Project description:Crop production under hydroponic environments has many advantages, yet the effects of solution flow rate on plant growth remain unclear. We conducted a hydroponic cultivation study using different flow rates under light-emitting diode lighting to investigate plant growth, nutrient uptake, and root morphology under different flow rates. Swiss chard plants were grown hydroponically under four nutrient solution flow rates (2 L/min, 4 L/min, 6 L/min, and 8 L/min). After 21 days, harvested plants were analyzed for root and shoot fresh weight, root and shoot dry weight, root morphology, and root cellulose and hemicellulose content. We found that suitable flow rates, acting as a eustress, gave the roots appropriate mechanical stimulation to promote root growth, absorb more nutrients, and increase overall plant growth. Conversely, excess flow rates acted as a distress that caused the roots to become compact and inhibited root surface area and root growth. Excess flow rate thereby resulted in a lower root surface area that translated to reduced nutrient ion absorption and poorer plant growth compared with plans cultured under a suitable flow rate. Our results indicate that regulating flow rate can regulate plant thigmomorphogenesis and nutrient uptake, ultimately affecting hydroponic crop quality.

Project description:While the majority of studies explore soot formation in relatively simple, one-dimensional flames, most real-world flames consist of complex flows defined by large-scale turbulent eddies, recirculating flow patterns, and buoyancy effects. The effects of complex flow on soot physicochemical properties are poorly understood. This work employs an inverted gravity flame reactor (IGFR) to compare differences in soot growth between a one-dimensional laminar diffusion flame and a recirculating flame. Computational fluid dynamics (CFD) and experimental observations show particle oscillations between (i) a rich region with a high concentration of surface growth species, and (ii) a high-temperature oxidation region. Transmission electron microscopy (TEM) shows a significant difference in final primary particle diameter, where the one-dimensional flame produces primary particles 10 to 25 nm in diameter and the recirculating flame produces primary particles 25 to 75 nm in diameter. Additionally, larger primary particles from the recirculating flame contain both single and multiple cores. We propose that due to the spheroidal shape of the large primary particles, the secondary surface growth is primarily a result of polyaromatic hydrocarbon (PAH) condensation during re-entrainment of mature soot into the fuel-rich region followed by subsequent liquid layer carbonization in the high-temperature environment of the flame front. The recirculating flow patterns in the IGFR and repeated particle growth/oxidation cycle can serve as a model for soot formation in the large-scale flames with complex flow patterns, such as forest fires, coal fire plants, and other sources.

Project description:Surface waves process the turbulent disturbances which drive dynamics in many space, astrophysical and laboratory plasma systems, with the outer boundary of Earth's magnetosphere, the magnetopause, providing an accessible environment to study them. Like waves on water, magnetopause surface waves are thought to travel in the direction of the driving solar wind, hence a paradigm in global magnetospheric dynamics of tailward propagation has been well-established. Here we show through multi-spacecraft observations, global simulations, and analytic theory that the lowest-frequency impulsively-excited magnetopause surface waves, with standing structure along the terrestrial magnetic field, propagate against the flow outside the boundary. Across a wide local time range (09-15h) the waves' Poynting flux exactly balances the flow's advective effect, leading to no net energy flux and thus stationary structure across the field also. Further down the equatorial flanks, however, advection dominates hence the waves travel downtail, seeding fluctuations at the resonant frequency which subsequently grow in amplitude via the Kelvin-Helmholtz instability and couple to magnetospheric body waves. This global response, contrary to the accepted paradigm, has implications on radiation belt, ionospheric, and auroral dynamics and potential applications to other dynamical systems.

