Project description:Cobalt (CoII ) ions have been an attractive candidate for the biomedical modification of orthopedic implants for decades. However, limited research has been performed into how immobilized CoII ions affect the physical properties of implant devices and how these changes regulate cellular behavior. In this study we modified biocompatible poly(vinyl alcohol) with terpyridine and catechol groups (PVA-TP-CA) to create a stable surface coating in which bioactive metal ions could be anchored, endowing the coating with improved broad-spectrum antibacterial activity against Escherichia coli and Staphylococcus aureus, as well as enhanced surface stiffness and cellular mechanoresponse manipulation. Strengthened by the addition of these metal ions, the coating elicited enhanced mechanosensing from adjacent cells, facilitating cell adhesion, spreading, proliferation, and osteogenic differentiation on the surface coating. This dual-functional PVA-TP-CA/Co surface coating offers a promising approach for improving clinical implantation outcomes.

Project description:Poly (vinyl butyral-co-vinyl alcohol-co-vinyl acetate) named further PVBA was investigated as a protective coating for copper corrosion in 0.9% NaCl solution using electrochemical measurements such as, electrochemical impedance spectroscopy (EIS) and potentiodynamic polarization associated with atomic force microscopy (AFM). The PVBA coating on the copper surface (Cu-PVBA) was modeled in methanol containing PVBA. Its inhibitory properties against corrosion was comparatively discussed with those of the copper sample treated in methanol without polymer (Cu-Me) and of untreated sample (standard copper). A protective performance of PVBA coating of 80% was computed from electrochemical measurements, for copper corrosion in NaCl solution. Moreover, AFM images designed a specific surface morphology of coated surface with PVBA, clearly highlighting a polymer film adsorbed on the copper surface, which presents certain deterioration after corrosion, but the metal surface was not significantly affected compared to those of untreated samples or treated in methanol, in the absence of PVBA.

Project description:Recently, tissue engineering became a very important medical alternative in patients who need to regenerate damaged or lost tissues through the use of scaffolds that support cell adhesion and proliferation. Carbon nanomaterials (carbon nanotubes, fullerenes, multi-wall fullerenes, and graphene) became a very important alternative to reinforce the mechanical, thermal, and antimicrobial properties of several biopolymers. In this work, five different formulations of chitosan/poly(vinyl alcohol)/oxidized carbon nano-onions (CS/PVA/ox-CNO) were used to prepare biodegradable scaffolds with potential biomedical applications. Film characterization consisted of Fourier-transform infrared (FTIR) spectroscopy, thermogravimetric analysis (TGA), differential scanning calorimetry (DSC), tension strength, Young's modulus, X-ray diffraction spectroscopy (XRD), scanning electron microscopy (SEM), and energy-dispersive spectroscopy (EDS). The degradation in a simulated body fluid (FBS) demonstrated that all the formulations lost between 75% and 80% of their weight after 15 days of treatment, but the degradation decreased with the ox-CNO content. In vivo tests after 90 days of subdermal implantation of the nanocomposite films in Wistar rats' tissue demonstrated good biocompatibility without allergenic reactions or pus formation. There was a good correlation between FBS hydrolytic degradation and degradation in vivo for all the samples, since the ox-CNO content increased the stability of the material. All these results indicate the potential of the CS/PVA/ox-CNO nanocomposite films in tissue engineering, especially for long-term applications.

Project description:Spectroscopic measurements in the infrared range combined with electrochemistry are a powerful technique for investigation of organic semiconductors to track changes during oxidation and reduction (p- and n-doping) processes. For these measurements it is important that the studied material, mostly deposited as a thin film on an internal reflection element, does not dissolve during this characterization. In this study we introduce a technique that allows infrared spectroelectrochemical characterization of films of these materials for the first time. In many cases so far this has been impossible, due to solubility in the oxidized and/or reduced form. This novel technique is shown on thin films of quinacridone by adding a protection layer of poly(vinyl alcohol) (PVA).

