Project description:As one of the most critical molecular parameters, molecular weight distribution has a profound impact on the structure and properties of polymers. Quantitative and comprehensive understanding, however, has yet to be established, mainly due to the challenge in the precise control and regulation of molecular weight distribution. In this work, we demonstrated a robust and effective approach to artificially engineer the molecular weight distribution through precise recombination of discrete macromolecules. The width, symmetry, and other characteristics of the distribution can be independently manipulated to achieve absolute control, serving as a model platform for highlighting the importance of chain length heterogeneity in structural engineering. Different from their discrete counterparts, each individual component in dispersed samples experiences a varied degree of supercooling at a specific crystallization temperature. Non-uniform crystal nucleation and growth kinetics lead to distinct molecular arrangements. This work could bridge the gap between discrete and dispersed macromolecules, providing fundamental perspectives on the critical role of molecular weight distribution.

Project description:Thyroid cancer is the most prevalent malignancy of the endocrine system. We and others have shown that several microRNAs, which are ~21-24nt post-transcriptional gene regulators, are aberrantly expressed in anaplastic thyroid cancer (ATC) and papillary thyroid cancer (PTC) tissues and cell lines. In the cell, miRNAs are bound to Argonaute (AGO) proteins as low molecular weight RNA Induced Silencing Complexes (LMW-RISCs) that can then assemble into high molecular weight RISCs (HMW-RISCs) that also include additional proteins and mRNA. In this study, we sought to analyze the association of miRNAs across RISC complexity in ATC and PTC. For both ATC and PTC lines, miRNAs were enriched in HMW-RISC and LMW-RISC fractions compared with intermediate fractions or very low molecular weight (AGO-poor) fractions. Furthermore, 60% of all miRNAs were more abundant in LMW-RISC versus HMW- RISC fractions by ~2-4 fold. Surprisingly, miR-21-5p, one of the most abundant miRNAs in both ATC and PTC lines, was consistently one the least abundant miRNAs in HMW-RISC and the most enriched miRNA in LMW-RISC fractions. These findings suggest that miR-21 may be uniquely distributed in RISCs relative to other miRNAs. Furthermore, the methodology described here is a useful way to assess the relative magnitude of miR-21association with HMW and LMW-RISCs and may help to reveal the true roles of this miRNA in thyroid cancer development, progression, and treatment.

Project description:Concentrated solutions of low-molecular-weight DNA (Mw=35000) with a known molecular-weight distribution can be prepared in several hours, and require no additional fractionation procedures. This is achieved by sonication of the DNA in 1.0 M-NaCl at high power at 0--2 degrees C. No denaturation of the DNA is detectable, even after 8h of continuous sonication. After 2h, the molecular-weight distribution of the total DNA sample is that of the most probably Schulz distribution, described by-Mn:-Mw:-Mz ...=1:2:3 ...etc. Such a molecular-weight distribution is expected for a random break-up of indefinitely long macromolecules and indicates that the sonication process is essentially by random double-strand scission. DNA was also sonicated in the presence of ligands capable of modifying the DNA tertiary structure. The results support the idea that inflexibility of the DNA is required for efficient sonic degradation.

Project description:In computational studies of glycosaminoglycans (GAGs), a group of anionic, periodic linear polysaccharides, so far there has been very little discussion about the role of solvent models in the molecular dynamics simulations of these molecules. Predominantly, the TIP3P water model is commonly used as one of the most popular explicit water models in general. However, there are numerous alternative explicit and implicit water models that are neglected in the computational research of GAGs. Since solvent-mediated interactions are particularly important for GAG dynamic and structural properties, it would be of great interest for the GAG community to establish the solvent model that is suited the best in terms of the quality of theoretically obtained GAG parameters and, at the same time, would be reasonably demanding in terms of computational resources required. In this study, heparin (HP) was simulated using five implicit and six explicit solvent models with the aim to find out how different solvent models influence HP's molecular descriptors in the molecular dynamics simulations. Here, we initiate the search for the most appropriate solvent representation for GAG systems and we hope to encourage other groups to contribute to this highly relevant subject.

Project description:The present study aimed to identify the required characteristics of binder pitches in the filler-binder mixing process to effectively manufacture graphite blocks. To this end, a binder pitch was separated into pitch fractions of varying molecular-weight segments. The role and effectiveness of each pitch fraction were then analyzed with respect to their molecular-weight distribution. As a result, the optimal molecular-weight distribution was determined. More specifically, a coal-tar pitch was separated into solvent-soluble and solvent-insoluble fractions. The molecular-weight distribution was determined according to this classification, and the characteristics of each pitch fraction were examined. The pitch separation process was conducted using three solvents: hexane, toluene, and quinoline. The resulting pitch was separated into the following pitch fractions: hexane-soluble (HS), hexane-insoluble-toluene-soluble (HI-TS), toluene-insoluble-quinoline-soluble (TI-QS), and quinoline-insoluble (QI). Fourier transform infrared (FT-IR) spectrum, matrix-assisted laser desorption ionization-time of flight (MALDI-TOF), and softening point of each pitch fraction were measured. Also, pitch samples were refabricated while varying the mixing ratio of these pitch fractions, and carbon blocks were then prepared using them. The compressive strength and porosity of these blocks were measured and compared. The P154_B pitch with a high content of TI-QS was used to fabricate a green block. Due to the high viscosity of the binder used, the fluidity was not sufficiently high, and thus, the green block made of this pitch had relatively low strength. The other blocks had similar levels of strength. After the carbonization process, the carbon block with a high content of HS (P352_B-C) and the carbon block with the HS content removed (P073_B-C) showed lower compressive strength than their respective green-block counterparts (P352_B and P073_B). However, their strength was higher compared to those of the other carbon blocks. In the case of carbon block P073_B-C, the HS content was completely removed, and thus, the content of TI-QS (β-resin) was relatively high. Accordingly, this carbon block ended up with large amounts of components that had high coking values (CVs), and this contributed to limiting the formation of pores. Therefore, the compressive strength of this carbon block was high. In the case of the carbon block with a high content of HS (P352_B-C), a suitable level of viscosity was achieved because the HS components ensured high fluidity. As a result, blocks with higher density and compressive strength could be fabricated. The major findings of the present study confirm that producing carbon blocks with high mechanical properties requires binder pitches with a balanced combination of suitable viscosity to ensure sufficiently high fluidity and a proper level of CV to effectively suppress the formation of pores in the mixing and molding process.

