Pupillometry tracks errors in interval timing.

ABSTRACT: Recent primate studies suggest a potential link between pupil size and subjectively elapsed duration. Here, we sought to investigate the relationship between pupil size and perceived duration in human participants performing two temporal bisection tasks in the subsecond and suprasecond interval ranges. In the subsecond task, pupil diameter was greater during stimulus processing when shorter intervals were overestimated but also during and after stimulus offset when longer intervals were underestimated. By contrast, in the suprasecond task, larger pupil diameter was observed only in the late stimulus offset phase prior to response prompts when longer intervals were underestimated. This pattern of results suggests that pupil diameter relates to an error monitoring mechanism in interval timing. These results are at odds with a direct relationship between pupil size and the perception of duration but suggest that pupillometric variation might play a key role in signifying errors related to temporal judgments. Effects of low and moderate refractive errors on chromatic pupillometry.

Project description:Chromatic pupillometry is an emerging modality in the assessment of retinal and optic nerve disorders. Herein, we evaluate the effect of low and moderate refractive errors on pupillary responses to blue- and red-light stimuli in a healthy older population. This study included 139 participants (≥50 years) grouped by refractive error: moderate myopes (>-6.0D and ≤-3.0D, n = 24), low myopes (>-3.0D and <-0.5D, n = 30), emmetropes (≥-0.5D and ≤0.5D, n = 31) and hyperopes (>0.5D and <6.0D, n = 54). Participants were exposed to logarithmically ramping-up blue (462 nm) and red (638 nm) light stimuli, designed to sequentially activate rods, cones and intrinsically-photosensitive retinal ganglion cells. Pupil size was assessed monocularly using infra-red pupillography. Baseline pupil diameter correlated inversely with spherical equivalent (R = -0.26, P < 0.01), and positively with axial length (R = 0.37, P < 0.01) and anterior chamber depth (R = 0.43, P < 0.01). Baseline-adjusted pupillary constriction amplitudes to blue light did not differ between groups (P = 0.45), while constriction amplitudes to red light were greater in hyperopes compared to emmetropes (P = 0.04) at moderate to bright light intensities (12.25-14.0 Log photons/cm²/s). Our results demonstrate that low and moderate myopia do not alter pupillary responses to ramping-up blue- and red-light stimuli in healthy older individuals. Conversely, pupillary responses to red light should be interpreted cautiously in hyperopic eyes.

Temporal context calibrates interval timing.

Project description:We use our sense of time to identify temporal relationships between events and to anticipate actions. The degree to which we can exploit temporal contingencies depends on the variability of our measurements of time. We asked humans to reproduce time intervals drawn from different underlying distributions. As expected, production times were more variable for longer intervals. However, production times exhibited a systematic regression toward the mean. Consequently, estimates for a sample interval differed depending on the distribution from which it was drawn. A performance-optimizing Bayesian model that takes the underlying distribution of samples into account provided an accurate description of subjects' performance, variability and bias. This finding suggests that the CNS incorporates knowledge about temporal uncertainty to adapt internal timing mechanisms to the temporal statistics of the environment.

Pupillometry tracks cognitive load and salience network activity in a working memory functional magnetic resonance imaging task.

Project description:The diameter of the human pupil tracks working memory processing and is associated with activity in the frontoparietal network. At the same time, recent neuroimaging research has linked human pupil fluctuations to activity in the salience network. In this combined functional magnetic resonance imaging (fMRI)/pupillometry study, we recorded the pupil size of healthy human participants while they performed a blockwise organized working memory task (N-back) inside an MRI scanner in order to monitor the pupil fluctuations associated neural activity during working memory processing. We first confirmed that mean pupil size closely followed working memory load. Combining this with fMRI data, we focused on blood oxygen level dependent (BOLD) correlates of mean pupil size modeled onto the task blocks as a parametric modulation. Interrogating this modulated task regressor, we were able to retrieve the frontoparietal network. Next, to fully exploit the within-block dynamics, we divided the blocks into 1 s time bins and filled these with corresponding pupil change values (first-order derivative of pupil size). We found that pupil change within N-back blocks was positively correlated with BOLD amplitudes in the areas of the salience network (namely bilateral insula, and anterior cingulate cortex). Taken together, fMRI with simultaneous measurement of pupil parameters constitutes a valuable tool to dissect working memory subprocesses related to both working memory load and salience of the presented stimuli.

