Project description:In dual-energy computed tomography (DECT), low- and high-kVp data are collected often over a full-angular range (FAR) of 360°. While there exists strong interest in DECT with low- and high-kVp data acquired over limited-angular ranges (LARs), there remains little investigation of image reconstruction in DECT with LAR data.Objective: we investigate image reconstruction with minimized LAR artifacts from low- and high-kVp data over LARs of ≤180° by using a directional-total-variation (DTV) algorithm.Methods: image reconstruction from LAR data is formulated as a convex optimization problem in which data-ℓ2is minimized with constraints on image's DTVs along orthogonal axes. We then achieve image reconstruction by applying the DTV algorithm to solve the optimization problem. We conduct numerical studies from data generated over arcs of LARs, ranging from 14° to 180°, and perform visual inspection and quantitative analysis of images reconstructed.Results: monochromatic images of interest obtained with the DTV algorithm from LAR data show substantially reduced artifacts that are observed often in images obtained with existing algorithms. The improved image quality also leads to accurate estimation of physical quantities of interest, such as effective atomic number and iodine contrast concentration.Conclusion: our study reveals that from LAR data of low- and high-kVp, monochromatic images can be obtained that are visually, and physical quantities can be estimated that are quantitatively, comparable to those obtained in FAR DECT.Significance: as LAR DECT is of high practical application interest, the results acquired in the work may engender insights into the design of DECT with LAR scanning configurations of practical application significance.

Such results as integral representations, convolution properties, and coefficient estimates are proved. The results presented here would provide extensions of those given in earlier works. Several other results are also obtained.

Project description:We define a new class of multivalent meromorphic functions using the generalised hypergeometric function. We derived this class related to conic domain. It is also shown that this new class of functions, under certain conditions, becomes a class of starlike functions. Some results on inclusion and closure properties are also derived.

Project description:To isolate the genetic locus responsible for saline-alkaline stress tolerance, we developed a high-throughput activation tagging-based T-DNA insertion mutagenesis method using the model rice (Oryza sativa L.) variety Kitaake. One of the activation-tagged insertion lines, activation tagging 7 (AC7), showed increased tolerance to saline-alkaline stress. This phenotype resulted from the overexpression of a gene that encodes a SET DOMAIN GROUP 721 protein with H3K4 methyltransferase activity. Transgenic plants overexpressing OsSDG721 showed saline-alkaline stress-tolerant phenotypes, along with increased leaf angle, advanced heading and ripening dates. By contrast, ossdg721 loss-of-function mutants showed increased sensitivity to saline-alkaline stress characterized by decreased survival rates and reduction in plant height, grain size, grain weight and leaf angle. RNA sequencing (RNA-seq) analysis of wild-type Kitaake and ossdg721 mutants indicated that OsSDG721 positively regulates the expression level of HIGH-AFFINITY POTASSIUM (K+ ) TRANSPORTER1;5 (OsHKT1;5), which encodes a Na+ -selective transporter that maintains K+ /Na+ homeostasis under salt stress. Furthermore, we showed that OsSDG721 binds to and deposits the H3K4me3 mark in the promoter and coding region of OsHKT1;5, thereby upregulating OsHKT1;5 expression under saline-alkaline stress. Overall, by generating Kitaake activation-tagging pools, we established that the H3K4 methyltransferase OsSDG721 enhances saline-alkaline stress tolerance in rice.

Project description:In Arabidopsis, a complex of Polycomb-group (PcG) proteins functions in the female gametophyte to control the initiation of seed development. Mutations in the PcG genes, including MEDEA (MEA) and FERTILIZATION-INDEPENDENT SEED 2 (FIS2), produce autonomous seeds where endosperm proliferation occurs in the absence of fertilization. By using a yeast two-hybrid screen, we identified MEA and a related protein, SWINGER (SWN), as SET-domain partners of FIS2. Localization data indicated that all three proteins are present in the female gametophyte. Although single-mutant swn plants did not show any defects, swn mutations enhanced the mea mutant phenotype in producing autonomous seeds. Thus, MEA and SWN perform partially redundant functions in controlling the initiation of endosperm development before fertilization in Arabidopsis.

