Growth of hollow cell spheroids in microbead templated chambers.

ABSTRACT: Cells form hollow, spheroidal structures during the development of many tissues, including the ocular lens, inner ear, and many glands. Therefore, techniques for in vitro formation of hollow spheroids are valued for studying developmental and disease processes. Current in vitro methods require cells to self-organize into hollow morphologies; we explored an alternative strategy based on cell growth in predefined, spherical scaffolds. Our method uses sacrificial, gelatin microbeads to simultaneously template spherical chambers within a hydrogel and deliver cells into the chambers. We use mouse lens epithelial cells to demonstrate that cells can populate the internal surfaces of the chambers within a week to create numerous hollow spheroids. The platform supports manipulation of matrix mechanics, curvature, and biochemical composition to mimic in vivo microenvironments. It also provides a starting point for engineering organoids of tissues that develop from hollow spheroids.

Lanthanum Hydroxide Nanorod-Templated Graphitic Hollow Carbon Nanorods for Supercapacitors.

Project description:Lanthanum hydroxide nanorods were employed as both a template and catalyst for carbon synthesis by chemical vapor deposition. The resulting carbon possesses hollow nanorod shapes with graphitic walls. The hollow carbon nanorods were interconnected at some junctions forming a mazelike network, and the broken ends of the tubular carbon provide accessibility to the inner surface of the carbon, resulting in a surface area of 771 m2/g. The hollow carbon was tested as an electrode material for supercapacitors. A specific capacitance of 128 F/g, an energy density of 55 Wh/kg, and a power density of 1700 W/kg at 1 A/g were obtained using the ionic liquid, 1-ethyl-3-methylimidazolium bis(trifluoromethylsulfonyl)imide, as the electrolyte.

Robust Amino-Functionalized Mesoporous Silica Hollow Spheres Templated by CO2 Bubbles.

Project description:Hollow-structured mesoporous silica has wide applications in catalysis and drug delivery due to its high surface area, large hollow space, and short diffusion mesochannels. However, the synthesis of hollow structures usually requires sacrificial templates, leading to increased production costs and environmental problems. Here, for the first time, amino-functionalized mesoporous silica hollow spheres were synthesized by using CO2 gaseous bubbles as templates. The assembly of anionic surfactants, co-structure directing agents, and inorganic silica precursors around CO2 bubbles formed the mesoporous silica shells. The hollow silica spheres, 200-400 nm in size with 20-30 nm spherical shell thickness, had abundant amine groups on the surface of the mesopores, indicating excellent applications for CO2 capture, Knoevenagel condensation reaction, and the controlled release of Drugs.

Single-Micelle-Templated Synthesis of Hollow Barium Carbonate Nanoparticle for Drug Delivery

Project description:A laboratory-synthesized triblock copolymer poly(ethylene oxide-b-acrylic acid-b-styrene) (PEG-PAA-PS) was used as a template to synthesize hollow BaCO3 nanoparticles (BC-NPs). The triblock copolymer was synthesized using reversible addition–fragmentation chain transfer radical polymerization. The triblock copolymer has a molecular weight of 1.88 × 104 g/mol. Transmission electron microscopy measurements confirm the formation of spherical micelles with a PEG corona, PAA shell, and PS core in an aqueous solution. Furthermore, the dynamic light scattering experiment revealed the electrostatic interaction of Ba2+ ions with an anionic poly(acrylic acid) block of the micelles. The controlled precipitation of BaCO3 around spherical polymeric micelles followed by calcination allows for the synthesis of hollow BC-NPs with cavity diameters of 15 nm and a shell thickness of 5 nm. The encapsulation and release of methotrexate from hollow BC-NPs at pH 7.4 was studied. The cell viability experiments indicate the possibility of BC-NPs maintaining biocompatibility for a prolonged time.

Templated-Construction of Hollow MoS<sub>2</sub> Architectures with Improved Photoresponses.