Self-paced timing detects and tracks change in prodromal Huntington disease.

Project description:This study compares self-paced timing performance (cross-sectionally and longitudinally) between participants with prodromal Huntington's disease (pr-HD) and a comparison group of gene non-expanded participants from affected families (NC).Participants (747 pr-HD: 188 NC) listened to tones presented at 550-ms intervals, matched that pace by tapping response keys and continued the rhythm (self-paced) after the tone had stopped. Standardized cross-sectional and longitudinal linear models examined the relationships between self-paced timing precision and estimated proximity to diagnosis, and other demographic factors.Pr-HD participants showed significantly less timing precision than NC. Comparison of pr-HD and NC participants showed a significant performance difference on two task administration conditions (dominant hand: p < .0001; alternating thumbs: p < .0001). Additionally, estimated proximity to diagnosis was related to timing precision in both conditions, (dominant hand: t = -11.14, df = 920, p < .0001; alternating thumbs: t = -11.32, df = 918, p < .0001). Longitudinal modeling showed that pr-HD participants worsen more quickly at the task than the NC group, and rate of decline increases with estimated proximity to diagnosis in both conditions (dominant hand: t = -2.85, df = 417, p = .0045; alternating thumbs: t = -3.56, df = 445, p = .0004). Effect sizes based on adjusted mean annual change ranged from -0.34 to 0.25 in the longitudinal model.The self-paced timing paradigm has potential for use as a screening tool and outcome measure in pr-HD clinical trials to gauge therapeutically mediated improvement or maintenance of function.

Conceptually plausible Bayesian inference in interval timing.

Project description:In a world that is uncertain and noisy, perception makes use of optimization procedures that rely on the statistical properties of previous experiences. A well-known example of this phenomenon is the central tendency effect observed in many psychophysical modalities. For example, in interval timing tasks, previous experiences influence the current percept, pulling behavioural responses towards the mean. In Bayesian observer models, these previous experiences are typically modelled by unimodal statistical distributions, referred to as the prior. Here, we critically assess the validity of the assumptions underlying these models and propose a model that allows for more flexible, yet conceptually more plausible, modelling of empirical distributions. By representing previous experiences as a mixture of lognormal distributions, this model can be parametrized to mimic different unimodal distributions and thus extends previous instantiations of Bayesian observer models. We fit the mixture lognormal model to published interval timing data of healthy young adults and a clinical population of aged mild cognitive impairment patients and age-matched controls, and demonstrate that this model better explains behavioural data and provides new insights into the mechanisms that underlie the behaviour of a memory-affected clinical population.

Opposing Timing Constraints Severely Limit the Use of Pupillometry to Investigate Visual Statistical Learning.

Project description:Majority of visual statistical learning (VSL) research uses only offline measures, collected after the familiarization phase (i.e., learning) has occurred. Offline measures have revealed a lot about the extent of statistical learning (SL) but less is known about the learning mechanisms that support VSL. Studies have shown that prediction can be a potential learning mechanism for VSL, but it is difficult to examine the role of prediction in VSL using offline measures alone. Pupil diameter is a promising online measure to index prediction in VSL because it can be collected during learning, requires no overt action or task and can be used in a wide-range of populations (e.g., infants and adults). Furthermore, pupil diameter has already been used to investigate processes that are part of prediction such as prediction error and updating. While the properties of pupil diameter have the potentially to powerfully expand studies in VSL, through a series of three experiments, we find that the two are not compatible with each other. Our results revealed that pupil diameter, used to index prediction, is not related to offline measures of learning. We also found that pupil differences that appear to be a result of prediction, are actually a result of where we chose to baseline instead. Ultimately, we conclude that the fast-paced nature of VSL paradigms make it incompatible with the slow nature of pupil change. Therefore, our findings suggest pupillometry should not be used to investigate learning mechanisms in fast-paced VSL tasks.

