Biogeography of the Sunda shelf revisited: Insights from Macaranga section Pruinosae (Euphorbiaceae)

ABSTRACT: Data derived from GBS of 72 individuals representing six species in Macaranga section Pruinosae was used to determine phylogenetic relationships and discern population structure through Maximum Likelihood (RAxML) and STRUCTURE analyses. Bayesian inference based on secondary calibration points, using BEAST2, and ancestral area reconstruction, on RASP, were performed to infer spatial and temporal origins of the major lineages in the section using the same individuals but without hybrids (62 individuals). Intraspecific STRUCTURE and PCA analyses were performed using GBS data for M. gigantea and M. pruinosa. Nuclear microsatellite data derived from a much larger sample set of the two most widely distributed species, M. gigantea and M. pruinosa (480 and 119 accessions respectively), was used to calculate genetic diversity and discern population differentiation based on STRUCTURE analysis to substantiate results obtained from GBS data.
