Project description:Single unit recordings in the rat hippocampus have demonstrated shifts in the specificity of spiking activity during learning of a contextual item-reward association task. In this task, rats received reward for responding to different items dependent upon the context an item appeared in, but not dependent upon the location an item appears at. Initially, neurons in the rat hippocampus primarily show firing based on place, but as the rat learns the task this firing became more selective for items. We simulated this effect using a simple circuit model with discrete inputs driving spiking activity representing place and item followed sequentially by a discrete representation of the motor actions involving a response to an item (digging for food) or the movement to a different item (movement to a different pot for food). We implemented spiking replay in the network representing neural activity observed during sharp-wave ripple events, and modified synaptic connections based on a simple representation of spike-timing dependent synaptic plasticity. This simple network was able to consistently learn the context-dependent responses, and transitioned from dominant coding of place to a gradual increase in specificity to items consistent with analysis of the experimental data. In addition, the model showed an increase in specificity toward context. The increase of selectivity in the model is accompanied by an increase in binariness of the synaptic weights for cells that are part of the functional network.

Project description:Perceptual learning is an improvement in sensitivity due to practice on a sensory task and is generally specific to the trained stimuli and/or tasks. The present study investigated the effect of stimulus configuration and crowding on perceptual learning in contrast discrimination in peripheral vision, and the effect of perceptual training on crowding in this task. 29 normally-sighted observers were trained to discriminate Gabor stimuli presented at 9° eccentricity with either identical or orthogonally oriented flankers with respect to the target (ISO and CROSS, respectively), or on an isolated target (CONTROL). Contrast discrimination thresholds were measured at various eccentricities and target-flanker separations before and after training in order to determine any learning transfer to untrained stimulus parameters. Perceptual learning was observed in all three training stimuli; however, greater improvement was obtained with training on ISO-oriented stimuli compared to CROSS-oriented and unflanked stimuli. This learning did not transfer to untrained stimulus configurations, eccentricities or target-flanker separations. A characteristic crowding effect was observed increasing with viewing eccentricity and decreasing with target-flanker separation before and after training in both configurations. The magnitude of crowding was reduced only at the trained eccentricity and target-flanker separation; therefore, learning for contrast discrimination and for crowding in the present study was configuration and location specific. Our findings suggest that stimulus configuration plays an important role in the magnitude of perceptual learning in contrast discrimination and suggest context-specificity in learning.

Project description:With a series of novel arm-reaching tasks, we have shown that visuomotor sequence learning encompasses the acquisition of the order of sequence elements, and the ability to combine them in a single, skilled behavior. The first component, which is mostly declarative, is reflected by changes in movement onset time (OT); the second, which occurs without subject's awareness, is measured by changes in kinematic variables, including movement time (MT). Key-press-based serial reaction time tasks (SRTT) have been used to investigate sequence learning and results interpreted as indicative of the implicit acquisition of the sequence order. One limitation to SRT studies, however, is that only one measure is used, the response time, the sum of OT and MT: this makes interpretation of which component is learnt difficult and disambiguation of implicit and explicit processes problematic. Here, we used an arm-reaching version of SRTT to propose a novel interpretation of such results. The pattern of response time changes we obtained was similar to the key-press-based tasks. However, there were significant differences between OT and MT, suggesting that both partial learning of the sequence order and skill improvement took place. Further analyses indicated that the learning of the sequence order might not occur without subjects' awareness.

The reads count was used as a proxy of translation rate here to validate the estimated regulatory effect of motifs that we inferred from multiple datasets, proving the robustness of the sequence motifs.

Project description:Background and objectiveClinical observations indicate that implicit procedural learning, a central component of physical and psychosocial rehabilitation, is impeded following spinal cord injury. In accordance, previous research has revealed a specific deficit in implicit sequence learning among individuals with paraplegia using a standard, manual version of the serial reaction time task. To extend these findings and shed light on the underlying sources of potential spinal cord injury-related deficits in sequence learning, we used an ocular activated serial reaction time task to compare sequence learning performance between individuals with tetraplegia and healthy controls.Participants and measuresTwelve participants with spinal cord injury in C5-T1 were compared to 12 matched control participants on measures derived from an ocular activated serial reaction time task. Depression and additional cognitive measures were assessed to explore the source and specificity of potential sequence learning deficits.ResultsLike controls, and in contrast with previous findings in paraplegia, the spinal cord injury group showed intact implicit sequence learning, evidenced by declining reaction times and improved anticipation over the first six blocks of the serial reaction time task, and an advantage for the initial learning sequence over a novel interference sequence.ConclusionsThe ocular activated serial reaction time task elicited a performance pattern similar to standard motor versions, such that participants with tetraplegia demonstrated unimpaired sequence learning. This suggests that previously reported implicit sequence learning deficits in spinal cord injury directly involved motor functioning rather than cognitive aspects of the task, and that the ocular activated sequence learning task could be a valid alternative for assessing implicit sequence learning in populations that cannot perform spinal-cord dependent motor tasks. Implications for post-spinal cord injury rehabilitation and adjustment are discussed.

