Project description:The classic mapping of distinct aspects of working memory (WM) to mutually exclusive brain areas is at odds with the distributed processing mechanisms proposed by contemporary network science theory. Here, we use machine-learning to determine how aspects of WM are dynamically coded in the human brain. Using cross-validation across independent fMRI studies, we demonstrate that stimulus domains (spatial, number and fractal) and WM processes (encode, maintain, probe) are classifiable with high accuracy from the patterns of network activity and connectivity that they evoke. This is the case even when focusing on 'multiple demands' brain regions, which are active across all WM conditions. Contrary to early neuropsychological perspectives, these aspects of WM do not map exclusively to brain areas or processing streams; however, the mappings from that literature form salient features within the corresponding multivariate connectivity patterns. Furthermore, connectivity patterns provide the most precise basis for classification and become fine-tuned as maintenance load increases. These results accord with a network-coding mechanism, where the same brain regions support diverse WM demands by adopting different connectivity states.

Project description:Choice-consistency is considered a hallmark of rational value-based choice. However, because the cognitive apparatus supporting decision-making is imperfect, real decision-makers often show some degree of choice inconsistency. Cognitive models are necessary to complement idealized choice axioms with attention, perception and memory processes. Specifically, compelling theoretical work suggests that the (imperfect) retention of choice-relevant memories might be important for choice-consistency, but this hypothesis has not been tested directly. We used a novel multi-attribute visual choice paradigm to experimentally test the influence of memory retrieval of exemplars on choice-consistency. Our manipulation check confirmed that our retention interval manipulation successfully reduced memory representation strength. Given this, we found strong evidence against our hypothesis that choice-consistency decreases with increasing retention time. However, quality controls indicated that the choice-consistency of our participants was non-discernable from random behaviour. In addition, an exploratory analysis showed essentially no test-retest reliability of choice-consistency between two observations. Taken together, this suggests the presence of a floor effect in our data and, thus, low data quality for conclusively evaluating our hypotheses. Further exploration tentatively suggested a high difficulty of discriminating between the choice objects driving this floor effect.

Project description:No medications prevent or reverse age-related cognitive decline. Physical activity (PA) enhances memory in rodents, but findings are mixed in human studies. As a result, exercise guidelines specific for brain health are absent. Here, we re-examine results from human studies, and suggest the use of more sensitive tasks to evaluate PA effects on age-related changes in the hippocampus, such as relational memory and mnemonic discrimination. We discuss recent advances from rodent and human studies into the underlying mechanisms at both the central and peripheral levels, including neurotrophins and myokines that could contribute to improved memory. Finally, we suggest guidelines for future research to help expedite well-founded PA recommendations for the public.

Project description:To perform nontrivial, real-time computations on a sensory input stream, biological systems must retain a short-term memory trace of their recent inputs. It has been proposed that generic high-dimensional dynamical systems could retain a memory trace for past inputs in their current state. This raises important questions about the fundamental limits of such memory traces and the properties required of dynamical systems to achieve these limits. We address these issues by applying Fisher information theory to dynamical systems driven by time-dependent signals corrupted by noise. We introduce the Fisher Memory Curve (FMC) as a measure of the signal-to-noise ratio (SNR) embedded in the dynamical state relative to the input SNR. The integrated FMC indicates the total memory capacity. We apply this theory to linear neuronal networks and show that the capacity of networks with normal connectivity matrices is exactly 1 and that of any network of N neurons is, at most, N. A nonnormal network achieving this bound is subject to stringent design constraints: It must have a hidden feedforward architecture that superlinearly amplifies its input for a time of order N, and the input connectivity must optimally match this architecture. The memory capacity of networks subject to saturating nonlinearities is further limited, and cannot exceed square root N. This limit can be realized by feedforward structures with divergent fan out that distributes the signal across neurons, thereby avoiding saturation. We illustrate the generality of the theory by showing that memory in fluid systems can be sustained by transient nonnormal amplification due to convective instability or the onset of turbulence.

Project description:Estrogen-containing hormone therapy initiated during late postmenopause does not improve episodic memory (an important early symptom of Alzheimer's disease), and it increases dementia risk. Cognitive consequences of exogenous estrogen exposures during midlife are less certain. Observational evidence implies that use of hormone therapy at a younger age close to the time of menopause may reduce risk of Alzheimer's disease later in life. However, there are concerns that observational findings may be systematically biased. Partial insight on this critical issue may be gleaned from results of ongoing clinical trials involving midlife postmenopausal women (Early versus Late Intervention Trial with Estrogen; Kronos Early Estrogen Prevention Study). The effects of exogenous midlife estrogen exposures and Alzheimer risk can also be approached through better animal models, through carefully designed cohort studies, and through use of surrogate outcomes in randomized controlled trials in midlife women. Selective estrogen receptor modulators have the potential to affect cognitive outcomes and also merit additional study.

