Tube length of chironomid larvae as an indicator for dissolved oxygen in water bodies.

ABSTRACT: Tube-building larvae of non-biting midges, or chironomids, are considered bioindicators of water pollution. The larvae use benthic particles to make their tubes and create a respiratory current with the movement of their bodies inside the tubes. The tube length of the chironomid larvae varies depending on several physicochemical properties of the aquatic medium. Here we study the role of physicochemical parameters on the tube length from different field sites and in the laboratory. It appears that among different physicochemical factors, dissolved oxygen (DO) has a major role in determining the tube length of the larvae. A quantitative relationship between oxygen concentration and the tube length of larvae is presented here. Our study reveals a longer tube in aquatic media with oxygen deficiency and a shorter tube in those with higher oxygen. This result may help to assess the quality of water bodies and, in particular the status of DO.

Unforeseen decreases in dissolved oxygen levels affect tube formation kinetics in collagen gels.

Project description:The availability of oxygen (O(2)) is a critical parameter affecting vascular tube formation. In this study, we hypothesize that dissolved oxygen (DO) levels in collagen gels change during the three-dimensional (3D) culture of human umbilical vein endothelial cells (HUVECs) in atmospheric conditions and that such changes affect the kinetics of tube formation through the production of reactive oxygen species (ROS). We demonstrate a decrease in O(2) tension during 3D cultures of HUVECs. Noninvasive measurements of DO levels during culture under atmospheric conditions revealed a profound decrease that reached as low as 2% O(2) at the end of 24 h. After media replacement, DO levels rose rapidly and equilibrated at ∼15% O(2), creating a reoxygenated environment. To accurately estimate DO gradients in 3D collagen gels, we developed a 3D mathematical model and determined the Michaelis-Menten parameters, V(max) and K(m), of HUVECs in collagen gels. We detected an increase in ROS levels throughout the culture period. Using diphenyliodonium to inhibit ROS production resulted in the complete inhibition of tube formation. Interference RNA studies further showed that hypoxia-inducible factors (HIFs)-1α and -2α are not involved in the formation of 3D tubes in collagen gels. We conclude that ROS affect the tubulogenesis process through HIFα-independent pathways, where the levels of ROS are influenced by the uncontrolled variations in DO levels. This study is the first demonstration of the critical and unexpected role of O(2) during 3D in vitro culture models of tubulogenesis in atmospheric conditions.

Consequences of physical disturbance by tadpoles and snails on chironomid larvae.

Project description:Indirect interactions among community members impact on organisms. The effects of two snails, banded pond snail, Bellamya bengalensis (Lamarck), and Red-rimmed melania, Melanoides tuberculata (Müller), and tadpoles of Asian common toad, Duttaphrynus melanostictus (Schneider), on nonbiting midge larvae, Chironomus striatipennis Kieffer, were observed in experimental microcosm. Decrease in tube number and tube length of midge larvae was observed compared to control condition due to introduction of selected above mentioned organisms. The direct effects of non-predator organisms on the midge larvae are due to physical disturbance that destroys their tubes. This may result in vulnerability of midge larvae to predators in the wild. So the community structure may be altered by indirect effects, where one or more species, through their direct disturbance, indirectly change the abundance of other species.

A novel leaf-rolling chironomid, <i>Eukiefferiella endobryonia</i> sp. nov. (Diptera, Chironomidae, Orthocladiinae), highlights the diversity of underwater chironomid tube structures.

Project description:The non-biting midges, Chironomidae (Diptera), are dominant components of most freshwater ecosystems. Many chironomids construct tubes or cases as larvae out of various materials bound together with silk. The structures of tubes show a wide range of variation, and some are morphologically comparable to those of caddisflies. Herein a new species is described, Eukiefferiella endobryonia sp. nov., which exhibits a very unusual behavior in which it constructs tubes from aquatic mosses. This species' fourth-instar larvae construct their cases exclusively from the leaves of Fontinalis mosses (Hypnales: Fontinalaceae) and exhibit a stereotyped behavior in which they remain attached to the apical shoot of the moss stem. The larvae then pupate within the case. The case of E. endobryonia sp. nov. represents one of only a few examples of chironomid tubes made exclusively out of plants. Based on the species delimitation analyses using the partial COI sequences, together with some morphological and behavioral characteristics, this species is hypothesized to be a member of devonica group, and especially may have a close affinity to E. dittmari (Lehman). A provisional typology for the diversity of chironomid tube structures is provided, with a summary of different tube structures, which can be used for future research.

The protective role of endogenous bacterial communities in chironomid egg masses and larvae.

