Self-organized criticality in a mesoscopic model of excitatory-inhibitory neuronal populations by short-term and long-term synaptic plasticity.

ABSTRACT: Dynamics of an interconnected population of excitatory and inhibitory spiking neurons wandering around a Bogdanov-Takens (BT) bifurcation point can generate the observed scale-free avalanches at the population level and the highly variable spike patterns of individual neurons. These characteristics match experimental findings for spontaneous intrinsic activity in the brain. In this paper, we address the mechanisms causing the system to get and remain near this BT point. We propose an effective stochastic neural field model which captures the dynamics of the mean-field model. We show how the network tunes itself through local long-term synaptic plasticity by STDP and short-term synaptic depression to be close to this bifurcation point. The mesoscopic model that we derive matches the directed percolation model at the absorbing state phase transition.

Self-organized criticality in developing neuronal networks.

Project description:Recently evidence has accumulated that many neural networks exhibit self-organized criticality. In this state, activity is similar across temporal scales and this is beneficial with respect to information flow. If subcritical, activity can die out, if supercritical epileptiform patterns may occur. Little is known about how developing networks will reach and stabilize criticality. Here we monitor the development between 13 and 95 days in vitro (DIV) of cortical cell cultures (n = 20) and find four different phases, related to their morphological maturation: An initial low-activity state (?19 DIV) is followed by a supercritical (?20 DIV) and then a subcritical one (?36 DIV) until the network finally reaches stable criticality (?58 DIV). Using network modeling and mathematical analysis we describe the dynamics of the emergent connectivity in such developing systems. Based on physiological observations, the synaptic development in the model is determined by the drive of the neurons to adjust their connectivity for reaching on average firing rate homeostasis. We predict a specific time course for the maturation of inhibition, with strong onset and delayed pruning, and that total synaptic connectivity should be strongly linked to the relative levels of excitation and inhibition. These results demonstrate that the interplay between activity and connectivity guides developing networks into criticality suggesting that this may be a generic and stable state of many networks in vivo and in vitro.

Developing neuronal networks: self-organized criticality predicts the future.

Project description:Self-organized criticality emerged in neural activity is one of the key concepts to describe the formation and the function of developing neuronal networks. The relationship between critical dynamics and neural development is both theoretically and experimentally appealing. However, whereas it is well-known that cortical networks exhibit a rich repertoire of activity patterns at different stages during in vitro maturation, dynamical activity patterns through the entire neural development still remains unclear. Here we show that a series of metastable network states emerged in the developing and "aging" process of hippocampal networks cultured from dissociated rat neurons. The unidirectional sequence of state transitions could be only observed in networks showing power-law scaling of distributed neuronal avalanches. Our data suggest that self-organized criticality may guide spontaneous activity into a sequential succession of homeostatically-regulated transient patterns during development, which may help to predict the tendency of neural development at early ages in the future.

Involvement of Cdk5 activating subunit p35 in synaptic plasticity in excitatory and inhibitory neurons.

Project description:Cyclin-dependent kinase 5 (Cdk5) /p35 is involved in many developmental processes of the central nervous system. Cdk5/p35 is also implicated in synaptic plasticity, learning and memory. Several lines of conditional Cdk5 knockout mice (KO) have been generated and have shown different outcomes for learning and memory. Here, we present our analysis of p35 conditional KO mice (p35cKO) in hippocampal pyramidal neurons or forebrain GABAergic neurons using electrophysiological and behavioral methods. In the fear conditioning task, CamKII-p35cKO mice showed impaired memory retention. Furthermore, NMDAR-dependent long-term depression (LTD) induction by low-frequency stimuli in hippocampal slices from CamkII-p35cKO mice was impaired compared to that in control mice. In contrast, Dlx-p35cKO mice showed no abnormalities in behavioral tasks and electrophysiological analysis in their hippocampal slices. These results indicated that Cdk5/p35 in excitatory neurons is important for the hippocampal synaptic plasticity and associative memory retention.

Dopaminergic modulation of short-term synaptic plasticity at striatal inhibitory synapses.

