A Chinese multi-modal neuroimaging data release for increasing diversity of human brain mapping.

ABSTRACT: The big-data use is becoming a standard practice in the neuroimaging field through data-sharing initiatives. It is important for the community to realize that such open science effort must protect personal, especially facial information when raw neuroimaging data are shared. An ideal tool for the face anonymization should not disturb subsequent brain tissue extraction and further morphological measurements. Using the high-resolution head images from magnetic resonance imaging (MRI) of 215 healthy Chinese, we discovered and validated a template effect on the face anonymization. Improved facial anonymization was achieved when the Chinese head templates but not the Western templates were applied to obscure the faces of Chinese brain images. This finding has critical implications for international brain imaging data-sharing. To facilitate the further investigation of potential culture-related impacts on and increase diversity of data-sharing for the human brain mapping, we released the 215 Chinese multi-modal MRI data into a database for imaging Chinese young brains, namely'I See your Brains (ISYB)', to the public via the Science Data Bank ( https://doi.org/10.11922/sciencedb.00740 ).

Project description:Fusing complementary information from different modalities can lead to the discovery of more accurate diagnostic biomarkers for psychiatric disorders. However, biomarker discovery through data fusion is challenging since it requires extracting interpretable and reproducible patterns from data sets, consisting of shared/unshared patterns and of different orders. For example, multi-channel electroencephalography (EEG) signals from multiple subjects can be represented as a third-order tensor with modes: subject, time, and channel, while functional magnetic resonance imaging (fMRI) data may be in the form of subject by voxel matrices. Traditional data fusion methods rearrange higher-order tensors, such as EEG, as matrices to use matrix factorization-based approaches. In contrast, fusion methods based on coupled matrix and tensor factorizations (CMTF) exploit the potential multi-way structure of higher-order tensors. The CMTF approach has been shown to capture underlying patterns more accurately without imposing strong constraints on the latent neural patterns, i.e., biomarkers. In this paper, EEG, fMRI, and structural MRI (sMRI) data collected during an auditory oddball task (AOD) from a group of subjects consisting of patients with schizophrenia and healthy controls, are arranged as matrices and higher-order tensors coupled along the subject mode, and jointly analyzed using structure-revealing CMTF methods [also known as advanced CMTF (ACMTF)] focusing on unique identification of underlying patterns in the presence of shared/unshared patterns. We demonstrate that joint analysis of the EEG tensor and fMRI matrix using ACMTF reveals significant and biologically meaningful components in terms of differentiating between patients with schizophrenia and healthy controls while also providing spatial patterns with high resolution and improving the clustering performance compared to the analysis of only the EEG tensor. We also show that these patterns are reproducible, and study reproducibility for different model parameters. In comparison to the joint independent component analysis (jICA) data fusion approach, ACMTF provides easier interpretation of EEG data by revealing a single summary map of the topography for each component. Furthermore, fusion of sMRI data with EEG and fMRI through an ACMTF model provides structural patterns; however, we also show that when fusing data sets from multiple modalities, hence of very different nature, preprocessing plays a crucial role.

Project description:Alzheimer's disease (AD) and Parkinson's disease (PD) are influenced by genetic and environmental factors. Using data from UK Biobank, SAIL Biobank, and FinnGen, we conducted an unbiased, population-scale study to: 1) Investigate how 155 endocrine, nutritional, metabolic, and digestive system disorders are associated with AD and PD risk prior to their diagnosis, considering known genetic influences; 2) Assess plasma biomarkers' specificity for AD or PD in individuals with these conditions; 3) Develop a multi-modal classification model integrating genetics, proteomics, and clinical data relevant to conditions affecting the gut-brain axis. Our findings show that certain disorders elevate AD and PD risk before AD and PD diagnosis including: insulin and non-insulin dependent diabetes mellitus, noninfective gastro-enteritis and colitis, functional intestinal disorders, and bacterial intestinal infections, among others. Polygenic risk scores revealed lower genetic predisposition to AD and PD in individuals with co-occurring disorders in the study categories, underscoring the importance of regulating the gut-brain axis to potentially prevent or delay the onset of neurodegenerative diseases. The proteomic profile of AD/PD cases was influenced by comorbid endocrine, nutritional, metabolic, and digestive systems conditions. Importantly, we developed multi-modal prediction models integrating clinical, genetic, proteomic and demographic data, the combination of which performs better than any single paradigm approach in disease classification. This work aims to illuminate the intricate interplay between various physiological factors involved in the gut-brain axis and the development of AD and PD, providing a multifactorial systemic understanding that goes beyond traditional approaches.
