Direct Dehydrogenative Access to Unsymmetrical Phenones.

ABSTRACT: The first non-directed dehydrogenative phenone coupling method of methylarenes with aromatic C-H bonds, displaying a large substrate scope, is herein reported. This reaction represents a far more direct atom- and step-efficient alternative to the classical Friedel-Crafts or Suzuki-Miyaura derived acylation reactions. The method can be carried out on a gram scale and was successfully applied to the synthesis of several Ketoprofen drug analogues.

Direct Synthesis of Unsymmetrical Dithioacetals.

Project description:Dithioacetals are a frequently used motif in synthetic organic chemistry and have recently seen increasing attention as structural motif in promising antiviral agents against plant pathogens. Most existing reports, however, only discuss symmetrical dithioacetals. Examples of mixed dithioacetals are scarce and no general method for the selective synthesis of these compounds exists. Herein, a synthetically simple general one-step protocol was developed for the synthesis of a broad range of unsymmetrical dithioacetals consisting of one aromatic and one aliphatic thiol moiety from the corresponding aldehyde/thiol mixture. The mixed S,S-acetals were obtained in high yields, and a great variety of functional groups was tolerated. Kinetic control enabled an excellent selectivity in regard to the unsymmetrical dithioacetal.

Direct electrochemical generation of organic carbonates by dehydrogenative coupling.

Project description:Organic carbonates are an important source for polycarbonate synthesis. However, their synthesis generally requires phosgene, sophisticated catalysts, harsh reaction conditions, or other highly reactive chemicals. We present the first direct electrochemical generation of mesityl methyl carbonate by C-H activation. Although this reaction pathway is still challenging concerning scope and efficiency, it outlines a new strategy for carbonate generation.

Project description:A one-pot, Cu-catalyzed direct C-H arylselenation protocol using elemental Se and aryl iodides was developed for nitro-substituted, N-alkylated pyrazoles, imidazoles and other heterocycles including 4H-chromen-4-one. This general and concise method allows one to obtain a large number of unsymmetrical heteroaryl selenides bearing a variety of substituents. The presence of the nitro group was confirmed to be essential for the C-H activation and can also be used for further functionalisation and manipulation. Several examples of heteroannulated benzoselenazines were also synthesized using the developed synthetic protocol.

Nickel-catalyzed dehydrogenative cross-coupling: direct transformation of aldehydes into esters and amides.

Project description:By exploring a new mode of nickel-catalyzed cross-coupling, a method to directly transform both aromatic and aliphatic aldehydes into either esters or amides has been developed. The success of this oxidative coupling depends on the appropriate choice of catalyst and organic oxidant, including the use of either α,α,α-trifluoroacetophenone or excess aldehyde. Mechanistic data that supports a catalytic cycle involving oxidative addition into the aldehyde C-H bond is also presented.

Direct dehydrogenative alkyl Heck-couplings of vinylarenes with umpolung aldehydes catalyzed by nickel.

Project description:Alkenes are fundamental functionalities in nature and highly useful intermediates in organic synthesis, medicinal chemistry and material sciences. Transition-metal-catalyzed Heck couplings with organic halides as electrophiles have been established as a powerful protocol for the synthesis of this valuable building block. However, the requirement of organic halides and the generation of stoichiometric hazardous halide wastes may cause significant sustainable concerns. The halide-free oxidative Heck alkenylations involving organometallics or arenes as the coupling partners provide a facile and alternative pathway. Nonetheless, stoichiometric amounts of extra oxidant are essential in most cases. Herein, we present a direct dehydrogenative alkyl Heck-coupling reaction under oxidant-free conditions, liberating hydrogen, nitrogen and water as the side products. Excellent regioselectivity is achieved via neighboring oxygen atom coordination. Broad substrate scope, great functional group (ketone, ester, phenol, free amine, amide etc) tolerance and modification of pharmaceutical candidates and biological molecules exemplified its generality and practicability.

Project description:A highly efficient silver-catalyzed dibenzylation of acetanilides by employing the benzhydrol derivatives as the coupling partners to yield triphenylmethane derivatives has been developed. Various functional groups were tolerated, leading to the corresponding products in moderate to good yields. Cross-dehydrogenative coupling enables enantioselective access to CF<sub>3</sub>-substituted all-carbon quaternary stereocenters.

Project description:A cross-dehydrogenative coupling strategy for enantioselective access to acyclic CF3-substituted all-carbon quaternary stereocenters has been established. By using catalytic DDQ with MnO2 as an inexpensive terminal oxidant, asymmetric cross coupling of racemic δ-CF3-substituted phenols with indoles proceeded smoothly, providing CF3-bearing all-carbon quaternary stereocenters with excellent chemo- and enantioselectivities. The generality of the strategy is further demonstrated by efficient construction of all-carbon quaternary stereocenters bearing other polyfluoroalkyl and perfluoroalkyl groups such as CF2Cl, C2F5, and C3F7.

Acceptorless Dehydrogenative Coupling Using Ammonia: Direct Synthesis of N-Heteroaromatics from Diols Catalyzed by Ruthenium.

Project description:The synthesis of N-heteroaromatic compounds via an acceptorless dehydrogenative coupling process involving direct use of ammonia as the nitrogen source was explored. We report the synthesis of pyrazine derivatives from 1,2-diols and the synthesis of N-substituted pyrroles by a multicomponent dehydrogenative coupling of 1,4-diols and primary alcohols with ammonia. The acridine-based Ru-pincer complex 1 is an effective catalyst for these transformations, in which the acridine backbone is converted to an anionic dearomatized PNP-pincer ligand framework.

Catalytic Mechanism of Liquid-Metal Indium for Direct Dehydrogenative Conversion of Methane to Higher Hydrocarbons.

Project description:There is a great interest in direct conversion of methane to valuable chemicals. Recently, we reported that silica-supported liquid-metal indium catalysts (In/SiO2) were effective for direct dehydrogenative conversion of methane to higher hydrocarbons. However, the catalytic mechanism of liquid-metal indium has not been clear. Here, we show the catalytic mechanism of the In/SiO2 catalyst in terms of both experiments and calculations in detail. Kinetic studies clearly show that liquid-metal indium activates a C-H bond of methane and converts methane to ethane. The apparent activation energy of the In/SiO2 catalyst is 170 kJ mol-1, which is much lower than that of SiO2, 365 kJ mol-1. Temperature-programmed reactions in CH4, C2H6, and C2H4 and reactivity of C2H6 for the In/SiO2 catalyst indicate that indium selectively activates methane among hydrocarbons. In addition, density functional theory calculations and first-principles molecular dynamics calculations were performed to evaluate activation free energy for methane activation, its reverse reaction, CH3-CH3 coupling via Langmuir-Hinshelwood (LH) and Eley-Rideal mechanisms, and other side reactions. A qualitative level of interpretation is as follows. CH3-In and H-In species form after the activation of methane. The CH3-In species wander on liquid-metal indium surfaces and couple each other with ethane via the LH mechanism. The solubility of H species into the bulk phase of In is important to enhance the coupling of CH3-In species to C2H6 by decreasing the formation of CH4 though the coupling of CH3-In species and H-In species. Results of isotope experiments by combinations of CD4, CH4, D2, and H2 corresponded to the LH mechanism.
