Project description:This data article is related to the research article entitled, "Antioxidant potential in non-extractable fraction of dried persimmon (Diospyros kaki Thunb.)" (Matsumura et al., 2016) [1]. We investigated antioxidant activities of the non-extractable fraction of dried persimmon fruits in vitro and in vivo. We evaluated both extracted fraction and non-extractable fraction, and reported that non-extractable fraction may possess significantly antioxidant potential in vivo on the basis of the oxygen radical absorbance capacity (ORAC). We showed our experimental raw data about antioxidant capacity of dried persimmon, plasma triglycerides (TG) and HDL-cholesterol (HDL-C), and this data article might contribute to evaluate real antioxidant capacity of other fruits and vegetables.

Project description:The aim of this study was to determine the antioxidant activity of the extractable and non-extractable phenolics of Terminalia. Sericea Burch. Ex DC. Free, ester bound, ether or glycoside bound and insoluble phenolics were extracted from the fruit, leaves, stem, and root samples. Follin Ciocalteu was used to estimate the phenolic content while DPPH (2,2-diphenyl-1-picrylhydrazyl) assay was used to determine the antioxidant activity. The data obtained were subjected to multivariate analysis for relationships. The result indicated that the highest average total phenolic contents and antioxidant activities were found in the free (14.8 mgGAE/g; IC50 6.8 ?g/mL) and ester bound (15.1 mgGAE/g; IC50 6.4 ?g/mL) extractable phenolics. There was a strong negative correlation between TPC and DPPH (r = -0.828). Agglomerative hierarchical clustering revealed three clusters. Cluster one contained the insoluble and glycoside phenolics while cluster 2 contained only free phenolic acid of the root. The third cluster was predominantly free and ester bound phenolic extracts. The principal component analysis score plot indicated two major clusters with factor 1 (F1) explaining 61% of the variation. The nuclear magnetic resonance spectroscopy spectra indicated that gallic acid and resveratrol are the major phenolic compounds present in the root. This study has demonstrated that extractable phenolics contributed more to the antioxidant activities compared to the non-extractables.

Project description:The antioxidant properties of foods are crucial in nutrition, food chemistry, and medicine studies but are often underestimated, with significant amounts of bioactive compounds containing physiological and biochemical properties remaining in the residue from extraction as non-extractable antioxidants. Over the last decade, extractable and non-extractable compounds have become key in the evaluation/determination of the antioxidant properties of food matrices because of their relevance in human health. This has led to the need to include extractable and non-extractable antioxidants in comprehensive and harmonized food composition databases for a wide range of applications within research, food, pharmaceutical, nutraceutical, and cosmeceutical areas. Additionally, the databases are invaluable as part of the health claims application process. eBASIS, (Bioactive Substances in Food Information System) a comprehensive database containing quality-evaluated scientific data, covering the composition of bioactive compounds present in foods, has flexible structures, allowing it to be extended to include newly emerging data on extractable and non-extractable compounds. Search criteria were developed and defined for compiling suitable peer-reviewed literature. Data quality assessment methods were established for the addition of composition data and antioxidant activity, with a focus on various parameters including: the extraction procedure, the antioxidant measurements, the expression of results. A total of 437 quality-evaluated datapoints on the composition of extractable and/or non-extractable compounds were entered into the database. This database update represents one of the first examples of building a database dedicated to antioxidant properties. This expansion of eBASIS provides a novel and unique tool for nutritionists, dietitians, researchers to use for a wide range of applications, such as dietary assessment, exposure studies and epidemiological studies, and may contribute to an increase in high-bioactive food consumption by consumers.

Project description:1. Retina in vitro showed progressive uptake of non-esterified fatty acid, given in the form of complexes with bovine serum albumin. The rate of influx was related linearly to the concentration of non-esterified fatty acid in the incubation medium and was enhanced by glucose. 2. There was some exchange between medium and tissue non-esterified fatty acid, particularly in the early stages of incubation, and this probably corresponded to the rapid labelling of one component of the tissue non-esterified fatty acid pool. A much slower exchange with tissue non-esterified fatty acid was also evident, not reaching equilibrium even after 7(1/2) hr. incubation. Within the tissue, non-esterified fatty acid also found its way into the fat esters, and about 10% of the uptake was oxidized to carbon dioxide. 3. At high concentrations of non-esterified fatty acid in the medium, disproportionately more of the ingoing non-esterified fatty acid was found in the fat esters of the tissue, whereas the proportion in the tissue non-esterified fatty acid pool or oxidized to carbon dioxide did not change. 4. The role of non-esterified fatty acid as an energy-producing oxidizable substrate is discussed and the regulating influence of glucose on the metabolism of non-esterified fatty acid considered.

