Changes in aquaporins mRNA expression and liquid storage at 17°C: A potential biomarker of boar sperm quality?

ABSTRACT: Artificial insemination (AI) for pigs relies on liquid storage of extended semen at 17°C, which preserves sperm quality and ensures its fertilizing capacity. Routine quality controls include the evaluation of sperm motility, viability and capacitation status. The physiological functions of all these features depend on transmembrane aquaporins (AQPs), proteins playing key roles in osmoadaptation. In this study, we made a relative quantification, using RT-qPCR, of the mRNA of several sperm AQPs in AI-liquid semen doses before and after a 48-hr incubation period, aiming to determine possible quantitative compromising expression changes during the process that could serve as a diagnostic tool. Our results showed a decrease in classical sperm motility variables (total and progressive motility and velocity) and sperm viability after 48-hr storage, whereas capacitation status increased overtime. mRNA expression increased in the orthodox AQP4 and AQP6 after 48-hr incubation, relative to control (0 hr) and 24-hr time-points. Moreover, mRNA expression of aquaglyceroporins AQP3, AQP7 and AQP10 was higher after 48-hr incubation, confirmed by AQP7-protein validation using Western blot. Our results indicate that expression levels of AQPs-mRNA can change in ejaculated pig spermatozoa under conditions of ex-vivo incubation that could modify sperm homeostasis, suggesting it could eventually become a relevant molecular biomarker to assess the efficiency of liquid storage of pig semen.

Project description:Storage and transport of liquid boar sperm for artificial insemination (AI) requires the addition of solutions called extenders, which increase the volume of the ejaculate and help preserve its functional characteristics. Yet, the quality of sperm decreases over time primarily due to the increased production of reactive oxygen species (ROS) that damage the plasma membrane. Many commercial extenders are supplemented with additives that mitigate this effect. In semen, zinc is supplied at high concentration on the seminal plasma and helps protect the plasma membrane of sperm. However, zinc in the seminal plasma is diluted and chelated upon addition of extenders for storage, potentially reducing its antioxidant effect. Here we characterize viability, motility, mitochondrial activity, DNA integrity and ROS content of boar sperm diluted with Sus (Medi Nova, Italy) extender supplemented with different concentrations of ZnCl2, at intervals after dilution during 3 days. The ability of sperm supplemented with 2 mM ZnCl2 to fertilize oocytes in vivo of was also tested. Sperm viability was over 82% for all treatments. Mitochondrial integrity analysis, measured by Cytochrome c activity, indicated a protector effect of Zn, noted as a reduced number of sperm with extensive loss of mitochondrial activity. Acrosomal integrity was improved by treatment with all concentrations of ZnCl2 tested. Sperm kinematics were affected by treatment with ZnCl2, showing higher percentage of progressive and rapid sperm in doses supplemented with 2mM ZnCl2. ROS levels and chromatin integrity did not show differences between ZnCl2-supplemented doses and the control. Fertilization rate, total number, live, still born and mummified piglets did not change when sperm were diluted with extender containing 2 mM ZnCl2. The presented characterization indicates that Zn addition to Sus extender have a protective effect on mitochondrial sheath and acrosomal membranes; and provides the basis for further studies aimed to optimize sperm performance in AI.

Project description:Liquid preservation of boar sperm is crucial for artificial insemination application in pig production. However, time-dependent oxidative damage to sperm is one of the major challenges during the liquid preservation period. Caffeic acid phenethyl ester (CAPE) possesses excellent antioxidant properties and has potential therapeutic use in reproductive organ injury linked to oxidative stress. Adenosine monophosphate (AMP)-activated protein kinase (AMPK) involves in modulating the cellular redox state and exerts a beneficial effect on sperm preservation. In the present study, we firstly assessed different concentrations of CAPE that affect sperm quality during liquid storage to determine the appropriate addition. To further investigate whether CAPE exerts protective effects on boar sperm through modulation of AMPK activity, sperm quality parameters, antioxidant capacity, and marker protein expressions were evaluated under co-incubation with H2O2. The results showed that sperm treated with 210 μmol/L CAPE exhibited the highest motion parameters (total motility and progressive motility) and best functional integrity (mitochondrial activity, plasma membrane integrity, and acrosomal integrity). Even in the presence of H2O2, the addition of 210 μmol/L CAPE not only significantly improved sperm quality parameters, but also elevated CAT, SOD, and GSH-Px activities to enhance sperm antioxidant capacity. In addition, we found that CAPE could affect the protein activities of AMPK, phospho-AMPK α (p-AMPK), SOD, and Caspase-3 regardless of whether H2O2 is present or not. Our findings suggested that CAPE has potential application in liquid preservation of boar sperm and preliminary indicated that CAPE-induced improvement of sperm quality and antioxidant capacity should be mediated through conservation of AMPK activity. Further studies are required to illustrate the specific mechanism by which CAPE attenuates oxidative stress-mediated damages dependent on AMPK activity.

Project description:The preservation of semen is pivotal in animal reproduction to ensure successful fertilization and genetic improvement of livestock and poultry. However, investigating the underlying causes of differences in sperm liquid preservation ability and identifying relevant biomarkers remains a challenge. This study utilized liquid chromatography-mass spectrometry (LC-MS) to analyze the metabolite composition of seminal plasma (SP) from two groups with extreme differences in sperm liquid preservation ability. The two groups namely the good liquid preservation ability (GPA) and the poor preservation ability (PPA). The aim was to explore the relationship between metabolite composition in SP and sperm liquid preservation ability, and to identify candidate biomarkers associated with this ability of sperm. The results revealed the identification of 756 metabolites and 70 differentially expressed metabolites (DEM) in the SP from two groups of boar semen with differing liquid preservation abilities at 17 °C. The majority of identified metabolites in the SP belonged to organic acids and derivatives as well as lipids and lipid-like molecules. The DEM in the SP primarily consisted of amino acids, peptides, and analogs. The Kyoto Encyclopedia of Genes and Genomes analysis also demonstrated that the DEM are mainly concentrated in amino acid synthesis and metabolism-related pathways (P < 0.05). Furthermore, eleven key metabolites were identified and six target amino acids were verified, and the results were consistent with the non-targeted metabolic analysis. These findings indicated that amino acids and their associated pathways play a potential role in determining boar sperm quality and liquid preservation ability. D-proline, arginine, L-citrulline, phenylalanine, leucine, DL-proline, DL-serine, and indole may serve as potential biomarkers for early assessment of boar sperm liquid preservation ability. The findings of this study are helpful in understanding the causes and mechanisms of differences in the liquid preservation ability of boar sperm, and provide valuable insights for improving semen quality assessment methods and developing novel extenders or protocols.
