Project description:Background: High stocking density and intensive feeding in aquaculture systems lead to the accumulation of organic waste, which results in an increase in ammonia, nitrite, and nitrite concentrations in culture media. Biofloc is a potential technology to overcome this problem. The starter is a crucial carbon source for bacteria in the formation of biofloc. The objective of the present study aimed to explore the best starter of biofloc in a red tilapia Oreochromis niloticus culture system. Methods: A completely randomized design with four levels of treatment was used in this study. The tested starter was (A) control treatment, biofloc without starter, (B) biofloc with molasses starter, (C) biofloc with tapioca starter, and (D) biofloc with sucrose starter. The floc was cultured in 100-L tanks with a salinity of 17 ppt. The tanks were stocked with O. niloticus with a size of 3.71±0.11 cm at a stocking density of 30 fish per tank. The fish were fed on a commercial diet two times a day at satiation for 40 days. The ammonia, nitrite, and nitrite concentrations were measured for an interval of 8 days. Results: The study showed that the NH 3-N range was 0.02-0.07 mg L -1 (mean, 0.03 ± 0.02 mg L -1), NO 2-N range was 0.20-0.43 mg L -1 (mean, 0.25 ± 0.12 mg L -1), and NO 3-N range was 0.90-3.20 mg L -1 (mean, 1.42 ± 1.19 mg L -1). Conclusion: Among the starters tested, molasses was found to be the best for biofloc in tilapia culture.

Project description:Oreochromis niloticus (genetically improved farmed tilapia, GIFT) often bites the root of Polygonum cuspidatum when it is used as a floating bed, and resveratrol (RES) is mainly accumulated in the root of P. cuspidatum. Blood acts as a pipeline for the fish immune system. Generating blood transcriptomic resources is crucial for understanding molecular mechanisms underlying blood immune responses. In this study, we determined the effects of RES administration on blood transcriptomic response in GIFT. With increasing RES concentration, 133 (0.025 vs. 0.05 g/kg RES), 155 (0.025 vs. 0.1 g/kg RES), and 123 (0.05 vs. 0.1 g/kg RES) genes were detected as significant differentially expressed genes (DEGs). Three and ninety-five shared significant DEGs were found to be enriched among the three (except 0.1 g/kg RES) and four groups (0, 0.025, 0.05, and 0.1 g/kg RES), respectively. To determine the relationship between mitochondrial regulation and RES supplementation, the results of RNA-Seq were analyzed and nine mitochondria-related genes (ATP synthase or mitochondrial-function-related genes) were verified. The results revealed the same expression pattern: cytochrome c isoform X2 (cox2), katanin p60 ATPase-containing subunit A1 isoform X1 (katna1), plasma membrane calcium-transporting ATPase 1-like (atp2b1) and GTP-binding protein A-like (gtpbpal) showed the highest expression in the 0.1 g/kg RES group, while NADH dehydrogenase [ubiquinone] iron-sulfur protein 2 mitochondrial (nad7), ATP synthase subunit beta, mitochondrial (atpb), ATP synthase subunit alpha, mitochondrial-like (atpal), ATP synthase subunit alpha, mitochondrial (atpa) and ATP-dependent Clp protease proteolytic subunit, mitochondrial (clpp) revealed a dose-dependent expression following RES supplementation. Blood Ca2+-ATPase activity, and malondialdehyde, glutathione, and ATP content were significantly increased in the 0.05 (except Ca2+-ATPase activity), 0.1 g/kg RES group when compared with the controls. Eighty-nine shared DGEs were mainly enriched in antigen processing and presentation, cell adhesion molecules and phagosome pathways, based on the comparison between previous reported hepatic and the present blood transcriptome. Our study demonstrated that RES supplementation might improve the resistance to metabolism dysfunction via mitochondrial energy synthesis and/or the respiratory chain (e.g., ATPase).

