Project description:Rice husk is a base adsorbent for pollutant removal. It is a cost-effective material and a renewable resource. This study provides the physicochemical characterization of chemically and thermally treated rice husk adsorbents for phenol removal from aqueous solutions. We revealed new functional groups on rice husk adsorbents by Fourier transform infrared spectroscopy, and observed major changes in the pore structure (from macro-mesopores to micro-mesopores) of the developed rice husk adsorbents using scanning electron microscopy. Additionally, we studied their surface area and pore size distribution, and found a greater enhancement of the morphological structure of the thermally treated rice husk compared with that chemically treated. Thermally treated adsorbents presented a higher surface area (24-201 m2.g-1) than those chemically treated (3.2 m2.g-1). The thermal and chemical modifications of rice husk resulted in phenol removal efficiencies of 36%-64% and 28%, respectively. Thus, we recommend using thermally treated rice husk as a promising adsorbent for phenol removal from aqueous solutions.

Project description:We report increased stimulation of dendritic cells via heterodimers of immunostimulants formed at a discrete molecular distance. Many vaccines present spatially organized agonists to immune cell receptors. These receptors cluster suggesting that signaling is increased by spatial organization and receptor proximity, but this has not been directly tested for multiple, unique receptors. In this study we probe the spatial aspect of immune cell activation using heterodimers of two covalently attached immunostimulants.

Project description:Engineering antibodies to utilize non-canonical amino acids (NCAA) should greatly expand the utility of an already important biological reagent. In particular, introducing crosslinking reagents into antibody complementarity determining regions (CDRs) should provide a means to covalently crosslink residues at the antibody-antigen interface. Unfortunately, finding the optimum position for crosslinking two proteins is often a matter of iterative guessing, even when the interface is known in atomic detail. Computer-aided antibody design can potentially greatly restrict the number of variants that must be explored in order to identify successful crosslinking sites. We have therefore used Rosetta to guide the introduction of an oxidizable crosslinking NCAA, l-3,4-dihydroxyphenylalanine (l-DOPA), into the CDRs of the anti-protective antigen scFv antibody M18, and have measured crosslinking to its cognate antigen, domain 4 of the anthrax protective antigen. Computed crosslinking distance, solvent accessibility, and interface energetics were three factors considered that could impact the efficiency of l-DOPA-mediated crosslinking. In the end, 10 variants were synthesized, and crosslinking efficiencies were generally 10% or higher, with the best variant crosslinking to 52% of the available antigen. The results suggest that computational analysis can be used in a pipeline for engineering crosslinking antibodies. The rules learned from l-DOPA crosslinking of antibodies may also be generalizable to the formation of other crosslinked interfaces and complexes.

Project description:In the present study, four biopolymer-based materials consisting of native corn starch (CS), phosphate corn starch (PS), starch nanocrystals (SNCs), and phosphate corn starch nanocrystals (PSNCs) were synthesized and used for methylene blue (MB) removal as a function of various parameters, including initial MB concentration (C0, 10-500 mg L-1), adsorbent dosage (Cs, 0.02-0.15 g), contact time (t, 5-15 min), solution pH (2-11), and temperature (25-45 °C). The removal percentage of MB increased dramatically upon increasing the biopolymer dosage, temperature, and pH; while it decreased upon increasing the initial MB concentration. The adsorption behavior of biopolymer-based materials towards MB was found to be accurately described by the pseudo-second-order kinetic and Langmuir isotherm models. According to the Langmuir model, the maximum adsorption capacities of the adsorbents were ordered as follows: PSNCs (88.53 mg g-1) > SNCs (79.55 mg g-1) > PS (73.17 mg g-1) > CS (63.02 mg g-1). PSNCs was able to remove 96.8% and 76.5% of 20 mg L-1 MB in greywater and petrochemical wastewater, respectively, at an optimum pH of 9 and retained 86.42% of its usability even after five adsorption-desorption cycles. The analysis of the surface charge of the adsorbents before and after MB adsorption, combined with the FTIR spectrum of MB-saturated biopolymer-based materials, provided evidence that electrostatic interactions was the primary mechanism involved in the adsorption of MB. Meanwhile, hydrogen bonding and π-π interactions were found to have a minor contribution to the adsorption process. Based on the results, it can be inferred that PSNCs has promising potential as an adsorbent for the treatment of MB-containing wastewater, owing to its exceptional properties, which include high adsorption capacity, low cost, and applicability for multiple reuses.

Project description:Nitric oxide (NO) is an important molecule that exerts multiple functions in biological systems. Because of the short-lived nature of NO, S-nitrosothiols (RSNOs) are believed to act as stable NO carriers. Recently, sulfhydryl (SH) containing macromolecules have been shown to be promising NO carriers. In the present study, we aimed to synthesize and characterize a potential NO carrier based on bovine Cu,Zn-superoxide dismutase (bSOD). To prepare S-nitrosated bSOD, the protein was incubated with S-nitrosoglutathione (GSNO) under varied experimental conditions. The results show that significant S-nitrosation of bSOD occurred only at high temperature (50 °C) for prolonged incubation time (>2 h) S-nitrosation efficiency increased with reaction time and reached a plateau at ~4 h. The maximum amount of NO loaded was determined to be about 0.6 mol SNO/mol protein (~30% loading efficiency). The enzymatic activity of bSOD, however, decreased with reaction time. Our data further indicate that NO functionality can only be measured in the presence of extremely high concentrations of Hg2+ or when the protein was denatured by guanidine. Moreover, mildly acidic pH was shown to favor S-nitrosation of bSOD. A model based on unfolding and refolding of bSOD during preparation was proposed to possibly explain our observation.