Project description:In this study, multiphysics simulations were carried out to understand the convection mechanisms of the top seeded solution growth (TSSG) of SiC. Experimental melting tests and crystal growth were conducted to verify the simulation results in the growing temperatures between 1700 and 1900 °C with rf induction heating furnace. From the solidified melt of Si-Cr solution after the melting test, the melt flow in the simulation was successfully verified. In the given experimental conditions, the electromagnetic convection was found to govern the global fluid flow, while other mechanisms including the Marangoni convection, the buoyancy convection and the centrifugal forced convection influence the fluid flow near the crystal. Based on an understanding of the fluid flow obtained with the simulations, a structural flow modifier (FM) was applied to enhance the growth rate of the SiC crystal. The growth rates of SiC with/without FM were successfully estimated from simulations showing good agreements with the experimental values. After the experimental crystal growth using FM, a remarkable enhancement in the growth rate was found in an FM configuration, which suggests a way to improve the growth rate by the TSSG method based on the efficient use of the dissolved C in the melt.

Project description:The aim was to evaluate the effect of different feed additives on intake, performance, and fecal consistency index (FCI) of dairy calves from 6-60 d of age and its residual effect 15 d after weaning. Fifty Holstein calves (38 ± 1.0 kg BW) were fed 5 L/d of milk plus starter feed until weaning, and corn silage and concentrate after weaning. The treatments were: control (CON), monensin (MON; 30 mg/kg of starter), probiotic E. faecium (PROB; 70 mg/kg of starter), essential oils (EO; 300 mg/kg of starter), or PROB + EO (EOPROB). Fecal score and dry matter intake (DMI) were measured daily, and animals were weighed every 15 d. A DNA extraction from feces was performed to identify the presence of microorganisms (E. coli, Hafnia, Shiguella, Lactobacillus spp, Enterococcus spp, and Enterococcus faecium NCIMB 10415) by PCR. Two 72-h digestibility trials were performed at days 20-28 and 50-56, by total fecal collection. The DMI before weaning was greater for EO (903.0 g/d) compared with MON (794.3 g/d) and EOPROB (783.1 g/d). The FCI decreased during pre-weaning for EO and MON. Average daily gain (ADG) and feed efficiency (FE) did not differ among treatments before weaning. After weaning, DMI and FCI did not differ among treatments. The EO had greater ADG (917.5 g/d) compared with CON (615.8 g/d) and PROB (592.6 g/d). The FE improved with EO (0.72 g/g) over CON (0.36 g/g), MON (0.49 g/g), and PROB (0.36 g/g). The PCR results showed absence of E. faecium NCIMB 10415 in animals fed PROB and CON. Animals fed PROB had greater intake of CP and NDF than animals fed EOPROB. The EO can be added to the dairy calf ration to improve fecal score and increase DMI. The pre-weaning FCI decrease with MON and increase with PROB.

Project description:As crystallization processes are often rapid, it can be difficult to monitor their growth mechanisms. In this study, we made use of the fact that crystallization proceeds more slowly in small volumes than in bulk solution to investigate the effects of the soluble additives Mg2+ and poly(styrene sulfonate) (PSS) on the early stages of growth of calcite crystals. Using a "Crystal Hotel" microfluidic device to provide well-defined, nanoliter volumes, we observed that calcite crystals form via an amorphous precursor phase. Surprisingly, the first calcite crystals formed are perfect rhombohedra, and the soluble additives have no influence on the morphology until the crystals reach sizes of 0.1-0.5??m for Mg2+ and 1-2??m for PSS. The crystals then continue to grow to develop morphologies characteristic of these additives. These results can be rationalized by considering additive binding to kink sites, which is consistent with crystal growth by a classical mechanism.

