Project description:Nanoparticles with diameters in the range of a few nanometers, consisting of gold and vanadium oxide, are synthesized by sequential doping of cold helium droplets in a molecular beam apparatus and deposited on solid carbon substrates. After surface deposition, the samples are removed and various measurement techniques are applied to characterize the created particles: scanning transmission electron microscopy (STEM) at atomic resolution, temperature dependent STEM and TEM up to 650 °C, energy-dispersive X-ray spectroscopy (EDXS) and electron energy loss spectroscopy (EELS). In previous experiments we have shown that pure V2O5 nanoparticles can be generated by sublimation from the bulk and deposited without affecting their original stoichiometry. Interestingly, our follow-up attempts to create Au@V2O5 core@shell particles do not yield the expected encapsulated structure. Instead, Janus particles of Au and V2O5 with diameters between 10 and 20 nm are identified after deposition. At the interface of the Au and the V2O5 parts we observe an epitaxial-like growth of the vanadium oxide next to the Au structure. To test the temperature stability of these Janus-type particles, the samples are heated in situ during the STEM measurements from room temperature up to 650 °C, where a reduction from V2O5 to V2O3 is followed by a restructuring of the gold atoms to form a Wulff-shaped cluster layer. The temperature dependent dynamic interplay between gold and vanadium oxide in structures of only a few nanometer size is the central topic of this contribution to the Faraday Discussion.

Project description:With a hyperbranched poly(amidoamine) core and many water-soluble poly(ethylene glycol) monomethyl ether arms connected by pH-sensitive acylhydrazone bonds, multiarm hyperbranched polymer was used as nanoreactor and reductant to prepare metal nanoparticles endowed with intelligence and biocompatibility. The multiarm hyperbranched polymer encapsulated nanoparticles (NPs) showed excellent catalytic activity for hydrogenation, thus an excellent catalyst system for hydrogenation was established. The rate constants could reach as high as 3.48 L·s-1·m-2, which can be attributed to the lack of surface passivation afforded by the multiarm hyperbranched polymer.

Project description:Metal co-catalysts tipped at a photocatalyst surface form a crucial component in the nanoheterostructures designed for the photocatalytic hydrogen evolution reaction. To examine the intermetallic differences and size effects at these interfaces, we use spin-polarized density functional theory to study single-atom, 13-atom, and 55-atom cluster depositions of Ni, Pd, Pt, and Au on the CdS(101?0) surface. For the single metal atoms, the ground-state configuration was the same site for all of the elements. Analysis of the metal-CdS bonding and of the charge transfers revealed a Ni-Cd bonding complex leading to depletion of electronic charge at the Ni single atom and at deposited Ni clusters, in contrast to charge accumulation observed for the other three metals Pd, Pt, and Au. For scaling up sizes of the metal deposition, six subnanometer cluster types were selected over a wide range of cluster's effective coordination number, and their interfaces were differentiated by charge redistributions, structure and adhesion energies, highest occupied molecular orbital-lowest occupied molecular orbital (HOMO-LUMO) gaps, and Schottky barrier heights. Although all considered clusters are semiconducting in the gas phase, 9 out of 28 clusters became (semi)metallic after deposition on the CdS semiconductor surface. Intermetallic differences and common trends are discussed.

Project description:Virus-like particles (VLPs) have been created from luminescent Pt(II) complex amphiphiles, able to form supramolecular structures in water solutions, that can be encapsulated or act as templates of cowpea chlorotic mottle virus capsid proteins. By virtue of a bottom-up molecular design, icosahedral and nonicosahedral (rod-like) VLPs have been constructed through diverse pathways, and a relationship between the molecular structure of the complexes and the shape and size of the VLPs has been observed. A deep insight into the mechanism for the templated formation of the differently shaped VLPs was achieved, by electron microscopy measurements (TEM and STEM) and bulk analysis (FPLC, DLS, photophysical investigations). Interestingly, the obtained VLPs can be visualized by their intense emission at room temperature, generated by the self-assembly of the Pt(II) complexes. The encapsulation of the luminescent species is further verified by their higher emission quantum yields inside the VLPs, which is due to the confinement effect of the protein cage. These hybrid materials demonstrate the potential of tailor-made supramolecular systems able to control the assembly of biological building blocks.

Project description:A facile synthetic route was developed to make Au nanowires (NWs) from surfactant-mediated bio-mineralization of a genetically engineered M13 phage with specific Au binding peptides. From the selective interaction between Au binding M13 phage and Au ions in aqueous solution, Au NWs with uniform diameter were synthesized at room temperature with yields greater than 98 % without the need for size selection. The diameters of Au NWs were controlled from 10 nm to 50 nm. The Au NWs were found to be active for electrocatalytic oxidation of CO molecules for all sizes, where the activity was highly dependent on the surface facets of Au NWs. This low-temperature high yield method of preparing Au NWs was further extended to the synthesis of Au/Pt core/shell NWs with controlled coverage of Pt shell layers. Electro-catalytic studies of ethanol oxidation with different Pt loading showed enhanced activity relative to a commercial supported Pt catalyst, indicative of the dual functionality of Pt for the ethanol oxidation and Au for the anti-poisoning component of Pt. These new one-dimensional noble metal NWs with controlled compositions could facilitate the design of new alloy materials with tunable properties.

