Survey of ex vivo drug combination effects in chronic lymphocytic leukemia reveals synergistic drug effects and genetic dependencies

ABSTRACT: This study investigates the effect of 352 drug combination pairs in multiple concentrations by analysing ex vivo drug response of 52 primary chronic lymphocytic leukemia (CLL) samples. Additional omics profiling of the samples is available from the Bioconductor package BloodCancerMultiOmics2017. The combination screen comprises combinations of 32 library drugs (measured at 5 concentrations) and 11 combination drugs (measured at 1-2 concentrations). Drugs include clinically used drugs and inhibitors of pathways of importance in CLL such as B-cell receptor (BCR) inhibitors, Bcl-2 inhibitors, chemotherapeutic drugs. For selected samples and drug combinations additional measurements in a 10x10 concentration matrix are available.