Project description:The brain has evolved to extract behaviourally meaningful information from the environment. For example, it has been shown that visual perceptual learning (VPL) can occur for task-irrelevant stimulus features when those features are consistently paired with internal or external reinforcement signals. It is, however, unclear whether or not task-irrelevant VPL is influenced by stimulus features that are unrelated to reinforcement in a given sensory context. To address this question, we exposed participants to task-irrelevant and subliminal coherent motion stimuli in the background while they performed a central character identification task. A specific motion direction was consistently paired with the task-targets, while two other directions occurred only with distractors and, thus, were unrelated to reinforcement. We found that the magnitude of VPL of the target-paired direction was significantly greater when the distractor-paired directions were close to the target-paired direction, compared to when they were farther. Thus, even very weak signals that are both subliminal and unrelated to reinforcement are processed and exert an influence on VPL. This finding suggests that the outcome of VPL depends on the sensory context in which learning takes place and calls for a refinement of VPL theories to incorporate exposure-based influences on learning.

Project description:Environmental context can have a profound influence on the efficacy of intervention protocols designed to eliminate undesirable behaviors. This is clearly seen in drug rehabilitation clinics where patients often relapse soon after leaving the context of the treatment facility. A similar pattern is commonly observed in controlled laboratory studies of context-dependent savings in instrumental conditioning, where simply placing an animal back into the original conditioning chamber can renew an extinguished instrumental response. Surprisingly, context-dependent savings in human procedural learning has not been carefully examined in the laboratory. Here, we provide the first known empirical demonstration of context-dependent savings in a perceptual categorization task known to recruit procedural learning. We also present a computational account of these savings using a biologically detailed model in which a key role is played by cholinergic interneurons in the striatum.

Project description:Effective learning requires using errors in a task-dependent manner, for example adjusting to errors that result from unpredicted environmental changes but ignoring errors that result from environmental stochasticity. Where and how the brain represents errors in a task-dependent manner and uses them to guide behavior are not well understood. We imaged the brains of human participants performing a predictive-inference task with two conditions that had different sources of errors. Their performance was sensitive to this difference, including more choice switches after fundamental changes versus stochastic fluctuations in reward contingencies. Using multi-voxel pattern classification, we identified task-dependent representations of error magnitude and past errors in posterior parietal cortex. These representations were distinct from representations of the resulting behavioral adjustments in dorsomedial frontal, anterior cingulate, and orbitofrontal cortex. The results provide new insights into how the human brain represents errors in a task-dependent manner and guides subsequent adaptive behavior.

Project description:It is assumed that learning on the Serial Reaction Time (SRT) task is related to learning involved in social skill development affected in autism, but this assumption has hardly been investigated. We have therefore examined associations between SRT task learning and social impairment measured by the Social Responsiveness Scale in 72 autistic and non-autistic adults. Results revealed a positive correlation between deterministic sequence learning, putatively involving explicit learning, and social impairment in autistic adults but not in non-autistic adults. No correlations with probabilistic learning were found. These results suggest that the type of learning that helps autistic adults during a deterministic SRT task hinders them during social development, and call for further investigating the ecological validity of the SRT task.

Project description:When environments lack compelling goals, humans often let their minds wander to thoughts with greater personal relevance; however, we currently do not understand how this context-dependent prioritisation process operates. Dorsolateral prefrontal cortex (DLPFC) maintains goal representations in a context-dependent manner. Here, we show this region is involved in prioritising off-task thought in an analogous way. In a whole brain analysis we established that neural activity in DLPFC is high both when 'on-task' under demanding conditions and 'off-task' in a non-demanding task. Furthermore, individuals who increase off-task thought when external demands decrease, show lower correlation between neural signals linked to external tasks and lateral regions of the DMN within DLPFC, as well as less cortical grey matter in regions sensitive to these external task relevant signals. We conclude humans prioritise daydreaming when environmental demands decrease by aligning cognition with their personal goals using DLPFC.
