter paper to prepare a test paper, which represents an on-site, sensitive, rapid and cost-effective method for the visual determination of Hg2+ by the naked eye.

A Simple and Effective Visual Fluorescent Sensing Paper-Based Chip for the Ultrasensitive Detection of Mercury Ions in Environmental Water.

Project description:Traces of mercury ions in environmental water can harm humans and animals. Paper-based visual detection methods have been widely developed for the rapid detection of mercury ions; however, existing methods are not sensitive enough to be used in real environments. Here, we developed a novel, simple and effective visual fluorescent sensing paper-based chip for the ultrasensitive detection of mercury ions in environmental water. CdTe-quantum-dots-modified silica nanospheres were firmly absorbed by and anchored to the fiber interspaces on the paper's surface to effectively avoid the unevenness caused by liquid evaporation. The fluorescence of quantum dots emitted at 525 nm can be selectively and efficiently quenched with mercury ions, and the ultrasensitive visual fluorescence sensing results attained using this principle can be captured using a smartphone camera. This method has a detection limit of 2.83 µg/L and a fast response time (90 s). We successfully achieved the trace spiking detection of seawater (from three regions), lake water, river water and tap water with recoveries in the range of 96.8-105.4% using this method. This method is effective, low-cost, user-friendly and has good prospects for commercial application. Additionally, the work is expected to be utilized in the automated big data collection of large numbers of environmental samples.

A novel highly fluorescent S, N, O co-doped carbon dots for biosensing and bioimaging of copper ions in live cells.

Project description:In this study, novel highly fluorescent sulfur, nitrogen, and oxygen co-doped carbon dots (S, N, O-CDs) were prepared from m-phenylenediamine and sulfamide by using the hydrothermal method. The prepared S, N, O-CDs show high doping rate and fluorescence yield as well as long-term fluorescence stability. In addition, S, N, O-CDs show good fluorescence response towards Cu2+ over a concentration range of 2-60 μM with a detection limit of 0.29 μM. Taking advantages of the properties of S, N, O-CDs including high selectivity and low cytotoxicity, S, N, O-CDs were successfully applied for Cu2+ sensing and imaging in the cells and O2˙--induced Cu2+ increase in the cells was observed. Furthermore, on account of strong complexation between Cu2+ and pyrophosphate ion (PPi) as well as specific hydrolysis ability of alkaline phosphatase (ALP) towards PPi, PPi and ALP were further detected with high selectivity based on S, N, O-CDs/Cu2+ system. The prepared S, N, O-CDs showed good detection results for PPi and ALP with detection limits of 0.44 μM and 1.03 U L-1, respectively. Moreover, the developed method also realized PPi and ALP detection in real samples.

Cyan Fluorescent Carbon Quantum Dots with Amino Derivatives for the Visual Detection of Copper (II) Cations in Sea Water.

Project description:Amino- and carboxyl-functionalized carbon quantum dots (Amino-CQDs) were synthesized through fast and simple microwave treatment of a citric acid, ethylenediamine and ethylenediaminetetraacetic acid (EDTA) mix. The reproducible and stable optical properties from newly synthesized CQD dispersion with a maximum absorbance spectra at 330 nm and the symmetric emission maximum at 470 nm made the Amino-CQDs a promising fluorescence material for analytical applications. The highly aminated and chelate moieties on the CQDs was appropriate for a copper (Cu2+) cation sensor in the linear range from 1 × 10-4 mg/mL to 10 mg/mL with a limit of detection at 0.00036 mg/mL by static fluorescence quenching effects. Furthermore, Amino-CQDs demonstrated stable fluorescence parameters for assays in diluted alkali metal solution (Na+ and K+) and sea water. Finally, a visual sensor, based on Amino-CQDs, was successfully created for the 0.01-100 mg/mL range to produce a colorimetric effect that can be registered by computer vision software (Open CV Python).

pH and redox dual-sensitive drug delivery system constructed based on fluorescent carbon dots.

Project description:Herein, a pH and redox dual-responsive drug delivery system (CDs-Pt(iv)-PEG) was developed based on fluorescence carbon dots (CDs). In this system, cisplatin(iv) prodrug (Pt(iv)) was selected as a model drug to reduce toxic side effects. The aldehyde-functionalized monomethoxy polyethylene glycol (mPEG-CHO) was conjugated to CDs-Pt(iv) to form pH sensitive benzoic imine bond. Owing to the slightly acidic tumor extracellular microenvironment (pH 6.8), the benzoic imine bond was then hydrolyzed, leading to charge reversal and decrease in the hydration radius of the drug-carrying, which facilitated in vivo circulation and tumor targeting. Notably, the cytotoxicity of the drug delivery system on cancer cells was comparable to that of cisplatin, while the side effects on normal cells were significantly reduced. In addition, the system realized recognition of cancer cells by the high-contrast fluorescent imaging. In conclusion, the CDs-Pt(iv)-PEG system provided a promising potential for effective delivery of anticancer drugs and cancer cells screening.

Highly Sensitive Ratiometric Fluorescent Paper Sensors for the Detection of Fluoride Ions.

Project description:Two sensitive and ratiometric fluorescent probes (probe I and probe II) were developed for the detection of fluoride ions. Probe I can detect fluoride ions quantitatively within a range of 0-6 μM and a detection limit of 73 nM, while probe II has a range of 0-40 μM and a detection limit of 138 nM. The test strips from probe I are quickly able to recognize F- (5 min) inside of the F- safety level in drinking water (1.0 mg/L, ∼5 μM) under 254 nm ultraviolet light, and the test strips from probe II quickly recognize F- (12 min) in dangerously high F- levels in water (4.0 mg/L, ∼21 μM) under 254 nm ultraviolet light. This combination of fluorescent paper sensors from probe I and probe II can be used as a simple and convenient tool to determine whether water is safe to drink or dangerous.

Cobalt-modulated dual emission carbon dots for ratiometric fluorescent vancomycin detection.

Project description:This work presents a simple yet selective fluorometric protocol for the quantification of vancomycin, an important antibiotic for treating infections caused by Gram-positive bacteria. A novel ratiometric fluorometric method for the determination of vancomycin is developed based on dual emissive carbon dots (DECDs) with emission at 382 nm and 570 nm in combination with Co2+ ions. Upon addition of Co2+ions, the fluorescence at 382 nm of DECDs is enhanced while emission at 570 nm remains constant. In the presence of vancomycin, it complexes with Co2+ leading to quenching of the 382 nm fluorescence due to strong binding with Co2+ in the Co@DECDs system. The DECDs are fully characterized by TEM and different spectroscopic techniques. The proposed ratiometric method is based on measuring fluorescence ratio (F570/F382) against vancomycin concentration and the method exhibits a good linearity range from 0.0 to 120.0 ng mL-1 with a low limit of detection (S/N = 3) of 0.31 ng mL-1. The method shows good selectivity with minimal interference from potential interfering species. This ratiometric fluorometric approach provides a promising tool for sensitive and specific vancomycin detection in clinical applications.
