Differential effects of temperature on a membrane adenosine triphosphatase and associated phosphatase.

ABSTRACT: Arrhenius plots of a membrane (Na+ + K+)-dependent ATPase (adenosine triphosphatase) activity showed characteristic discontinuities, whereas those of the associated K+-dependent phosphatase activity did not. These findings support the contention that the phosphatase activity does not depend on phospholipid in the same way as does the ATPase activity.

Differential effects of lipid depletion on membrane sodium-plus-potassium ion-dependent adenosine triphosphatase and potassium ion-dependent phosphatase.

Project description:The phospholipid-dependence of the (Na-++K-+)-dependent ATPase (adenosine triphosphatase) (EC 3.6.1.3) and associated K-+-dependent phosphatase activity (EC 3.6.1.7) have been compared. Unlike the (Na-++K-+)-dependent ATPase activities, the K-+-dependent phosphatase activities of a number of different preparations were not closely correlated with their total phospholipid contents. After partial lipid depletion with a single extraction in Lubrol W the residual ATPase and phosphatase activities were correlated, but their magnitudes were quite different: on average only about 5% of the former remained compared with 50% of the latter. A similar differential effect on these activities was found after extraction with deoxycholate. In contrast with the ATPase, consistent restoration of the phosphatase activity of Lubrol-extracted enzymes by added exogenous phospholipids was not observed. We conclude that, although the K-+-dependent phosphatase may be lipid-dependent, the lipid requirement must be different from that of the complete ATPase system, and this difference should help investigations of their relationship.

The effects of environmental temperature on the properties of myofibrillar adenosine triphosphatase from various species of fish.

Project description:1. Myofibrillar adenosine triphosphatase (ATPase) activities were measured for white myotomal muscle of 19 species of fish. 2. The activity was measured at different temperatures and after periods of preincubation at 37 degrees C. 3. The inactivation half-life at 37 degrees C depended on environmental temperature, increasing as the temperature increased. 4. Cold-water fish had higher myofibrillar adenosine triphosphatase activity at low temperatures than had warm-water fish. 5. The significance of these results is discussed.

The mother-cell-membrane adenosine triphosphatase of sporulating Clostridium pasteurianum.

Project description:1. Sporulation of Clostridium pasteurianum effects several changes in its proton-translocating cell-membrane H(+)-ATPase. Notable among these are the acquisition of susceptibility to activation by trypsin and a changed protein subunit composition. 2. A protein was isolated from the mother-cell membrane that inhibited the ATP phosphohydrolase activity of purified vegetative-cell-membrane H(+)-ATPase [BF(0)F(1) complex, which consists of soluble ATPase (BF(1)) and the proton-channel component (BF(0))] and rendered it susceptible to trypsin activation. 3. This trypsin-sensitive inhibitor protein had a molecular weight of 10000 and on sodium dodecyl sulphate/polyacrylamide-gel electrophoresis was indistinguishable from the novel protein subunit e of the mother-cell-membrane ATPase 4. In bacteriorhodopsin-containing everted membrane vesicles, the specific ATP synthetase activity of the mother-cell-membrane ATPase was significantly greater than that of the vegetative-cell-membrane ATPase. 5. Treatment with trypsin-sensitive inhibitor protein of artificial proteoliposomes containing bacteriorhodopsin and vegetative-cell-membrane H(+)-ATPase (BF(0)F(1)) significantly increased the specific ATP synthetase activity of this enzyme. 6. The ATP synthetase activity of crude cell-membrane preparations from cultures of Clostridium pasteurianum increased during that period in the course of sporulation when the membrane ATP phosphohydrolase was both most rapidly decreasing in specific activity and acquiring its susceptibility to activation by trypsin.

Influence of temperature acclimatization on the temperature-dependence and ouabain-sensitizing of goldfish intestinal adenosine triphosphatase.

Project description:1. Homogenates of goldfish intestinal mucosa were separated into various fractions by differential centrifugation. Both adenosine-triphosphatase and beta-glycerophosphatase activities were found to be concentrated mainly in a membrane fraction which sedimented after 1200000g-min. 2. This membrane adenosine-triphosphatase system was activated by Na(+)+K(+) and inhibited by ouabain. 3. The ouabain-sensitive adenosine-triphosphatase activity was high and the ouabain-insensitive activity low in membrane fractions prepared from fish acclimatized previously to 8 degrees . The opposite was true for fish acclimatized to 30 degrees . 4. The Arrhenius plots of ouabain-sensitive and ouabain-insensitive adenosine-triphosphatase activities, measured from 5 degrees to 30 degrees , showed discontinuities at incubation temperatures that varied with the previous acclimatization temperature of the fish. 5. It is considered that modification of the membrane adenosine-triphosphatase system in goldfish intestinal mucosa may serve to regulate Na(+) transport at different environmental temperatures.

