Project description:Carbon nanohorns (CNHs), closed cone-shaped cages of sp 2-hybridized carbons, are a promising nanomaterial to improve carbon-fiber microelectrode (CFME) dues to their high specific surface area and edge planes, but few studies have tested their electrochemical properties. Here, we tested the dopamine detection at electrodeposited CNHs on CFME (CNH/CFME). The optimized concentration of CNHs in the deposition solution is 0.5 mg/mL, and the optimized electrodeposition waveform is 10 cycles of triangular waveform scanned from -1.0 V and +1.0 V at 50 mV/s. Using fast-scan cyclic voltammetry, the optimized CNH/CFME enhances dopamine peak current to 2.3 ± 0.2 times that of the CFME. To further increase the current, CNH/CFMEs were oxidized in NaOH (ox-CNH/CFME), which creates more defects and surface oxide groups to adsorb dopamine. The oxidative etching further increases the peak current to 3.5 ± 0.2 times of the CFME, and ox-CNH/CFME had a limit of detection of 6 ± 2 nM. The dopamine anodic current at ox-CNH/CFME was stable for 8 h of continuous scanning. The ox-CNH/CFME enhanced the anodic peak current for other cationic neurotransmitters including epinephrine, norepinephrine, and serotonin, but less enhancement was found for ascorbic acid, showing higher selectivity for cationic molecules. CNHs also decreased tissue biofouling at CFME. Thus, electrodeposited CNHs are a promising new method for increasing the surface area and current of CFMEs for dopamine detection.

Project description:We present a method to modify carbon-fiber microelectrodes (CFME) with porous carbon nanofibers (PCFs) to improve detection and to investigate the impact of porous geometry for dopamine detection with fast-scan cyclic voltammetry (FSCV). PCFs were fabricated by electrospinning, carbonizing, and pyrolyzing poly(acrylonitrile)-b-poly(methyl methacrylate) (PAN-b-PMMA) block copolymer nanofiber frameworks. Commonly, porous nanofibers are used for energy storage applications, but we present an application of these materials for biosensing which has not been previously studied. This modification impacted the topology and enhanced redox cycling at the surface. PCF modifications increased the oxidative current for dopamine 2.0 ± 0.1-fold (n = 33) with significant increases in detection sensitivity. PCF are known to have more edge plane sites which we speculate lead to the two-fold increase in electroactive surface area. Capacitive current changes were negligible providing evidence that improvements in detection are due to faradaic processes at the electrode. The ΔEp for dopamine decreased significantly at modified CFMEs. Only a 2.2 ± 2.2 % change in dopamine current was observed after repeated measurements and only 10.5 ± 2.8% after 4 hours demonstrating the stability of the modification over time. We show significant improvements in norepinephrine, ascorbic acid, adenosine, serotonin, and hydrogen peroxide detection. Lastly, we demonstrate that the modified electrodes can detect endogenous, unstimulated release of dopamine in living slices of rat striatum. Overall, we provide evidence that porous nanostructures significantly improve neurochemical detection with FSCV and echo the necessity for investigating the extent to which geometry impacts electrochemical detection.

Project description:Carbon fiber-microelectrodes (CFMEs) are one of the standards for the detection of neurotransmitters such as dopamine (DA). In this study, we demonstrate that CFMEs electrodeposited with poly (3,4-ethylenedioxythiophene) (PEDOT) in the presence of Nafion exhibit enhanced sensitivity for DA detection. Scanning electron microscopy (SEM) revealed the smooth outer surface morphologies of polymer coatings, which filled in the ridges and grooves of the bare unmodified carbon electrode and energy-dispersive X-ray spectroscopy (EDX) confirmed PEDOT:Nafion incorporation. PEDOT:Nafion coated CMFEs exhibited a statistically enhanced two-fold increase in DA sensitivity compared to unmodified microelectrodes, with stability and integrity of the coated microelectrodes maintained for at least 4 h. A scan rate test revealed a linear relationship with peak DA oxidative current (5 μM), indicating adsorption control of DA to the surface of the PEDOT:Nafion electrode. As proof of principle, PEDOT:Nafion coated electrodes were used to detect potassium chloride (KCl)-induced DA release in zebrafish (Danio rerio) retinal tissue ex vivo, thus illustrating their applicability as biosensors.