Project description:We have cloned and functionally characterized a Na(+)-coupled dicarboxylate transporter, SdcS, from Staphylococcus aureus. This carrier protein is a member of the divalent anion/Na(+) symporter (DASS) family and shares significant sequence homology with the mammalian Na(+)/dicarboxylate cotransporters NaDC-1 and NaDC-3. Analysis of SdcS function indicates transport properties consistent with those of its eukaryotic counterparts. Thus, SdcS facilitates the transport of the dicarboxylates fumarate, malate, and succinate across the cytoplasmic membrane in a Na(+)-dependent manner. Furthermore, kinetic work predicts an ordered reaction sequence with Na(+) (K(0.5) of 2.7 mM) binding before dicarboxylate (K(m) of 4.5 microM). Because this transporter and its mammalian homologs are functionally similar, we suggest that SdcS may serve as a useful model for DASS family structural analysis.

Project description:Therapeutic gene editing is becoming more feasible with the emergence of the Clustered Regularly Interspaced Short Palindromic Repeats (CRISPR)/CRISPR-associated protein (Cas) system. However, the successful implementation of CRISPR/Cas9-based therapeutics requires a safe and efficient in vivo delivery of the CRISPR components, which remains challenging. This study presents successful preparation, optimization, and characterization of alginate nanoparticles (ALG NPs), loaded with two CRISPR plasmids, using electrospray technique. The aim of this delivery system is to edit a target gene in another plasmid (green fluorescent protein (GFP)). The effect of formulation and process variables were evaluated. CRISPR ALG NPs showed mean size and zeta potential of 228 nm and -4.42 mV, respectively. Over 99.0% encapsulation efficiency was achieved while preserving payload integrity. The presence of CRISPR plasmids in the ALG NPs was confirmed by Attenuated Total Reflectance-Fourier Transform Infrared Spectroscopy. The tests revealed that the nanoparticles were cytocompatible and successfully introduced the Cas9 transgene in HepG2 cells. Nanoparticles-transfected HepG2 was able to edit its target plasmid by introducing double-strand break (DSB) in GFP gene, indicating the bioactivity of CRISPR plasmids encapsulated in alginate nanoparticles. This suggests that this method is suitable for biomedical application in vitro or ex vivo. Future investigation of theses nanoparticles might result in nanocarrier suitable for in vivo delivery of CRISPR/Cas9 system.

Project description:Validated and easily applicable analytical tools are required to develop and implement regulatory frameworks and an appropriate risk assessment for engineered nanoparticles (ENPs). Concerning metal-based ENPs, two main aspects are the quantification of the absolute mass concentration and of the "dissolved" fraction in, e.g., (eco)toxicity and environmental studies. To provide information on preparative aspects and on potential uncertainties, preferably simple off-line methods were compared to determine (1) the total concentration of suspensions of five metal-based ENP materials (Ag, TiO2, CeO2, ZnO, and Au; two sizes), and (2) six methods to quantify the "dissolved" fraction of an Ag ENP suspension. Focusing on inductively coupled plasma-mass spectrometry, the total concentration of the ENP suspensions was determined by direct measurement, after acidification and after microwave-assisted digestion. Except for Au 10 nm, the total concentrations determined by direct measurements were clearly lower than those measured after digestion (between 61.1 % for Au 200 nm and 93.7 % for ZnO). In general, acidified suspensions delivered better recoveries from 89.3 % (ZnO) to 99.3 % (Ag). For the quantification of dissolved fractions two filtration methods (ultrafiltration and tangential flow filtration), centrifugation and ion selective electrode were mainly appropriate with certain limitations, while dialysis and cloud point extraction cannot be recommended. With respect to precision, time consumption, applicability, as well as to economic demands, ultrafiltration in combination with microwave digestion was identified as best practice.

Project description:The supply of inorganic carbon (Ci) at the site of fixation by Rubisco is a key parameter for efficient CO2 fixation in aquatic organisms including the green alga, Chlamydomonas reinhardtii. Chlamydomonas reinhardtii cells, when grown on limiting CO2, have a CO2-concentrating mechanism (CCM) that functions to concentrate CO2 at the site of Rubisco. Proteins thought to be involved in inorganic carbon uptake have been identified and localized to the plasma membrane or chloroplast envelope. However, current CCM models suggest that additional molecular components are involved in Ci uptake. In this study, the gene Cia8 was identified in an insertional mutagenesis screen and characterized. The protein encoded by Cia8 belongs to the sodium bile acid symporter subfamily. Transcript levels for this gene were significantly up-regulated when the cells were grown on low CO2. The cia8 mutant exhibited reduced growth and reduced affinity for Ci when grown in limiting CO2 conditions. Prediction programs localize this protein to the chloroplast. Ci uptake and the photosynthetic rate, particularly at high external pH, were reduced in the mutant. The results are consistent with the model that CIA8 is involved in Ci uptake in C. reinhardtii.

Project description:The literature shows that inorganic phase change materials (PCM) have been very seldom microencapsulated, so this study aims to contribute to filling this research gap. Bischofite, a by-product from the non-metallic industry identified as having good potential to be used as inorganic PCM, was microencapsulated by means of a fluidized bed method with acrylic as polymer and chloroform as solvent, after compatibility studies of both several solvents and several polymers. The formation of bischofite and pure MgCl₂·6H₂O microcapsules was investigated and analyzed. Results showed an efficiency in microencapsulation of 95% could be achieved when using 2 min of fluidization time and 2 kg/h of atomization flow. The final microcapsules had excellent melting temperatures and enthalpy compared to the original PCM, 104.6 °C and 95 J/g for bischofite, and 95.3 and 118.3 for MgCl₂·6H₂O.