Project description:Circuit properties, such as the selection of motor synergies, have been posited as relevant tasks for the recurrent inhibitory synapses between spiny projection neurons of the neostriatum, a nucleus of the basal ganglia participating in procedural learning and voluntary motor control. Here we show how the dopaminergic system regulates short-term plasticity (STP) in these synapses. STP is thought to endow neuronal circuits with computational powers such as gain control, filtering, and the emergence of transitory net states. But little is known about STP regulation. Employing unitary and population synaptic recordings, we observed that activation of dopamine receptors can modulate STP between spiny neurons. A D(1)-class agonist enhances, whereas a D(2)-class agonist decreases, short-term depression most probably by synaptic redistribution. Presynaptic receptors appear to be responsible for this modulation. In contrast, STP between fast-spiking interneurons and spiny projection neurons is largely unregulated despite expressing presynaptic receptors. Thus, the present experiments provide an explanation for dopamine actions at the circuit level: the control of STP between lateral connections of output neurons and the reorganization of the balance between different forms of inhibitory transmission. Theoretically, D(1) receptors would promote a sensitive, responsive state for temporal precision (dynamic component), whereas D(2) receptors would sense background activity (static component).

Self-organized criticality occurs in non-conservative neuronal networks during Up states.

Project description:During sleep, under anesthesia and in vitro, cortical neurons in sensory, motor, association and executive areas fluctuate between Up and Down states (UDS) characterized by distinct membrane potentials and spike rates [1, 2, 3, 4, 5]. Another phenomenon observed in preparations similar to those that exhibit UDS, such as anesthetized rats [6], brain slices and cultures devoid of sensory input [7], as well as awake monkey cortex [8] is self-organized criticality (SOC). This is characterized by activity "avalanches" whose size distributions obey a power law with critical exponent of about [Formula: see text] and branching parameter near unity. Recent work has demonstrated SOC in conservative neuronal network models [9, 10], however critical behavior breaks down when biologically realistic non-conservatism is introduced [9]. We here report robust SOC behavior in networks of non-conservative leaky integrate-and-fire neurons with short-term synaptic depression. We show analytically and numerically that these networks typically have 2 stable activity levels corresponding to Up and Down states, that the networks switch spontaneously between them, and that Up states are critical and Down states are subcritical.

Neddylation regulates excitatory synaptic transmission and plasticity.

Project description:Post-translational modifications, like phosphorylation, ubiquitylation, and sumoylation, have been shown to impact on synaptic neurotransmission by modifying pre- and postsynaptic proteins and therefore alter protein stability, localization, or protein-protein interactions. Previous studies showed that post-translational modifications are essential during the induction of synaptic plasticity, defined by a major reorganization of synaptic proteins. We demonstrated before that neddylation, a post-translational modification that covalently binds Nedd8 to lysine-residues, strongly affects neuronal maturation and spine stability. We now analysed the consequences of inhibiting neddylation on excitatory synaptic transmission and plasticity, which will help to narrow down possible targets, to make educated guesses, and test specific candidates. Here, we show that acute inhibition of neddylation impacts on synaptic neurotransmission before morphological changes occur. Our data indicate that pre- and postsynaptic proteins are neddylated since the inhibition of neddylation impacts on presynaptic release probability and postsynaptic receptor stabilization. In addition, blocking neddylation during the induction of long-term potentiation and long-term inhibition abolished both forms of synaptic plasticity. Therefore, this study shows the importance of identifying synaptic targets of the neddylation pathway to understand the regulation of synaptic transmission and plasticity.

microRNA-218-5p Coordinates Scaling of Excitatory and Inhibitory Synapses during Homeostatic Synaptic Plasticity

Project description:Homeostatic synaptic plasticity (HSP) is a fundamental neuronal mechanism that allows networks to compensate for prolonged changes in activity by adjusting synaptic strength. This process is crucial for maintaining stable brain function and has been implicated in memory consolidation during sleep. While scaling of both excitatory and inhibitory synapses plays an important role during homeostatic synaptic plasticity, molecules coordinating both of these processes are unknown. In this study, we investigate the role of miR-218-5p as a regulator of inhibitory and excitatory synapses in the context of picrotoxin (PTX)-induced homeostatic synaptic downscaling (HSD) in rat hippocampal neurons. Using enrichment analysis of miRNA-binding sites in differentially expressed genes changing upon PTX-induced HSD, we bioinformatically predicted and experimentally validated increased miR-218-5p activity upon PTX-treatment in the process compartment. By monitoring synapse structure in vitro with confocal microscopy, we demonstrate that miR-218-5p exerts a dual effect in HSD: it prevents the downscaling of excitatory synapses and dendritic spines, while at the same time blocking inhibitory synapse upscaling. Furthermore, we identify the Neuroligin2 interacting molecule Mdga1 as a crucial target of miR-218-5p in the context of homeostatic upscaling of inhibitory synapses. By performing long-term electroencephalographic (EEG) recordings, we further revealed that local inhibition of miR-218-5p in the somatosensory cortex reduced local slow-wave activity (SWA) during non-rapid-eye-movement (NREM) sleep. In summary, this study uncovers miR-218-5p as a key player in coordinating inhibitory and excitatory synapses during homeostatic plasticity and sleep. Our findings contribute to a deeper understanding of how neural circuits maintain stability in the face of activity-induced perturbations, with potential implications for both physiological and pathological conditions.

