Project description:Prostate cancer cells control energy metabolism by chelating intracellular zinc. Thus, zinc delivery has been a popular therapeutic approach for prostate cancer. Here, we propose the use of the membrane-penetrating peptide Novicidin connected to zinc-Schiff base as a carrier vehicle for the delivery of zinc to prostate cells. Mass spectrometry, electrochemistry and spectrophotometry confirmed the formation/stability of this complex and provided insight regarding the availability of zinc for complex interactions. This delivery system showed minor toxicity in normal PNT1A cells and high potency towards PC3 tumor cells. The complex preferentially penetrated PC3 tumor cells in contrast to confinement to the membranes of PNT1A. Furthermore, zinc uptake was confirmed in both cell lines. Molecular analysis was used to confirm the activation of zinc stress (e.g., ZnT-1) and apoptosis (e.g., CASP-1). Our results strongly suggest that the zinc-Schiff base-Novicidin complex has great potential as a novel anticancer drug.

Project description:The synthesis of the new cofacial binuclear zinc complex [Zn(2)(L)] of a Schiff-base pyrrole macrocycle is reported. It was discovered that the binuclear microenvironment between the two metals of [Zn(2)(L)] is suited for the encapsulation of anions, leading to the formation of [K(THF)(6)][Zn(2)(μ-Cl)(L)]·2THF and [Bu(n)(4)N][Zn(2)(μ-OH)(L)] which were characterized by X-ray crystallography. Unusually obtuse Zn-X-Zn angles (X = Cl: 150.54(9)° and OH: 157.4(3)°) illustrate the weak character of these interactions and the importance of the cleft preorganization to stabilize the host. In the absence of added anion, aggregation of [Zn(2)(L)] was inferred and investigated by successive dilutions and by the addition of coordinating solvents to [Zn(2)(L)] solutions using NMR spectroscopy as well as isothermal microcalorimetry (ITC). On anion addition, evidence for deaggregation of [Zn(2)(L)], combined with the formation of the 1:1 host-guest complex, was observed by NMR spectroscopy and ITC titrations. Furthermore, [Zn(2)(L)] binds to Cl(-) selectively in THF as deduced from the ITC analyses, while other halides induce only deaggregation. These conclusions were reinforced by density functional theory (DFT) calculations, which indicated that the binding energies of OH(-) and Cl(-) were significantly greater than for the other halides.

Project description:BackgroundThe study was carried out to assess the gastroprotective effect of the zinc (II) complex against ethanol-induced acute hemorrhagic lesions in rats.Methodology/principal findingThe animals received their respective pre-treatments dissolved in tween 20 (5% v/v), orally. Ethanol (95% v/v) was orally administrated to induce superficial hemorrhagic mucosal lesions. Omeprazole (5.790×10(-5) M/kg) was used as a reference medicine. The pre-treatment with the zinc (II) complex (2.181×10(-5) and 4.362×10(-5) M/kg) protected the gastric mucosa similar to the reference control. They significantly increased the activity levels of nitric oxide, catalase, superoxide dismutase, glutathione and prostaglandin E2, and decreased the level of malondialdehyde. The histology assessments confirmed the protection through remarkable reduction of mucosal lesions and increased the production of gastric mucosa. Immunohistochemistry and western blot analysis indicated that the complex might induced Hsp70 up-regulation and Bax down-regulation. The complex moderately increased the gastroprotectiveness in fine fettle. The acute toxicity approved the non-toxic characteristic of the complex (<87.241×10(-5) M/kg).Conclusion/significanceThe gastroprotective effect of the zinc (II) complex was mainly through its antioxidant activity, enzymatic stimulation of prostaglandins E2, and up-regulation of Hsp70. The gastric wall mucus was also a remarkable protective mechanism.

Project description:A zinc(ii) complex, [ZnL(H2O)]·H2O {H2L = 2,2'-[(2,2-dimethyl-1,3-propanediyl)bis(nitrilomethylidyne)]bis[6-ethoxyphenol]} has been synthesized and characterized by UV-vis and IR spectroscopy. The structure of the complex has been confirmed by X-ray crystallography and the noncovalent interactions characterized using Hirshfeld surface analysis. In addition to the conventional H-bonds involving the Zn-coordinated and non-coordinated water molecules, interesting C-H⋯π interactions between the H-atoms belonging to aliphatic part of the ligand (2,2-dimethyl-1,3-propanediyl) and the Zn-coordinated aromatic rings are established. These interactions have been studied using DFT calculations (PBE0-D3/def2-TZVP) and characterized using molecular electrostatic potential (MEP) surfaces and the noncovalent interaction (NCI) plot index analyses.

