Elephant facial motor control.

ABSTRACT: We studied facial motor control in elephants, animals with muscular dexterous trunks. Facial nucleus neurons (~54,000 in Asian elephants, ~63,000 in African elephants) outnumbered those of other land-living mammals. The large-eared African elephants had more medial facial subnucleus neurons than Asian elephants, reflecting a numerically more extensive ear-motor control. Elephant dorsal and lateral facial subnuclei were unusual in elongation, neuron numerosity, and a proximal-to-distal neuron size increase. We suggest that this subnucleus organization is related to trunk representation, with the huge distal neurons innervating the trunk tip with long axons. African elephants pinch objects with two trunk tip fingers, whereas Asian elephants grasp/wrap objects with larger parts of their trunk. Finger "motor foveae" and a positional bias of neurons toward the trunk tip representation in African elephant facial nuclei reflect their motor strategy. Thus, elephant brains reveal neural adaptations to facial morphology, body size, and dexterity.

Efficient recognition of facial expressions does not require motor simulation.

Project description:What mechanisms underlie facial expression recognition? A popular hypothesis holds that efficient facial expression recognition cannot be achieved by visual analysis alone but additionally requires a mechanism of motor simulation - an unconscious, covert imitation of the observed facial postures and movements. Here, we first discuss why this hypothesis does not necessarily follow from extant empirical evidence. Next, we report experimental evidence against the central premise of this view: we demonstrate that individuals can achieve normotypical efficient facial expression recognition despite a congenital absence of relevant facial motor representations and, therefore, unaided by motor simulation. This underscores the need to reconsider the role of motor simulation in facial expression recognition.

olig2-Expressing hindbrain cells are required for migrating facial motor neurons.

Project description:The complicated trajectory of facial motor neuron migration requires coordination of intrinsic signals and cues from the surrounding environment. Migration begins in rhombomere (r) 4 where facial motor neurons are born and proceeds in a caudal direction. Once facial motor neurons reach their target rhombomeres, they migrate laterally and radially from the ventral neural tube. In zebrafish, as facial motor neurons migrate through r5/r6, they pass near cells that express olig2, which encodes a bHLH transcription factor. In this study, we found that olig2 function is required for facial motor neurons to complete their caudal migration into r6 and r7 and form stereotypical clusters. Additionally, embryos that lack mafba function, in which facial motor neurons also fail to complete caudal migration, lack olig2 expression in r5 and r6. Our data raise the possibility that cells expressing olig2 are intermediate targets that help guide facial motor neuron migration.

Shox2 is required for the proper development of the facial motor nucleus and the establishment of the facial nerves.

Project description:BackgroundAxons from the visceral motor neurons (vMNs) project from nuclei in the hindbrain to innervate autonomic ganglia and branchial arch-derived muscles. Although much is known about the events that govern specification of somatic motor neurons, the genetic pathways responsible for the development of vMNs are less well characterized. We know that vMNs, like all motor neurons, depend on sonic hedgehog signaling for their generation. Similarly, the paired-like homeobox 2b (Phox2b) gene, which is expressed in both proliferating progenitors and post-mitotic motor neurons, is essential for the development of vMNs. Given that our previous study identified a novel role for the short stature homeobox 2 (Shox2) gene in the hindbrain, and since SHOX2 has been shown to regulate transcription of islet 1 (Isl1), an important regulator of vMN development, we sought to determine whether Shox2 is required for the proper development of the facial motor nucleus.ResultsUsing a Nestin-Cre driver, we show that elimination of Shox2 throughout the brain results in elevated cell death in the facial motor nucleus at embryonic day 12.5 (E12.5) and E14.5, which correlates with impaired axonal projection properties of vMNs. We also observed changes in the spatial expression of the vMN cell fate factors Isl1 and Phox2b, and concomitant defects in Shh and Ptch1 expression in Shox2 mutants. Furthermore, we demonstrate that elimination of Shox2 results in the loss of dorsomedial and ventromedial subnuclei by postnatal day 0 (P0), which may explain the changes in physical activity and impaired feeding/nursing behavior in Shox2 mutants.ConclusionsCombined, our data show that Shox2 is required for development of the facial motor nucleus and its associated facial (VII) nerves, and serves as a new molecular tool to probe the genetic programs of this understudied hindbrain region.

Inducing a concurrent motor load reduces categorization precision for facial expressions.