Project description:This study reports the potential of TEMPO-oxidized cellulose nanofibrils (T-CNF)/poly(vinyl alcohol) (PVA) coatings to develop functionalized membranes in the ultrafiltration regime with outstanding antifouling performance and dimensional/pH stability. PVA acts as an anchoring phase interacting with the polyethersulfone (PES) substrate and stabilizing for the hygroscopic T-CNF via crosslinking. The T-CNF/PVA coated PES membranes showed a nano-textured surface, a change in the surface charge, and improved mechanical properties compared to the original PES substrate. A low reduction (4%) in permeance was observed for the coated membranes, attributable to the nanometric coating thickness, surface charge, and hydrophilic nature of the coated layer. The coated membranes exhibited charge specific adsorption driven by electrostatic interaction combined with rejection due to size exclusion (MWCO 530 kDa that correspond to a size of ∼35-40 nm). Furthermore, a significant reduction in organic fouling and biofouling was found for T-CNF/PVA coated membranes when exposed to BSA and E. coli. The results demonstrate the potential of simple modifications using nanocellulose to manipulate the pore structure and surface chemistry of commercially available membranes without compromising on permeability and mechanical stability.

Project description:The biocompatible, viscoelastic properties of poly(vinyl alcohol) (PVA) in combination with the antimicrobial and antioxidant natural polyphenolic, tannic acid (TA), and the natural flavonoid and antioxidant curcumin (Cur), were used in the preparation of PVA:TA and PVA:TA:Cur cryogel composites using cryotropic gelation to combine the individually beneficial properties. The effect of TA content on the antioxidant and antimicrobial activities of PVA:TA cryogel composites and the antioxidant activities of PVA:TA:Cur cryogel composites was determined using Trolox equivalent antioxidant capacity (TEAC) and total phenol content (TPC) assays, and were compared. The PVA:TA:Cur cryogel composite showed the highest antioxidant activity, with a TEAC value of 2.10 ± 0.24 and a TPC value of 293 ± 12.00. The antibacterial capacity of the PVA:TA and PVA:TA:Cur 1:1:0.1 cryogel composites was examined against two different species of bacteria, E. coli and S. aureus. It was found that the minimum inhibition concentration (MIC) value of the PVA:TA:Cur 1:1:0.1 cryogel composites varied between 5 and 10 mg/mL based on the type of microorganism, and the minimum bactericidal concentration (MBC) value was 20 mg/mL irrespective of the type of microorganism. Furthermore, the hemocompatibility of the PVA:TA cryogel composites was evaluated by examining their hemolytic and coagulation behaviors. PVA:TA 1:1 cryogels with a value of 95.7% revealed the highest blood clotting index value amongst all of the synthesized cryogels, signifying the potential for blood contacting applications. The release of TA and Cur from the cryogel composites was quantified at different pH conditions, i.e., 1.0, 7.4, and 9.0, and additionally in ethanol (EtOH) and an ethanol-water (EtOH:Wat) mixture. The solution released from the PVA:TA cryogels in PBS was tested for inhibition capability against α-glucosidase (E.C. 3.2.1.20). Concentration-dependent enzyme inhibition was observed, and 70 µL of 83 µg/mL PVA:TA (1:1) cryogel in PBS inhibited α-glucosidase enzyme solution of 0.03 unit/mL in 70 µL by 81.75 ± 0.96%.

Project description:Poly(vinyl alcohol) (PVA) displays ice recrystallization inhibition (IRI) properties as many antifreeze proteins found in cold tolerant organisms. The molecular architecture and composition (molecular weight and distribution of pendant OH and acetate groups) have been studied to improve the antifreezing properties of PVA, suggesting that the molecular architecture of PVA plays an important role in IRI activity. The present work deals with the preparation of PVA microparticles using an alkaline treatment. The effect of PVA molecular weight on the morphology and antifreezeing properties of PVA microparticles was investigated. The antifreezeing property of PVA microparticles on the susceptibility of flower bud tissues to freeze damage was also evaluated. The alkaline treatment of an aqueous PVA solution produced stable polymer chain aggregates with spherical shapes. The average size of the PVA microparticles increased significantly with the increasing molecular weight of the PVA macromolecule precursor. The PVA microparticles inhibited the growth of ice crystals and blocked ice growth at concentrations as low as 0.01 % w/v. The effect of impeding ice crystal growth by preventing the joining of adjacent ice crystals is attributed to the larger size of the PVA particles adsorbed on the ice surface compared to the aggregated PVA macromolecules in saline solution. The thermal hysteresis activity of PVA macromolecules and microparticles was not detected by differential scanning calorimetry analysis. The PVA microparticles reduced the incidence of freeze injuries in flower bud tissues by 55% and their application, considering the low toxicity of PVA, has a high potential for freeze protection in fruit crops.