Project description:Studies of the ammonia oxidation reaction (AOR) for the synthesis of nitrite and nitrate (NO2/3 - ) have been limited to a small number of catalytic materials, majorly Pt based. As the demand for nitrate-based products such as fertilisers continues to grow, exploration of alternative catalysts is needed. Herein, 19 metals immobilised as particles on carbon fibre electrodes were tested for their catalytic activity for the ammonia electrooxidation to NO2/3 - under alkaline conditions (0.1 m KOH). Nickel-based electrodes showed the highest overall NO2/3 - yield with a rate of 5.0±1.0 nmol s-1 cm-2 , to which nitrate contributed 62±8 %. Cu was the only catalyst that enabled formation of nitrate, at a rate of 1.0±0.4 nmol s-1 cm-2 , with undetectable amounts of nitrite produced. Previously unexplored in this context, Fe and Ag also showed promise and provided new insights into the mechanisms of the process. Ag-based electrodes showed strong indications of activity towards NH3 oxidation in electrochemical measurements but produced relatively low NO2/3 - yields, suggesting the formation of alternate oxidation products. NO2/3 - production over Fe-based electrodes required the presence of dissolved O2 and was more efficient than with Ni on longer timescales. These results highlight the complexity of the AOR mechanism and provide a broad set of catalytic activity and nitrate versus nitrite yield data, which might guide future development of a practical process for the distributed sustainable production of nitrates and nitrites at low and medium scales.

Project description:Understanding population structure and genetic diversity within and between local Nile tilapia lines cultured in Tanzania is important for sustainable aquaculture production. This study investigated the genetic structure and diversity among seven Nile tilapia populations in Tanzania (Karanga, Igunga, Ruhila, Fisheries Education and Training Agency, Tanzania Fisheries Research Institute, Kunduchi, and Lake Victoria). Double-digest restriction site-associated DNA (ddRAD) libraries were prepared from 140 individual fish (20 per population) and sequenced using an Illumina HiSeq 4000 resulting in the identification of 2,180 informative single nucleotide polymorphisms (SNPs). Pairwise Fst values revealed strong genetic differentiation between the closely related populations; FETA, Lake Victoria, and Igunga and those from TAFIRI and Karanga with values ranging between 0.45 and 0.55. Population structure was further evaluated using Bayesian model-based clustering (STRUCTURE) and discriminant analysis of principal components (DAPC). Admixture was detected among Karanga, Kunduchi, and Ruhila populations. A cross-validation approach (25% of individual fish from each population was considered of unknown origin) was conducted in order to test the efficiency of the SNP markers to correctly assign individual fish to the population of origin. The cross-validation procedure was repeated 10 times resulting in 77% of the tested individual fish being allocated to the correct population. Overall our results provide a new database of informative SNP markers for both conservation management and aquaculture activities of Nile tilapia strains in Tanzania.

Project description:The genus Aeromonas has been recognised as an important pathogenic species in aquaculture that causes motile Aeromonas septicaemia (MAS) or less severe, chronic infections. This study compares the pathogenicity of the different Aeromonas spp. that were previously isolated from freshwater fish with signs of MAS. A total of 124 isolates of Aeromonas spp. were initially screened for the ability to grow on M9 agar with myo-inositol as a sole carbon source, which is a discriminatory phenotype for the hypervirulent A. hydrophila (vAh) pathotype. Subsequently, LD50 of six selected Aeromonas spp. were determined by intraperitoneal injection of bacterial suspension containing 103, 105, and 107 CFU/mL of the respective Aeromonas sp. to red hybrid tilapias. The kidneys, livers and spleens of infected moribund fish were examined for histopathological changes. The screening revealed that only A. dhakensis 1P11S3 was able to grow using myo-inositol as a sole carbon source, and no vAh strains were identified. The LD50-240h of A. dhakensis 1P11S3 was 107 CFU/mL, while the non-myo-inositol utilizing A. dhakensis 4PS2 and A. hydrophila 8TK3 was lower at 105 CFU/mL. Similarly, tilapia challenged with the myo-inositol A. dhakensis 1P11S3 showed significantly (p < 0.05) less severe signs, gross and histopathological lesions, and a lower mortality rate than the non-myo-inositol A. dhakensis 4PS2 and A. hydrophila 8TK3. These findings suggested that myo-inositol utilizing A. dhakensis 1P11S3 was not a hypervirulent Aeromonas sp. under current experimental disease challenge conditions, and that diverse Aeromonas spp. are of concern in aquaculture farmed freshwater fish. Therefore, future study is warranted on genomic level to further elucidate the influence of myo-inositol utilizing ability on the pathogenesis of Aeromonas spp., since this ability correlates with hypervirulence in A. hydrophila strains.

