From Thioureas to Thioquinolines through Isolated Benzothiazines by Gold Catalysis.

ABSTRACT: New benzothiazine heterocycles have been formed from thiourea derivatives by using different gold catalysts. The catalyst and the conditions were optimised towards the selective synthesis of six-membered benzothiazine heterocycles, characterised by X-ray diffraction. Interestingly, these organic compounds evolved under gold catalysis in basic medium to achieve the formation of amino thioquinolines through an unprecedented aromatisation process of the heterocycle. The reaction was also carried out stoichiometrically by reaction with gold complexes to afford thioquinolines coordinated to the gold fragment. Benzothiazine, amino thioquinoline heterocycles and gold-derived species have a great potential for biological applications.

Non-Diazo C-H Insertion Approach to Cyclobutanones through Oxidative Gold Catalysis.

Project description:Cyclobutanones are synthetically versatile compounds that often require extensive effort to access. Herein, we report a facile synthesis of cyclobutanones based on the C(sp3 )-H insertion chemistry of oxidatively generated gold carbenes. Various cyclobutanones were obtained in synthetically useful yields from substrates with minimal structural prefunctionalization. This discovery reveals new synthetic utilities of gold-catalyzed oxidative transformations of alkynones.

One-pot synthesis of fused pyrroles through a key gold-catalysis-triggered cascade.

Project description:A two-step, one-pot synthesis of fused pyrroles is realized by firstly condensing an N-alkynylhydroxammonium salt with a readily enolizable ketone under mild basic conditions and then subjecting the reaction mixture to a gold catalyst, which triggers a cascade reaction involving a facile initial [3.3]-sigmatropic rearrangement of the gold-catalysis product, that is, an N,O-dialkenylhydroxamine. The reaction provides a facile access to polycyclic pyrroles in moderate to good yields.

Alkyne Trifunctionalization via Divergent Gold Catalysis: Combining π-Acid Activation, Vinyl-Gold Addition, and Redox Catalysis.

Project description:Here we report the first example of alkyne trifunctionalization through simultaneous construction of C-C, C-O, and C-N bonds via gold catalysis. With the assistance of a γ-keto directing group, sequential gold-catalyzed alkyne hydration, vinyl-gold nucleophilic addition, and gold(III) reductive elimination were achieved in one pot. Diazonium salts were identified as both electrophiles (N source) and oxidants (C source). Vinyl-gold(III) intermediates were revealed as effective nucleophiles toward diazonium, facilitating nucleophilic addition and reductive elimination with high efficiency. The rather comprehensive reaction sequence was achieved with excellent yields (up to 95%) and broad scope (>50 examples) under mild conditions (room temperature or 40 °C).

Project description:Golden Cascade: With a tethered, electron-rich arene as the internal nucleophile, a gold-catalyzed amide cyclization to an alkyne initiates a cascade process that ends with a Ferrier rearrangement. Electron-rich arene-bearing hexahydroquinolizin-2-ones are formed in good yields and can be converted into indole alkaloids in only a few steps.

Gold Redox Catalysis through Base-Initiated Diazonium Decomposition toward Alkene, Alkyne, and Allene Activation.

Project description:The discovery of photoassisted diazonium activation toward gold(I) oxidation greatly extended the scope of gold redox catalysis by avoiding the use of a strong oxidant. Some practical issues that limit the application of this new type of chemistry are the relative low efficiency (long reaction time and low conversion) and the strict reaction condition control that is necessary (degassing and inert reaction environment). Herein, an alternative photofree condition has been developed through Lewis base induced diazonium activation. With this method, an unreactive AuI catalyst was used in combination with Na2 CO3 and diazonium salts to produce a AuIII intermediate. The efficient activation of various substrates, including alkyne, alkene and allene was achieved, followed by rapid AuIII reductive elimination, which yielded the C-C coupling products with good to excellent yields. Relative to the previously reported photoactivation method, our approach offered greater efficiency and versatility through faster reaction rates and broader reaction scope. Challenging substrates such as electron rich/neutral allenes, which could not be activated under the photoinitiation conditions (<5 % yield), could be activated to subsequently yield the desired coupling products in good to excellent yield.

Total Syntheses of the Reported Structures of Curcusones I and J through Tandem Gold Catalysis.

Project description:Total syntheses of the reported structures of the rhamnofolane diterpene natural products curcusones I and J in racemic form were achieved. The synthetic strategy features a novel tandem gold-catalyzed furan formation and furan-allene [4+3] cycloaddition to build the 5,7-fused ring system with an oxa-bridge in one step, and a stereoselective exo-Diels-Alder reaction to form the 6-membered ring. The newly developed tandem gold catalysis is quite general and can be scaled up. Our syntheses suggest that structural revisions of curcusones I and J are needed.

The dome of gold nanolized for catalysis.

Project description:Gold is noble in bulk but turns out to be a superior catalyst at the nanoscale when supported on oxides, in particular titania. The critical thickness for activity, namely two-layer gold particles on titania, observed two decades ago represents one of the most influential mysteries in the recent history of heterogeneous catalysis. By developing a Bayesian optimization controlled global potential energy surface exploration tool with machine learning potential, here we determine the atomic structures of gold particles within ∼2 nm on a TiO2 surface. We show that the smallest stable Au nanoparticle is Au24 which is pinned on the oxygen-rich TiO2 and exhibits an unprecedented dome architecture made by a single-layer Au sheet but with an apparent two-atomic-layer height. Importantly, this has the highest activity for CO oxidation at room temperature. The physical origin of the high activity is the outstanding electron storage ability of the nano-dome, which activates the lattice oxygen of the oxide. The determined CO oxidation mechanism, the simulated rate and the fitted apparent energy barrier are consistent with known experimental facts, providing key evidence for the presence of both the high-activity Au dome and the low-activity close-packed Au particles in real catalysts. The future direction for the preparation of active and stable Au-based catalysts is thus outlined.

Catalytic Enantioselective Protonation of Gold Enolates Enabled by Cooperative Gold(I) Catalysis.

Project description:Enantioselective protonation is a versatile approach to the construction of tertiary α-stereocenters, which are common structural motifs in various natural products and biologically relevant compounds. Herein we report a mild access to these chiral centers using cooperative gold(I) catalysis. From cyclic ketone enol carbonates, this asymmetric catalysis provides highly enantioselective access to cyclic ketones featuring an α tertiary chiral center, including challenging 2-methylsuberone. In combination with the gold-catalyzed formation of cyclopentadienyl carbonates in a one-pot, two-step process, this chemistry enables expedient access to synthetically versatile α'-chiral cyclopentenones with excellent enantiomeric excesses from easily accessible enynyl carbonate substrates.
