Project description:Cognitive aging is associated with widespread neural reorganization processes in the human brain. However, the behavioral impact of such reorganization is not well understood. The current neuroimaging study investigated age differences in the functional network architecture during semantic word retrieval in young and older adults. Combining task-based functional connectivity, graph theory and cognitive measures of fluid and crystallized intelligence, our findings show age-accompanied large-scale network reorganization even when older adults have intact word retrieval abilities. In particular, functional networks of older adults were characterized by reduced decoupling between systems, reduced segregation and efficiency, and a larger number of hub regions relative to young adults. Exploring the predictive utility of these age-related changes in network topology revealed high, albeit less efficient, performance for older adults whose brain graphs showed stronger dedifferentiation and reduced distinctiveness. Our results extend theoretical accounts on neurocognitive aging by revealing the compensational potential of the commonly reported pattern of network dedifferentiation when older adults can rely on their prior knowledge for successful task processing. However, we also demonstrate the limitations of such compensatory reorganization and show that a youth-like network architecture in terms of balanced integration and segregation is associated with more economical processing.

Project description:Aging has a multi-faceted impact on brain structure, brain function and cognitive task performance, but the interaction of these different age-related changes is largely unexplored. We hypothesize that age-related structural changes alter the functional connectivity within the brain, resulting in altered task performance during cognitive challenges. In this neuroimaging study, we used independent components analysis to identify spatial patterns of coordinated functional activity involved in the performance of a verbal delayed item recognition task from 75 healthy young and 37 healthy old adults. Strength of functional connectivity between spatial components was assessed for age group differences and related to speeded task performance. We then assessed whether age-related differences in global brain volume were associated with age-related differences in functional network connectivity. Both age groups used a series of spatial components during the verbal working memory task and the strength and distribution of functional network connectivity between these components differed across the age groups. Poorer task performance, i.e. slower speed with increasing memory load, in the old adults was associated with decreases in functional network connectivity between components comprised of the supplementary motor area and the middle cingulate and between the precuneus and the middle/superior frontal cortex. Advancing age also led to decreased brain volume; however, there was no evidence to support the hypothesis that age-related alterations in functional network connectivity were the result of global brain volume changes. These results suggest that age-related differences in the coordination of neural activity between brain regions partially underlie differences in cognitive performance.

Project description:PurposeTo study age-related changes in corneal deformation response to air-puff applanation tonometry.MethodsFifty healthy subjects were recruited for a prospective study and divided into two equal age groups (≤ 28 and ≥ 50 years old). Up to three measurements by a corneal deformation analyser based on the Scheimpflug principle were performed on the left eye of each subject. Raw Scheimpflug images were used to extract changes in anterior and posterior corneal profiles, which were further modelled by an orthogonal series of Chebyshev polynomial functions. Time series of the polynomial coefficients of even order exhibited a dynamic behavior in which three distinct stages were recognized. A bilinear function was used to model the first and the third stage of corneal dynamics. Slope parameters of the bilinear fit were then tested between the two age groups using Wilcoxon rank sum test and two-way non-parametric ANOVA (Friedman) test.ResultsStatistically significant changes (Wilcoxon test, P<0.05) between the age groups were observed in the phase of the second applanation dynamics for the posterior corneal profile. In a two-way comparison, in which the corneal profile was used as a dependent variable, statistically significant changes (ANOVA/Friedman test, P = 0.017) between the groups were also observed for that phase.ConclusionCorneal biomechanics depend on age. The changes in corneal deformation dynamics, which correspond to mostly free return of the cornea to its original shape after the air pulse, indicate that the age related differences in corneal biomechanics are subtle but observable with high speed imaging.