Project description:Platinum electrodes were modified with polymers of the (±)-trans-N,N'-bis(salicylidene)-1,2-cyclohexanediaminenickel(II) ([Ni(salcn)]) and (±)-trans-N,N'-bis(3,3'-tert-Bu-salicylidene)-1,2-cyclohexanediaminenickel(II) ([Ni(salcn(Bu))]) complexes to study their electrocatalytic and electroanalytical properties. Poly[Ni(salcn)] and poly[Ni(salcn(Bu))]) modified electrodes catalyze the oxidation of catechol, aspartic acid and NO2-. In the case of poly[Ni(salcn)] modified electrodes, the electrocatalysis process depends on the electroactive surface coverage. The films with low electroactive surface coverage are only a barrier in the path of the reducer to the electrode surface. The films with more electroactive surface coverage ensure both electrocatalysis inside the film and oxidation of the reducer directly on the electrode surface. In the films with the most electroactive surface coverage, electrocatalysis occurs only at the polymer-solution interface. The analysis was based on cyclic voltammetry, EQCM (electrochemical quartz crystal microbalance) and rotating disc electrode method.

Project description:A novel nanocatalyst, denoted as UiO-66/Sal-ZnCl2, has been synthesized and systematically characterized employing a range of analytical techniques, including Fourier-transform infrared spectroscopy (FT-IR), X-ray diffraction (XRD), transmission electron microscopy (TEM), scanning electron microscopy (SEM), energy-dispersive X-ray spectroscopy (EDX), thermogravimetric analysis (TGA), Brunauer-Emmett-Teller (BET) surface area analysis, and inductively coupled plasma (ICP) analysis. The comprehensive analyses collectively affirm the effective coordination of zinc chloride onto the functionalized UiO-66. Subsequently, the catalytic efficacy of UiO-66/Sal-ZnCl2 was assessed in a one-pot, three-component click reaction involving terminal alkynes, alkyl halides, and sodium azide, conducted in an aqueous medium. The catalyst demonstrated remarkable catalytic activity, showcasing the capability to facilitate the reaction with high yields and exceptional regioselectivity. Noteworthy attributes of this nanocatalyst and the method include its elevated efficiency, recyclability, convenient product workup, and, significantly, the utilization of a sustainable solvent medium. The synthesis, characterization, and catalytic performance of this catalyst collectively contribute to its potential as an innovative and reusable nanocatalyst for diverse synthetic transformations.

Project description:The development of chemosensors able to detect analytes in a variety of sample matrices through a low-cost, fast, and direct approach is of current interest in food, health, industrial, and environmental fields. This contribution presents a simple approach for the selective and sensitive detection of Cu2+ ions in aqueous solution based on a transmetalation process of a fluorescent substituted Zn(salmal) complex. Transmetalation is accompanied by relevant optical absorption changes and quenching of the fluorescence emission, leading to high selectivity and sensitivity of the chemosensor, with the advantage of not requiring any sample pretreatment or pH adjustment. Competitive experiments demonstrate a high selectivity of the chemosensor towards Cu2+ with respect to the most common metal cations as potential interferents. A limit of detection down to 0.20 μM and a dynamic linear range up to 40 μM are achieved from fluorometric data. By exploiting the fluorescence quenching upon formation of the copper(II) complex, simple paper-based sensor strips, visible to naked eyes under UV light, are used for the rapid, qualitative, and quantitative in situ detection of Cu2+ ions in aqueous solution over a wide concentration range, up to 10.0 mM, in specific environments, such as in industrial wastewater, where higher concentrations of Cu2+ ions can occur.

Project description:A bench-stable cobalt(II) complex, with 3N-donor socket-type benzimidazole-imine-2H-imidazole ligand is reported as a precatalyst for regioselective hydrosilylation of terminal alkynes. Both aromatic and aliphatic alkynes could be effectively hydrosilylated with primary, secondary, and tertiary silane to give α-vinylsilanes in high yields with excellent Markovnikov selectivity and extensive functional-group tolerance. Catalyst loading varies within 0.5-0.05 mol %, which is one of the most efficient reported so far in the literature on cobalt-catalyzed alkyne hydrosilylation.

