Synthesis of dye/fluorescent functionalized dendrons based on cyclotriphosphazene.

ABSTRACT: Functionalized phenols based on tyramine were synthesized in order to be selectively grafted on to hexachlorocyclotriphosphazene, affording a variety of functionalized dendrons of type AB(5). The B functions comprised fluorescent groups (dansyl) or dyes (dabsyl), whereas the A function was provided by either an aldehyde or an amine. The characterization of these dendrons is reported. An unexpected behaviour of a fluorescent and water-soluble dendron based on dansyl groups in mixtures of dioxane/water was observed.

A Ratiometric Fluorescent Sensor Based on Dye/Tb (III) Functionalized UiO-66 for Highly Sensitive Detection of TDGA.

Project description:Thiodiglycolic acid (TDGA) is a biomarker for monitoring vinyl chloride exposure. Exploring a facile, rapid and precise analysis technology to quantify TDGA is of great significance. In this research, we demonstrate a fluorescent sensor based on dual-emissive UiO-66 for TDGA detection. This ratiometric fluorescent material named C460@Tb-UiO-66-(COOH)2 was designed and synthesized by introducing organic dye 7-diethylamino-4-methylcoumarin (C460) and Tb3+ into UiO-66-(COOH)2. The as-obtained C460@Tb-UiO-66-(COOH)2 samples showed highly selective recognition, excellent anti-interference and rapid response characteristics for the recognition of TDGA. The detection limit is 0.518 mg·mL-1, which is much lower than the threshold of 20 mg·mL-1 for a healthy person. In addition, the mechanism of TDGA-induced fluorescence quenching is discussed in detail. This sensor is expected to detect TDGA content in human urine.

Total chemical synthesis of biologically active fluorescent dye-labeled Ts1 toxin.

Project description:Ts1 toxin is a protein found in the venom of the Brazilian scorpion Tityus serrulatus. Ts1 binds to the domain II voltage sensor in the voltage-gated sodium channel Nav and modifies its voltage dependence. In the work reported here, we established an efficient total chemical synthesis of the Ts1 protein using modern chemical ligation methods and demonstrated that it was fully active in modifying the voltage dependence of the rat skeletal muscle voltage-gated sodium channel rNav1.4 expressed in oocytes. Total synthesis combined with click chemistry was used to label the Ts1 protein molecule with the fluorescent dyes Alexa-Fluor 488 and Bodipy. Dye-labeled Ts1 proteins retained their optical properties and bound to and modified the voltage dependence of the sodium channel Nav. Because of the highly specific binding of Ts1 toxin to Nav, successful chemical synthesis and labeling of Ts1 toxin provides an important tool for biophysical studies, histochemical studies, and opto-pharmacological studies of the Nav protein.

Pyrene-Functionalized Fluorescent Nanojars: Synthesis, Mass Spectrometric, and Photophysical Studies.

Project description:Nanojars are a class of supramolecular coordination complexes based on pyrazolate, Cu2+, and OH- ions that self-assemble around highly hydrophilic anions and serve as efficient anion binding and extraction agents. In this work, the synthesis, characterization, and photophysical properties of pyrene-functionalized fluorescent nanojars are presented. Three pyrene derivatives, 4-(pyren-1-yl)pyrazole (HL1), 4-(5-(pyren-1-yl)pent-4-yn-1-yl)pyrazole (HL2), and 4-(3-(pyrazol-4-yl)propyl)-1-(pyren-1-yl)-1,2,3-triazole (HL3), and the corresponding nanojars were synthesized and characterized using nuclear magnetic resonance spectroscopy and mass spectrometry. Electronic absorption, steady-state, and time-resolved fluorescence measurements were carried out to understand the interaction between the pyrene fluorophore and copper nanojars. Optical absorption measurements have shown minor ground state interaction between the fluorophore and nanojars. The fluorescence of pyrene is significantly quenched when attached to nanojars, suggesting strong contribution from the paramagnetic Cu2+ ions. Significant static quenching is observed in the case of L1, when pyrene is directly bound to the nanojar, whereas in the case of L2 and L3, when pyrene is attached to the nanojars using flexible tethers, both static and dynamic quenching are observed.

Green Electrospun Membranes Based on Chitosan/Amino-Functionalized Nanoclay Composite Fibers for Cationic Dye Removal: Synthesis and Kinetic Studies.

