Project description:1. The syntheses of ubiquinone-9 and plastoquinone-9 were used as parameters respectively of mitochondrial and proplastid development in the germinating wheat grain. 2. The changes in the amounts of the tocopherols were also studied and the possible biological significance of these changes is discussed. During germination, the dimethyl tocopherols of the resting grain are probably not utilized for the synthesis of alpha-tocopherol. 3. It was demonstrated that ubiquinone synthesis, and hence probably mitochondrial development, in the aleurone cells during germination, is independent of control by gibberellic acid from the embryo. 4. The influence of light on the syntheses of the isoprenoid quinones in the etiolated wheat shoot was investigated. In particular, illumination did not stimulate the synthesis of either alpha-tocopherol or alpha-tocopherolquinone.

Project description:Background and aimsGluten proteins are the major storage protein fraction in the mature wheat grain. They are restricted to the starchy endosperm, which forms white flour on milling, and interact during grain development to form large polymers which form a continuous proteinaceous network when flour is mixed with water to give dough. This network confers viscosity and elasticity to the dough, enabling the production of leavened products. The starchy endosperm is not a homogeneous tissue and quantitative and qualitative gradients exist for the major components: protein, starch and cell wall polysaccharides. Gradients in protein content and composition are the most evident and are of particular interest because of the major role played by the gluten proteins in determining grain processing quality.MethodsProtein gradients in the starchy endosperm were investigated using antibodies for specific gluten protein types for immunolocalization in developing grains and for western blot analysis of protein extracts from flour fractions obtained by sequential abrasion (pearling) to prepare tissue layers.Key resultsDifferential patterns of distribution were found for the high-molecular-weight subunits of glutenin (HMW-GS) and γ-gliadins when compared with the low-molecular-weight subunits of glutenin (LMW-GS), ω- and α-gliadins. The first two types of gluten protein are more abundant in the inner endosperm layers and the latter more abundant in the subaleurone. Immunolocalization also showed that segregation of gluten proteins occurs both between and within protein bodies during protein deposition and may still be retained in the mature grain.ConclusionsQuantitative and qualitative gradients in gluten protein composition are established during grain development. These gradients may be due to the origin of subaleurone cells, which unlike other starchy endosperm cells derive from the re-differentiation of aleurone cells, but could also result from the action of specific regulatory signals produced by the maternal tissue on specific domains of the gluten protein gene promoters.

Project description:Elemental selenium is one of the dominant selenium species in soil, but the mechanism of its uptake by plants is still unclear. In this study, nanoparticles of elemental selenium (SeNPs) with different sizes were prepared, and their uptake and transformation in wheat (Triticum aestivum L.) were analyzed in hydroponic experiments by HPLC-ICP-MS. We found that the SeNPs can be absorbed by wheat seedlings, and the process is energy independent. The addition of aquaporins inhibitor caused 92.5 and 93.4% inhibition of chemosynthesized SeNPs (CheSeNPs) and biosynthesized SeNPs (BioSeNPs) absorption by wheat roots, respectively. The 40 nm SeNPs uptake by wheat roots was 1.8-fold and 2.2-fold higher than that of 140 and 240 nm, respectively. The rate of SeNPs uptake in wheat was much slower than that of selenite [Se (IV)], and CheSeNPs were more efficiently absorbed than BioSeNPs. The SeNPs were rapidly oxidized to Se (IV) and converted to organic forms [selenocystine (SeCys2), se-methyl-selenocysteine (MeSeCys), and selenomethionine (SeMet)] after they were absorbed by wheat roots. Additionally, we demonstrated that the aquaporin function in some way is related to the absorption of SeNPs. The particle size and synthesis method of the SeNPs affected their uptake rates by plants. Taken together, our results provide a deep understanding of the SeNPs uptake mechanism in plants.

Project description:Wheat grain end-use value is determined by complex molecular interactions that occur during grain development, including those in the cell nucleus. However, our knowledge of how the nuclear proteome changes during grain development is limited. Here, we analyzed nuclear proteins of developing wheat grains collected during the cellularization, effective grain-filling, and maturation phases of development, respectively. Nuclear proteins were extracted and separated by two-dimensional gel electrophoresis. Image analysis revealed 371 and 299 reproducible spots in gels with first dimension separation along pH 4-7 and pH 6-11 isoelectric gradients, respectively. The relative abundance of 464 (67%) protein spots changed during grain development. Abundance profiles of these proteins clustered in six groups associated with the major phases and phase transitions of grain development. Using nano liquid chromatography-tandem mass spectrometry to analyse 387 variant and non-variant protein spots, 114 different proteins were identified that were classified into 16 functional classes. We noted that some proteins involved in the regulation of transcription, like HMG1/2-like protein and histone deacetylase HDAC2, were most abundant before the phase transition from cellularization to grain-filling, suggesting that major transcriptional changes occur during this key developmental phase. The maturation period was characterized by high relative abundance of proteins involved in ribosome biogenesis. Data are available via ProteomeXchange with identifier PXD002999.

