Project description:Polycystic kidney disease (PKD), a monogenic disease with an autosomal dominant or an autosomal recessive form of inheritance (ARPKD), is the most common genetic cause of renal dysfunction and end-stage renal failure. In addition to the development of cysts, the autosomal form of PKD is associated with vascular endothelial dysfunction, a marker of vascular disease. Whether vascular endothelial dysfunction is also present in ARPKD, and its relationship with renal dysfunction remain to be determined.ARPKD rats (PCK model) and controls were studied at 6 and 10 weeks of age, and mean arterial pressure and renal function were measured. Aortic endothelial function was assessed using organ chamber techniques. Aortic endothelial cells (ECs) were isolated, characterized and their function studied.Compared to controls, ARPKD animals had a decrease in the vasorelaxation to endothelium-dependent vasodilators, even prior to changes in mean arterial pressure or renal function. The abnormal vasoreactivity was corrected with L-arginine (a precursor of nitric oxide, NO), while the expression of endothelial NO synthase (eNOS) was unchanged. Furthermore, isolated ECs from 6-week-old ARPKD animals showed increased oxidative stress, with preserved eNOS expression and abnormal patterns of migration and angiogenic capacity (measured by the scratch and tube formation assays, respectively).ARPKD leads to impairments in aortic vascular function and ECs at an early stage, which can have significant functional consequences, potentially representing a novel therapeutic target in this disease.

Project description:Events and objects in the world must be inferred from sensory signals to support behavior. Because sensory measurements are temporally and spatially local, the estimation of an object or event can be viewed as the grouping of these measurements into representations of their common causes. Perceptual grouping is believed to reflect internalized regularities of the natural environment, yet grouping cues have traditionally been identified using informal observation and investigated using artificial stimuli. The relationship of grouping to natural signal statistics has thus remained unclear, and additional or alternative cues remain possible. Here, we develop a general methodology for relating grouping to natural sensory signals and apply it to derive auditory grouping cues from natural sounds. We first learned local spectrotemporal features from natural sounds and measured their co-occurrence statistics. We then learned a small set of stimulus properties that could predict the measured feature co-occurrences. The resulting cues included established grouping cues, such as harmonic frequency relationships and temporal coincidence, but also revealed previously unappreciated grouping principles. Human perceptual grouping was predicted by natural feature co-occurrence, with humans relying on the derived grouping cues in proportion to their informativity about co-occurrence in natural sounds. The results suggest that auditory grouping is adapted to natural stimulus statistics, show how these statistics can reveal previously unappreciated grouping phenomena, and provide a framework for studying grouping in natural signals.

Project description:The way the visual system processes different scales of spatial information has been widely studied, highlighting the dominant role of global over local processing. Recent studies addressing how the auditory system deals with local-global temporal information suggest a comparable processing scheme, but little is known about how this organization is modulated by long-term musical training, in particular regarding musical sequences. Here, we investigate how non-musicians and expert musicians detect local and global pitch changes in short hierarchical tone sequences structured across temporally-segregated triplets made of musical intervals (local scale) forming a melodic contour (global scale) varying either in one direction (monotonic) or both (non-monotonic). Our data reveal a clearly distinct organization between both groups. Non-musicians show global advantage (enhanced performance to detect global over local modifications) and global-to-local interference effects (interference of global over local processing) only for monotonic sequences, while musicians exhibit the reversed pattern for non-monotonic sequences. These results suggest that the local-global processing scheme depends on the complexity of the melodic contour, and that long-term musical training induces a prominent perceptual reorganization that reshapes its initial global dominance to favour local information processing. This latter result supports the theory of "analytic" processing acquisition in musicians.

Project description:S. E. Guttman, L. A. Gilroy, and R. Blake (2005) investigated whether observers could perform temporal grouping in multi-element displays where each local element was stochastically modulated over time along one of several potential dimensions--or "messenger types"--such as contrast, position, orientation, or spatial scale. Guttman et al.'s data revealed that grouping discards messenger type and therefore support a single-pathway model that groups elements with similar temporal waveforms. In the current study, we carried out three experiments in which temporal-grouping information resided either in the carrier, the envelope, or the combined carrier and envelope of each messenger's timecourse. Results revealed that grouping is highly specific for messenger type if carrier envelopes lack grouping information but largely messenger nonspecific if carrier envelopes contain grouping information. These imply that temporal grouping is mediated by several messenger-specific carrier pathways as well as by a messenger-nonspecific envelope pathways. Findings also challenge simple temporal-filtering accounts of perceptual grouping (E. H. Adelson & H. Farid, 1999).