Influence of temperature acclimatization on the ionic activation of goldfish intestinal adenosine triphosphatase.

Project description:1. An adenosine triphosphatase membrane system, dependent on Mg(2+) and activated further by Na(+)+K(+), was prepared from goldfish anterior intestine by differential centrifugation of homogenized intestinal scrapings. 2. The affinity of this preparation for Na(+) in the presence of K(+)+Mg(2+), for K(+) in the presence of Na(+)+Mg(2+) and for Mg(2+) alone, measured at 37 degrees , did not depend on the previous environmental temperature of the fish. When Na(+)+K(+) were added to preparations from 8 degrees -acclimatized fish the affinity for Mg(2+) increased; this was not seen with preparations from 30 degrees -acclimatized fish. 3. Part of the Mg(2+)-activated adenosine triphosphatase was inhibited by Na(+) and the amount of inhibition appeared to increase at high acclimatization temperatures. 4. This Na(+)-inhibited adenosine triphosphatase was separated from the (Na(+)+K(+))-activated enzyme by centrifugation on sucrose density gradients. 5. Preparations from 8 degrees -acclimatized fish contained less Mg(2+)-activated and more (Na(+)+K(+))-activated adenosine triphosphatase than did similar fractions from 30 degrees -acclimatized fish. 6. Acclimatization to different environmental temperatures might involve one form of adenosine triphosphatase changing to another. The origin of various membranes seen in microsomal fractions must, however, be established before this hypothesis can be tested further.

The mode of inhibition by calcium of cell-membrane adenosine-triphosphatase activity.

Project description:1. The mechanism of the inhibition of Na(+)-plus-K(+)-activated adenosine triphosphatase by calcium was investigated with an enzyme preparation from rabbit kidney cortex and with membranes of human erythrocytes. 2. CaATP, rather than ionic Ca(2+), acts as a competitive inhibitor, competing with MgATP in the Na(+)-plus-K(+)-activated adenosine-triphosphatase reaction. 3. There appears to be no competition between calcium and Na(+) for the activation of adenosine triphosphatase. 4. The inhibition of Na(+)-plus-K(+)-activated adenosine triphosphatase of cell membranes by low concentrations of CaATP and the consequent need of intact cells to keep the cytoplasmic concentration of calcium low relative to that of magnesium suggests a raison d'être for the mitochondrial calcium pump.

Effects of triphenylsulphonium ions on mitochondria. Inhibition of adenosine triphosphatase activity.

Project description:Triphenylsulphonium ions inhibit mitochondrial oxidative phosphorylation and adenosine triphosphatase activity. The site of action is on the soluble F1 adenosine triphosphatase component. Triphenylsylphonium ions also inhibit electron transfer in the NAD-cytochrome b region of the respiratory chain. In both types of inhibition, triphenylsulphonium ions are effective at low concentrations, half-maximal inhibition being produced by a concentration of about 20-30 muM. These effects resemble the effects of alkylguanidines on mitochondria and are discussed in relation to the effects of alkylguanidines and other lipophilic cations such as ethidium and dibenzyldimethylammonium ions. A modification of the purification procedure for the soluble mitochondrial adenosine triphosphatase [Beechey, Hubbard, Linnett, Mitchell & Munn (1975) Biochem. Synonymous mutation in adenosine triphosphatase copper-transporting beta causes enhanced exon skipping in Wilson disease.

Project description:Wilson disease (WD) is caused by biallelic pathogenic variants in adenosine triphosphatase copper-transporting beta (ATP7B); however, genetic testing identifies only one or no pathogenic ATP7B variant in a number of patients with WD. Synonymous single-nucleotide sequence variants have been recognized as pathogenic in individual families. The aim of the present study was to evaluate the prevalence and disease mechanism of the synonymous variant c.2292C>T (p.Phe764=) in WD. A cohort of 280 patients with WD heterozygous for a single ATP7B variant was investigated for the presence of c.2292C>T (p.Phe764=). In this cohort of otherwise genetically unexplained WD, the allele frequency of c.2292C>T (p.Phe764=) was 2.5% (14 of 560) compared to 7.1 × 10-6 in the general population (2 of 280,964 in the Genome Aggregation Database; p < 10-5 ; Fisher exact test). In an independent United Kingdom (UK) cohort, 2 patients with WD homozygous for p.Phe764= were identified. RNA analysis of ATP7B transcripts from patients homozygous or heterozygous for c.2292C>T and control fibroblasts showed that this variant caused high expression of an ATP7B transcript variant lacking exon 8. Conclusion: The synonymous ATP7B variant c.2292C>T (p.Phe764=) causes abnormal messenger RNA processing of ATP7B transcripts and is associated with WD in compound heterozygotes and homozygotes.