Project description:Metal coordination is a key structural and functional component of a large fraction of proteins. Given this dual role we considered the possibility that metal coordination may have played a templating role in the early evolution of protein folds and complexes. We describe here a rational design approach, Metal Templated Interface Redesign (MeTIR), that mimics the time course of a hypothetical evolutionary pathway for the formation of stable protein assemblies through an initial metal coordination event. Using a folded monomeric protein, cytochrome cb(562), as a building block we show that its non-self-associating surface can be made self-associating through a minimal number of mutations that enable Zn coordination. The protein interfaces in the resulting Zn-directed, D(2)-symmetrical tetramer are subsequently redesigned, yielding unique protein architectures that self-assemble in the presence or absence of metals. Aside from its evolutionary implications, MeTIR provides a route to engineer de novo protein interfaces and metal coordination environments that can be tuned through the extensive noncovalent bonding interactions in these interfaces.

Project description:Carbon based nanomaterials offer the potential to provide solutions to key technological challenges. This work describes the preparation of luminescent carbon nanofibers by template-assisted microwave pyrolysis of environmentally friendly precursors, citric acid and polyethyleneimine, in aqueous solution. SEM reveals a dense forest of vertically aligned cylindrical carbon nanofibers with an average diameter of ca. 200 nm, which are shown by TEM to be amorphous. Compositional analysis indicated the incorporation of amino and pyrrolic nitrogen, and carbon-oxygen moieties. These species contribute to UV light absorption with an absorption shoulder and tail towards visible wavelengths. UV excitation gave visible (blue) emission at ca. 450 nm with a quantum yield of ca. 5%; emission decay under pulsed excitation was predominantly mono-exponential with a lifetime of ca. 1 ns. The emission maximum is largely excitation wavelength independent suggesting the involvement of citrazinic acid-type functionalities in the fiber photophysics. Reversible pH-dependent excitation and emission behaviour was observed, with maximum emission at ca. pH 7. Nanofiber emission was also quenched in aqueous solutions of metal cations, in a concentration-dependent manner. Single nanofiber emission intensity was quite stable under continuous excitation permitting single fiber quenching-based metal ion detection whereby a significant (>90%) and prompt (sub-10 s) quenching was observed upon exposure to sub-millimolar Fe(iii) solutions. The introduction of these new 1D luminescent carbon nanofibers offers the potential for exciting developments across a range of applications.

Project description:Selective binding by metalloproteins to their cognate metal ions is essential to cellular survival. How proteins originally acquired the ability to selectively bind metals and evolved a diverse array of metal-centered functions despite the availability of only a few metal-coordinating functionalities remains an open question. Using a rational design approach (Metal-Templated Interface Redesign), we describe the transformation of a monomeric electron transfer protein, cytochrome cb(562), into a tetrameric assembly ((C96)RIDC-1(4)) that stably and selectively binds Zn(2+) and displays a metal-dependent conformational change reminiscent of a signaling protein. A thorough analysis of the metal binding properties of (C96)RIDC-1(4) reveals that it can also stably harbor other divalent metals with affinities that rival (Ni(2+)) or even exceed (Cu(2+)) those of Zn(2+) on a per site basis. Nevertheless, this analysis suggests that our templating strategy simultaneously introduces an increased bias toward binding a higher number of Zn(2+) ions (four high affinity sites) versus Cu(2+) or Ni(2+) (two high affinity sites), ultimately leading to the exclusive selectivity of (C96)RIDC-1(4) for Zn(2+) over those ions. More generally, our results indicate that an initial metal-driven nucleation event followed by the formation of a stable protein architecture around the metal provides a straightforward path for generating structural and functional diversity.

Project description:Highly stable and facile one-pot copper nanoclusters (Cu NCs) coated with poly(allylamine hydrochloride) (PAH) have been synthesized for selectively sensing deferasirox (DFX) in β-thalassemia plasma. DFX is an important drug used for treating iron overloading in β-thalassemia, but needs to be monitored due to certain toxicity. In this study, the PAH-Cu NCs showed highly stable fluorescence with emission wavelengths at 450 nm. The DFX specifically interacted with the copper nanocluster to turn off the fluorescence of the PAH-Cu NCs, and could be selectively quantified through the fluorescence quenching effect. The linear range of DFX in plasma analyzed by PAH-Cu NCs was 1.0-100.0 µg/mL (r = 0.985). The relative standard deviation (RSD) and relative error (RE) were lower than 6.51% and 7.57%, respectively, showing excellent reproducibility of PAH-Cu NCs for sensing DFX in plasma. This method was also successfully applied for an analysis of three clinical plasma samples from β-thalassemia patients taking DFX. The data presented high similarity with that obtained through a capillary electrophoresis method. According to the results, the PAH-Cu NCs could be used as a tool for clinically sensing DFX in human plasma for clinical surveys.

Project description:Highly luminescent gold clusters simultaneously synthesized and stabilized by protein molecules represent a remarkable category of nanoscale materials with promising applications in bionanotechnology as sensors. Nevertheless, the atomic structure and luminescence mechanism of these gold clusters are still unknown after several years of developments. Herein, we report findings on the structure, luminescence and biomolecular self-assembly of gold clusters stabilized by the large globular protein, bovine serum albumin. We highlight the surprising identification of interlocked gold-thiolate rings as the main gold structural unit. Importantly, such gold clusters are in a rigidified state within the protein scaffold, offering an explanation for their highly luminescent character. Combined free-standing cluster synthesis (without protecting protein scaffold) with rigidifying and un-rigidifying experiments, were designed to further verify the luminescence mechanism and gold atomic structure within the protein. Finally, the biomolecular self-assembly process of the protein-stabilized gold clusters was elucidated by time-dependent X-ray absorption spectroscopy measurements and density functional theory calculations.