Project description:Healthy aging is associated with brain changes that reflect an alteration to a functional unit in response to the available resources and architecture. Even before the onset of noticeable cognitive decline, the neural scaffolds underlying cognitive function undergo considerable change. Prior studies have suggested a disruption of the connectivity pattern within the "default-mode" network (DMN), and more specifically a disruption of the anterio-posterior connectivity. In this study, we explored the effects of aging on within-network connectivity of three DMN subnetworks: a posterior DMN (pDMN), an anterior DMN (aDMN), and a ventral DMN (vDMN); as well as between-network connectivity during resting-state. Using groupICA on 43 young and 43 older healthy adults, we showed a reduction of network co-activation in two of the DMN subnetworks (pDMN and aDMN) and demonstrated a difference in between-component connectivity levels. The older group exhibited more numerous high-correlation pairs (Pearson's rho > 0.3, Number of comp-pairs = 46) in comparison to the young group (Number of comp-pairs = 34), suggesting a more connected/less segregated cortical system. Moreover, three component-pairs exhibited statistically significant differences between the two populations. Visual areas V2-V1 and V2-V4 were more correlated in the older adults, while aDMN-pDMN correlation decreased with aging. The increase in the number of high-correlation component-pairs and the elevated correlation in the visual areas are consistent with the prior hypothesis that aging is associated with a reduction of functional segregation. However, the aDMN-pDMN dis-connectivity may be occurring under a different mechanism, a mechanism more related to a breakdown of structural integrity along the anterio-posterior axis.

Project description:Self-regulation is a dynamic process wherein executive processes (EP) delay, minimize or desist prepotent responses (PR) that arise in situations that threaten well-being. It is generally assumed that, over the course of early childhood, children expand and more effectively deploy their repertoire of EP-related strategies to regulate PR. However, longitudinal tests of these assumptions are scarce in part because self-regulation has been mostly studied as a static construct. This study engages dynamic systems modeling to examine developmental changes in self-regulation between ages 2 and 5 years. Second-by-second time-series data derived from behavioral observations of 112 children (63 boys) faced with novel laboratory-based situations designed to elicit wariness, hesitation, and fear were modeled using differential equation models designed to capture age-related changes in the intrinsic dynamics and bidirectional coupling of PR (fear/wariness) and EP (strategy use). Results revealed that dynamic models allow for the conceptualization and measurement of fear regulation as intrinsic processes as well as direct and indirect coupling between PR and EP. Several patterns of age-related changes were in line with developmental theory suggesting that PR weakened and was regulated more quickly and efficiently by EP at age 5 than at age 2. However, most findings were in the intrinsic dynamics and moderating influences between PR and EP rather than direct influences. The findings illustrate the precision with which specific aspects of self-regulation can be articulated using dynamic systems models, and how such models can be used to describe the development of self-regulation in nuanced and theoretically meaningful ways.

Project description:Human brains experience whole-brain anatomic and functional changes throughout the lifespan. Age-related whole-brain network changes have been studied with functional magnetic resonance imaging (fMRI) to determine their low-frequency spatial and temporal characteristics. However, little is known about age-related changes in whole-brain fast dynamics at the scale of neuronal events. The present study investigated age-related whole-brain dynamics in resting-state electroencephalography (EEG) signals from 73 healthy participants from 6 to 65 years old via characterizing transient neuronal coactivations at a resolution of tens of milliseconds. These uncovered transient patterns suggest fluctuating brain states at different energy levels of global activations. Our results indicate that with increasing age, shorter lifetimes and more occurrences were observed in the brain states that show the global high activations and more consecutive visits to the global highest-activation brain state. There were also reduced transitional steps during consecutive visits to the global lowest-activation brain state. These age-related effects suggest reduced stability and increased fluctuations when visiting high-energy brain states and with a bias toward staying low-energy brain states. These age-related whole-brain dynamics changes are further supported by changes observed in classic alpha and beta power, suggesting its promising applications in examining the effect of normal healthy brain aging, brain development, and brain disease.

Project description:Individual differences in semantic categorization are commonplace. Individuals apply a word like SPORTS to different instances because they employ different conditions for category membership (vagueness in criteria) or because they differ regarding the extent to which they feel the term can be applied given fixed conditions (vagueness in degree). Three individuals may, for instance, disagree as to whether chess and hiking are SPORTS, because one believes SPORTS are competitive in nature, while the other two require SPORTS to be effortful (vagueness in criteria). On the basis of whether they consider hiking sufficiently effortful or not, the latter two individuals might still disagree as to whether to call it a SPORT (vagueness in degree). We investigated whether there are systematic age-related differences in semantic categorization by analyzing the categorization decisions of 1,868 adults for eight semantic categories with a formal model that allows the two sources of categorization differences to be disentangled. We found that young and older adults assess instances differently with respect to the categorization conditions and that older adults employ a lower threshold for category membership than young adults do. We recommend that these criteria and degree differences are taken into account in studies of age-related semantic processing.

