Project description:Exposure to environmental chemicals has been linked to various health disorders, including obesity, type 2 diabetes, cancer and dysregulation of the immune and reproductive systems, whereas the gastrointestinal microbiota critically contributes to a variety of host metabolic and immune functions. We aimed to evaluate the bidirectional relationship between gut bacteria and environmental pollutants and to assess the toxicological relevance of the bacteria-xenobiotic interplay for the host. We examined studies using isolated bacteria, faecal or caecal suspensions-germ-free or antibiotic-treated animals-as well as animals reassociated with a microbiota exposed to environmental chemicals. The literature indicates that gut microbes have an extensive capacity to metabolise environmental chemicals that can be classified in five core enzymatic families (azoreductases, nitroreductases, β-glucuronidases, sulfatases and β-lyases) unequivocally involved in the metabolism of >30 environmental contaminants. There is clear evidence that bacteria-dependent metabolism of pollutants modulates the toxicity for the host. Conversely, environmental contaminants from various chemical families have been shown to alter the composition and/or the metabolic activity of the gastrointestinal bacteria, which may be an important factor contributing to shape an individual's microbiotype. The physiological consequences of these alterations have not been studied in details but pollutant-induced alterations of the gut bacteria are likely to contribute to their toxicity. In conclusion, there is a body of evidence suggesting that gut microbiota are a major, yet underestimated element that must be considered to fully evaluate the toxicity of environmental contaminants.

Project description:Humans are chronically exposed to multiple exogenous substances, including environmental pollutants, drugs and dietary components. Many of these compounds are suspected to impact human health, and their combination in complex mixtures could exacerbate their harmful effects. Here we demonstrate that a pharmaceutical oestrogen and a persistent organochlorine pesticide, both exhibiting low efficacy when studied separately, cooperatively bind to the pregnane X receptor, leading to synergistic activation. Biophysical analysis shows that each ligand enhances the binding affinity of the other, so the binary mixture induces a substantial biological response at doses at which each chemical individually is inactive. High-resolution crystal structures reveal the structural basis for the observed cooperativity. Our results suggest that the formation of 'supramolecular ligands' within the ligand-binding pocket of nuclear receptors contributes to the synergistic toxic effect of chemical mixtures, which may have broad implications for the fields of endocrine disruption, toxicology and chemical risk assessment.

Project description:The incomplete mineralization of contaminants of emerging concern (CECs) during the advanced oxidation processes can generate transformation products that exhibit toxicity comparable to or greater than that of the original contaminant. In this study, we demonstrated the application of a novel, fast, and cost-effective quantitative toxicogenomics-based approach for the evaluation of the evolution and nature of toxicity along the electro-Fenton oxidative degradation of three representative CECs whose oxidative degradation pathways have been relatively well studied, bisphenol A, triclosan, and ibuprofen. The evolution of toxicity as a result of the transformation of parent chemicals and production of intermediates during the course of degradation are monitored, and the quantitative toxicogenomics assay results revealed the dynamic toxicity changes and mechanisms, as well as their association with identified intermediates during the electro-Fenton oxidation process of the selected CECs. Although for the three CECs, a majority (>75%) of the parent compounds disappeared at the 15 min reaction time, the nearly complete elimination of toxicity required a minimal 30 min reaction time, and they seem to correspond to the disappearance of identified aromatic intermediates. Bisphenol A led to a wide range of stress responses, and some identified transformation products containing phenolic or quinone group, such as 1,4-benzoquinone and hydroquinone, likely contributed to the transit toxicity exhibited as DNA stress (genotoxicity) and membrane stress during the degradation. Triclosan is known to cause severe oxidative stress, and although the oxidative damage potential decreased concomitantly with the disappearance of triclosan after a 15 min reaction, the sustained toxicity associated with both membrane and protein stress was likely attributed at least partially to the production of 2,4-dichlorophenol that is known to cause the production of abnormal proteins and affect the cell membrane. Ibuprofen affects the cell transporter function and exhibited significantly high membrane stress related to both membrane structure and function. Oxidative degradation of ibuprofen led to a shift in its toxicity profile from mainly membrane stress to one that exhibited not only sustained membrane stress but also protein stress and DNA stress. The information-rich and high-resolution toxicogenomics results served as "fingerprints" that discerned and revealed the toxicity mechanism at the molecular level among the CECs and their oxidation transformation products. This study demonstrated that the quantitative toxicogenomics assay can serve as a useful tool for remediation technology efficacy assessment and provide guidance about process design and optimization for desired toxicity elimination and risk reduction.

