ABSTRACT: Maize stalk rot caused by Fusarium graminearum can reduce the yield of maize and efficiency of mechanized harvesting. Besides, deoxynivalenol and zearalenone toxins produced by F. graminearum can also affect domestic animals and human health. As chemical fungicides are expensive and exert negative effects on the environment, the use of biological control agents has become attractive in recent years. In the present study, we collected rhizosphere soil with severe stalk rot disease (ZDD), the rhizosphere soil with disease-free near by the ZDD (ZDH), and measured rhizosphere microbial diversity and microbial taxonomic composition by amplicon sequencing targeting either bacteria or fungi. The results showed that Fusarium stalk rot caused by the Fusarium species among which F. graminearum is frequent and can reduce the abundance and alpha diversity of rhizosphere microbial community, and shift the beta diversity of microorganisms. Furthermore, a bacterial strain, Bacillus siamensis GL-02, isolated from ZDD, was found to significantly affect growth of F. graminearum. In vitro and in vivo assays demonstrated that B. siamensis GL-02 had good capability to inhibit F. graminearum. These results revealed that B. siamensis GL-02 could be a potential biocontrol agent for the control of maize stalk rot.

Project description:BackgroundGibberella stalk rot caused by Fusarium graminearum is one of the most destructive soil-borne diseases of maize (Zea mays L.). Chemical means of controlling Gibberella stalk rot are not very effective; development of highly resistant hybrids is the best choice for disease control. Hence, understanding of the molecular basis underlying maize resistance against Gibberella stalk rot would undoubtedly facilitate the resistance breeding for stalk rot.ResultsTwo quantitative trait loci (QTL), qRfg1 and qRfg2, conferring resistance to Gibberella stalk rot were detected in our previous study. Three near-isogenic lines (NILs) of maize with either qRfg1 (NIL1) or qRfg2 (NIL2), or neither (NIL3) were generated and subjected to RNA sequencing to study the transcriptional changes after F. graminearum inoculation at 0 (control), 6, and 18 h post-inoculation (hpi). In total, 536,184,652 clean reads were generated, and gene expression levels were calculated using FPKM (fragments per kilobase of exon model per million mapped reads). A total of 7252 differentially expressed genes (DEGs) were found in the three NILs after F. graminearum inoculation. As many as 2499 DEGs were detected between NIL1 and NIL3 at 0 hpi, of which 884 DEGs were more abundant in NIL1 and enriched in defense responses. After F. graminearum inoculation, 1070 and 751 genes were exclusively up- and downregulated, respectively, in NIL1 as compared to NIL3. The 1070 upregulated DEGs were enriched in growth/development, photosynthesis/biogenesis, and defense-related responses. Genes encoding putative auxin-induced proteins and GH3 family proteins in auxin signaling pathway were highly induced and lasted longer in NIL3. Genes involved in polar auxin transport (PAT) were more abundant in NIL3 as compared with NIL2.ConclusionsThe qRfg1 confers its resistance to Gibberella stalk rot through both constitutive and induced high expression of defense-related genes; while qRfg2 enhances maize resistance to the disease via relatively lower induction of auxin signaling and repression of PAT. The defense-related transcriptional changes underlying each QTL will undoubtedly facilitate our understanding of the resistance mechanism and resistance breeding for maize stalk rot.

Project description:IntroductionMaize stalk rot (MSR), caused by Fusarium graminearum, is the most serious soil borne disease in maize production, seriously affecting maize yield and quality worldwide. Microbial biocontrol agents are the best means of controlling MSR and reducing the use of chemical fungicides, such as Bacillus spp.Methods and resultsIn the study, a soil-isolated strain B105-8 was identified as B. velezensis (accession No. PP325775.1 and No. PP869695.1), demonstrated a broad spectrum against various pathogens causing maize diseases, which effectively controlled MSR, exhibited a high control efficacy of more than 60% and growth-promoting effect in the pot plant. B105-8 could effectively improve soil urease (S-UE), invertase (S-SC), and catalase (S-CAT) activities. S-NP activity showed an initial increase with a peak of 20,337 nmol/h/g, followed by a decrease, but activity remained significantly better than control treatment with chemical fungicides. The application of B105-8 repaired the damage caused by F. graminearum on soil activity. The antifungal compound B-1, extracted from B105-8, was purified using a protein purifier, revealing inhibitory effects against F. graminearum. Mass spectrometry analysis indicated the potential presence of C14 Bacillomycin, C15 Iturin, C15 Mycosubtilin, C17, and C15 fengycin in B-1. In pot experiments, a 5 μL/mL concentration of B-1 exhibited 69% control on MSR, enhancing maize root elongation, elevation, and fresh weight. At 10 μL/mL, B-1 showed 89.0 and 82.1% inhibition on spore production and mycelial growth, causing hyphal deformities.DiscussionThis study presents the innovative use of B. velezensis, isolated from maize rhizosphere in cold conditions to effectively control MSR caused by F. graminearum. The findings highlight the remarkable regional and adaptive characteristics of this strain, making it an excellent candidate to fight MSR in diverse environments. In conclusion, B. velezensis B105-8 demonstrated potential as a biocontrol agent for MSR.