Project description:Despite the outstanding optoelectronic properties of MoS2 and its analogues, synthesis of such materials with desired features including fewer layers, arbitrary hollow structures, and particularly specifically customized morphologies, via inorganic reactions has always been challenging. Herein, using predesigned lanthanide-doped upconversion luminescent materials (e.g., NaYF4:Ln) as templates, arbitrary MoS2 hollow structures with precisely defined morphologies, widely variable dimensions, and very small shell thickness (≈2.5 nm) are readily constructed. Most importantly, integration of the near-infrared-responsive template significantly improves the photoresponse of up to 600 fold in device made of NaYF4:Yb/Er@MoS2 compared with that of MoS2 nanosheets under 980 nm laser illumination. Multichannel optoelectronic device is further fabricated by simply changing luminescent ions in the template, e.g., NaYF4:Er@MoS2, operating at 1532 nm light excitation with a 276-fold photoresponse enhancement. The simple chemistry, easy operation, high reliability, variable morphologies, and wide universality represent the most important advantages of this novel strategy that has not been accessed before.

Natural Halloysite-Templated Synthesis of Highly Graphitic Boron-Doped Hollow Carbon Nanocapsule Webs.

Project description:Hollow carbon nanocapsules have been attracting growing interest due to their fascinating characteristics and extensive potential applications. In this work, a novel natural halloysite-templated synthesis approach for highly graphitic boron-doped hollow carbon nanocapsule webs (B-HCNCWs) using glucose as the carbon source and boric acid as the heteroatom dopant was first reported. The formation process and physicochemical properties of B-HCNCWs were revealed by SEM, TEM, XRD, Raman, Brunauer-Emmett-Teller (BET), and XPS characterization techniques. The outcomes showed that the as-obtained B-HCNCWs with hollow nanocapsule network architecture had a specific surface area of 263 m2 g-1, a pore volume of 0.8 cm3 g-1, a high degree of graphitization (81.4%), graphite-like interplanar spacing (0.3370 nm), and B-containing functional groups (0.77 at%). The density function theory (DFT) calculation demonstrated that the adsorption energies of Li on B-HCNCWs were much higher than that of HCNCWs, which proved that B-doping in a carbon matrix could increase the lithium intercalation capacity.

Pickering-emulsion-templated synthesis of 3D hollow graphene as an efficient oil absorbent.

Project description:The preparation of graphene in three-dimensional mode represents an alternative method to maintain its characteristically large surface area, which, under normal circumstances, is diminished by the restacking of the individual sheets. Sufficiently stable 3D graphene enables the high surface area characteristic of monoatomic graphene layers to be obtained. Based on the coupling of the high surface area and the void spaces that are thus created, which act as pores, 3D graphene is anticipated to have potential as a sorbent material. In this study, lightweight 3D hollow graphene featuring a unique thin skeletal framework was developed using the Pickering emulsion route for oil absorbent applications. In this technique, toluene droplets stabilized by graphene oxide layers in a water system were used as the template, and upon the removal of the solvent by freeze-drying and microwave-assisted reduction, 3D hollow graphene was obtained. The produced 3D graphene demonstrates excellent sorption efficiencies of 84 to 145 g g-1 for different types of oil and organic solvents in the first absorption. This excellence can be attributed to its multi-level porosity as elucidated by mercury intrusion porosimetry (MIP) and Brunauer-Emmett-Teller (BET) surface area analysis, which indicated a bimodal pore size distribution with macroporosity and mesoporosity and a surface area of 127 m2 g-1. The 3D hollow graphene prepared using the Pickering emulsion template technique incorporating microwave treatment can be readily recycled using a solvent extraction process for a total of ten sorption-desorption cycles without significant losses in its efficiency, making it promising for further consideration as an appropriate material for oil spill incidents.

Hierarchical hollow tubular fibrous brucite-templated carbons obtained by KOH activation for supercapacitors.

