Underlying Mechanisms of Reductive Amination on Pd-Catalysts: The Unique Role of Hydroxyl Group in Generating Sterically Hindered Amine.

ABSTRACT: Pd nanospecies supported on porous g-C₃N₄ nanosheets were prepared for efficient reductive amination reactions. The structures of the catalysts were characterized via FTIR, XRD, XPS, SEM, TEM, and TG analysis, and the mechanisms were investigated using in situ ATR-FTIR spectroscopic analysis complemented by theoretical calculation. It transpired that the valence state of the Pd is not the dominating factor; rather, the hydroxyl group of the Pd(OH)₂ cluster is crucial. Thus, by passing protons between different molecules, the hydroxyl group facilitates both the generation of the imine intermediate and the reduction of the C=N unit. As a result, the sterically hindered amines can be obtained at high selectivity (>90%) at room temperature.

Amine dehydrogenases: efficient biocatalysts for the reductive amination of carbonyl compounds.

Project description:Amines constitute the major targets for the production of a plethora of chemical compounds that have applications in the pharmaceutical, agrochemical and bulk chemical industries. However, the asymmetric synthesis of α-chiral amines with elevated catalytic efficiency and atom economy is still a very challenging synthetic problem. Here, we investigated the biocatalytic reductive amination of carbonyl compounds employing a rising class of enzymes for amine synthesis: amine dehydrogenases (AmDHs). The three AmDHs from this study - operating in tandem with a formate dehydrogenase from Candida boidinii (Cb-FDH) for the recycling of the nicotinamide coenzyme - performed the efficient amination of a range of diverse aromatic and aliphatic ketones and aldehydes with up to quantitative conversion and elevated turnover numbers (TONs). Moreover, the reductive amination of prochiral ketones proceeded with perfect stereoselectivity, always affording the (R)-configured amines with more than 99% enantiomeric excess. The most suitable amine dehydrogenase, the optimised catalyst loading and the required reaction time were determined for each substrate. The biocatalytic reductive amination with this dual-enzyme system (AmDH-Cb-FDH) possesses elevated atom efficiency as it utilizes the ammonium formate buffer as the source of both nitrogen and reducing equivalents. Inorganic carbonate is the sole by-product.

Tuning Pd-nanoparticle@MIL-101(Cr) Catalysts for Tandem Reductive Amination.

Project description:AbstractThe versatility of MOFs as highly porous Lewis acidic supports for precious metal nanoparticles has been exploited for one-pot tandem reductive amination catalysis. MIL-101(Cr) loaded with Pd nanoparticles ca. 3 nm in size at 0.2-1 wt% has been used to catalyse the reaction of 4'-fluoroacetophenone with benzylamine under 10 bar of H2 to give the secondary amine, 4'-fluoro-α-methyl-N-phenylmethylbenzenemethanamine. For the highest Pd loading, major hydrogenolysis of the secondary amine occurs in a second tandem reaction, but by changing the ratio of Pd to Lewis acidic Cr3+ active sites it is possible to tune the catalytic selectivity to the desired 2° amine product. An empirical kinetic analysis was performed to demonstrate this active site complementarity.Graphical abstract

Ferrocene-Containing Sterically Hindered Phosphonium Salts.

Project description:The synthesis and physical properties of the series of the ferrocenyl-containing sterically hindered phosphonium salts based on di(tert-butyl)ferrocenylphosphine is reported. Analysis of voltamogramms of the obtained compounds revealed some correlations between their structures and electrochemical properties. The elongation of the alkyl chain at the P atom as well as replacement of the Br- anion by [BF₄]- shifts the ferrocene/ferrocenium transition of the resulting salts into the positive region. DFT results shows that in the former case, the Br- anion destabilizes the corresponding ion pair, making its oxidation easier due to increased highest occupied molecular orbital (HOMO) energy. Increased HOMO energy for ion pairs with the Br- ion compared to BF₄- are caused by contribution of bromide atomic orbitals to the HOMO. The observed correlations can be used for fine-tuning the properties of the salts making them attractive for applications in multicomponent batteries and capacitors.

Activity and Thermal Stability of Cobalt(II)-Based Olefin Polymerization Catalysts Adorned with Sterically Hindered Dibenzocycloheptyl Groups.