Project description:This study evaluated the lethal, sublethal, and toxic of a commercial formulation of cypermethrin in the anuran species Physalaemus gracilis. In the acute test, concentrations of 100-800 μg L-1 were tested over 96 h. In the chronic test, cypermethrin concentrations recorded in nature (1, 3, 6, and 20 μg L-1) were tested for mortality and then used for the micronucleus test and erythrocyte nuclear abnormalities over a 7-days period. The LC50 determined for P. gracilis for the commercial cypermethrin formulation was 273.41 μg L-1. In the chronic test, a mortality of more than 50% was observed at the highest concentration (20 μg L-1), as it caused half of the tadpoles studied to die. The micronucleus test showed significant results at concentrations of 6 and 20 μg L-1 and recorded the presence of several nuclear abnormalities, indicating the genotoxic potential of the commercial cypermethrin formulation for P. gracilis. Cypermethrin presented a high risk to the species, indicating that it has the potential to cause several problems in the short and long term and to affect the dynamics of this ecosystem. Therefore, it can be concluded that the commercial formulation of cypermethrin had toxicological effects on P. gracilis.

Project description:Rhipicephalus microplus is widely distributed in tropical and subtropical areas of the world where livestock is a principal activity with great veterinary and economic importance. In Ecuador, this hematophagous ectoparasite has been observed between 0 and 2600 masl. One of the main tick control measures is the use of acaricides, which have been indiscriminately used worldwide and in Ecuador. In this country, no studies on acaricide resistance in Rhipicephalus microplus have been published. The current study aims to characterise the level of resistance of R. microplus against three main acaricides commonly used in Ecuador i.e. amitraz, alpha-cypermethrin and ivermectin to determine the level and pattern of dose-responses for R. microplus in 12 field populations (farms). The level of acaricide resistance was evaluated using three different bioassays: adult immersion test (AIT), larval package test (LPT) and larval immersion test (LIT), as recommended by the FAO. The predictive dose-responses were analysed by binomial logistics regression of the larval survival rate (resistance). In general, we found resistance of 67% for amitraz; 50% for alpha-cypermethrin and from 25 to 42% for ivermectin in the twelve field populations analysed. Resistance levels were studied in larval and adult bioassays, respectively, which were slightly modified for this study. For larval bioassays based on corrected mortality i.e. high (above 51%), medium (21-50%) and low (11-20%) resistance, percentages less than 10% were considered as susceptible. For the adult test, two resistance levels were used i.e. high (more than 76%) and medium (51 to 75%) resistance. Percentages lower than 50% were considered as susceptible. In this context, for larval bioassays, amitraz showed 21%, 38% and 8% for high, medium and low resistance, respectively. Alpha-cypermethrin presented 8%, 4 and 38% for high, medium and low resistance, respectively. Ivermectin presented 8%, 25% and 8% for high, medium and low resistance, respectively. For adult tests with amitraz 50% and 17% of the field populations showed average and high resistance, with evidences of average resistance to alpha-cypermethrin in 50% of the samples and average resistance against ivermectin in 25% of the farms. No statistical difference amongst the three bioassays was found and acaricide resistance was confirmed by logistic regression analysis; hence resistance (dose-responses) in each field populations differed, depending on the choice of the acaricide, frequent usage, frequency of treatment and farm management. The effective estimated dose needed to eliminate 99% of ticks is higher than dose recommended by the manufacturer. In conclusion, amitraz showed the highest resistance followed by ivermectin and alpha-cypermethrin and reveals differences on resistance in each individual field population. This information is important in order to establish the monitoring of resistance on each farm individually, contributing to the rational use of acaricides included in an integrated control program for R. microplus.

Project description:Cypermethrin (Cyp), a persistent synthetic pyrethroid insecticide widely used for insect control. The persistence of Cyp creates toxicity to both humans and the environment This study investigates biochar and Bacillus cereus distinct and collective effects on Cyp -contaminated soil during a 90-day incubation. This study also investigates the effects of different concentrations of Cyp (50, 100, ,500 to 1000 mg kg-1) on soil physicochemical and biological activities during a 90-day incubation period. Microbial biomass carbon and soil respiration rates decreased significantly across all cypermethrin concentrations, with the most substantial reductions observed at 1000 mg kg-1. However noticeable variations in soil enzymes and MBC over time during the entire incubation period. On 1st day, the GMean Enz and MBC rate for Cyp treatments (50, 100, ,500 to 1000 mg kg-1) ranged from 0.98 to 0.63, and 9.06, to 5.03, respectively. Under Cyp pollution, microbial biomass carbon exhibited significant decreases, with the highest inhibition (86.2%) at 1000 mg kg-1 on 1st day of incubation. Soil respiration rates dropped 77%, at 1000 mg kg-1, and Integrated biomarker response (IBR) values peaked on day 30, indicating environmental stress. Biochar and Bacillus cereus effectively facilitated the degradation of Cyp, achieving approximately 85% degradation within the first 45 days of the experiment. The combined application of biochar and Bacillus cereus increased soil pH to a neutral level from 5.9, to 7.1, reduced electrical conductivity from 1.41 µS cm- 1 to 1.20 µS cm-1, and elevated cation exchange capacity from 1.54 ± 0.04 to 6.18 C mol kg-1, while also improving organic carbon content to 3.135%. However, the dehydrogenase activity was decresed upto 47% in the combined application and all other enzymes including urasese catlayse and phostasese enzymes with Gmean enzymeatic activities were significantly improved. These findings suggest biochar and bacterial interaction for soil management to enhance soil resilience against pesticide stress.

