High Levels of Policosanols and Phytosterols from Sugar Mill Waste by Subcritical Liquefied Dimethyl Ether.

ABSTRACT: Extracting nutraceuticals with high value from bagasse, filter mud, and sugarcane leaves discarded as sugar mill by-products, is crucial for the development of a sustainable bio-economy. These by-products are important sources of policosanols and phytosterols, which have a cholesterol-lowering effect. This research focused on using a promising green technology, subcritical liquefied dimethyl ether extraction, with a low pressure of 0.8 MPa, to extract policosanols and phytosterols and on application of pretreatments to increase their contents. For direct extraction by subcritical liquefied dimethyl ether without sample pretreatment, the highest extraction yield (7.4%) and policosanol content were found in sugarcane leaves at 2888 mg/100 g, while the highest and lowest phytosterol contents were found in filter mud at 20,878.75 mg/100 g and sugarcane leaves at 10,147.75 mg/100 g, respectively. Pretreatment of filter mud by ultrasonication in hexane solution together with transesterification before the second subcritical liquefied dimethyl ether extraction successfully increased the policosanol content, with an extract purity of 60%, but failed to increase the phytosterol content.

Project description:BackgroundRecent studies of lipid extraction from microalgae have focused primarily on dewatered or dried samples, and the processes are simple with high lipid yield. Yet, the dewatering with drying step is energy intensive, which makes the energy input during the lipid production more than energy output from obtained lipid. Thus, exploring an extraction technique for just a thickened sample without the dewatering, drying and auxiliary operation (such as cell disruption) is very significant. Whereas lipid extraction from the thickened microalgae is complicated by the high water content involved, and traditional solvent, hence, cannot work well. Dimethyl ether (DME), a green solvent, featuring a high affinity for both water and organic compounds with an ability to penetrate the cell walls has the potential to achieve this goal.ResultsThis study investigated an energy-saving method for lipid extraction using DME as the solvent with an entrainer solution (ethanol and acetone) for flocculation-thickened microalgae. Extraction efficiency was evaluated in terms of extraction time, DME dosage, entrainer dosage, and ethanol:acetone ratio. Optimal extraction occurred after 30 min using 4.2 mL DME per 1 mL microalgae, with an entrainer dosage of 8% at 1:2 ethanol:acetone. Raw lipid yields and its lipid component (represented by fatty acid methyl ester) contents were compared against those of common extraction methods (Bligh and Dryer, and Soxhlet). Thermal gravimetry/differential thermal analysis, Fourier-transform infrared spectroscopy, and C/H/N elemental analyses were used to examine differences in lipids extracted using each of the evaluated methods. Considering influence of trace metals on biodiesel utilization, inductively coupled plasma mass spectrometry and inductively coupled plasma atomic emission spectroscopy analyses were used to quantify trace metals in the extracted raw lipids, which revealed relatively high concentrations of Mg, Na, K, and Fe.ConclusionsOur DME-based method recovered 26.4% of total raw lipids and 54.4% of total fatty acid methyl esters at first extraction with remnants being recovered by a 2nd extraction. In additional, the DME-based approach was more economical than other methods, because it enabled simultaneous dewatering with lipid extraction and no cell disruption was required. The trace metals of raw lipids indicated a purification demand in subsequent refining process.

Project description:A key challenge in chemicals biomanufacturing is the maintenance of stable, highly productive microbial strains to enable cost-effective fermentation at scale. A "cookie-cutter" approach to microbial engineering is often used to optimize host stability and productivity. This can involve identifying potential limitations in strain characteristics followed by attempts to systematically optimize production strains by targeted engineering. Such targeted approaches however do not always lead to the desired traits. Here, we demonstrate both 'hit and miss' outcomes of targeted approaches in attempts to generate a stable Escherichia coli strain for the bioproduction of the monoterpenoid linalool, a fragrance molecule of industrial interest. First, we stabilized linalool production strains by eliminating repetitive sequences responsible for excision of pathway components in plasmid constructs that encode the pathway for linalool production. These optimized pathway constructs were then integrated within the genome of E. coli in three parts to eliminate a need for antibiotics to maintain linalool production. Additional strategies were also employed including: reduction in cytotoxicity of linalool by adaptive laboratory evolution and modification or homologous gene replacement of key bottleneck enzymes GPPS/LinS. Our study highlights that a major factor influencing linalool titres in E. coli is the stability of the genetic construct against excision or similar recombination events. Other factors, such as decreasing linalool cytotoxicity and changing pathway genes, did not lead to improvements in the stability or titres obtained. With the objective of reducing fermentation costs at scale, the use of minimal base medium containing paper mill wastewater secondary paper fiber as sole carbon source was also investigated. This involved simultaneous saccharification and fermentation using either supplemental cellulase blends or by co-expressing secretable cellulases in E. coli containing the stabilized linalool production pathway. Combined, this study has demonstrated a stable method for linalool production using an abundant and low-cost feedstock and improved production strains, providing an important proof-of-concept for chemicals production from paper mill waste streams. For scaled production, optimization will be required, using more holistic approaches that involve further rounds of microbial engineering and fermentation process development.
