Effect of dietary chitosan oligosaccharide supplementation on the pig ovary transcriptome.

ABSTRACT: Fecundity improvement is one of the most important economic traits for the swine industry as it significantly increases production efficiency. Intriguingly, chitosan oligosaccharide (COS), a biomaterial with an active amino group, could promote sow reproductive performance. Therefore, we investigated the effects of dietary COS supplementation on the gene expression differences in the ovaries of sows using the RNA-Seq method. This analysis obtained 13 960 051 and 14 564 863 clean reads in control ovary and COS ovary libraries, respectively. A total of 486 differentially expressed genes (DEGs) were thereby identified (FDR ≤ 0.001, |log₂ ratio| ≥ 1). There were 234 up-regulated and 252 down-regulated genes in the COS ovary samples compared with the control ovary samples. A large number of these DEGs were involved in the terms cellular process, cell & cell part and binding. Furthermore, pathway analysis indicated that these DEGs were significantly enriched in 34 Kyoto Encyclopedia of Genes and Genomes (KEGG) pathways, including cell cycle, progesterone-mediated oocyte maturation, metabolic pathways, oocyte meiosis, and hematopoietic cell lineage among others. These results provided the molecular mechanisms of using COS feed additive for improving sow litter size and prolificacy.

Project description:According to a previous study, we had found that early weaning causes harm to growth performance, intestinal morphology, activity of digestive enzymes, and antioxidant status in pigeon squabs (Columba livia). Chitosan oligosaccharides (COS) and Clostridium butyricum have been reported to have great potential to improve the growth performance and intestinal health of early-weaned animals. Therefore, the aim of this study is to explore whether dietary supplementation with COS-C. butyricum synbiotic could relieve early-weaned stress by evaluating its effects on growth performance and intestinal health in pigeon squabs. A total of 160 squabs (weaned at 7 days of age) were randomly divided into 5 groups: the control group, fed with artificial crop milk; the COS supplementation group, fed with artificial crop milk + 150 mg/kg COS; and three synbiotic supplementation groups, fed with artificial crop milk + 150 mg/kg COS + 200, 300, and 400 mg/kg C. butyricum. The results showed that a diet supplemented with COS-C. butyricum synbiotic benefitted the growth performance of early-weaned squabs; even so the differences were not significant among the five groups (p > 0.05). In addition, dietary supplementation of 150 mg/kg COS + 300~400 mg/kg C. butyricum significantly improved the intestinal morphology (especially villus surface area and the ratio of villus height to crypt depth), the activity of digestive enzymes (lipase, trypsin, and leucine aminopeptidase) in duodenum contents, and the production of total short-chain fatty acids and acetic acid in ileum content (p < 0.05). Additionally, dietary supplementation of 150 mg/kg COS + 400 mg/kg C. butyricum benefitted gut health by improving the antioxidant capacity (glutathione peroxidase and total antioxidant capacity) and cytokine status (IL-4 and IL-10) (p < 0.05), as well as by improving the intestinal microbiota diversity. In conclusion, our results revealed that dietary supplementation with synbiotic (150 mg/kg COS + 300~400 mg/kg C. butyricum) could relieve early-weaned stress by maintaining intestinal health in pigeon squabs.

Project description:Reproduction performance is one of the most important economic traits for the poultry industry. Intriguingly, apple pectic oligosaccharide (APO) could promote gastrointestinal function and immune function to improve performance; however, literature about APO on reproduction performance in breeders is limited. This study aimed to determine whether APO administration can improve reproduction performance and ovary function of broiler breeders with different egg laying rates. Two hundred and fifty six Arbor Acres broiler breeders (48-week-old) were used in a 2 × 2 factorial design with 2 egg laying rates (average [AR] and low [LR]) and 2 dietary levels of APO (0 and 200 mg/kg APO). Results showed that the LR breeders presented higher egg weight but lower egg laying rate, qualified egg rate, and feed efficiency than the AR breeders (P(laying) < 0.05). Also, the LR breeders had decreased serum Anti-Müllerian hormone, leptin, and antioxidant enzyme (superoxide dismutase, total antioxidant capacity) levels than the AR breeders (P(laying) ≤ 0.05). Dietary supplementation with APO improved egg weight, feed efficiency, as well as egg albumen quality (higher albumen height and Haugh unit) (P(APO) < 0.05), and decreased the concentration of pro-inflammatory cytokine levels (interleukin [IL]-1β, IL-8) in serum (P(APO) ≤ 0.05). The apoptosis rate and pro-apoptosis-related gene expression (caspase 9 and Bax) in the ovary of LR breeders were higher, while the anti-apoptosis-related gene expression (Bcl-2, PCNA) was lower in LR compared with the AR breeders (P(laying) < 0.05). Dietary supplementation with APO decreased the caspase 9 and Bax expression in LR breeders (P(interaction) < 0.05), and increased the Bcl-2 and PCNA expression in the 2 breeders (P(APO) < 0.05). These findings indicate that breeders with a lower egg laying rate exhibit lower antioxidant capacity and high cell apoptosis in the ovary. Dietary supplementation with APO might improve albumen quality and antioxidant capacity, and decrease the inflammatory factors and ovary apoptosis-related genes expression to improve ovary function. Moreover, the effect of APO on decreasing ovarian pro-apoptosis-related gene expression was more pronounced in lower reproductive breeders.