Project description:BackgroundBased on the potential of Schiff base compounds to act as sources for the development of cancer chemotherapeutic agents, this in vivo study was performed to investigate the inhibitory properties of the synthetic Schiff base compound Cu(BrHAP)2 on colonic aberrant crypt foci (ACF).MethodologyThis study involved five groups of male rats. The negative control group was injected with normal saline once a week for 2 weeks and fed 10% Tween 20 for 10 weeks, the cancer control group was subcutaneously injected with 15 mg/kg azoxymethane once per week for two consecutive weeks, the positive control group was injected with 15 mg/kg azoxymethane once per week for two consecutive weeks and 35 mg/kg 5-fluorouracil (injected intra-peritoneally) for 4 weeks, and the experimental groups were first injected with 15 mg/kg azoxymethane once per week for two consecutive weeks and then fed 2.5 or 5 mg/kg of the Schiff base compound once a day for 10 weeks. Application of the Schiff base compound suppressed total colonic ACF formation by up to 72% to 74% (P<0.05) when compared with the cancer control group. Analysis of colorectal specimens revealed that treatments with the Schiff base compound decreased the mean crypt scores in azoxymethane-treated rats. Significant elevations of superoxide dismutase, glutathione peroxidase and catalase activities and a reduction in the level of malondialdehyde were also observed. Histologically, all treatment groups exhibited significant decreases in dysplasia compared to the cancer control group (P<0.05). Immunohistochemical staining demonstrated down-regulation of the PCNA protein. Comparative western blot analysis revealed that COX-2 and Bcl2 were up-regulated and Bax was down-regulated compared with the AOM control group.ConclusionThe current study demonstrated that the Cu(BrHAP)2 compound has promising chemoprotective activities that are evidenced by significant decreases in the numbers of ACFs in azoxymethane-induced colon cancer.

Project description:Condensation of 2-hydroxybenzaldehyde (salicylaldehyde) or 2-hydroxy-1-naphthaldehyde with 2-ethylaniline yields the Schiff base compound of (E)-2-(((2-ethylphenyl)imino)methyl)phenol (HL1) or (E)-1-(((2-ethylphenyl)imino)methyl)naphthalen-2-ol (HL2), which in turn react with the dinuclear complex of [Rh(η4-cod)(µ-O2CCH3)]2 (cod = cycloocta-1,5-diene) to afford the mononuclear (η4-cod){(E)-2-(((2-ethylphenyl)imino)methyl)phenolato-κ2N,O}rhodium(I), [Rh(η4-cod)(L1)] (1) or (η4-cod){(E)-1-(((2-ethylphenyl)imino)methyl)naphthalen-2-olato-κ2N,O}rhodium(I), [Rh(η4-cod)(L2)] (2) (L1 or L2 = deprotonated Schiff base ligand). The X-ray structure determination revealed that the HL2 exists in the solid state not as the usual (imine)N···H-O(phenol) form (enolamine form) but as the zwitterionic (imine)N-H+···-O(phenol) form (ketoamine form). 1H NMR spectra for HL2 in different solvents demonstrated the existence of keto-enol tautomerism (i.e., keto ⇆ enol equilibrium) in solution. The structure for 1 and 2 showed that the deprotonated Schiff base ligand coordinates to the Rh(η4-cod)-fragment as a six-membered N^O-chelate around the rhodium atom with a close-to-square-planar geometry. Two symmetry-independent molecules (with Rh1 and Rh2) were found in the asymmetric unit in 1 in a structure with Z' = 2. The supramolecular packing in HL2 was organized by π-π and C-H···π contacts, while only two recognized C-H···π contacts were revealed in 1 and 2. Remarkably, there were reciprocal or pairwise C-H···π contacts between a pair of each of the symmetry-independent molecules in 1. This pairwise C-H contact to the Rh-N^O chelate (metalloaromatic) ring may be a reason for the two symmetry-independent molecules in 1. Differential scanning calorimetry (DSC) analyses revealed an irreversible phase transformation from the crystalline-solid to the isotropic-liquid phase and subsequently confirmed the thermal stability of the compounds. Absorption spectra in solution were explained by excited state properties from DFT/TD-DFT calculations.