Project description:Chitosan/poly(vinyl alcohol)/amino-functionalized montmorillonite nanocomposite electrospun membranes with enhanced adsorption capacity and thermomechanical properties were fabricated and utilized for the removal of a model cationic dye (Basic Blue 41). Effects of nanofiller concentrations (up to 3.0 wt %) on the morphology and size of the nanofibers as well as the porosity and thermomechanical properties of the nanocomposite membranes are studied. It is shown that the incorporation of the nanoclay particles with ∼10 nm lateral sizes into the polymer increases the size of the pores by about 80%. To demonstrate the efficiency of the adsorbents, the dye removal rate is investigated as a function of pH, adsorbent dosage, dye concentration, and nanofiller loading. The highest and fastest dye removal occurs for the nanofibrous membranes containing 2 wt % nanofiller, where about 80% of the cationic dye is removed after 15 min. This performance is at least 20% better than the pristine chitosan/poly(vinyl alcohol) membrane. The thermal stability and compression resistance of the nanocomposite membranes are found to be higher than those of the pristine membrane. In addition, reusability studies show that the dye removal performance of this nanocomposite membrane reduces by only about 5% over four cycles. The adsorption kinetics is explained by the Langmuir isotherm model and is expressed by a pseudo-second-order kinetic mechanism that determines a spontaneous chemisorption process. The results of this study provide a valuable perspective on the fabrication of high-performance, reusable, and efficient electrospun fibrous nanocomposite adsorbents.

A Ratiometric Fluorescent Nano-Probe for Rapid and Specific Detection of Tetracycline Residues Based on a Dye-Doped Functionalized Nanoscaled Metal-Organic Framework.

Project description:Tetracycline (TC) residues are harmful to the environment and human body, so it is necessary to develop a highly sensitive probe for rapid detection of tetracycline residues. In the present paper, a novel dye-doped porous metal-organic framework (UiO-66)-based multi-color fluorescent nano-probe was designed for sensitive ratiometric detection of tetracycline (TC). In this probe, dye-molecules doped UiO-66 was used as a fluorescent internal standard, and the externally grafted lanthanide Eu3+ complex was used as response signals. The fluorescence of the Eu3+ complex was selectively enhanced with increasing concentrations of TC, which was accompanied by a visual blue-to-red color switch. The nano-probe had a linear response between 0.1 and 6 μM with a lowest detection limit of 17.9 nM, which was much lower than the maximum residue limits set by the United States Food and Drug Administration (676 nM) and the European Union (225 nM). The applicability of this method in the analysis of actual samples was evaluated by the determination of TC in honey and milk samples, indicating satisfactory recovery and good reproducibility. In addition, a cost-effective paper-based probe for rapid and visual detection of TC was developed by fixing the nano-probe on filter papers. With the help of a smartphone camera to capture the fluorescence color, and chromaticity analysis software, the calculation and analysis of red (R) and blue (B) values can be realized, which has the potential for real-time visual detection of TC.

Synthesis and properties of fluorescent 4'-azulenyl-functionalized 2,2':6',2?-terpyridines.

Project description:4'-Azulenyl-substituted terpyridines were efficiently synthesized following the Kröhnke methodology via azulenylchalcone intermediates. These azulenyl-containing terpyridines showed fluorescent emission with a fluorescence quantum yield varying from 0.14, in the case of parent terpyridine, to 0.64 when methyl groups are grafted on the azulenyl seven-membered ring. According to the crystal structures and TDDFT calculations, different twisting of the aromatic constituents is responsible for the observed fluorescent behavior. The electrochemical profile contains one-electron oxidation/reduction steps, which can only be explained on the basis of the redox behavior of the azulene unit. The ability of the 4'-azulenyl 2,2':6',2?-terpyridine to bind poisoning metal cations was studied by UV-vis titrations using aqueous solutions of Hg(II) and Cd(II) chlorides as illustrative examples.

Fluorescent DNA nanotags: supramolecular fluorescent labels based on intercalating dye arrays assembled on nanostructured DNA templates.

