Project description:Two hundred and thirty-two bacterial strains were isolated from the rhizospheric soil of Populus euphratica which is the dominant tree living in extreme arid regions in northwest China. Some strains with plant growth-promoting bacteria related metabolic characteristics were able to promote drought resistance in plants after inoculation. Ten strains with the greatest effects increased the dry weight of wheat shoots from 0.5 to 34.4 %, and the surface area of the root systems from 12.56 to 212.17 % compared to the control after drought treatment whereas no obvious promoting effect was observed in normal water conditions. These 10 strains were identified to be of the genera Pseudomonas, Bacillus, Stenotrophomonas and Serratia by 16S rRNA (rrs) gene sequence alignment. Among these strains, Serratia sp. 1-9 and Pseudomonas sp. 5-23 were the two most effective strains. Both of them produced auxin and the production increased significantly when cultured under simulated drought conditions which are inferred to be the most plausible mechanism for their plant growth-promoting effect under drought stress.

Project description:The application of plant growth promoting rhizobacteria (PGPR) to agro-ecosystems is considered to have the potential for improving plant growth in extreme environments featured by water shortage. Herein, we isolated bacterial strains from foxtail millet (Setaria italica L.), a drought-tolerant crop cultivated in semiarid regions in the northeast of China. Four isolates were initially selected for their ability to produce ACC deaminase as well as drought tolerance. The isolates were identified as Pseudomonas fluorescens, Enterobacter hormaechei, and Pseudomonas migulae on the basis of 16S rRNA sequence analysis. All of these drought-tolerant isolates were able to produce EPS (exopolysaccharide). Inoculation with these strains stimulated seed germination and seedling growth under drought stress. Pseudomonas fluorescens DR7 showed the highest level of ACC deaminase and EPS-producing activity. DR7 could efficiently colonize the root adhering soil, increased soil moisture, and enhance the root adhering soil/root tissue ratio. These results suggest drought tolerant PGPR from foxtail millet could enhance plant growth under drought stress conditions and serve as effective bioinoculants to sustain agricultural production in arid regions.

Project description:Tree allometry in semi-arid forests is characterized by short height but large canopy. This pattern may be important for maintaining water-use efficiency and carbon sequestration simultaneously, but still lacks quantification. Here we use terrestrial laser scanning to quantify allometry variations of Quercus mongolica in semi-arid forests. With tree height (Height) declining, canopy area (CA) decreases with substantial variations. The theoretical CA-Height relationship in dynamic global vegetation models (DGVMs) matches only the 5th percentile of our results because of CA underestimation and Height overestimation by breast height diameter (DBH). Water supply determines Height variation (P = 0.000) but not CA (P = 0.2 in partial correlation). The decoupled functions of stem, hydraulic conductance and leaf spatial arrangement, may explain the inconsistency, which may further ensure hydraulic safety and carbon assimilation, avoiding forest dieback. Works on tree allometry pattern and determinant will effectively supply tree drought tolerance studying and support DGVM improvements.

Project description:Understanding how patterns of leaf nutrient traits respond to groundwater depth is crucial for modeling the nutrient cycling of desert riparian ecosystems and forecasting the responses of ecosystems to global changes. In this study, we measured leaf nutrients along a transect across a groundwater depth gradient in the downstream Heihe River to explore the response of leaf nutrient traits to groundwater depth and soil properties. We found that leaf nutrient traits of dominant species showed different responses to groundwater depth gradient. Leaf C, leaf N, leaf P, and leaf K decreased significantly with groundwater depth, whereas patterns of leaf C/N and leaf N/P followed quadratic relationships with groundwater depth. Meanwhile, leaf C/P did not vary significantly along the groundwater depth gradient. Variations in leaf nutrient traits were associated with soil properties (e.g., soil bulk density, soil pH). Groundwater depth and soil pH jointly regulated the variation of leaf nutrient traits; however, groundwater depth explained the variation of leaf nutrient traits better than did soil pH. At the local scale in the typical desert riparian ecosystem, the dominant species was characterized by low leaf C, leaf N, and leaf P, but high leaf N/P and leaf C/P, indicating that desert riparian plants might be more limited by P than N in the growing season. Our observations will help to reveal specific adaptation patterns in relation to the groundwater depth gradient for dominant desert riparian species, provide insights into adaptive trends of leaf nutrient traits, and add information relevant to understanding the adaptive strategies of desert riparian forest vegetation to moisture gradients.

