Virus Diversity, Abundance, and Evolution in Three Different Bat Colonies in Switzerland.

ABSTRACT: Bats are increasingly recognized as reservoirs for many different viruses that threaten public health, such as Hendravirus, Ebolavirus, Nipahvirus, and SARS- and MERS-coronavirus. To assess spillover risk, viromes of bats from different parts of the world have been investigated in the past. As opposed to most of these prior studies, which determined the bat virome at a single time point, the current work was performed to monitor changes over time. Specifically, fecal samples of three endemic Swiss bat colonies consisting of three different bat species were collected over three years and analyzed using next-generation sequencing. Furthermore, single nucleotide variants of selected DNA and RNA viruses were analyzed to investigate virus genome evolution. In total, sequences of 22 different virus families were found, of which 13 are known to infect vertebrates. Most interestingly, in a Vespertilio murinus colony, sequences from a MERS-related beta-coronavirus were consistently detected over three consecutive years, which allowed us to investigate viral genome evolution in a natural reservoir host.

Project description:Woodpecker diversity is usually higher in natural forests rich in dead wood and old trees than in managed ones, thus this group of birds is regarded as an indicator of forest biodiversity. Woodpeckers excavate cavities which can be subsequently used by several bird species. As a consequence, their abundance indicates high avian abundance and diversity in forests. However, woodpecker-made holes may be also important for other animals, for example, mammals but it has seldom been investigated so far. Here, we examine how well one species, the Great Spotted Woodpecker, predicts species richness, occurrence and acoustic activity of bats in Polish pine forests. In 2011 we conducted woodpecker and bat surveys at 63 point-count sites in forests that varied in terms of stand age, structure and amount of dead wood. From zero to five Great Spotted Woodpeckers at a point-count site were recorded. The total duration of the echolocation calls during a 10-min visit varied from 0 to 542 s and the number of bat species/species groups recorded during a visit ranged between zero to five. The local abundance of the woodpecker was positively correlated with bat species richness (on the verge of significance), bat occurrence and pooled bat activity. The occurrence of Eptesicus and Vespertilio bats and Nyctalus species was positively related with the abundance of the Great Spotted Woodpecker. The activity of Pipistrellus pygmaeus, Eptesicus and Vespertilio bats and a group of Myotis species was not associated with the woodpecker abundance, but echolocation calls of Nyctalus species, P. nathusii and P.pipistrellus were more often at sites with many Great Spotted Woodpeckers. Moreover, the probability of bat presence and the activity of bats was generally higher shortly after dusk and in middle of the summer than in late spring. We suggest that the observed correlations can be driven by similar roosting habitats (e.g., woodpeckers can provide breeding cavities for bats) or possibly by associated invertebrate food resources of woodpeckers and bats. The abundance of Great Spotted Woodpecker seems to be a good positive indicator of bat species richness, occurrence and activity, thus adding a group of relatively cryptic forest species that are indicated by the presence of the Great Spotted Woodpecker.

Project description:BackgroundStudies on mosquito species diversity in Europe often focus on a specific habitat, region or country. Moreover, different trap types are used for these sampling studies, making it difficult to compare and validate results across Europe. To facilitate comparisons of trapping sites and community analysis, the present study used two trap types for monitoring mosquito species diversity in three habitat types for three different countries in Europe.MethodsMosquitoes were trapped using Biogents Sentinel (BGS), and Mosquito Magnet Liberty Plus (MMLP) traps at a total of 27 locations in Sweden, the Netherlands and Italy, comprising farm, peri-urban and wetland habitats. From July 2014 to June 2015 all locations were sampled monthly, except for the winter months. Indices of species richness, evenness and diversity were calculated, and community analyses were carried out with non-metric multidimensional scaling (NMDS) techniques.ResultsA total of 11,745 female mosquitoes were trapped during 887 collections. More than 90% of the mosquitoes belonged to the genera Culex and Aedes, with Culex pipiens being the most abundant species. The highest mosquito diversity was found in Sweden. Within Sweden, species diversity was highest in wetland habitats, whereas in the Netherlands and Italy this was highest at farms. The NMDS analyses showed clear differences in mosquito communities among countries, but not among habitat types. The MMLP trapped a higher diversity of mosquito species than the BGS traps. Also, MMLP traps trapped higher numbers of mosquitoes, except for the genera Culex and Culiseta in Italy.ConclusionsA core mosquito community could be identified for the three countries, with Culex pipiens as the most abundant species. Differences in mosquito species communities were more defined by the three countries included in the study than by the three habitat types. Differences in mosquito community composition across countries may have implications for disease emergence and further spread throughout Europe. Future research should, therefore, focus on how field data of vector communities can be incorporated into models, to better assess the risk of mosquito-borne disease outbreaks.

Project description:BackgroundMosquito-borne dengue virus (DENV) is maintained in a sylvatic, enzootic cycle of transmission between canopy-dwelling non-human primates and Aedes mosquitoes in Borneo. Sylvatic DENV can spill over into humans living in proximity to forest foci of transmission, in some cases resulting in severe dengue disease. The most likely vectors of such spillover (bridge vectors) in Borneo are Ae. albopictus and Ae. niveus. Borneo is currently experiencing extensive forest clearance. To gauge the effect of this change in forest cover on the likelihood of sylvatic DENV spillover, it is first necessary to characterize the distribution of bridge vectors in different land cover types. In the current study, we hypothesized that Ae. niveus and Ae. albopictus would show significantly different distributions in different land cover types; specifically, we predicted that Ae. niveus would be most abundant in forests whereas Ae. albopictus would have a more even distribution in the landscape.ResultsMosquitoes were collected from a total of 15 sites using gravid traps and a backpack aspirator around Kampong Puruh Karu, Sarawak, Malaysian Borneo, where sylvatic DENV spillover has been documented. A total of 2447 mosquitoes comprising 10 genera and 4 species of Aedes, were collected over the three years, 2013, 2014 and 2016, in the three major land cover types in the area, homestead, agriculture and forest. Mosquitoes were identified morphologically, pooled by species and gender, homogenized, and subject to DNA barcoding of each Aedes species and to arbovirus screening. As predicted, Ae. niveus was found almost exclusively in forests whereas Ae. albopictus was collected in all land cover types. Aedes albopictus was significantly (P = 0.04) more abundant in agricultural fields than forests. Sylvatic DENV was not detected in any Aedes mosquito pools, however genomes of 14 viruses were detected using next generation sequencing.ConclusionsLand cover type affects the abundance and distribution of the most likely bridge vectors of sylvatic DENV in Malaysia Borneo. Conversion of forests to agriculture will likely decrease the range and abundance of Ae. niveus but enhance the abundance of Ae. albopictus.