Project description:The thermal conductivity enhancement of neat poly(vinyl alcohol) and poly(vinyl alcohol) (PVA)/cellulose nanocrystal (CNC) composite was attempted via electrospinning. The suspended microdevice technique was applied to measure the thermal conductivity of electrospun nanofibers (NFs). Neat PVA NFs and PVA/CNC NFs with a diameter of approximately 200 nm showed thermal conductivities of 1.23 and 0.74 W/m-K, respectively, at room temperature, which are higher than that of bulk PVA by factors of 6 and 3.5, respectively. Material characterization by Fourier transform infrared spectroscopy, differential scanning calorimetry, and thermogravimetric analysis confirmed that the thermal conductivity of the PVA/CNC NFs was enhanced by the reinforcement of their backbone rigidity, while that of the neat PVA NFs was attributed to the increase in their crystallinity that occurred during the electrospinning.

Project description:Hydrogels have broad application prospects in bone repair. Pure poly(vinyl alcohol) (PVA) hydrogels have limited applications because of their low hardness and poor mechanical properties. This study found that resveratrol (Res) and PVA self-assembled and cross-linked through the formation of strong hydrogen bonds after freeze-thawing, forming an easily available PVA-Res supramolecular hydrogel through a green process. PVA-Res hydrogels with different Res wt %:10 wt % PVA ratios were prepared through freeze-thawing and designated as 0.4, 1.2, and 2.0 wt % PVA-Res hydrogels. Rheological studies demonstrated that the viscoelastic modulus of the PVA-Res hydrogels was significantly improved compared to pure PVA hydrogels. The viscoelastic modulus G' of 1.2% PVA-Res hydrogel was 2299 Pa, which was 8.5-fold that of the pure PVA hydrogel. We conducted a study on cell proliferation and osteogenic differentiation using MC3T3-E1 (preosteoblasts from newborn mouse calvaria). The results showed that the 0.4% PVA-Res hydrogel promotes alkaline phosphatase activity and mineral deposition. Real-time quantitative PCR (RT-qPCR) analysis demonstrated that the 0.4% PVA-Res hydrogel upregulated the expression of osteogenic differentiation-related genes (BMP-9, OCN, and ALP). Furthermore, RT-qPCR and flow cytometry demonstrated that the 0.4% PVA-Res hydrogel could effectively promote the M2 transformation and polarization of mouse mononuclear macrophage leukemia cells (Raw 264.7). The expression of related genes, such as Arg-1 and CD206, significantly increased, whereas that of M1 polarization-related genes, such as iNOS and TNF-α, significantly decreased. In summary, PVA-Res supramolecular hydrogels are potential materials for use in bone repair.

Project description:Poly(vinyl alcohol) (PVA) films annealed at different temperatures are used to explore the effects of the water absorption on the formation of PVA-iodine complexes. It's found that the higher the annealing temperature, the stronger the interaction force between PVA segments, and the smaller the free volume of the PVA films. These mainly lead to the reduction of the amount of PVA segments with a moderate degree of hydration (i.e., PVA segments with moderate mobility), which are the major segments participating in the formation of PVA-iodine complexes. Therefore, PVA films with higher water absorption not only possess faster complexation speed and form more PVA-iodine complexes, but also increase the proportion of polyiodide ions with a longer length. Moreover, the complexation restricts the PVA segments with high mobility, resulting in the formation of the intermolecular ordered structure. The water absorption dependence may guide the dyeing process to obtain PVA polarizers with excellent optical performance.