Project description:We examined the effect of soil microbial communities on plant physiological responses to drought. Bouteloua gracilis seeds were planted in sterilized sand with (inoculated) and without (controls) soil microbial communities. After substantial growth, drought was imposed by completely withholding water. Before soil moisture declined to zero, inoculated plants germinated faster, were significantly taller, and maintained greater soil moisture than controls. The greater soil moisture of the inoculated plants allowed greater photosynthesis but also induced lower tissue drought tolerance (as indicated by turgor loss point) compared to controls. The inoculated plants were more susceptible to severe drought compared to control plants as indicated by significantly lower mean stomatal conductance, as well as marginally significantly greater mean wilting score, for the entire severe drought period after soil moisture declined to zero. Inoculated plants exhibited enhanced growth and photosynthesis and dampened drought stress over short timescales, but also increased susceptibility to drought over long timescales. This work demonstrates (1) an unexpected insight that microbes can have positive initial effects on plant performance, but negative impacts on plant performance during severe drought, and (2) that microbially altered effects on plant function during well-watered and moderate drought conditions can influence plant function under subsequent severe drought.

Project description:Plant-plant interactions are among the fundamental processes that shape structure and functioning of arid and semi-arid plant communities. Despite the large amount of studies that have assessed the relationship between plant-plant interactions (i.e., facilitation and competition) and diversity, often researchers forget a third kind of interaction, known as allelopathy. We examined the effect of plant-plant interactions of three dominant species: the perennial grass Lygeum spartum, the allelopathic dwarf shrub Artemisia herba-alba, and the nurse shrub Salsola vermiculata, on plant diversity and species composition in a semi-arid ecosystem in NE Spain. Specifically, we quantified the interaction outcome (IO) based on species co-occurrence, we analyzed diversity by calculation of the individual species-area relationship (ISAR), and compositional changes by calculation of the Chao-Jaccard similarity index. We found that S. vermiculata had more positive IO values than L. spartum, and A. herba-alba had values between them. Lygeum spartum and A. herba-alba acted as diversity repellers, whereas S. vermiculata acted as a diversity accumulator. As aridity increased, A. herba-alba transitioned from diversity repeller to neutral and S. vermiculata transitioned from neutral to diversity accumulator, while L. spartum remained as diversity repeller. Artemisia herba-alba had more perennial grass species in its local neighborhood than expected by the null model, suggesting some tolerance of this group to its "chemical neighbor". Consequently, species that coexist with A. herba-alba were very similar among different A. herba-alba individuals. Our findings highlight the role of the nurse shrub S. vermiculata as ecosystem engineer, creating and maintaining patches of diversity, as well as the complex mechanism that an allelopathic plant may have on diversity and species assemblage. Further research is needed to determine the relative importance of allelopathy and competition in the overall interference of allelopathic plants.

Project description:Soil drying causes leaf rolling in rice, but the relationship between leaf rolling and drought tolerance has historically confounded selection of drought-tolerant genotypes. In this study on tropical japonica and aus diversity panels (170-220 genotypes), the degree of leaf rolling under drought was more affected by leaf morphology than by stomatal conductance, leaf water status, or maintenance of shoot biomass and grain yield. A range of canopy temperature and leaf rolling (measured as change in normalized difference vegetation index [ΔNDVI]) combinations were observed among aus genotypes, indicating that some genotypes continued transpiration while rolled. Association mapping indicated colocation of genomic regions for leaf rolling score and ΔNDVI under drought with previously reported leaf rolling genes and gene networks related to leaf anatomy. The relatively subtle variation across these large diversity panels may explain the lack of agreement of this study with earlier reports that used small numbers of genotypes that were highly divergent in hydraulic traits driving leaf rolling differences. This study highlights the large range of physiological responses to drought among rice genotypes and emphasizes that drought response processes should be understood in detail before incorporating them into a varietal selection programme.

Project description:Climate change and anthropogenic activities are responsible for extensive crop yield losses, with negative impact on global agricultural production. The occurrence of extreme weather events such as drought is a big challenge for agriculture, negatively impacting crops. Thus, methodologies reducing crop dependence on water will be a great advantage. Plant roots are colonized by soil bacteria, that can establish beneficial associations with plants, increasing crop productivity and plant tolerance to abiotic stresses. The aim of this study was to promote plant growth and to increase crop tolerance to drought by inoculation with osmotolerant bacterial strains. For that, bacteria were isolated from plants growing in Sal Island (Cape Verde) and identified. The osmotolerance and plant-growth promotion (PGP) abilities of the strains were determined. A maize seed cultivar tolerant to drought was inoculated with the strains evidencing best PGP capacity and osmo-tolerance. Results evidenced the ability of some bacterial strains increasing the development and inducing osmotolerance in plants. These results evidence the potential of osmotolerant bacteria to further increase the level of tolerance of maize varieties tolerant to drought, decreasing the dependence of this crop on irrigation, and open new perspectives to growth maize in drought affected areas and to use water more efficiently.