Project description:Insects of the family Chironomidae, also known as chironomids, are distributed worldwide in a variety of water habitats. These insects display a wide range of tolerance toward metals and organic pollutions. Bacterial species known for their ability to degrade toxicants were identified from chironomid egg masses, leading to the hypothesis that bacteria may contribute to the survival of chironomids in polluted environments. To gain a better understanding of the bacterial communities that inhabit chironomids, the endogenous bacteria of egg masses and larvae were studied by 454-pyrosequencing. The microbial community of the egg masses was distinct from that of the larval stage, most likely due to the presence of one dominant bacterial Firmicutes taxon, which consisted of 28% of the total sequence reads from the larvae. This taxon may be an insect symbiont. The bacterial communities of both the egg masses and the larvae were found to include operational taxonomic units, which were closely related to species known as toxicant degraders. Furthermore, various bacterial species with the ability to detoxify metals were isolated from egg masses and larvae. Koch-like postulates were applied to demonstrate that chironomid endogenous bacterial species protect the insect from toxic heavy metals. We conclude that chironomids, which are considered pollution tolerant, are inhabited by stable endogenous bacterial communities that have a role in protecting their hosts from toxicants. This phenomenon, in which bacteria enable the continued existence of their host in hostile environments, may not be restricted only to chironomids.

Artificially decreased dissolved oxygen increases the persistence of <i>Trichomonas gallinae</i> in water.

Project description:Water containing organic material has been shown to increase the persistence of the avian pathogenic protozoa, Trichomonas gallinae. We hypothesized that the decrease in dissolved oxygen due to microbes in the organic material could increase persistence of the microaerophilic trichomonads. Using simulated birdbaths, we determined 1) the levels of dissolved oxygen in distilled water with various amounts of organic material, 2) the concentration of the oxygen-scavenging enzyme Oxyrase® needed to achieve the dissolved oxygen levels obtained in organic material contaminated water, and finally, 3) the persistence of two T. gallinae isolates in Oxyrase®-supplemented water. An average of 9.6% dissolved oxygen was obtained with the addition of 15 g organic material to 500 ml of distilled water, whereas organic material-free water had 86.2% dissolved oxygen. The addition of 0.5% and 1.0% (vol/vol) Oxyrase® to organic material-free water yielded dissolved oxygen of 18.6% and 6.9%, respectively. Using 0.5% and 1.0% concentrations of Oxyrase®, we evaluated the persistence of two trichomonad isolates by inoculating ∼1 million trichomonads into 500 ml distilled water in triplicate. At various time-points, 0.5 ml aliquots of trichomonad-inoculated water were obtained and placed into Hollander Fluid media, incubated at 37 °C, and read by light microscopy every other day for 5 days. In our 1% Oxyrase® treatments, the longest recorded persistence of broad-winged hawk 1 increased from the previously reported 4hrs to 30hrs and Cooper's hawk 4 from 16hrs to 30hrs. These results indicate that the mechanism for organic material-mediated trichomonad persistence is associated with decreased dissolved oxygen, further demonstrating the importance of keeping birdbaths free of organic debris to discourage trichomonad persistence.

Warming Decreases Bioconversion of Polyunsaturated Fatty Acids in Chironomid Larvae Maintained on Cyanobacterium Microcystis.

Project description:Cyanobacteria dominance and warming have been suggested to decrease the production of polyunsaturated fatty acids (PUFA) in freshwater ecosystems. Physiological adaptations of poikilothermic animals to higher temperatures may further decrease PUFA levels in aquatic food webs. We conducted diet manipulation experiments to investigate the combined effects of dietary PUFA and warming on the proportions of eicosapentaenoic acid (EPA) and arachidonic acid (ARA) in Chironomus riparius. The experimental diet consisted of a nontoxic cyanobacterium Microcystis, which contained C20 PUFA: 20:3n-3, 20:4n-3, and 20:3n-6, but no EPA or ARA. Additionally, we used TetraMin® fish flakes as a control treatment. A temperature increase from 20 °C to 25 °C decreased the proportion of n-3 C20 PUFA and the n-3/n-6 ratio in Microcystis. Diet manipulation experiments indicated that Chironomus desaturated dietary C20 precursors to EPA and ARA, but warming decreased this bioconversion and resulted in lower levels of EPA and ARA in Chironomus. Warming did not alter the proportions of EPA and ARA in Chironomus larvae if these PUFA were readily available in the diet (TetraMin® control treatment). In conclusion, warming and cyanobacteria dominance may decrease the production and trophic transfer of physiologically important PUFA in freshwaters by (1) decreasing the n-3/n-6 ratio and the abundance of n-3 C20 precursors in Microcystis, and (2) decreasing the bioconversion of n-3 and n-6 C20 precursors to EPA and ARA in chironomids. These changes may have cascading effects throughout the food web and decrease the content of EPA in fish, potentially affecting its availability to humans.