Project description:In this study, cypermethrin toxicity was investigated using physiological, biochemical and cytogenetic parameters, and more than one organ and cell type was preferred to determine these effects. In this multifaceted study, the genotoxicity mechanism of cypermethrin was elucidated by molecular docking. In addition, comet assay technique was applied to detect and quantify DNA damage at the cell level. For this aim, body and organ weights, aspartate aminotransferase (AST), alanine aminotransferase (ALT), malondialdehyde (MDA), glutathione (GSH), blood urea nitrogen (BUN) and creatinine levels, mitotic index (MI), DNA fragmentation, frequency of micronucleus (MN) and chromosomal aberrations (CAs) were used as indicators of toxicity. Mice were divided into 4 groups. The control group was fed with tap water and the administration groups were orally exposed to 62.5, 125 and 250 mg/kg b.w cypermethrin for 28 days. Then, the mice were sacrificed and tissue samples were collected. Cypermethrin caused a decrease in body and organ weights, GSH levels and MI and an increase in AST, ALT, MDA, BUN, creatinine levels and the frequency of MN and CAs (break, ring, gap, acentric, etc.). Cypermethrin promoted MN formation in leukocyte, erythrocyte, buccal mucosa epithelial cells. CAs and MN formation promoted by cypermethrin have been associated with DNA-cypermethrin interactions. This interaction has been demonstrated by simulation with molecular docking method and experimentally by spectral measurements of DNA. As a result, all three doses of cypermethrin caused toxicity in different cell types. In other words, the effect of cypermethrin taken into the body was not limited to only one cell type or region. Therefore, cypermethrin is a pyrethroid insecticide that promotes multifaceted toxicity in non-target organisms.

Project description:Bioassay is a system for monitoring toxicity of chemicals in the environment via the biological responses of experimental organisms. These responses can be detected by analysis of genome-wide changes in mRNA expression levels using DNA microarray. We applied this system for evaluation of synergistic toxicity by cadmium and thiuram, as this combination showed mutual growth inhibition in yeast. Hierarchical cluster analysis using the mRNA expression profiles suggested the response of yeast to this combination is similar to that seen with cadmium treatment. Functional characterization of induced genes by this combination treatment also suggests the enhanced toxicity of cadmium. This toxicity was observed as the damage to mitochondrial functions which were not observed with either cadmium or thiuram treatments alone. The potential toxicity to mitochondria by this combinational treatment was confirmed as the result of mitochondrial curing. We could evaluate the synergistic toxicity by cadmium and thiuram and show the possible use of transcriptome bioassay for synergistic toxicity. Keywords: stress response

Project description:Previous studies have suggested an association between air pollution and lung disease. However, few studies have explored the relationship between chronic lung diseases classified by lung function and environmental parameters. This study aimed to comprehensively investigate the relationship between chronic lung diseases, air pollution, meteorological factors, and anthropometric indices. We conducted a cross-sectional study using the Taiwan Biobank and the Taiwan Air Quality Monitoring Database. A total of 2889 participants were included. We found a V/U-shaped relationship between temperature and air pollutants, with significant effects at both high and low temperatures. In addition, at lower temperatures (<24.6 °C), air pollutants including carbon monoxide (CO) (adjusted OR (aOR):1.78/Log 1 ppb, 95% CI 0.98-3.25; aOR:5.35/Log 1 ppb, 95% CI 2.88-9.94), nitrogen monoxide (NO) (aOR:1.05/ppm, 95% CI 1.01-1.09; aOR:1.11/ppm, 95% CI 1.07-1.15), nitrogen oxides (NOx) (aOR:1.02/ppm, 95% CI 1.00-1.05; aOR:1.06/ppm, 95% CI 1.04-1.08), and sulfur dioxide (SO2) (aOR:1.29/ppm, 95% CI 1.01-1.65; aOR:1.77/ppm, 95% CI 1.36-2.30) were associated with restrictive and mixed lung diseases, respectively. Exposure to CO, NO, NO2, NOx and SO2 significantly affected obstructive and mixed lung disease in southern Taiwan. In conclusion, temperature and air pollution should be considered together when evaluating the impact on chronic lung diseases.