Project description:We investigated, herein, the effects of dietary inclusion of sugarcane bagasse powder (SB) on Nile tilapia development, mucosal and serum immunities, and relative immune and antioxidant genes. Fish (15.12 ± 0.04 g) were provided a basal diet (SB0) or basal diet incorporated with SB at 10 (SB10), 20 (SB20), 40 (SB40), or 80 (SB80) g kg-1 for 8 weeks. Our results demonstrated that the dietary incorporation of sugarcane bagasse powder (SB) at 20 and 40 g kg-1 significantly ameliorated FW, WG, and SGR as opposed to fish fed basal, SB10, and SB80 diets. However, no significant changes in FCR and survivability were observed between the SB supplemented diets and the control (basal diet). The mucosal immunity exhibited significantly higher SMLA and SMPA activities (p < 0.005) in fish treated with SB diets after eight weeks. The highest SMLA and SMPA levels were recorded in fish fed SB80 followed by SB20, SB40, and SB10, respectively. For serum immunity, fish fed SB incorporated diets significantly ameliorated SL and RB levels (p < 0.05) compared with the control. However, SP was not affected by the inclusion of SB in any diet throughout the experiment. The expression of IL1, IL8, LBP, GSTa, GPX, and GSR genes in the fish liver was significantly increased in fish fed the SB20 and SB10 diets relative to the basal diet fed fish (p < 0.05); whereas only the IL8, LBP, and GPX genes in the intestines were substantially augmented via the SB20 and SB80 diets (p < 0.05). IL1 and GSR were not influenced by the SB incorporated diets (p > 0.05). In summary, sugarcane bagasse powder (SB) may be applied as a feed additive to improve growth performance, immune response, and immune and antioxidant-related gene expression in Nile tilapia.

Project description:Macrobrachium nipponense is one of the commonest species threatened by ambient superfluous nitrite. The mechanism of nitrite stress at the molecular level was studied using de novo RNA-Seq to explore the molecular pathways in M. nipponense exposed to the acute nitrite stress (26.05 mg/L nitrite-N) for 24h and the chronic nitrite stress (6.58 mg/L nitrite-N) for 21d. A total of 175.13 million reads were obtained and assembled into 58,871 unigenes with an average length of 1,028.7 bp and N50 of 1,294bp. 2,824 and 2,610 unigenes in the acute and chronic nitrite stress were significantly differentially expressed respectively. Based on the change in GO analysis and KEGG pathway analysis, pathways both in the acute and chronic nitrite stress were glycosphingolipid biosynthesis - ganglio series, alanine aspartate and glutamate metabolism, biotin metabolism and amino sugar and nucleotide sugar metabolism which revealed the commonly functional pathways in acute and chronic nitrite stress. The markedly altered pathways were divided into four sections of immunity, metabolism, cell and others. Immunity section contained the most pathways among the classifications as phagosome, folate biosynthesis, glycerolipid metabolism, glycine, serine and threonine metabolism, selenoamino acid metabolism, cysteine and methionine metabolism, amino sugar and nucleotide sugar metabolism and taurine and hypotaurine metabolism in the acute nitrite stress and lysosome, alanine, aspartate and glutamate metabolism, arginine and proline metabolism, glycosaminoglycan degradation and amino sugar and nucleotide sugar metabolism in the chronic nitrite stress. This is the first report of whole molecular responses of M. nipponense under acute and chronic nitrite stress through de novo transcriptome sequencing. The findings of this study will further promote the understanding of the underlying molecular mechanisms of the nitrite stress for crustacean species.