Electron generation in water induced by magnetic effect and its impact on dissolved oxygen concentration

Project description:pH, oxidation-reduction potential (ORP) and dissolved oxygen (DO) concentration are important parameters in water quality surveillance and treatment. The changes of these parameters are associated with electron density in water. Several techniques including electrolysis and catalysis which require redox reactions and electron exchange are employed to improve these parameters. In recent years, studies reported that magnetic effects can impart considerable changes on the pH, ORP and DO concentration of water. However, the correlation between electron density and magnetic effects on these parameters has yet to be disclosed despite the fact that increased electron density in water could improve water’s reductive properties, heat capacity and hydrogen bonding characteristics. In this study, the magnetic effects on pH, ORP and DO concentration were investigated using different magnets arrangements and water flow rates based on reversed electric motor principle. Results showed that the improvement of pH, ORP and DO concentration from 5.40–5.42 to 5.58–5.62 (+ 3.5%), 392 to 365 mV (− 6.9%), and 7.30 to 7.71 mg L− 1 (+ 5.6%), respectively were achieved using combined variables of non-reversed polarity magnet arrangement (1000–1500 G magnetic strength) and water flow rate of 0.1–0.5 mL s− 1. Such decrement in ORP value also corresponded to 8.0 × 1013 number of electron generation in water. Furthermore, Raman analysis revealed that magnetic effect could strengthen the intermolecular hydrogen bonding of water molecules and favor formation of smaller water clusters. The findings of this study could contribute to potential applications in aquaculture, water quality control and treatment of cancer attributed to free radical induced-oxidative stress. Dissolved Oxygen and Water Temperature Drive Vertical Spatiotemporal Variation of Phytoplankton Community: Evidence from the Largest Diversion Water Source Area.

Project description:In order to study the vertical distribution characteristics of phytoplankton in the Danjiangkou Reservoir, the water source of the Middle Route Project of the South-North Water Diversion, seven sampling sites were set up in the Reservoir for quarterly sampling from 2017 to 2019, and water environment surveys were conducted simultaneously. The results showed that 157 species (including varieties) were identified, belonging to 9 phyla and 88 genera. In terms of species richness, Chlorophyta had the largest number of species, accounting for 39.49% of the total species. The Bacillariophyta and Cyanobacteria accounted for 28.03% and 13.38% of the total species, respectively. From the whole Danjiangkou Reservoir, the total phytoplankton abundance varied from 0.09 × 102 to 20.01 × 106 cells/L. In the vertical distribution, phytoplankton were mainly observed in the surface-thermospheric layer (I-II layer) and the bottom layer, while the Shannon-Wiener index showed a trend of gradually decreasing from the I-V layer. The Surfer model analysis showed that there was no significant stratification in the Q site's water temperature (WT) and dissolved oxygen (DO) levels in the water diversion area during the dynamic water diversion process. A canonical correspondence analysis (CCA) showed that DO, WT, pH, electrical conductivity (Cond), chemical oxygen demand (CODMn), total phosphorus (TP), ammonia nitrogen (NH4+-N), and total nitrogen (TN) had significant effects on the vertical distribution of phytoplankton (p < 0.05). A partial Mantel analysis showed that the vertical distribution of the phytoplankton community was related to WT, and the phytoplankton community structure at the other sites, except for Heijizui (H) and Langhekou (L), was affected by DO. This study has positive significance for exploring the vertical distribution characteristics of a phytoplankton community in a deep-water dynamic water diversion reservoir.

S-type Dissolved Oxygen Distribution along Water Depth in a Canyon-shaped and Algae Blooming Water Source Reservoir: Reasons and Control.

Project description:Dissolved oxygen (DO) is a crucial indicator of water quality. DO usually shows a monotonic decrease along water depth during thermal stratification in reservoir, whereas metalimnetic oxygen minimum (MOM) is observed in some cases. Although MOM phenomena have been reported in different areas, the characteristics of different reservoirs are greatly different, and few comprehensive studies have been published regarding MOM in Chinese drinking water source reservoirs. The DO distribution along water depth was determined and the detailed reasons were clarified by two-years of field monitoring. In addition the effect of water lifting aerators (WLAs) on DO improvement was investigated in the Lijiahe Reservoir in Northwest China. A typical S-type DO distribution with two anaerobic water layers, below the epilimnion (10⁻25 m water depth) and above the sediment (bottom water), was observed derived from the decomposition of dead algae or organic matter and the restriction of DO vertical exchange. Moreover, after WLAs' operation since 10 June 2018, the water body was completely mixed and DO was rich and uniform along water depth by eliminating the water stratification and inhibiting algae growth. The deep understanding of the DO distribution in a deep canyon-shaped reservoir and the technical support for reservoir restoration are meaningful for optimizing reservoir management.