Project description:Five examples of unsymmetrical 2-(2,4-bis(dibenzocycloheptyl)-6-methylphenyl- imino)ethyl)-6-(1-(arylyimino)ethyl)pyridine derivatives (aryl = 2,6-Me2C6H3 in L1; 2,6-Et2C6H3 in L2; 2,6-i-Pr2C6H3 in L3; 2,4,6-Me3C6H2 in L4 and 2,6-Et2-4-MeC6H2 in L5) were prepared and characterized. Treatment with CoCl2 offered the corresponding cobalt precatalysts Co1-Co5, which were characterized by FT-IR and NMR spectroscopy as well as elemental analysis. The molecular structures of Co3 and Co4 determined by single crystal X-ray diffraction revealed distorted square pyramidal geometries with τ5 values of 0.052-0.215. Activated with either MAO or MMAO, the precatalysts displayed high activities in ethylene polymerization, where Co1 with the least bulky substituents exhibited a peak activity of 1.00 × 107 g PE mol-1 (Co) h-1 at 60 °C. With MAO as a cocatalyst, the activity was reduced only by one order of magnitude at 90 °C, which implies thermally stable active sites. The polymerization product was highly linear polyethylene with vinyl end groups. Co3 with the most sterically hindered active sites was capable of generating polyethylene of high molecular weight, reaching 6.46 × 105 g mol-1. Furthermore, high melting point and unimodal molecular weight distribution were observed in the resulting polyethylene. It must be stressed that the thermal stability of the catalyst and the molecular weight of the obtained polyethylene attain the highest values reported for the unsymmetrical 2,6-bis(imino)pyridylcobalt (II) chloride precatalysts.

Project description:The synthesis of ruthenium complexes incorporating an overcrowded pentaarylcyclopentadienyl ligand has been investigated, and higher efficiency has been reached using chlorine-functionalised precursors when compared with their brominated counterparts. A new methodology for the preparation of chlorocyclopentadienes has been developed which is well adapted for highly sterically hindered compounds and works with either electron rich or poor systems.

Enhanced clickability of doubly sterically-hindered aryl azides.

Project description:Steric character is one of the most fundamental factors to determine the reactivity of the substrate in organic synthesis. In bimolecular reaction, the sterically-bulky group situated close to the reactive center generally prevents the approach of the reaction partner retarding the bond formation. This report describes, to the contrary, significantly enhanced reactivity of 2,6-disubstituted phenyl azides observed in catalyst-free 1,3-dipolar cycloaddition with alkynes, unexpectedly reacting faster than unsubstituted phenyl azide and even more faster than unhindered alkyl azide, despite the steric hindrance adjacent to the reactive azido group. Experimental and computational studies have indicated that the steric hindrance eliciting the inhibition of resonance between azido group and the aromatic ring is the primary cause of this apparently-paradoxical phenomenon. This is the first type of steric acceleration, indicating a possibility of designing a highly reactive functional group by strategically locating it in the sterically-congested environment.

Flow carbonylation of sterically hindered ortho-substituted iodoarenes.

Project description:The flow synthesis of ortho-substituted carboxylic acids, using carbon monoxide gas, has been studied for a number of substrates. The optimised conditions make use of a simple catalyst system compromising of triphenylphosphine as the ligand and palladium acetate as the pre-catalyst. Carbon monoxide was introduced via a reverse "tube-in-tube" flow reactor at elevated pressures to give yields of carboxylated products that are much higher than those obtained under normal batch conditions.

Cu-Catalyzed C-N Coupling with Sterically Hindered Partners.

Project description:Copper, an earth-abundant metal, has reemerged as a viable alternative to the versatile Pd-catalyzed C-N coupling. Coupling sterically hindered reaction partners, however, remains challenging. Herein, we disclose the discovery and development of a pyrrole-ol ligand to facilitate the coupling of ortho-substituted aryl iodides with sterically hindered amines. The ligand was discovered through a library screening approach and highlights the value of mining heteroatom-rich pharmaceutical libraries for useful ligand motifs. Further evaluation revealed that this ligand is uniquely effective in these challenging transformations. The reaction enables the coupling of sterically hindered primary and secondary amines, anilines, and amides with broad functional group tolerance.