Project description:Ammonia oxidation is performed by both ammonia-oxidizing bacteria (AOB) and ammonia-oxidizing archaea (AOA). To explore the effect of ammonia concentration on the population dynamic changes of ammonia-oxidizing microorganisms, we examined changes in the abundance and community composition of AOA and AOB in different layers. Most of the archaeal amoA sequences were Nitrosotalea-related and the proportion that Nitrosotalea cluster occupied decreased in the surface layer and increased in the deep layer during the cultivation process. Nitrosopumilus-related sequences were only detected in the deep layer in the first stage and disappeared later. Both phylogenetic and quantitative analysis showed that there were increased Nitrosomonas-related sequences appeared in the surface layer where the ammonia concentration was the highest. Both AOA and AOB OTU numbers in different layers decreased under selective pressure and then recovered. The potential nitrification rates were 25.06 µg · N · L(-1) · g(-1) dry soil · h(-1) in the mid layer which was higher than the other two layers. In general, obvious population dynamic changes were found for both AOA and AOB under the selective pressure of exogenous ammonia and the changes were different in three layers of the soil column.

Project description:In spring 2019, diseased four-month-old tilapia (Oreochromis spp.) from an aquaculture farm in Southern California, USA were received for diagnostic evaluation with signs of lethargy, anorexia, abnormal swimming, and low-level mortalities. At necropsy, non-specific external lesions were noted including fin erosion, cutaneous melanosis, gill pallor, and coelomic distension. Internal changes included ascites, hepatomegaly, renomegaly, splenomegaly, and multifocal yellow-white nodules in the spleen and kidney. Cultures of spleen and kidney produced bacterial colonies identified as Francisella orientalis. Homogenized samples of gill, brain, liver, spleen, and kidney inoculated onto Mozambique tilapia brain cells (OmB) developed cytopathic effects, characterized by rounding of cells and detaching from the monolayer 6-10 days post-inoculation at 25 °C. Transmission electron microscopy revealed 115.4 ± 5.8 nm icosahedral virions with dense central cores in the cytoplasm of OmB cells. A consensus PCR, targeting the DNA polymerase gene of large double-stranded DNA viruses, performed on cell culture supernatant yielded a sequence consistent with an iridovirus. Phylogenetic analyses based on the concatenated full length major capsid protein and DNA polymerase gene sequences supported the tilapia virus as a novel species within the genus Megalocytivirus, most closely related to scale drop disease virus and European chub iridovirus. An intracoelomic injection challenge in Nile tilapia (O. niloticus) fingerlings resulted in 39% mortality after 16 days. Histopathology revealed necrosis of head kidney and splenic hematopoietic tissues.

Project description:Fingerlings of Oreochromis niloticus collected in an artificial urban lake from Belo Horizonte, Minas Gerais, Brazil, were evaluated for natural infection with trematodes. Morphological taxonomic identification of four fluke species was performed in O. niloticus examined, and the total prevalence of metacercariae was 60.7% (37/61). Centrocestus formosanus, a heterophyid found in the gills, was the species with the highest prevalence and mean intensity of infection (31.1% and 3.42 (1-42), resp.), followed by the diplostomid Austrodiplostomum compactum (29.5% and 1.27 (1-2)) recovered from the eyes. Metacercariae of Drepanocephalus sp. and Ribeiroia sp., both found in the oral cavity of the fish, were verified at low prevalences (8.2% and 1.6%, resp.) and intensities of infection (only one metacercaria of each of these species per fish). These species of trematodes are reported for the first time in O. niloticus from South America. The potential of occurrence of these parasites in tilapia farming and the control strategies are briefly discussed.