Project description:BackgroundHigh post-anthesis (p.a) temperatures reduce mature grain weights in wheat and other cereals. However, the causes of this reduction are not entirely known. Control of grain expansion by the maternally derived pericarp of the grain has previously been suggested, although this interaction has not been investigated under high p.a. temperatures. Down-regulation of pericarp localised genes that regulate cell wall expansion under high p.a. temperatures may limit expansion of the encapsulated endosperm due to a loss of plasticity in the pericarp, reducing mature grain weight. Here the effect of high p.a. temperatures on the transcriptome of the pericarp and endosperm of the wheat grain during early grain-filling was investigated via RNA-Seq and is discussed alongside grain moisture dynamics during early grain development and mature grain weight.ResultsHigh p.a. temperatures applied from 6-days after anthesis (daa) and until 18daa reduced the grain's ability to accumulate water, with total grain moisture and percentage grain moisture content being significantly reduced from 14daa onwards. Mature grain weight was also significantly reduced by the same high p.a. temperatures applied from 6daa for 4-days or more, in a separate experiment. Comparison of our RNA-Seq data from whole grains, with existing data sets from isolated pericarp and endosperm tissues enabled the identification of subsets of genes whose expression was significantly affected by high p.a. temperature and predominantly expressed in either tissue. Hierarchical clustering and gene ontology analysis resulted in the identification of a number of genes implicated in the regulation of cell wall expansion, predominantly expressed in the pericarp and significantly down-regulated under high p.a. temperatures, including endoglucanase, xyloglucan endotransglycosylases and a β-expansin. An over-representation of genes involved in the 'cuticle development' functional pathway that were expressed in the pericarp and affected by high p.a. temperatures was also observed.ConclusionsHigh p.a. temperature induced down-regulation of genes involved in regulating pericarp cell wall expansion. This concomitant down-regulation with a reduction in total grain moisture content and grain weight following the same treatment period, adds support to the theory that high p.a. temperatures may cause a reduction in mature grain weight as result of decreased pericarp cell wall expansion.

Project description:Grain weight is one of the most important components of cereal yield and quality. A clearer understanding of the physiological and molecular determinants of this complex trait would provide an insight into the potential benefits for plant breeding. In the present study, the dynamics of dry matter accumulation, water uptake, and grain size in parallel with the expression of expansins during grain growth in wheat were analysed. The stabilized water content of grains showed a strong association with final grain weight (r(2)=0.88, P <0.01). Grain length was found to be the trait that best correlated with final grain weight (r(2)=0.98, P <0.01) and volume (r(2)=0.94, P <0.01). The main events that defined final grain weight occurred during the first third of grain-filling when maternal tissues (the pericarp of grains) undergo considerable expansion. Eight expansin coding sequences were isolated from pericarp RNA and the temporal profiles of accumulation of these transcripts were monitored. Sequences showing high homology with TaExpA6 were notably abundant during early grain expansion and declined as maturity was reached. RNA in situ hybridization studies revealed that the transcript for TaExpA6 was principally found in the pericarp during early growth in grain development and, subsequently, in both the endosperm and pericarp. The signal in these images is likely to be the sum of the transcript levels of all three sequences with high similarity to the TaExpA6 gene. The early part of the expression profile of this putative expansin gene correlates well with the critical periods of early grain expansion, suggesting it as a possible factor in the final determination of grain size.

Project description:The use of cereal microgreens is increasing because of increased consumer's interest in healthier products. Chlorophyll (Chl) and Carotenoids (Car) are suggested to correlate with health promoting components like phenolics and antioxidant potential of the plant-part. They also play role against clinical conditions like thalassemia and hemolytic anemia and reduce the risk of some chronic diseases, such as cancer, cardiovascular diseases, skin diseases and age-related eye diseases. This study was carried out for the comprehensive profiling of Chl and Car in wheat (Triticum aestivum L.), and barley (Hordeum vulgare L.) micro-greens between 7 and 16 days on dry basis. Chl and Car content strongly correlated with the number of days of growth. Significantly high correlations existed among Chl a, Chl b, total Chl and total Car with concomitant Chl a/b and Chl/Car ratios. The peaks for the rate of accumulation of pigments were between 7-10 days on wheat and 10-13 days on barley. The maximum content of Chl and Car were 616.63?±?18.45 mg/100 g DM and 54.80?±?1.72 mg/100 g DM on day 16 and statistically not significant to variety of grain. The Chl level was slightly lower than Chl rich vegetables like kale and comparable to spinach and Car level was comparable to carrots, which is higher than most of the daily consumable fruits and vegetables. Further cell-based or in vivo studies of cereal microgreens could be considered to draw more valuable information related to human health.

Project description:Cellular protein abundance results from the relative rates of protein synthesis and protein degradation. Through combining in vivo stable isotope labelling and in-depth quantitative proteomics, we created a protein turnover atlas of wheat grain proteins during grain development. Our data demonstrate that protein turnover rates for 1447 unique wheat grain protein groups have an apparent spatiotemporal pattern that aids explanation of the 60% of variation in protein abundances that are not attributable to gene expression. Protein synthesis rates of individual proteins vary over 100 fold and degradation rates over 20 fold. Storage proteins have both higher synthesis and degradation rates than the overarching average rates of grain proteins in other functional categories, while those proteins involved in photosynthesis, DNA synthesis and glycolysis, by contrast, are house-keeping proteins that show low synthesis and degradation rates at all times. Approximately 20% of total grain ATP production through respiration is used for grain proteome biogenesis and maintenance, and the grain invests nearly half of this budget in storage protein synthesis alone. Degradation of storage proteins as a class of grain proteins also consumed a significant amount of the total ATP allocated to protein degradation processes. This analysis suggests that 20% of newly synthesized storage proteins are turned over rather than stored suggesting that this process is not energetically optimal. This approach to measure protein turnover rates at the proteome scale shows how different functional categories of grain proteins accumulate, calculates the costs of futile cycling of protein turnover during wheat grain development and identifies the most and the least stable wheat grain proteins.