Project description:Hierarchical hollow tubular porous carbons have been widely used in applications of supercapacitors, batteries, CO2 capture and catalysis due to their hollow tubular morphology, large aspect ratio, abundant pore structure and superior conductivity. Herein, hierarchical hollow tubular fibrous brucite-templated carbons (AHTFBCs) were prepared using natural mineral fiber brucite as the template and KOH as the chemical activator. The effects of different KOH additions on the pore structure and capacitive performance of AHTFBCs were systematically studied. The specific surface area and micropore content of AHTFBCs after KOH activation were higher than those of HTFBC. The specific surface area of the HTFBC is 400 m2 g-1, while the activated AHTFBC5 has a specific surface area of up to 625 m2 g-1. In particular, compared with HTFBC (6.1%), a series of AHTFBCs (22.1% for AHTFBC2, 23.9% for AHTFBC3, 26.8% for AHTFBC4 and 22.9% for AHTFBC5) with significantly increased micropore content were prepared by controlling the amount of KOH added. The AHTFBC4 electrode displays a high capacitance of 197 F g-1 at 1 A g-1 and the capacitance retention of 100% after 10 000 cycles at 5 A g-1 in the three-electrode system. And an AHTFBC4//AHTFBC4 symmetric supercapacitor exhibits the capacitance of 109 F g-1 at 1 A g-1 in 6 M KOH and an energy density of 5.8 W h kg-1 at 199.0 W kg-1 in 1 M Na2SO4 electrolyte. In addition, the capacity retention of AHTFBC4 in the symmetric supercapacitor was maintained at 92% after 5000 cycles in both 6 M KOH and 1 M Na2SO4 electrolytes.

Cell Mimicking Microparticles Influence the Organization, Growth, and Mechanophenotype of Stem Cell Spheroids.

Project description:Substrate stiffness is known to alter cell behavior and drive stem cell differentiation, though most research in this area has been restricted to traditional, two-dimensional culture systems rather than more physiologically relevant, three-dimensional (3D) platforms. In this study, we utilized polymer-based, cell mimicking microparticles (CMMPs) to deliver distinct, stable mechanical cues to human adipose derived stem cells in 3D spheroid culture to examine changes in adipogenic differentiation response and mechanophenotype. After 21 days of adipogenic induction, spheroids containing CMMPs (composite spheroids) stiffened in accordance with CMMP elasticity such that spheroids containing the stiffest, ~ 10 kPa, CMMPs were over 27% stiffer than those incorporating the most compliant, ~ 0.25 kPa CMMPs. Adipogenically induced, cell-only spheroids were over 180% larger and 50% more compliant than matched controls. Interestingly, composite spheroids cultured without chemical induction factors dissociated when presented with CMMPs stiffer than ~ 1 kPa, while adipogenic induction factors mitigated this behavior. Gene expression for PPARG and FABP4 were upregulated more than 45-fold in adipogenically induced samples compared to controls but were unaffected by CMMP elasticity, attributed to insufficient cell-CMMP contacts throughout the composite spheroid. In summary, mechanically tuned CMMPs influenced whole-spheroid mechanophenotype and stability but minimally affected differentiation response.

Biomimetic branched hollow fibers templated by self-assembled fibrous polyvinylpyrrolidone structures in aqueous solution.

Project description:Branched hollow fibers are common in nature, but to form artificial fibers with a similar branched hollow structure is still a challenge. We discovered that polyvinylpyrrolidone (PVP) could self-assemble into branched hollow fibers in an aqueous solution after aging the PVP solution for about two weeks. On the basis of this finding, we demonstrated two approaches by which the self-assembly of PVP into branched hollow fibers could be exploited to template the formation of branched hollow inorganic fibers. First, inorganic material such as silica with high affinity against the PVP could be deposited on the surface of the branched hollow PVP fibers to form branched hollow silica fibers. To extend the application of PVP self-assembly in templating the formation of hollow branched fibers, we then adopted a second approach where the PVP molecules bound to inorganic nanoparticles (using gold nanoparticles as a model) co-self-assemble with the free PVP molecules in an aqueous solution, resulting in the formation of the branched hollow fibers with the nanoparticles embedded in the PVP matrix constituting the walls of the fibers. Heating the resultant fibers above the glass transition temperature of PVP led to the formation of branched hollow gold fibers. Our work suggests that the self-assembly of the PVP molecules in the solution can serve as a general method for directing the formation of branched hollow inorganic fibers. The branched hollow fibers may find potential applications in microfluidics, artificial blood vessel generation, and tissue engineering.