Project description:Signals in one sensory modality can influence perception of another, for example the bias of visual timing by audition: temporal ventriloquism. Strong accounts of temporal ventriloquism hold that the sensory representation of visual signal timing changes to that of the nearby sound. Alternatively, underlying sensory representations do not change. Rather, perceptual grouping processes based on spatial, temporal, and featural information produce best-estimates of global event properties. In support of this interpretation, when feature-based perceptual grouping conflicts with temporal information-based in scenarios that reveal temporal ventriloquism, the effect is abolished. However, previous demonstrations of this disruption used long-range visual apparent-motion stimuli. We investigated whether similar manipulations of feature grouping could also disrupt the classical temporal ventriloquism demonstration, which occurs over a short temporal range. We estimated the precision of participants' reports of which of two visual bars occurred first. The bars were accompanied by different cross-modal signals that onset synchronously or asynchronously with each bar. Participants' performance improved with asynchronous presentation relative to synchronous - temporal ventriloquism - however, unlike the long-range apparent motion paradigm, this was unaffected by different combinations of cross-modal feature, suggesting that featural similarity of cross-modal signals may not modulate cross-modal temporal influences in short time scales.

Project description:Decoding the rich temporal dynamics of complex sounds such as speech is constrained by the underlying neuronal-processing mechanisms. Oscillatory theories suggest the existence of one optimal perceptual performance regime at auditory stimulation rates in the delta to theta range (< 10 Hz), but reduced performance in the alpha range (10-14 Hz) is controversial. Additionally, the widely discussed motor system contribution to timing remains unclear. We measured rate discrimination thresholds between 4 and 15 Hz, and auditory-motor coupling strength was estimated through a behavioral auditory-motor synchronization task. In a Bayesian model comparison, high auditory-motor synchronizers showed a larger range of constant optimal temporal judgments than low synchronizers, with performance decreasing in the alpha range. This evidence for optimal processing in the theta range is consistent with preferred oscillatory regimes in auditory cortex that compartmentalize stimulus encoding and processing. The findings suggest, remarkably, that increased auditory-motor synchronization might extend such an optimal range towards faster rates.

Project description:BackgroundAuditory hallucinations (AH) are typically associated with schizophrenia (SZ), but they are also prevalent in bipolar disorder (BD). Despite the large body of research on the neural correlates of AH in SZ, the pathophysiology underlying AH remains unclear. Few studies have examined the neural substrates associated with propensity for AH in BD. Investigating AH across the psychosis spectrum has the potential to inform about the neural signature associated with the trait of AH, irrespective of psychiatric diagnosis.MethodsWe compared resting state functional magnetic resonance imaging data in psychosis patients with (n = 90 AH; 68 SZ, 22 BD) and without (n = 55 NAH; 16 SZ, 39 BD) lifetime AH. We performed region of interest (ROI)-to-ROI functional connectivity (FC) analysis using 91 cortical, 15 subcortical, and 26 cerebellar atlas-defined regions. The primary aim was to identify FC differences between patients with and without lifetime AH. We secondarily examined differences between AH and NAH within each diagnosis.ResultsCompared to the NAH group, patients with AH showed higher FC between cerebellum and frontal (left precentral gyrus), temporal [right middle temporal gyrus (MTG), left inferior temporal gyrus (ITG), left temporal fusiform gyrus)], parietal (bilateral superior parietal lobules), and subcortical (left accumbens, left palldium) brain areas. AH also showed lower FC between temporal lobe regions (between right ITG and right MTG and bilateral superior temporal gyri) relative to NAH.ConclusionsOur findings suggest that dysconnectivity involving the cerebellum and temporal lobe regions may be common neurofunctional elements associated with AH propensity across the psychosis spectrum. We also found dysconnectivity patterns that were unique to lifetime AH within SZ or bipolar psychosis, suggesting both common and distinct mechanisms underlying AH pathophysiology in these disorders.

