Combined Inhibitory Effect of Canada Goldenrod Invasion and Soil Microplastics on Rice Growth.

ABSTRACT: Alien plant invasion and residual soil microplastics (MPs) are growing threats to agricultural crop production. This study determined the adverse effects of Canadian goldenrod (Solidago canadensis L.) invasion and residual soil MPs on rice growth and development. The biomass, phenological indices, photosynthetic parameters, and antioxidant enzyme activities of rice were measured on the 50th and 80th day of post-plantation. Biomass and phenotypic results indicated the more harmful effects of the combination of S. canadensis invasion and residual soil MPs compared to S. canadensis invasion or residual soil MPs effects alone. Moreover, the interaction effect of S. canadensis invasion and residual soil MPs markedly reduced the ascorbate peroxidase and catalase belowground, while they increased in the aboveground parts of the rice. However, the S. canadensis invasion and residual soil MPs interactive treatments lowered the superoxide dismutase concentrations in the belowground parts of the rice plants while elevating the peroxidase and reactive oxygen species concentrations in both the belowground and aboveground parts compared to the other treatments. Among all treatments, S. canadensis invasion alone had the most negligible negative impact on rice biomass and growth indices. Our study suggests that soil MPs could negatively affect crop production with invasive alien plants, and the combined effects were more harmful than either of the single factors. Our findings will lay the groundwork for analyzing the impacts of invasive alien plants on rice crops.

Project description:Microplastics affect plants and soil biota and the processes they drive. However, the legacy effect of microplastics on plant-soil feedbacks is still unknown. To address this, we used soil conditioned from a previous experiment, where Daucus carota grew with 12 different microplastic types (conditioning phase). Here, we extracted soil inoculum from those 12 soils and grew during 4 weeks a native D. carota and a range-expanding plant species Calamagrostis epigejos in soils amended with this inoculum (feedback phase). At harvest, plant biomass and root morphological traits were measured. Films led to positive feedback on shoot mass (higher mass with inoculum from soil conditioned with microplastics than with inoculum from control soil). Films may decrease soil water content in the conditioning phase, potentially reducing the abundance of harmful soil biota, which, with films also promoting mutualist abundance, microbial activity and carbon mineralization, would positively affect plant growth in the feedback phase. Foams and fragments caused positive feedback on shoot mass likely via positive effects on soil aeration in the conditioning phase, which could have increased mutualistic biota and soil enzymatic activity, promoting plant growth. By contrast, fibers caused negative feedback on root mass as this microplastic may have increased soil water content in the conditioning phase, promoting the abundance of soil pathogens with negative consequences for root mass. Microplastics had a legacy effect on root traits: D. carota had thicker roots probably for promoting mycorrhizal associations, while C. epigejos had reduced root diameter probably for diminishing pathogenic infection. Microplastic legacy on soil can be positive or negative depending on the plant species identity and may affect plant biomass primarily via root traits. This legacy may contribute to the competitive success of range-expanding species via positive effects on root mass (foams) and on shoot mass (PET films). Overall, microplastics depending on their shape and polymer type, affect plant-soil feedbacks.

Project description:BackgroundThe application of beneficial microbes in agriculture is gaining increasing attention as a means to reduce reliance on chemical fertilizers. This approach can potentially mitigate negative impacts on soil, animal, and human health, as well as decrease climate-changing factors. Among these microbes, yeast has been the least explored, particularly within the phyllosphere compartment. This study addresses this knowledge gap by investigating the potential of phyllosphere yeast to improve rice yield while reducing fertilizer dosage.ResultsFrom fifty-two rice yeast phyllosphere isolates, we identified three yeast strains-Rhodotorula paludigena Y1, Pseudozyma sp. Y71, and Cryptococcus sp. Y72-that could thrive at 36 °C and possessed significant multifarious plant growth-promoting traits, enhancing rice root and shoot length upon seed inoculation. These three strains demonstrated favorable compatibility, leading to the creation of a yeast consortium. We assessed the combined effect of foliar application of this yeast consortium and individual strains with two distinct recommended doses of chemical fertilizers (RDCFs) (75 and 100%), as well as RDCFs alone (75 and 100%), in rice maintained in pot-culture and field experiments. The pot-culture experiment investigated the leaf microbial community, plant biochemicals, root and shoot length during the stem elongation, flowering, and dough phases, and yield-related parameters at harvest. The field experiment determined the actual yield. Integrated results from both experiments revealed that the yeast consortium with 75% RDCFs was more effective than the yeast consortium with 100% RDCFs, single strain applications with RDCFs (75 and 100%), and RDCFs alone (75 and 100%). Additionally, this treatment improved leaf metabolite levels compared to control rice plants.ConclusionsOverall, a 25% reduction in soil chemical fertilizers combined with yeast consortium foliar application improved rice growth, biochemicals, and yield. This study also advances the field of phyllosphere yeast research in agriculture.

Project description:Accurate and effective analysis of microplastics depends on the purification of any biogenic matter present in the environmental sample. Within the soil environment, the presence of biogenic materials (living and non-living) obstruct reliable identification of microplastics. However, while conventional acidic, alkaline, and wet peroxidation methods are often used for microplastics purification, they can result in significant alteration to the polymer integrity. To overcome this issue of polymer damage, we have developed a more efficient protocol using enzymes to eliminate organic materials from soil media without affecting the synthetic polymers. This method describes a simple enzymatic protocol that eliminates the organic matter based on the application of enzymes that target the three natural components of lignocellulosic biomass (cellulose, hemicellulose, and lignin). The enzymatic protocol introduced in this study allows for the use of an oxidizing agent for the pre-treatment of samples and density separation step. Additionally, this method was used to determine the enzymatic digestion efficiencies of soil organic matter and recovery rates of microplastics. Finally, the recovered microplastics were characterized by techniques including stereomicroscopy, FT-IR, and SEM. This method proved to be effective in reducing approximately >90% of organic materials in soil samples. After showing a high digestion efficiency the method was applied to spiked microplastic soil samples where high recovery rates were established and plastic characteristics were conserved. Despite being a time consuming and expensive method when compared to other purification methods. The key benefits of this methodology are:•Minimal sample preparation•Representative purification of microplastic from diverse soil media; and•Most importantly, preservation of polymer integrity aiding requirements needed for particle identification (e.g.,% mass polymer remaining, SEM images revealing no obvious polymer morphology change after enzyme treatment).
