Project description:Herein, we report the electropolymerization of crystal violet (CRV) on a bare glassy carbon electrode (GCE) for the detection of adrenaline (AD). Electropolymerization parameters such as electrolyte pH, scan rate and monomer concentrations were optimized using cyclic voltammetry (CV) and electrochemical impedance spectroscopy (EIS). The characterization of CRV and poly(crystal violet) (PCV) was done using FT-IR, UV-visible spectroscopy and EIS. More importantly, the charge transfer resistance (Rct) and other EIS data recorded from the EIS of various forms of the poly(crystal violet) (PCV) modified glassy carbon electrode (GCE) in AD were used for identifying the best PCV modified electrode. Subsequent application of the electrode prepared at optimum conditions (PGCE) for AD detection using the square wave voltammetry (SWV) gave a limit of detection (LOD) of 2.86 μM over a linear range of 10.3-102.7 μM. This sensor also showed considerable stability, good AD recovery from the real sample (98.9%), and excellent reproducibility, making it a suitable analytical tool for AD detection at the micromolar level.

Project description:Ferulic acid (FA), a monohydroxycinnamic acid, is an antioxidant with multiple beneficial effects on human health, presenting also importance in the food and cosmetics industry. Its electrochemical behavior was investigated at the disposable and cost-effective pencil graphite electrode (PGE). Cyclic voltammetry emphasized its pH-dependent, diffusion-controlled oxidation. Using the optimized conditions (HB type PGE, Britton Robinson buffer pH 4.56) differential pulse and square-wave voltammetric techniques were applied for its quantitative determination in the range 4.00 × 10-7-1.00 × 10-3 mol/L FA. The developed methods were employed for the rapid and simple assessment of the FA content from a commercially available powder designed for cosmetic use.

Project description:BackgroundIntegrative analysis between dynamical modeling of metabolic networks and data obtained from high throughput technology represents a worthy effort toward a holistic understanding of the link among phenotype and dynamical response. Even though the theoretical foundation for modeling metabolic network has been extensively treated elsewhere, the lack of kinetic information has limited the analysis in most of the cases. To overcome this constraint, we present and illustrate a new statistical approach that has two purposes: integrate high throughput data and survey the general dynamical mechanisms emerging for a slightly perturbed metabolic network.Methodology/principal findingsThis paper presents a statistic framework capable to study how and how fast the metabolites participating in a perturbed metabolic network reach a steady-state. Instead of requiring accurate kinetic information, this approach uses high throughput metabolome technology to define a feasible kinetic library, which constitutes the base for identifying, statistical and dynamical properties during the relaxation. For the sake of illustration we have applied this approach to the human Red blood cell metabolism (hRBC) and its capacity to predict temporal phenomena was evaluated. Remarkable, the main dynamical properties obtained from a detailed kinetic model in hRBC were recovered by our statistical approach. Furthermore, robust properties in time scale and metabolite organization were identify and one concluded that they are a consequence of the combined performance of redundancies and variability in metabolite participation.Conclusions/significanceIn this work we present an approach that integrates high throughput metabolome data to define the dynamic behavior of a slightly perturbed metabolic network where kinetic information is lacking. Having information of metabolite concentrations at steady-state, this method has significant relevance due its potential scope to analyze others genome scale metabolic reconstructions. Thus, I expect this approach will significantly contribute to explore the relationship between dynamic and physiology in other metabolic reconstructions, particularly those whose kinetic information is practically nulls. For instances, I envisage that this approach can be useful in genomic medicine or pharmacogenomics, where the estimation of time scales and the identification of metabolite organization may be crucial to characterize and identify (dis)functional stages.

Project description:In this work, a novel electrochemical assay for characterizing both lipases and lipase inhibitors as well as for the determination of lipase activity is described. It is based on a carbon paste electrode, modified with cobalt(II)phthalocyanine, and multi-walled carbon nanotubes (MWCNTs). As reaction media, a sodium borate buffer was used (0.1 M, pH 9). The measurements were carried out in a batch system using differential pulse voltammetry (DPV) and 1,3-dilinolein as standard substrate. The activity assay showed a linearity for porcine pancreas lipase activity in a range between 20 and 300 U L-1 (per min) with a limit of detection (LOD) of 7 U L-1 and a limit of quantification (LOQ) of 20 U L-1. The kinetic behavior of the lipase reaction was investigated, resulting in a KM value of 0.29 mM. The applicability of the activity assay could be shown by investigating the activity of lipases from Aspergillus oryzae and Candida rugosa, and the results were confirmed by a reference method. The inhibitory effects were characterized with Orlistat.

Project description:Building on diamond characteristics such as hardness, chemical inertness and low electron emission threshold voltage, the current microscopic, spectroscopic and voltammetric investigations are directed towards improving the properties of electrode coating materials for their future use in clinical studies of deep brain stimulation via fast-scan cyclic voltammetry (FSCV). In this study we combine the capabilities of confocal Raman mapping in providing detailed and accurate analysis of local distributions of material constituents in a series of boron-doped polycrystalline diamond films grown by chemical vapor deposition, with information from the more conventional techniques of scanning electron microscopy (SEM) and infrared absorption spectroscopy. Although SEM images show a uniform distribution of film crystallites, they have the limitation of being unable to differentiate the distribution of boron in the diamond. Values of 1018-1021 atoms/cm³ of boron content have been estimated from the absorption coefficient of the 1290 cm-1 infrared absorption band and from the 500 cm-1 Raman vibration. The observed accumulation of boron atoms and carbon sp² impurities at the grain boundaries suggests that very high doping levels do not necessarily contribute to improvement of the material's conductivity, corroborating with voltammetric data. FSCV results also indicate an enhanced stability of analyte detection.