Project description:We have compared the properties and resistance to DA fouling of a carbon nanotube fiber (CNTF) microelectrode to a traditional carbon fiber (CF) microelectrode. These two materials show comparable electrochemical activities for outer-sphere and inner-sphere redox reactions. Although the CNTF might have a higher intrinsic RC constant, thus limiting its high-frequency behavior, the CNTF shows a significantly higher durability than the CF in terms of electrode stability. During constant oxidation of 100 ?M DA, the signal measured by the CNTF microelectrode shows a 2-h window over which no decrease in current is observed. Under the same conditions, the current obtained at the CF microelectrode decreases by almost 50%. A model of the fouling process, assuming the formation of growing patches of insulator on the surface, has been compared to the data. This model is found to be in good agreement with our results and indicates a growth rate of the patches in the 0.1-2 nm s(-1) range.

Project description:Exocytosis is an ultrafast cellular process which facilitates neuron-neuron communication in the brain. Microelectrode electrochemistry has been an essential tool for measuring fast exocytosis events with high temporal resolution and high sensitivity. Due to carbon fiber's irreproducible and inhomogeneous surface conditions, however, it is often desirable to develop simple and reproducible modification schemes to enhance a microelectrode's analytical performance for single-cell analysis. Here we present carbon-fiber microelectrodes (CFEs) modified with a thin film of electrodeposited gold for the detection of exocytosis from rat pheochromocytoma cells (PC12), a model cell line for neurosecretion. These new probes are made by a novel voltage-pulsing deposition procedure and demonstrate improved electron-transfer characteristics for catecholamine oxidation, and their fabrication is tractable for many different probe designs. When we applied the probes to the detection of catecholamine release, we found that they outperformed unmodified CFEs. Further, the improved performance was conserved at cells incubated with L-DOPA (l-3,4-dihydroxyphenylalanine), a precursor to dopamine that increases the quantal size of the release events. Future use of this method may allow nanoelectrodes to be modified for highly sensitive detection of exocytosis from chemical synapses.

Project description:The mechanisms that control extracellular serotonin levels in vivo are not well-defined. This shortcoming makes it very challenging to diagnose and treat the many psychiatric disorders in which serotonin is implicated. Fast-scan cyclic voltammetry (FSCV) can measure rapid serotonin release and reuptake events but cannot report critically important ambient serotonin levels. In this Article, we use fast-scan controlled adsorption voltammetry (FSCAV), to measure serotonin's steady-state, extracellular chemistry. We characterize the "Jackson" voltammetric waveform for FSCAV and show highly stable, selective, and sensitive ambient serotonin measurements in vitro. In vivo, we report basal serotonin levels in the CA2 region of the hippocampus as 64.9 ± 2.3 nM (n = 15 mice, weighted average ± standard error). We electrochemically and pharmacologically verify the selectivity of the serotonin signal. Finally, we develop a statistical model that incorporates the uncertainty in in vivo measurements, in addition to electrode variability, to more critically analyze the time course of pharmacological data. Our novel method is a uniquely powerful analysis tool that can provide deeper insights into the mechanisms that control serotonin's extracellular levels.

Project description:Carbon fiber-microelectrodes (CFMEs) are considered to be one of the standard electrodes for neurotransmitter detection such as dopamine (DA). DA is physiologically important for many pharmacological and behavioral states, but is readily metabolized on a fast, subsecond timescale. Recently, DA metabolites such as 3-methoxytyramine (3-MT) and 3,4-dihydroxyphenylacetaldehyde (DOPAL) were found to be involved in physiological functions, such as movement control and progressive neuro degeneration. However, there is no current assay to detect and differentiate them from DA. In this study, we demonstrate the co-detection of similarly structured neurochemicals such as DA, 3-MT, and DOPAL. We accomplished this through electrodepositing CFMEs with polyethyleneimine (PEI) and poly(3,4-ethylenedioxythiophene) polystyrene sulfonate (PEDOT:PSS) polymers. This endowed the bare unmodified CFMEs with surface charge, physical, and chemical differences, which resulted in the improved sensitivity and selectivity of neurotransmitter detection. The differentiation and detection of 3-MT, DOPAL, and DA will potentially help further understand the important physiological roles that these dopaminergic metabolites play in vivo.