Firing clamp: a novel method for single-trial estimation of excitatory and inhibitory synaptic neuronal conductances.

Project description:Understanding non-stationary neuronal activity as seen in vivo requires estimation of both excitatory and inhibitory synaptic conductances from a single trial of recording. For this purpose, we propose a new intracellular recording method, called "firing clamp." Synaptic conductances are estimated from the characteristics of artificially evoked probe spikes, namely the spike amplitude and the mean subthreshold potential, which are sensitive to both excitatory and inhibitory synaptic input signals. The probe spikes, timed at a fixed rate, are evoked in the dynamic-clamp mode by injected meander-like current steps, with the step duration depending on neuronal membrane voltage. We test the method with perforated-patch recordings from isolated cells stimulated by external application or synaptic release of transmitter, and validate the method with simulations of a biophysically-detailed neuron model. The results are compared with the conductance estimates based on conventional current-clamp recordings.

LTS and FS inhibitory interneurons, short-term synaptic plasticity, and cortical circuit dynamics.

Project description:Somatostatin-expressing, low threshold-spiking (LTS) cells and fast-spiking (FS) cells are two common subtypes of inhibitory neocortical interneuron. Excitatory synapses from regular-spiking (RS) pyramidal neurons to LTS cells strongly facilitate when activated repetitively, whereas RS-to-FS synapses depress. This suggests that LTS neurons may be especially relevant at high rate regimes and protect cortical circuits against over-excitation and seizures. However, the inhibitory synapses from LTS cells usually depress, which may reduce their effectiveness at high rates. We ask: by which mechanisms and at what firing rates do LTS neurons control the activity of cortical circuits responding to thalamic input, and how is control by LTS neurons different from that of FS neurons? We study rate models of circuits that include RS cells and LTS and FS inhibitory cells with short-term synaptic plasticity. LTS neurons shift the RS firing-rate vs. current curve to the right at high rates and reduce its slope at low rates; the LTS effect is delayed and prolonged. FS neurons always shift the curve to the right and affect RS firing transiently. In an RS-LTS-FS network, FS neurons reach a quiescent state if they receive weak input, LTS neurons are quiescent if RS neurons receive weak input, and both FS and RS populations are active if they both receive large inputs. In general, FS neurons tend to follow the spiking of RS neurons much more closely than LTS neurons. A novel type of facilitation-induced slow oscillations is observed above the LTS firing threshold with a frequency determined by the time scale of recovery from facilitation. To conclude, contrary to earlier proposals, LTS neurons affect the transient and steady state responses of cortical circuits over a range of firing rates, not only during the high rate regime; LTS neurons protect against over-activation about as well as FS neurons.

Learning excitatory-inhibitory neuronal assemblies in recurrent networks.

Project description:Understanding the connectivity observed in the brain and how it emerges from local plasticity rules is a grand challenge in modern neuroscience. In the primary visual cortex (V1) of mice, synapses between excitatory pyramidal neurons and inhibitory parvalbumin-expressing (PV) interneurons tend to be stronger for neurons that respond to similar stimulus features, although these neurons are not topographically arranged according to their stimulus preference. The presence of such excitatory-inhibitory (E/I) neuronal assemblies indicates a stimulus-specific form of feedback inhibition. Here, we show that activity-dependent synaptic plasticity on input and output synapses of PV interneurons generates a circuit structure that is consistent with mouse V1. Computational modeling reveals that both forms of plasticity must act in synergy to form the observed E/I assemblies. Once established, these assemblies produce a stimulus-specific competition between pyramidal neurons. Our model suggests that activity-dependent plasticity can refine inhibitory circuits to actively shape cortical computations.