Project description:Fluorescence detection and imaging are vital technologies in the life sciences and clinical diagnostics. The key to obtaining high-resolution images and sensitive detection is to use fluorescent molecules or particles that absorb and emit visible light with high efficiency. We have synthesized supramolecular complexes consisting of a branched DNA template and fluorogenic intercalating dyes. Because dyes can intercalate up to every other base pair, high densities of fluorophores are assembled yet the DNA template keeps them far enough away from each other to prevent self-quenching. The efficiency with which these noncovalent assemblies absorb light is more than 10-fold greater than that of the individual dye molecules. Förster resonance energy transfer from the intercalated dyes to covalently attached acceptor dyes is very efficient, allowing for wavelength shifting of the emission spectrum. Simple biotinylation of the DNA template allows for labeling of streptavidin-coated synthetic microspheres and mouse T-cells.

Direct and sensitive detection of sulfide ions based on one-step synthesis of ionic liquid functionalized fluorescent carbon nanoribbons.

Project description:Despite widely reported fluorescence sensors for cations, direct detection of anions is nevertheless still rare. In this work, ionic liquid-functionalized fluorescent carbon nanoribbons (IL-CNRs) are one-step synthesized and serve as the fluorescent probes for direct and sensitive detection of sulfide ions (S2-). The IL-CNRs are synthesized based on electrochemical exfoliation of graphite rods in a water-IL biphasic system. The as-prepared IL-CNRs exhibit uniform structure, high crystallinity, strong blue fluorescence (absolute photoluminescence quantum yield of 11.4%), and unique selectivity towards S2-. Based on the fluorescence quenching of IL-CNRs by S2-, a fluorescence sensor is developed for direct, rapid and sensitive detection of S2- in the range of 100 nM to 1 μM and 1-300 μM with a low detection limit (LOD, 85 nM). Moreover, detection of S2- in a real sample (tap water) is also demonstrated.

Synthesis of [18F]2B-SRF101: A Sulfonamide Derivative of the Fluorescent Dye Sulforhodamine 101.

Project description:BackgroundThe red fluorescent dye Sulforhodamine 101 (SR101) has been used in neuroscience research as a useful tool for staining of astrocytes, since it has been reported as a marker of astroglia in the neocortex of rodents in vivo. The aim of this work is to label SR101 with positron emission radionuclides, in order to provide a radiotracer to study its biological behavior. This is the first attempt to label SR101 by [18F], using a chemical derivatization via a sulfonamidelinker and a commercially available platform.MethodsThe synthesis of SR101 N-(3-Bromopropyl) sulfonamide and SR101 N-(3- Fluoropropyl) sulfonamide (2B-SRF101) was carried out. The radiosynthesis of SR101 N-(3- [18F]Fluoropropyl) sulfonamide ([18F]2B-SRF101) was performed in a TRACERlab® FX-FN. Different labeling conditions were tested. Three pilot batches were produced and quality control was performed. Lipophilicity, plasma protein binding and radiochemical stability of [18F]2BSRF101 in final formulation and in plasma were determined.ResultsSR101 N-(3-Bromopropyl) sulfonamide was synthetized as a precursor for radiolabeling with [18F]. 2B-SRF101 was prepared for analytical purpose. [18F]2B-SRF101 was obtained with radiochemical purity of (97.0 ± 0.6%). The yield of the whole synthesis was (11.9 ± 1.7 %), nondecay corrected. [18F]2B-SRF101 was found to be stable in final formulation and in plasma. The octanol-water partition coefficient was (Log POCT = 1.88 ± 0.14). The product showed a high percentage of plasma protein binding.ConclusionsThe derivatization of SR101 via sulfonamide-linker and the first radiosynthesis of [18 F]2B-SRF101 were performed. It was obtained in accordance with quality control specifications. In vitro stability studies verified that [18F]2B-SRF101 was suitable for preclinical evaluations.

Facile One-Pot Synthesis of Functionalized Quinoline-Fused Fluorescent Dihydro/Spiro-quinazolinone Derivatives.

Project description:A facile and efficient method has been developed for the synthesis of quinoline-fused fluorescent dihydro/spiro-quinazolinones. A plausible mechanism involving an acid-mediated enaminone intermediate is provided. The reaction proceeded using p-toluene sulfonic acid as a green promoter. The methodology was successful in synthesizing various quinoline-appended spiro-quinazolinones 4a-o. The synthetic utility of compounds 4a-o was demonstrated by synthesizing compounds 6a-d via Suzuki coupling as a key reaction. Significantly, the π-π* electronic transition of compounds 4c and 4k showed a blue shift. The molar extinction coefficient (ε), Stoke's shift (Δu̅), and quantum yield (Φf)c were calculated for these derivatives (4c and 4k).