Project description:Normal ageing is associated with gradual brain atrophy. Determining spatial and temporal patterns of change can help shed light on underlying mechanisms. Neuroimaging provides various measures of brain structure that can be used to assess such age-related change but studies to date have typically considered single imaging measures. Although there is consensus on the notion that brain structure deteriorates with age, evidence on the precise time course and spatial distribution of changes is mixed. We assessed grey matter (GM) and white matter (WM) structure in a group of 66 adults aged between 23 and 81. Multimodal imaging measures included voxel-based morphometry (VBM)-style analysis of GM and WM volume and diffusion tensor imaging (DTI) metrics of WM microstructure. We found widespread reductions in GM volume from middle age onwards but earlier reductions in GM were detected in frontal cortex. Widespread age-related deterioration in WM microstructure was detected from young adulthood onwards. WM decline was detected earlier and more sensitively using DTI-based measures of microstructure than using markers of WM volume derived from conventional T1-weighted imaging.

Project description:Push-off intensity is largely governed by the forces generated by the triceps surae (TS) muscles (gastrocnemius-GAS, soleus-SOL). During walking, the TS muscles undergo different fascicle kinematics and contribute differently to biomechanical subtasks. These differences may be facilitated by the Achilles tendon (AT), which is comprised of subtendons that originate from the TS muscles. We and others have revealed non-uniform displacement patterns within the AT-evidence for sliding between subtendons that may facilitate independent muscle actuation. However, in older adults, we have observed more uniform AT tissue displacements that correlate with reduced push-off intensity. Here, we employed dual-probe ultrasound imaging to investigate TS muscle length change heterogeneity (GAS-SOL) as a determinant of reduced push-off intensity in older adults. Compared to young, older adults walked with more uniform AT tissue displacements and reduced TS muscle length change heterogeneity. These muscle-level differences appeared to negatively impact push-off intensity-evidenced by between-group differences in the extent to which TS muscle length change heterogeneity correlates with mechanical output across walking tasks. Our findings suggest that the capacity for sliding between subtendons may facilitate independent TS muscle actuation in young adults but may restrict that actuation in older adults, likely contributing to reduced push-off intensity.

Project description:Behavioral and physiological evidence suggests that developmental changes lead to enhanced cortical differentiation and integration through maturation and learning, and that senescent changes during aging result in dedifferentiation and reduced cortical specialization of neural cell assemblies. We used electroencephalographic (EEG) recordings to evaluate network structure and network topology dynamics during rest with eyes closed and open, and during auditory oddball task across the lifespan. For this evaluation, we constructed a hyper-frequency network (HFN) based on within- and cross-frequency coupling (WFC and CFC, respectively) at 10 oscillation frequencies ranging between 2 and 20 Hz. We found that WFC increased monotonously across the lifespan, whereas CFC showed a U-shaped relationship. These changes in WFC and CFC strengths coevolve with changes in network structure and network topology dynamics, namely the magnitude of graph-theoretical topology measures increased linearly with age (except for characteristic path length, which is going shorter), while their standard deviation showed an inverse U-shaped relationship with a peak in young adults. Temporal as well as structural or nodal similarity of network topology (with some exceptions) seems to coincide with variability changes, i.e., stronger variability is related to higher similarity between consecutive time windows or nodes. Furthermore, network complexity measures showed different lifespan-related patterns, which depended on the balance of WFC and CFC strengths. Both variability and complexity of HFNs were strongly related to the perceptual speed scores. Finally, investigation of the modular organization of the networks revealed higher number of modules and stronger similarity of community structures across time in young adults as compared with children and older adults. We conclude that network variability and complexity measures reflect temporal and structural topology changes in the functional organization and reorganization of neuronal cell assemblies across the lifespan.
