C/N-Dependent Element Bioconversion Efficiency and Antimicrobial Protein Expression in Food Waste Treatment by Black Soldier Fly Larvae.

ABSTRACT: The black soldier fly (BSF), Hermetia illucens, has emerged as a promising species for waste bioconversion and source of antimicrobial proteins (AMPs). However, there is a scarcity of research on the element transformation efficiency and molecular characterization of AMPs derived from waste management. Here, food waste treatment was performed using BSF larvae (BSFL) in a C/N ratio of 21:1-10:1, with a focus on the C/N-dependent element bioconversion, AMP antimicrobial activity, and transcriptome profiling. The C-larvae transformation rates were found to be similar among C/Ns (27.0-35.5%, p = 0.109), while the N-larvae rates were different (p = 0.001), with C/N 21:1-16:1 (63.5-75.0%) being higher than C/N 14:1-10:1 (35.0-45.7%). The C/N ratio did not alter the antimicrobial spectrum of AMPs, but did affect the activities, with C/N 21:1 being significantly lower than C/N 18:1-10:1. The lysozyme genes were found to be significantly more highly expressed than the cecropin, defensin, and attacin genes in the AMP gene family. Out of 51 lysozyme genes, C/N 18:1 and C/N 16:1 up-regulated (p < 0.05) 14 and 12 genes compared with C/N 21:1, respectively, corresponding to the higher activity of AMPs. Overall, the element bioconversion efficiency and AMP expression can be enhanced through C/N ratio manipulation, and the C/N-dependent transcriptome regulation is the driving force of the AMP difference.

Project description:Significant economic, environmental, and social impacts are associated with the avoidable disposal of foods worldwide. Mass-rearing of black soldier fly (Hermetia illucens) larvae using organic wastes and food- and agro-industry side products is promising for recycling resources within the food system. One current challenge of this approach is ensuring a reliable and high conversion performance of larvae with inherently variable substrates. Research has been devoted to increasing rearing performance by optimizing substrate nutrient contents and ratios, while the potential of the substrate and larval gut microbiota to increase rearing performance remains untapped. Since previous research has focused on gut microbiota, here, we describe bacterial dynamics in the residue (i.e., the mixture of frass and substrate) of black soldier fly larvae reared on two food wastes (i.e., canteen and household waste). To identify members of the substrate and residue microbiota, potentially associated with rearing performance, bacterial dynamics were also studied in the canteen waste without larvae, and after inactivation by irradiation of the initial microbiota in canteen waste. The food waste substrates had similar microbiota; both were dominated by common lactic acid bacteria. Inactivation of the canteen waste microbiota, which was dominated by Leuconostoc, Bacillus, and Staphylococcus, decreased the levels of all rearing performance indicators by 31-46% relative to canteen waste with the native microbiota. In both food waste substrates, larval rearing decreased the bacterial richness and changed the physicochemical residue properties and composition over the rearing period of 12 days, and typical members of the larval intestinal microbiota (i.e., Providencia, Dysgonomonas, Morganella, and Proteus) became more abundant, suggesting their transfer into the residue through excretions. Future studies should isolate members of these taxa and elucidate their true potential to influence black soldier fly mass-rearing performance.

Project description:Mealworms and black soldier fly (BSF) larvae are two of the most reared insects at an industrial scale. Both may feed on by-products from agricultural and food industries. Feed efficiency is one of the most important aspects of such processes and varies between species and feed substrates and depends on the metabolic performance of the larvae. Compared to each other, both species may hold advantageous capabilities affecting their feed efficiency, likely depending on the feed substrate. We reared mealworms and BSF larvae on a diverse selection of by-products from agricultural and food industries, quantified major metabolic rates across their life spans, and compared their performances. The type of feed substrates had stronger effects on the growth of mealworms than on black soldier fly larvae. Generally, BSF larvae were advantageous in terms of the highest maximal specific growth rate (0.50-0.77 day-1) and feed assimilation rate (0.81-1.16 day-1) and shortest development period (23-25 days) but disadvantageous in terms of metabolic maintenance cost (0.07-0.21 day-1). In mealworms, the maximal specific growth rates were 0.02-0.11 day-1, the highest feed assimilation rates were 0.16-0.37 day-1, and the development period was at least 65-93 days, while maintenance was only 0.02-0.05 day-1. In contrast to the BSF larvae, the specific maintenance rate was weight dependent in the mealworms and lowest in the largest individuals. The combined outcome of these metabolic rates resulted in an average carbon net growth efficiency, NGE*avg of 0.16-0.40 in mealworms and 0.33-0.56 in BSF larvae across their life span. It thus seems that BSF larvae are more versatile and somewhat more efficient at converting diverse feed substrates into growth than mealworms. Differences in NGE*avg affected the substrate conversion efficiencies (i.e., the ratio of the weight gain of the larvae to the reduced weight of feed substrates) and may thus impact the overall outcome of insect farming.