Project description:In recent years, significant work has been devoted to the use of angle-resolved elastic scattering for the extraction of nuclear morphology in tissue. By treating the nucleus as a Mie scattering object, techniques such as angle-resolved low-coherence interferometry (a/LCI) have demonstrated substantial success in identifying nuclear alterations associated with dysplasia. Because optical biopsies are inherently noninvasive, only a small, discretized portion of the 4? scattering field can be collected from tissue, limiting the amount of information available for diagnostic purposes. In this work, we comprehensively characterize the diagnostic impact of variations in angular sampling, range and noise for inverse light scattering analysis of nuclear morphology, using a previously reported dataset from 40 patients undergoing a/LCI optical biopsy for cervical dysplasia. The results from this analysis are applied to a benchtop scanning a/LCI system which compromises angular range for wide-area scanning capability. This work will inform the design of next-generation optical biopsy probes by directing optical design towards parameters which offer the most diagnostic utility.

Project description:Metnase (also known as SETMAR) is a chimeric SET-transposase protein that plays essential role(s) in non-homologous end joining (NHEJ) repair and replication fork restart. Although the SET domain possesses histone H3 lysine 36 dimethylation (H3K36me2) activity associated with an improved association of early repair components for NHEJ, its role in replication restart is less clear. Here we show that the SET domain is necessary for the recovery from DNA damage at the replication forks following hydroxyurea (HU) treatment. Cells overexpressing the SET deletion mutant caused a delay in fork restart after HU release. Our In vitro study revealed that the SET domain but not the H3K36me2 activity is required for the 5' end of ss-overhang cleavage with fork and non-fork DNA without affecting the Metnase-DNA interaction. Together, our results suggest that the Metnase SET domain has a positive role in restart of replication fork and the 5' end of ss-overhang cleavage, providing a new insight into the functional interaction of the SET and the transposase domains.

Project description:Due to the nature of infinite dimensionality, the orbital angular momentum (OAM) has been considered as a new degree of freedom of light and widely expanded the scopes of substantial optical applications such as optical telecommunication, quantum information, particle manipulation and imaging. In recent years, the integrated photonic OAM emitters have been actively investigated due to both compactness and tunability. Essentially, the number of available OAM modes by dynamic switching should be large enough so that the dimensionality of OAM could be explored as much as possible. In this work, an integrated photonic emitter with a wide switching range of OAM modes is theoretically developed, numerically simulated, and experimentally verified. The independence of the micro-ring cavity and the scattering unit provides the flexibility to design the device and optimize the performance. Specifically, the dynamic switching of nine OAM modes (l = -4 ~ 4) with azimuthal polarization has been demonstrated by electrically controlled thermo-optic effect.

Project description:Islet-brain 1 (IB1 or JIP-1) is a scaffold protein that interacts with components of the c-Jun N-terminal kinase (JNK) signal-transduction pathway. IB1 is expressed at high levels in neurons and in pancreatic beta-cells, where it controls expression of several insulin-secretory components and secretion. IB1 has been shown to homodimerize, but neither the molecular mechanisms nor the function of dimerization have yet been characterized. Here, we show that IB1 homodimerizes through a novel and unique set of Src homology 3 (SH3)-SH3 interactions. X-ray crystallography studies show that the dimer interface covers a region usually engaged in PxxP-mediated ligand recognition, even though the IB1 SH3 domain lacks this motif. The highly stable IB1 homodimer can be significantly destabilized in vitro by three individual point mutations directed against key residues involved in dimerization. Each mutation reduces IB1-dependent basal JNK activity in 293T cells. Impaired dimerization also results in a reduction in glucose transporter type 2 expression and in glucose-dependent insulin secretion in pancreatic beta-cells. Taken together, these results indicate that IB1 homodimerization through its SH3 domain has pleiotropic effects including regulation of the insulin secretion process.