Project description:Nanocorrals with different size, shape, and orientation are created on covalently modified highly oriented pyrolytic graphite surfaces using scanning probe nanolithography, i.e., nanoshaving. Alkylated diacetylene molecules undergo laterally confined supramolecular self-assembly within these corrals. When nanoshaving is performed in situ, at the liquid-solid interface, the orientation of the supramolecular lamellae structure is directionally influenced by the gradual graphite surface exposure. Careful choice of the nanoshaving direction with respect to the substrate symmetry axes promotes alignment of the supramolecular lamellae within the corral. Self-assembly occurring inside corrals of different size and shape reveals the importance of geometric and kinetic constraints controlled by the nanoshaving process. Finally, seed-mediated crystallization studies demonstrate confinement control over nucleation and growth principles.

Project description:Many DNA methylome profiling methods cannot distinguish between 5-methylcytosine (5mC) and 5-hydroxymethylcytosine (5hmC). Because 5mC typically acts as a repressive mark whereas 5hmC is an intermediate form during active demethylation, the inability to separate their signals could lead to incorrect interpretation of the data. Is the extra information contained in 5hmC signals worth the additional experimental and computational costs? Here we combine whole-genome bisulfite sequencing (WGBS) and oxidative WGBS (oxWGBS) data in various human tissues to investigate the quantitative relationships between gene expression and the two forms of DNA methylation at promoters, transcript bodies, and immediate downstream regions. We find that 5mC and 5hmC signals correlate with gene expression in the same direction in most samples. Considering both types of signals increases the accuracy of expression levels inferred from methylation data by a median of 18.2% as compared to having only WGBS data, showing that the two forms of methylation provide complementary information about gene expression. Differential analysis between matched tumor and normal pairs is particularly affected by the superposition of 5mC and 5hmC signals in WGBS data, with at least 25%-40% of the differentially methylated regions (DMRs) identified from 5mC signals not detected from WGBS data. Our results also confirm a previous finding that methylation signals at transcript bodies are more indicative of gene expression levels than promoter methylation signals. Overall, our study provides data for evaluating the cost-effectiveness of some experimental and analysis options in the study of DNA methylation in normal and cancer samples.

Project description:Semi-batch emulsion polymerizations of vinylidene fluoride (VDF) are reported. The molar mass control is achieved via iodine transfer polymerization (ITP) using IC₄F₈I as chain transfer agent. Polymerizations carried out at 75 °C and pressures ranging from 10 to 30 bar result in low dispersity polymers with respect to the molar mass distribution (MMD). At higher pressures a significant deviation from the ideal behavior expected for a reversible deactivation transfer polymerization occurs. As identified by kinetic Monte Carlo (kMC) simulations of the activation⁻deactivation equilibrium, during the initialization period of the chain transfer agent already significant propagation occurs due to the higher pressure, and thus, the higher monomer concentration available. Based on the kMC modeling results, semi-batch emulsion polymerizations were carried out as a two pressure process, which resulted in very good control of the MMD associated with a comparably high polymerization rate.

Project description:Reactions of the [3Fe-4S] cluster and various metallated [M3Fe-4S] adducts co-ordinated in the ferredoxin from the hyperthermophile Pyrococcus furiosus have been studied by protein-film voltammetry, bulk-solution voltammetry, solution kinetics and magnetic CD (MCD). The [3Fe-4S] cluster exhibits two couples, [3Fe-4S]+/0 and [3Fe-4S]0/2-. Film voltammetry is possible over a wide pH range (2-8), revealing that the [3Fe-4S]+/0 couple shows a complex pH dependence with pKred1=2.8, pKox=4.9 and pKred2=6.7. From MCD, pKred1 corresponds with protonation of [3Fe-4S]0 to give a spectroscopically distinct species, as reported for ferredoxins from Azotobacter and Sulfolobus. The status of the disulphide/disulphydryl entity makes no significant difference to the data (given for the -S-S- form). Formation of the hyper-reduced [3Fe-4S]2- state is observed, requiring 3H+ for the overall 3e- reduction of [3Fe-4S]+, the change therefore being electroneutral. By comparison with the ferredoxin from Desulfovibrio africanus, uptake of Fe(II) and other M(II) by [3Fe-4S]0 to give [M3Fe-4S] clusters is slow (t1/2>10 min at room temperature, slower still if the protein is adsorbed on the electrode), whereas reaction with Tl(I) to produce [Tl3Fe-4S] is very rapid (t1/2<<1 s), suggesting that co-ordination of Tl does not require reorganization of the protein structure. Rates of formation of [3Fe-4S] from [M3Fe-4S] adducts increase sharply at high potentials, showing that metal release involves a labile 'super-oxidized' [M3Fe-4S]3+ state.