Project description:A dominant view of prefrontal cortex (PFC) function is that it stores task-relevant information in working memory. To examine this and determine how it applies when multiple pieces of information must be stored, we trained two subjects to perform a multi-item color change detection task and recorded activity of neurons in PFC. Few neurons encoded the color of the items. Instead, the predominant encoding was spatial: a static signal reflecting the item's position and a dynamic signal reflecting the subject's covert attention. These findings challenge the notion that PFC stores task-relevant information. Instead, we suggest that the contribution of PFC is in controlling the allocation of resources to support working memory. In support of this, we found that increased power in the alpha and theta bands of PFC local field potentials, which are thought to reflect long-range communication with other brain areas, was correlated with more precise color representations.

Project description:The chronology of the component processes subserving working memory (WM) and hemodynamic response lags has hindered the use of fMRI for exploring neural substrates of WM. In the present study, however, participants completed full trials that involved encoding two or six letters, maintaining the memory set over a delay, and then deciding whether a probe was in the memory set or not. Additionally, they completed encode-only, encode-and-maintain, and encode-and-decide partial trials intermixed with the full trials. The inclusion of partial trials allowed for the isolation of BOLD signal changes to the different trial periods. The results showed that only lateral and medial prefrontal cortex regions differentially responded to the 2- and 6-letter memory sets over the trial periods, showing greater activation to 6-letter sets during the encode and maintain trial periods. Thus, the data showed the differential involvement of PFC in the encoding and maintenance of supra- and sub-capacity memory sets and show the efficacy of using fMRI partial trial methods to study WM component processes.

Project description:Working memory performance is a key indicator of cognitive and developmental status. While recent evidence indicates that stabilizing neural gain supports the stabilization of working memory during adolescence, the computational mechanisms linking neural stabilization to behavior are poorly understood. We develop a mechanistic account of behavior during the memory-guided saccade task based on a stochastic accumulator framework. Results indicate that a specific balance of independent gain signals affecting working memory representations and oculomotor response thresholds can account for a peculiar U-shaped feature of the speed-accuracy relationship. Additionally, aspects of behavioral variability and mean behavioral performance, as well as subtle shifts in the shape of the speed-accuracy relationship across development, can be accounted for by the stabilization of these two sources of variability. Thus, the stabilization of neural variability can, in part, account for developmental improvements in behavioral variability as well as some improvement in mean behavioral performance.

Project description:Impaired social abilities are characteristics of a variety of psychiatric disorders such as schizophrenia, autism spectrum disorder, and bipolar disorder. Studies consistently implicated the relationship between the anterior insular cortex (aIC) and social ability, however, how the aIC involves in processing specific subtypes of social ability was uninvestigated. We, therefore, investigated whether the absence or presence of the aIC affects the social behaviors of mice. We found that electrolytic lesions of the aIC specifically impaired mice's ability to recognize a novel stranger mouse, while the sociability of the aIC-lesioned mice was intact. Interestingly, the aIC-lesioned mice were still distinguished between a mouse that had been housed together before the aIC lesion and a novel mouse, supporting that retrieval of social recognition memory may not involve the aIC. Additional behavioral tests revealed that this specific social ability impairment induced by the aIC lesion was not due to impairment in olfaction, learning and memory, locomotion, or anxiety levels. Together our data suggest that the aIC is specifically involved in processing social recognition memory, but not necessarily involved in retrieving it.

Project description:Magnetoencephalography (MEG) was used to compare memory processes in two experiments, one involving recognition of word pairs and the other involving recall of newly learned arithmetic facts. A combination of hidden semi-Markov models and multivariate pattern analysis was used to locate brief "bumps" in the sensor data that marked the onset of different stages of cognitive processing. These bumps identified a separation between a retrieval stage that identified relevant information in memory and a decision stage that determined what response was implied by that information. The encoding, retrieval, decision, and response stages displayed striking similarities across the two experiments in their duration and brain activation patterns. Retrieval and decision processes involve distinct brain activation patterns. We conclude that memory processes for two different tasks, associative recognition versus arithmetic retrieval, follow a common spatiotemporal neural pattern and that both tasks have distinct retrieval and decision stages.