Influence of Recent Trial History on Interval Timing.

Project description:Interval timing is involved in a variety of cognitive behaviors such as associative learning and decision-making. While it has been shown that time estimation is adaptive to the temporal context, it remains unclear how interval timing behavior is influenced by recent trial history. Here we found that, in mice trained to perform a licking-based interval timing task, a decrease of inter-reinforcement interval in the previous trial rapidly shifted the time of anticipatory licking earlier. Optogenetic inactivation of the anterior lateral motor cortex (ALM), but not the medial prefrontal cortex, for a short time before reward delivery caused a decrease in the peak time of anticipatory licking in the next trial. Electrophysiological recordings from the ALM showed that the response profiles preceded by short and long inter-reinforcement intervals exhibited task-engagement-dependent temporal scaling. Thus, interval timing is adaptive to recent experience of the temporal interval, and ALM activity during time estimation reflects recent experience of interval.

A model of interval timing by neural integration.

Project description:We show that simple assumptions about neural processing lead to a model of interval timing as a temporal integration process, in which a noisy firing-rate representation of time rises linearly on average toward a response threshold over the course of an interval. Our assumptions include: that neural spike trains are approximately independent Poisson processes, that correlations among them can be largely cancelled by balancing excitation and inhibition, that neural populations can act as integrators, and that the objective of timed behavior is maximal accuracy and minimal variance. The model accounts for a variety of physiological and behavioral findings in rodents, monkeys, and humans, including ramping firing rates between the onset of reward-predicting cues and the receipt of delayed rewards, and universally scale-invariant response time distributions in interval timing tasks. It furthermore makes specific, well-supported predictions about the skewness of these distributions, a feature of timing data that is usually ignored. The model also incorporates a rapid (potentially one-shot) duration-learning procedure. Human behavioral data support the learning rule's predictions regarding learning speed in sequences of timed responses. These results suggest that simple, integration-based models should play as prominent a role in interval timing theory as they do in theories of perceptual decision making, and that a common neural mechanism may underlie both types of behavior.

Accuracy and precision of visual stimulus timing in PsychoPy: no timing errors in standard usage.

Project description:In a recent report published in PLoS ONE, we found that the performance of PsychoPy degraded with very short timing intervals, suggesting that it might not be perfectly suitable for experiments requiring the presentation of very brief stimuli. The present study aims to provide an updated performance assessment for the most recent version of PsychoPy (v1.80) under different hardware/software conditions. Overall, the results show that PsychoPy can achieve high levels of precision and accuracy in the presentation of brief visual stimuli. Although occasional timing errors were found in very demanding benchmarking tests, there is no reason to think that they can pose any problem for standard experiments developed by researchers.

Temporal Context affects interval timing at the perceptual level.

Project description:There is now ample evidence that when observers are asked to estimate features of an object they take into account recent stimulation history and blend the current sensory evidence with the recent stimulus intensity according to their reliability. Most of this evidence has been obtained via estimation or production paradigms both of which entail a conspicuous post-perceptual decision stage. So it is an unsolved question, as to whether the trace of previous stimulation contributes at the decision stage or as early as the perceptual stage. To this aim we focused on duration judgments, which typically exhibit strong central tendency effects and asked a duration comparison between two intervals, one of which characterized by high uncertainty. We found that the perceived duration of this interval regressed toward the average duration, demonstrating a genuine perceptual bias. Regression did not transfer between the visual and the auditory modality, indicating it is modality specific, but generalized across passively observed and actively produced intervals. These findings suggest that temporal central tendency effects modulate how long an interval appears to us and that integration of current sensory evidence can occur as early as in the sensory systems.