Project description:The auditory system extracts behaviorally relevant information from acoustic stimuli. The average activity in auditory cortex is known to be sensitive to spectro-temporal patterns in sounds. However, it is not known whether the auditory cortex also processes more abstract features of sounds, which may be more behaviorally relevant than spectro-temporal patterns. Using recordings from three stations of the auditory pathway, the inferior colliculus (IC), the ventral division of the medial geniculate body (MGB) of the thalamus, and the primary auditory cortex (A1) of the cat in response to natural sounds, we compared the amount of information that spikes contained about two aspects of the stimuli: spectro-temporal patterns, and abstract entities present in the same stimuli such as a bird chirp, its echoes, and the ambient noise. IC spikes conveyed on average approximately the same amount of information about spectro-temporal patterns as they conveyed about abstract auditory entities, but A1 and the MGB neurons conveyed on average three times more information about abstract auditory entities than about spectro-temporal patterns. Thus, the majority of neurons in auditory thalamus and cortex coded well the presence of abstract entities in the sounds without containing much information about their spectro-temporal structure, suggesting that they are sensitive to abstract features in these sounds.

Project description:Whether the retinal process alone or retinal and cortical processes jointly determine afterimage (AI) formation has long been debated. Based on the retinal rebound responses, recent work proposes that afterimage signals are exclusively generated in the retina, although later modified by cortical mechanisms. We tested this notion with the method of "indirect proof". Each eye was presented with a 2-by-2 checkerboard of horizontal and vertical grating patches. Each corresponding patch of the two checkerboards was perpendicular to each other, which produces binocular rivalry, and can generate percepts ranging from complete interocular grouping to either monocular pattern. The monocular percepts became more frequent with higher contrast. Due to adaptation, the visual system is less sensitive during the AIs than during the inductions with AI-similar contrast. If the retina is the only origin of AIs, comparable contrast appearance would require stronger retinal signals in the AIs than in the inductions, thus leading to more frequent monocular percepts in the AIs than in the inductions. Surprisingly, subjects saw the fully coherent stripes significantly more often in AIs. Our results thus contradict the retinal generation notion, and suggest that in addition to the retina, cortex is directly involved in the generation of AI signals.

Project description:In human and animal auditory perception the perceived quality of sound streams changes depending on the duration of inter-sound intervals (ISIs). Here, we studied whether adaptation and the precision of temporal coding in the auditory periphery reproduce general perceptual boundaries in the time domain near 20, 100, and 400 ms ISIs, the physiological origin of which are unknown. In four experiments, we recorded auditory brainstem responses with five wave peaks (P1 -P5) in response to acoustic models of communication calls of house mice, who perceived these calls with the mentioned boundaries. The newly introduced measure of average standard deviations of wave latencies of individual animals indicate the waves' temporal precision (latency jitter) mostly in the range of 30-100 μs, very similar to latency jitter of single neurons. Adaptation effects of response latencies and latency jitter were measured for ISIs of 10-1000 ms. Adaptation decreased with increasing ISI duration following exponential or linear (on a logarithmic scale) functions in the range of up to about 200 ms ISIs. Adaptation effects were specific for each processing level in the auditory system. The perceptual boundaries near 20-30 and 100 ms ISIs were reflected in significant adaptation of latencies together with increases of latency jitter at P2-P5 for ISIs < ~30 ms and at P5 for ISIs < ~100 ms, respectively. Adaptation effects occurred when frequencies in a sound stream were within the same critical band. Ongoing low-frequency components/formants in a sound enhanced (decrease of latencies) coding of high-frequency components/formants when the frequencies concerned different critical bands. The results are discussed in the context of coding multi-harmonic sounds and stop-consonants-vowel pairs in the auditory brainstem. Furthermore, latency data at P1 (cochlea level) offer a reasonable value for the base-to-apex cochlear travel time in the mouse (0.342 ms) that has not been determined experimentally.