Project description:L-DOPA has been the gold standard for symptomatic treatment of Parkinson's disease. However, its efficacy wanes over time as motor complications develop. Very little is known about how L-DOPA therapy affects the dynamics of fluctuating dopamine concentrations in the striatum on a rapid time scale (seconds). Electrochemical studies investigating the effects of L-DOPA treatment on electrically evoked dopamine release have reported conflicting results with significant variability. We hypothesize that the uncertainty in the electrochemical data is largely due to electrode fouling caused by polymerization of L-DOPA and endogenous catecholamines on the electrode surface. Thus, we have systematically optimized the procedure for fabricating cylindrical, Nafion-coated, carbon-fiber microelectrodes. This has enabled rapid and reliable detection of L-DOPA's effects on striatal dopamine signaling in intact rat brain using fast-scan cyclic voltammetry. An acute dose of 5 mg/kg L-DOPA had no significant effect on dopamine dynamics, demonstrating the highly efficient regulatory mechanisms at work in the intact brain. In contrast, administration of 200 mg/kg L-DOPA significantly increased the amplitude of evoked dopamine release by ∼200%. Overall, this work describes a reliable tool that allows a better measure of L-DOPA augmented dopamine release in vivo, measured using fast-scan cyclic voltammetry. It provides a methodology that improves the stability and performance of the carbon-fiber microelectrode when studying the molecular mechanisms underlying L-DOPA therapy and also promises to benefit a wide variety of studies because Nafion is so commonly used in electroanalytical chemistry.

Project description:Adenosine is a neuromodulator that regulates neurotransmission. Adenosine can be monitored using fast-scan cyclic voltammetry at carbon-fiber microelectrodes and ATP is a possible interferent in vivo because the electroactive moiety, adenine, is the same for both molecules. In this study, we investigated carbon-fiber microelectrodes coated with Nafion and carbon nanotubes (CNTs) to enhance the sensitivity of adenosine and decrease interference by ATP. Electrodes coated in 0.05 mg mL(-1) CNTs in Nafion had a 4.2 ± 0.2 fold increase in current for adenosine, twice as large as for Nafion alone. Nafion-CNT electrodes were 6 times more sensitive to adenosine than ATP. The Nafion-CNT coating did not slow the temporal response of the electrode. Comparing different purine bases shows that the presence of an amine group enhances sensitivity and that purines with carbonyl groups, such as guanine and hypoxanthine, do not have as great an enhancement after Nafion-CNT coating. The ribose group provides additional sensitivity enhancement for adenosine over adenine. The Nafion-CNT modified electrodes exhibited significantly more current for adenosine than ATP in brain slices. Therefore, Nafion-CNT modified electrodes are useful for sensitive, selective detection of adenosine in biological samples.

Project description:Detection of ions is challenging due to their small size, rapid diffusion, and high mobility, especially for assaying in samples of low volumes. Among the traditional analytical methods, potentiometric ion-selective electrodes (ISE) have become a popular choice for detecting ions as they are cost-effective, user-friendly and can be miniaturized, making them useful for on-site analysis. In this context, radially aligned carbon nanotubes (RACNT) directly grown on glass fibers (GF) via the chemical vapor deposition method is investigated as a solid contact material for the fabrication of ion-selective microelectrodes (μISE) upon incorporating specific ionophores within a polymeric encapsulation membrane. As an illustration, sensitive detection of ammonium ions is accomplished by the fabricated μISE (plasticized PVC membrane containing nonactin ionophores), which yielded a LOD and a linear response range between 7.5 × 10-6 and 1.0 × 10-5 to 1.0 × 10-1 M, respectively. The μISE fabricated with RACNT-GF as an interface material exhibited improvements in LOD and enhanced the detection selectivity as compared to a conventional ISE fabricated using planar solid contact materials such as graphite. We hypothesize that the fabricated μISE with a high surface area and mechanical durability maximize the accommodation of ionophores in the barrier membrane for yielding improved potentiometric responses. Experimental results illustrate that the μISE possesses the potential to be utilized for the fabrication of selective and sensitive ISE upon incorporation of specific ionophores with RACNT-GF composites.