Project description:The presence of many salts, such as sodium chloride, can adversely affect the performance of native electrospray ionization mass spectrometry for the analysis of proteins and protein complexes by reducing the overall molecular ion abundances and distributing signal for any given charge state into many cationized forms with various numbers of adducts attached. Several solution additives, such as ammonium bromide, ammonium iodide, and NaSbF(6), can significantly lower the extent of sodium ion adduction to the molecular ions of proteins and protein complexes. For ubiquitin, addition of 25 mM ammonium bromide or ammonium iodide into aqueous solutions also containing 1.0 mM NaCl results in a factor of 72 and 56 increase, respectively, in the relative abundances of the fully protonated molecular ions compared to when these additives are not present. The effectiveness of this method for reducing sodium ion adduction is related to the low proton affinity (PA) values of the anions. Anions with very low PA also have a propensity to adduct as an acid molecule, but these adducts can be readily dissociated from the molecular ions either by activation in the source or subsequently by collisional activation in the mass spectrometer. This method of reducing sodium ion adduction to proteins is simple and requires no experimental modifications, making it an attractive alternative to other methods for desalting proteins prior to mass spectrometry analysis.

Project description:We use regular solution theory and implement a three-gradient model for a liquid/vapour system in contact with a complex surface topology to study the shape of a liquid drop in advancing and receding wetting scenarios. More specifically, we study droplets on an inverse opal: spherical cavities in a hexagonal pattern. In line with experimental data, we find that the surface may switch from hydrophilic (contact angle on a smooth surface ?Y < 90°) to hydrophobic (effective advancing contact angle ? > 90°). Both the Wenzel wetting state, that is cavities under the liquid are filled, as well as the Cassie-Baxter wetting state, that is air entrapment in the cavities under the liquid, were observed using our approach, without a discontinuity in the water front shape or in the water advancing contact angle ?. Therefore, air entrapment cannot be the main reason why the contact angle ? for an advancing water front varies. Rather, the contact line is pinned and curved due to the surface structures, inducing curvature perpendicular to the plane in which the contact angle ? is observed, and the contact line does not move in a continuous way, but via depinning transitions. The pinning is not limited to kinks in the surface with angles ?kink smaller than the angle ?Y. Even for ?kink > ?Y, contact line pinning is found. Therefore, the full 3D-structure of the inverse opal, rather than a simple parameter such as the wetting state or ?kink, determines the final observed contact angle.

Project description:Alginate is a promising biocompatible and biodegradable polymer for production of nanofibers for drug delivery and tissue engineering. However, alginate is difficult to electrospin due to its polyelectrolyte nature. The aim was to improve the 'electrospinability' of alginate with addition of exceptionally high molecular weight poly(ethylene oxide) (PEO) as a co-polymer. The compositions of the polymer-blend solutions for electrospinning were varied for PEO molecular weight, total (alginate plus PEO) polymer concentration, and PEO proportion in the dry alginate-PEO polymer mix used. These were tested for rheology (viscosity, complex viscosity, storage and loss moduli) and conductivity, and the electrospun nanofibers were characterized by scanning electron microscopy. One-parameter-at-a-time approach and response surface methodology (RSM) were used to optimize the polymer-blend solution composition to obtain defined nanofibers. Both approaches revealed that the major influence on nanofiber formation and diameter were total polymer concentration and PEO proportion. These polymer-blend solutions of appropriate conductivity and viscosity enabled fine-tuning of nanofiber diameter. PEO molecular weight of 2-4 million Da greatly improved the electrospinnability of alginate, producing nanofibers with >85% alginate. This study shows that RSM can be used to design nanofibers with optimal alginate and co-polymer contents to provide efficient scaffold material for regenerative medicine.

Project description:Almost 200 years ago, benzamide was reported as polymorphic with two of its forms (II and III) found to be difficult to crystallise. In a recent study, it was shown that benzamide form I can easily convert into benzamide form III using mechanochemistry in the presence of nicotinamide. Here we show, experimentally and computationally, that this transformation is the result of a thermodynamic switch between these two polymorphic forms driven by the formation of solid solutions with small amounts of nicotinamide. The presence of nicotinamide in the crystallisation environment promotes the robust and exclusive crystallisation of the elusive form III. These results represent a promising route to the synthesis and utilisation of elusive polymorphs of pharmaceutical interest.