Project description:Componential analysis of extractives is important for better understanding the structure and utilization of biomass. In this investigation, wheat straw (WS) was extracted with petroleum ether (PE) and carbon disulfide (CS2) sequentially, to afford extractable fractions EFPE and EFCS2, respectively. Detailed componential analyses of EFPE and EFCS2 were carried out with Fourier transform infrared (FTIR) spectroscopy, gas chromatography/mass spectrometry (GC/MS), X-ray photoelectron spectroscopy (XPS), transmission electron microscopy (TEM), energy dispersive spectrometry (EDS), and electron probe microanalysis (EPMA). Total extractives were quantified 4.96% by weight compared to the initial WS sample. FTIR and GC/MS analyses results showed that PE was effective for the extraction of ketones and waxes derived compounds; meanwhile CS2 preferred ketones and other species with higher degrees of unsaturation. Steroids were enriched into EFPE and EFCS2 with considerable high relative contents, namely, 64.52% and 79.58%, respectively. XPS analysis showed that most of the C atoms in extractives were contained in the structures of C-C, C-COOR, and C-O. TEM-EDS and EPMA analyses were used to detect trace amount elements, such as Al, Si, P, S, Cl, and Ca atoms. Detailed characterization of extractable species from WS can provide more information on elucidation of extractives in biomass.

Project description:1. Mitochondrial and cytoplasmic ribosomes of Euglena gracilis differ in their total RNA and protein content. 2. Mitochondrial ribosomes dissociate to subunits at higher Mg(2+) concentrations than do cytoplasmic ribosomes. 3. A separable 5S RNA is obtained from cytoplasmic and chloroplast ribosomes, but not from mitochondrial ribosomes. 4. For protein-synthesizing activity with a natural mRNA, mitochondrial ribosomes use tRNA from any cell compartment and are partly active with supernatant enzymes from cytoplasm. Cytoplasmic ribosomes are partly active with enzymes and tRNA from mitochondria or chloroplasts. 5. Both mitochondrial and cytoplasmic ribosomes show high specificity for the homologous salt-extractable ribosomal fraction for protein-synthesizing activity.

Project description:Solubilization of phosphatidylinositol (PtdIns) synthase (CDP-diacylglycerol: myo-inositol 3-phosphatidyltransferase, EC 2.7.8.11) from rat pituitary (GH3) tumours was investigated. PtdIns synthase activity was partially extracted from crude membranes by 3 M-KCl. Prior separation of membranes revealed that a greater proportion of plasma-membrane PtdIns synthase activity was salt-extractable than was endoplasmic reticulum activity. The activity of the salt-extracted enzyme was maximized by low concentrations of 3-(3-cholamidopropyl) dimethylammonio-1-propanesulphonate (CHAPS; 0.5 mM), Triton X-100 (0.1 mM) or a phospholipid mixture (0.05 mg/ml), but higher concentrations of detergents were inhibitory. The activity of salt-extracted PtdIns synthase was 0.25 +/- 0.08 nmol/min per mg of protein. Salt-extracted PtdIns synthase activity was dependent on Mg2+ (maximal at 0.1 mM) and Mn2+ (maximal at 5 mM), and its pH optimum was in the range 7.0-7.5. The apparent Km for myo-inositol (in the presence of 0.1 mM-CDP-diacylglycerol) was 0.06 mM, and that for CDP-diacylglycerol (at 0.1 mM-myo-inositol) was 0.21 mM. Salt-extracted PtdIns synthase activity was potently inhibited by Ca2+ (50% inhibition at 1 microM), with over 90% inhibition at 10 microM-Ca2+. These data imply the existence of two forms of membrane-associated PtdIns synthase, namely salt-extractable and salt-resistant, with different intracellular localizations. The salt-extractable form of this enzyme may be a useful preparation for further characterization and purification of mammalian PtdIns synthase.

Project description:1. Two polysaccharides were isolated from the interstitial matrix surrounding the photoreceptor cells of cattle retina. They were liberated from this region of the tissue in a soluble form after agitation of whole retinas in 0.9% sodium chloride. One, which comprises two-thirds of the polysaccharides present, is a hyaluronidase-sensitive ;half-sulphated' chondroitin sulphate containing uronic acid, galactosamine and sulphate in the molar proportions 1.27:1.0:0.54. The other is a hyaluronidase-resistant non-sulphated heteropolysaccharide for which the name sialoglycan is proposed. It contains galactose, glucosamine and sialic acid in the molar proportions 2.4:1.0:0.4. Both polysaccharides contain only small amounts of nitrogen in excess of the amount calculated from their amino sugar and sialic acid content. 2. A similar combination of mucopolysaccharides is associated with the pigment epithelial-cell layer but in quantities only one-fifth of those present in the adjacent matrix area. 3. The ease with which they are released into aqueous media is consistent with the assumption that they are present in the extracellular spaces in both of these tissue layers. 4. The retinal residue left after removal of the two soluble polysaccharides is rich in amino sugar- and sialic acid-containing polymers, which appear to be firmly bound to the tissue fragments. 5. About one-third of the sialic acid and one-tenth of the amino sugar could be extracted with chloroform-methanol. The components in this fraction were tentatively identified as gangliosides. 6. Digestion of the chloroform-methanol-insoluble residue with Pronase yielded as the principal product a heteropolysaccharide containing 16.5% of glucosamine, 24.3% of neutral sugar (galactose plus fucose) and 18.1% of sialic acid. This substance has been classified as a sialoglycan of composition similar to (but not identical with) that of the soluble one isolated from the matrix area of the tissue.