Project description:Motor theories of expression perception posit that observers simulate facial expressions within their own motor system, aiding perception and interpretation. Consistent with this view, reports have suggested that blocking facial mimicry induces expression labeling errors and alters patterns of ratings. Crucially, however, it is unclear whether changes in labeling and rating behavior reflect genuine perceptual phenomena (e.g., greater internal noise associated with expression perception or interpretation) or are products of response bias. In an effort to advance this literature, the present study introduces a new psychophysical paradigm for investigating motor contributions to expression perception that overcomes some of the limitations inherent in simple labeling and rating tasks. Observers were asked to judge whether smiles drawn from a morph continuum were sincere or insincere, in the presence or absence of a motor load induced by the concurrent production of vowel sounds. Having confirmed that smile sincerity judgments depend on cues from both eye and mouth regions (Experiment 1), we demonstrated that vowel production reduces the precision with which smiles are categorized (Experiment 2). In Experiment 3, we replicated this effect when observers were required to produce vowels, but not when they passively listened to the same vowel sounds. In Experiments 4 and 5, we found that gender categorizations, equated for difficulty, were unaffected by vowel production, irrespective of the presence of a smiling expression. These findings greatly advance our understanding of motor contributions to expression perception and represent a timely contribution in light of recent high-profile challenges to the existing evidence base.

Digital screener of socio-motor agency balancing motor autonomy and motor control.

Project description:Dyadic social interactions evoke complex dynamics between two agents that, while exchanging unequal levels of body autonomy and motor control, may find a fine balance to synergize, take turns, and gradually build social rapport. To study the evolution of such complex interactions, we currently rely exclusively on subjective pencil and paper means. Here, we complement this approach with objective biometrics of socio-motor behaviors conducive to socio-motor agency. Using a common clinical test as the backdrop of our study to probe social interactions between a child and a clinician, we demonstrate new ways to streamline the detection of social readiness potential in both typically developing and autistic children by uncovering a handful of tasks that enable quantification of levels of motor autonomy and levels of motor control. Using these biometrics of autonomy and control, we further highlight differences between males and females and uncover a new data type amenable to generalizing our results to any social setting. The new methods convert continuous dyadic bodily biorhythmic activity into spike trains and demonstrate that in the context of dyadic behavioral analyses, they are well characterized by a continuous Gamma process that can classify individual levels of our thus defined socio-motor agency during a dyadic exchange. Finally, we apply signal detection processing tools in a machine learning approach to show the validity of the streamlined version of the digitized ADOS test. We offer a new framework that combines stochastic analyses, non-linear dynamics, and information theory to streamline and facilitate scaling the screening and tracking of social interactions with applications to autism.

Enhanced emotional and motor responses to live versus videotaped dynamic facial expressions.

Project description:Facial expression is an integral aspect of non-verbal communication of affective information. Earlier psychological studies have reported that the presentation of prerecorded photographs or videos of emotional facial expressions automatically elicits divergent responses, such as emotions and facial mimicry. However, such highly controlled experimental procedures may lack the vividness of real-life social interactions. This study incorporated a live image relay system that delivered models' real-time performance of positive (smiling) and negative (frowning) dynamic facial expressions or their prerecorded videos to participants. We measured subjective ratings of valence and arousal and facial electromyography (EMG) activity in the zygomaticus major and corrugator supercilii muscles. Subjective ratings showed that the live facial expressions were rated to elicit higher valence and more arousing than the corresponding videos for positive emotion conditions. Facial EMG data showed that compared with the video, live facial expressions more effectively elicited facial muscular activity congruent with the models' positive facial expressions. The findings indicate that emotional facial expressions in live social interactions are more evocative of emotional reactions and facial mimicry than earlier experimental data have suggested.

Early maternal mirroring predicts infant motor system activation during facial expression observation.

Project description:Processing facial expressions is an essential component of social interaction, especially for preverbal infants. In human adults and monkeys, this process involves the motor system, with a neural matching mechanism believed to couple self- and other-generated facial gestures. Here, we used electroencephalography to demonstrate recruitment of the human motor system during observation and execution of facial expressions in nine-month-old infants, implicating this system in facial expression processing from a very young age. Notably, examination of early video-recorded mother-infant interactions supported the common, but as yet untested, hypothesis that maternal mirroring of infant facial gestures is central to the development of a neural matching mechanism for these gestures. Specifically, the extent to which mothers mirrored infant facial expressions at two months postpartum predicted infant motor system activity during observation of the same expressions at nine months. This suggests that maternal mirroring strengthens mappings between visual and motor representations of facial gestures, which increases infant neural sensitivity to particularly relevant cues in the early social environment.
