ABSTRACT: The clove (Syzygium aromaticum) is an important tropical spice crop in global trade. Evolving environmental pressures necessitate modern characterization and selection techniques that are currently inaccessible to clove growers owing to the scarcity of genomic and genetic information. Here, we present a 370-Mb high-quality chromosome-scale genome assembly for clove. Comparative genomic analysis between S. aromaticum and Eucalyptus grandis-both species of the Myrtaceae family-reveals good genome structure conservation and intrachromosomal rearrangements on seven of the eleven chromosomes. We report genes that belong to families involved in the biosynthesis of eugenol, the major bioactive component of clove products. On the basis of our transcriptomic and metabolomic findings, we propose a hypothetical scenario in which eugenol acetate plays a key role in high eugenol accumulation in clove leaves and buds. The clove genome is a new contribution to omics resources for the Myrtaceae family and an important tool for clove research.

Project description:Background/aimSyzygium aromaticum L. is a versatile plant traditionally used to treat digestive and respiratory issues, improve oral health, and relieve pain, particularly in regions such as Southeast Asia, South Asia, and parts of the Middle East. Its essential oil (EO), predominantly composed of eugenol, is rich in bioactive compounds. This study aims to clarify the specific contribution of eugenol to the antioxidant, antibacterial, antifungal, and insecticidal activities of S. aromaticum EO by comparing their individual effects. Additionally, density functional theory (DFT) calculations were employed to examine the antioxidant mechanism of eugenol and its role in enhancing the overall activity of the EO.Materials and methodsThe EO was obtained from S. aromaticum and analyzed using GC-MS to determine its composition. Antioxidant activity was assessed through the DPPH scavenging assay. Antibacterial and antifungal activities were evaluated using the disk diffusion method against various strains, while insecticidal and repellent effects were tested on Bruchus lentis at different concentrations and exposure times. Antioxidant mechanisms were investigated using DFT calculations.ResultsThe findings underscore the strong antioxidant, antibacterial, antifungal, and insecticidal properties of S. aromaticum EO, with eugenol identified as the primary active component driving the antioxidant and insecticidal effects. Additionally, eugenol has been found to exhibit moderate scavenging activity in lipid media. However, its activity is higher in polar media, with a k overall = 1.70 ⊠ 106 M-1s-1 comparable to that of ascorbic acid. The single-electron transfer mechanism from the deprotonated state was found to play a decisive role under these conditions.ConclusionS. aromaticum EO exhibits remarkable antioxidant, antibacterial, antifungal, and insecticidal activities. A significant portion of these properties can be attributed to the presence of eugenol. This suggests that eugenol plays a critical role in the EO's overall efficacy, making S. aromaticum a promising candidate for applications in natural health products, pharmaceuticals, and agricultural pest management.

Project description:ObjectiveConsidering the emergence of biofilm-associated pathogens with multidrug resistance, the objective(s) of the present study was to evaluate the anti-virulence property of Syzygium aromaticum extracts/fractions against 2 multidrug-resistant catheter isolates.Materials and methodsPulverized clove buds were subjected to bioactivity-guided isolation to identify the bioactive extract/fraction(s) with significant anti-virulence property. The clove bud powder was subjected to Soxhlet extraction and all these extracts were investigated in terms of anti-virulent efficacy using initial readout assays. Bioassay-guided partial purification was performed through column and size exclusion chromatography. Detailed testing of the anti-virulence and anti-biofilm activity of sub-minimum inhibitory concentration (sub-MIC) levels of the active fraction, was performed besides the cytotoxicity profiling in RAW 264.7 cells.ResultsGas chromatography-mass spectrometry (GC-MS) analysis of the clove bioactive fraction-2 (CBF-2), done after the bioassay-guided fractionation, revealed eugenol as the major bioactive ingredient present in CBF-2. Reduced mRNA levels of virulence factor genes after CBF-2 (700 μg/ml) treatment correlated well with the respective phenotypic assays (p<0.001). Similarly, CBF-2 (700 μg/ml) treatment exhibited significantly low mRNA levels of quorum sensing (QS) receptor genes compared to their cognate synthase genes (p<0.001). Crystal violet staining and scanning electron micrographs of CBF-2-treated biofilms showed lesser macrocolonies with remarkably simplified architecture. Cytotoxic evaluation of CBF-2 suggested a minute reduction in viability even at the highest tested concentration (1600 μg/ml, p<0.05).ConclusionThe present study indicated that eugenol-rich CBF-2 has potent anti-virulence and anti-biofilm activity against urinary catheter isolates and can be regarded as an alternative for treatment of catheter-associated urinary tract infections.
