LMWP from the larvae of Musca domestica alleviate diarrhea-predominant irritable bowel syndrome by impacting the gut microbiota

ABSTRACT: The Low Molecular Weight Peptides (LMWP) from the larvae of Musca domestica were proven to be effective in treating diarrhea. However, Its effects on Diarrhea-predominant Irritable Bowel Syndrome (D-IBS) and mechanisms are largely unknown. This study aimed to inspect the inhibition of D-IBS in mice by LMWP from the larvae of Musca domestica. The results revealed that LMWP from the larvae of Musca domestica had an excellent therapeutic effect against visceral hypersensitivity, diarrhea and GI dysfunction in D-IBS model mice. Moreover, LMWP from the larvae of Musca domestica significantly reduced the levels of 5-HT in colon tissues or serum. They also altered the levels of the following intestinal microbiota: Akkermansia, Lactobacillus and Acinetobacter. Furthermore, the results showed that the mechanisms involved in this process may have been related to 5-HT, SERT, 5-HT2AR, 5-HT3AR and 5-HT4R. The findings suggested that treatment with LMWP from the larvae of Musca domestica regulates the gut microbiota, thereby providing beneficial effects against diarrhea, high visceral sensitivity and excessive gastrointestinal motility. This also regulates the 5-HT levels in serum and colon as well as the expression of related pathway genes.
