(3R,8aS)-3-Ethyl-perhydro-pyrrolo[1,2-a]pyrazine-1,4-dione.

ABSTRACT: In the title compound, C(9)H(14)N(2)O(2), the pyrrolidine and piperazine rings adopt envelope and boat conformations, respectively. The chiral centers were assigned on the basis of the known stereogenic center of an enanti-omerically pure starting material and the trans relationship between the H atoms attached to these centers. The crystal packing is stabilized by an inter-molecular hydrogen bond between the N-H group and a carbonyl O atom of the diketopiperazine group, forming zigzag C(5) chains along [010].

Project description:The crystal structure of the title compound, C(17)H(16)N(2)O(3)S, exhibits intra-molecular C-H⋯S and inter-molecular C-H⋯S and C-H⋯O hydrogen bonds, C-S⋯N [S⋯N = 3.033 (2) Å and C-S⋯N = 142.76 (9)°] inter-actions, and C-H⋯π inter-actions; these inter-actions generate S(4), S(6) and R(2) (2)(14) ring motifs. The isoxazole ring adopts an envelope conformation, with the N atom displaced by 0.672 (2) Å from the plane of the other ring atoms. The thio-phene ring is oriented with respect to the succinimide and phenyl rings at dihedral angles of 40.03 (12) and 5.21 (13)°, respectively. The dihedral angle between the succinimide and phenyl rings is 39.38 (12)°.

Presence of antioxidative agent, Pyrrolo[1,2-a]pyrazine-1,4-dione, hexahydro- in newly isolated Streptomyces mangrovisoli sp. nov.

Project description:A novel Streptomyces, strain MUSC 149(T) was isolated from mangrove soil. A polyphasic approach was used to study the taxonomy of MUSC 149(T), which shows a range of phylogenetic and chemotaxonomic properties consistent with those of the members of the genus Streptomyces. The diamino acid of the cell wall peptidoglycan was LL-diaminopimelic acid. The predominant menaquinones were identified as MK9(H8) and MK9(H6). Phylogenetic analysis indicated that closely related strains include Streptomyces rhizophilus NBRC 108885(T) (99.2% sequence similarity), S. gramineus NBRC 107863(T) (98.7%) and S. graminisoli NBRC 108883(T) (98.5%). The DNA-DNA relatedness values between MUSC 149(T) and closely related type strains ranged from 12.4 ± 3.3% to 27.3 ± 1.9%. The DNA G + C content was determined to be 72.7 mol%. The extract of MUSC 149(T) exhibited strong antioxidant activity and chemical analysis reported identification of an antioxidant agent, Pyrrolo[1,2-a]pyrazine-1,4-dione, hexahydro-. These data showed that metabolites of MUSC 149(T) shall be useful as preventive agent against free-radical associated diseases. Based on the polyphasic study of MUSC 149(T), the strain merits assignment to a novel species, for which the name S. mangrovisoli sp. nov. is proposed. The type strain is MUSC 149(T) (=MCCC 1K00699(T)=DSM 100438(T)).

The dihedral angle between the aromatic rings is 85.53 (5)°. The fused rings adopt envelope and twist conformations.

3-Benzyl-Hexahydro-Pyrrolo[1,2-a]Pyrazine-1,4-Dione Extracted From Exiguobacterium indicum Showed Anti-biofilm Activity Against Pseudomonas aeruginosa by Attenuating Quorum Sensing.

Project description:Bacterial cell-to-cell communication promotes biofilm formation and can potentially lead to multidrug resistance development. Quorum sensing inhibition (QSI) is an effective and widely employed strategy against biofilm formation. The extract from Exiguobacterium indicum SJ16, a gram-positive bacterium, isolated from the rhizosphere of Cyperus laevigatus showed significant anti-quorum sensing activity (about 99%) against the reference Chromobacterium violaceum CV026 strain without exerting any antibacterial effect. The potentially active QSI compound identified in the SJ16 extract was 3-Benzyl-hexahydro-pyrrolo[1, 2-a]pyrazine-1,4-dione. The SJ16 extract containing this active compound showed significant anti-quorum sensing activity against a model quorum sensing bacterium strain Pseudomonas aeruginosa PAO1 and a clinical isolate P. aeruginosa PAH by preventing biofilm formation without attenuating the cell growth within the biofilm. More specifically, the SJ16 extract changed the topography and architecture of the biofilm, thus preventing bacterial adherence and further development of the biofilm. Furthermore, it decreased virulence factors (rhamnolipid and pyocyanin), the bacterial motility, as well as the elastase, and protease activities in P. aeruginosa. Microarray analysis revealed the differential expression of quorum sensing regulatory genes. Based on these results, we herein propose a hypothetical model, characterizing the role of this QSI agent in the transcriptional regulation of quorum sensing in P. aeruginosa PAO1, demonstrating that this compound has significant drug-development potential. Further research is required to delineate its possible applications in therapeutics in the context of biofilm forming bacterial infections.

An antibiotic agent pyrrolo[1,2-a]pyrazine-1,4-dione,hexahydro isolated from a marine bacteria Bacillus tequilensis MSI45 effectively controls multi-drug resistant Staphylococcus aureus.

Project description:Sponge associated bacteria are a rich source of bioactive secondary metabolites. This study aims to isolate bacteria producing antimicrobial agents from a marine sponge, Callyspongia diffusa. A total of fifty-six bacteria were isolated and screened for antibacterial activity against multidrug resistant S. aureus. Based on the 16S rRNA sequence and phylogenetic analysis the antimicrobial producer strain MSI45 was identified as a novel Bacillus tequilensis. The culture conditions of strain MSI45 were optimized to enhance the yield of the antimicrobial compound. The antimicrobial compound was purified using a silica gel column chromatography and high performance liquid chromatography. On the basis of spectroscopic analysis such as FT-IR, NMR and GC-MS, the bioactive metabolite was identified as pyrrolo[1,2-a]pyrazine-1,4-dione,hexahydro. The extracted compound MSI45 showed a potent inhibitory effect on multidrug resistant S. aureus with an MIC of 15 ± 0.172 mg L-1 and MBC of 20 ± 0.072 mg L-1. The compound was non-hemolytic and showed high antioxidant activity. The antioxidant activity may increase the efficacy and safety of the molecule in drug development. Hence, this compound produced by Bacillus tequilensis MSI45 could have potent antimicrobial and antioxidant activity against S. aureus infection.