Project description:Heparin, a commonly used anticoagulant drug, is a mixture of highly sulfated polysaccharides with various molecular weights (MWs). The unique sulfation pattern dictates the anticoagulant activity of heparin. Commercial heparins are categorized into three forms according to their average MW: unfractionated heparin (UFH, MWavg 14,000), low-MW heparin (LMWH, MWavg 3500-6500) and the synthetic pentasaccharide (fondaparinux, MW 1508.3). UFH is isolated from porcine intestine while LMWH is derived from UFH by various methods of depolymerization, which generate a wide range of oligosaccharide chain lengths. Different degradation methods result in structurally distinct LMWH products, displaying different pharmacological and pharmacokinetic properties. In this report, we utilized a chemoenzymatic method to synthesize LMWH with the emphasis on controlling the size distribution of the oligosaccharides. A tetrasaccharide primer and a controlled enzyme-based polymerization were employed to build a narrow size oligosaccharide backbone. The oligosaccharide backbones were further modified by a series of sulfation and epimerization steps in order to obtain a full anticoagulation activity. Determination of the anticoagulation activity in vitro and ex vivo indicated that the synthetic LMWH has higher potency than enoxaparin, a commercial LMWH drug in clinical usage.

Project description:Pea protein isolate (PPI, from Pisum sativum L.) was fermented with six different lactic acid bacteria strains for 24 ?h and 48 ?h. The fermented samples were analyzed regarding their retronasal aroma and taste, their protein solubility, emulsifying and foaming capacity. Changes in the molecular weight distribution were analyzed to monitor potential effects of fermentation on the main allergenic protein fractions of PPI. After 24-h fermentation, PPI's characteristic aroma attributes and bitter taste decreased for all fermented PPI. However, after 48-h fermentation, cheesy aroma, and acid and salty tastes were increased. The PPI fermented with L. plantarum showed the most neutral taste and the panel's highest preference; instead, fermentation with L. fermentum led to a fecal aroma and was the least preferred. The protein solubility and emulsifying capacity decreased after PPI fermentation, while foaming capacity remained constant in comparison to the untreated PPI. The electrophoretic results showed a reduction in the intensity of the allergenic protein fractions; however, these changes might be attributed to the reduced protein solubility rather than to a high proteolytic effect of the strains. Fermentation of PPI for 24 ?h and 48 ?h might not be a suitable method for the production of highly functional pea proteins. Further modification methods have to be investigated in the future.

Project description:The breadth and the shape of molecular weight distributions can significantly influence fundamental polymer properties that are critical for various applications. However, current approaches require the extensive synthesis of multiple polymers, are limited in dispersity precision and are typically incapable of simultaneously controlling both the dispersity and the shape of molecular weight distributions. Here we report a simplified approach, whereby on mixing two polymers (one of high Đ and one of low Đ), any intermediate dispersity value can be obtained (e.g. from 1.08 to 1.84). Unrivalled precision is achieved, with dispersity values obtained to even the nearest 0.01 (e.g. 1.37→1.38→1.39→1.40→1.41→1.42→1.43→1.44→1.45), while maintaining fairly monomodal molecular weight distributions. This approach was also employed to control the shape of molecular weight distributions and to obtain diblock copolymers with high dispersity accuracy. The straightforward nature of our methodology alongside its compatibility with a wide range of polymerisation protocols (e.g. ATRP, RAFT), significantly expands the toolbox of tailored polymeric materials and makes them accessible to all researchers.

Project description:We characterized the molecular weight distribution of polymeric proteins (PP) of bread wheat grains using asymmetric flow field flow fractionation (A4F). The experiment, involving six environmental conditions and 130 cultivars, offered the opportunity to approach the phenotypic values of the polymer characteristics and their contribution of the rheological properties of flours and/or doughs. The contents of high-molecular-weight polymers (MW > 2 × 106 g·mol-1) that can be considered as "rheologically active polymers" (RAPP) for their major contribution to dough baking strength and mixing tolerance were mainly controlled by environmental factors. Under the influence of the growing conditions, at the cellular level, the redox status of non-protein free thiol, such as glutathione, is modified and leads to the formation of polymeric protein-bound glutathione conjugates (PPSSG). The accumulation of these conjugates reduces the formation of the RAPP by limiting the intermolecular interactions between PP in the grain during desiccation. This phenomenon is, therefore, potentially responsible for decreases in the technological properties of the wheat genotypes concerned. These first results invite us to continue our investigations to fully confirm this phenomenon, with emphasis on the behavior of wheat genotypes under various growing conditions.
