Diversity of manota williston (Diptera, mycetophilidae) in ulu temburong national park, brunei.

ABSTRACT: A total of 15 species of Manota Williston, 1896 are recorded from Brunei, based on the investigations in 2013-2014. Thirteen species are recorded from Ulu Temburong National Park and three species from the Universiti Brunei Darussalam Campus in Tungku. Six species are described as new to science: Manota belalongensis sp. n., M. kaspraki sp. n., M. macrothrix sp. n., M. megachaeta sp. n. and M. pileata sp. n. from Ulu Temburong, and M. ricina sp. n. from Tungku. New records of the following species are given: Manota bifida Hippa & Papp, M. bruneiensis Hippa & Šev?ík, M. hyboloma Hippa & Šev?ík, M. oligochaeta Hippa, M. pappi Hippa, M. perangulata Hippa & Šev?ík, M. pollex Hippa, M. procera Hippa and M. simplex Hippa.

Oral microbiome diversity in chimpanzees from Gombe National Park.

Project description:Historic calcified dental plaque (dental calculus) can provide a unique perspective into the health status of past human populations but currently no studies have focused on the oral microbial ecosystem of other primates, including our closest relatives, within the hominids. Here we use ancient DNA extraction methods, shotgun library preparation, and next generation Illumina sequencing to examine oral microbiota from 19 dental calculus samples recovered from wild chimpanzees (Pan troglodytes schweinfurthii) who died in Gombe National Park, Tanzania. The resulting sequences were trimmed for quality, analyzed using MALT, MEGAN, and alignment scripts, and integrated with previously published dental calculus microbiome data. We report significant differences in oral microbiome phyla between chimpanzees and anatomically modern humans (AMH), with chimpanzees possessing a greater abundance of Bacteroidetes and Fusobacteria, and AMH showing higher Firmicutes and Proteobacteria. Our results suggest that by using an enterotype clustering method, results cluster largely based on host species. These clusters are driven by Porphyromonas and Fusobacterium genera in chimpanzees and Haemophilus and Streptococcus in AMH. Additionally, we compare a nearly complete Porphyromonas gingivalis genome to previously published genomes recovered from human gingiva to gain perspective on evolutionary relationships across host species. Finally, using shotgun sequence data we assessed indicators of diet from DNA in calculus and suggest exercising caution when making assertions related to host lifestyle. These results showcase core differences between host species and stress the importance of continued sequencing of nonhuman primate microbiomes in order to fully understand the complexity of their oral ecologies.

Data on alpine grassland diversity in Gran Paradiso National Park, Italy.

Project description:The diversity of alpine grassland species and their functional traits constitute alpine ecosystem functioning and services that support human-wellbeing. However, alpine grassland diversity is threatened by land use and climate change. Field surveys and monitoring are necessary to understand and preserve such endangered ecosystems. Here we describe data on abundances (percentage cover) of 247 alpine plant species (including mosses and lichens) inside nine 20 m by 20 m plots that were subdivided into 2 m by 2 m subplots. The nine plots are located in Gran Paradiso National Park, Italy. They cover three distinct alpine vegetation subtypes ('pure' natural grassland, sparsely vegetated 'rocky' grassland, and wetland) in each of three valleys (Bardoney, Colle de Nivolet and Levionaz) between 2200 and 2700 m a.s.l., i.e. above the treeline. The vegetation survey was conducted in 2015 at the peak of vegetation development during August. The dataset is provided as supplementary material and associated with the research article "Optimizing sampling effort and information content of biodiversity surveys: a case study of alpine grassland" [1]. Distribution and diversity of myxomycetes in Tiantangzhai National Forest Park, China.

Project description:Although myxomycetes are ubiquitous in terrestrial ecosystems, studies on their distribution and diversity in subtropical humid forests are still lacking. Field collections and moist chamber cultures were conducted from May to October within a two-year period in the Tiantangzhai National Forest Park of China. A total of 1,492 records representing 73 species belonging to 26 genera were obtained, of which 243 records/37 species were from field collections, and 1,249 records/52 species were from moist chamber cultures. Among the specimens obtained by culturing, 896 records/38 species and 353 records/37 species were obtained from living bark and ground litter, respectively. ANOVA showed that the sampling months had significant impacts on collection of myxomycetes from field and those that inhabit litter. An LEfSe analysis indicated that Arcyria was significantly abundant in August, while Stemonitis and Physarum were more abundant in July when collected from field. An RDA analysis showed that temperature was the main factor that affected the litter-inhabiting myxomycetes. The ANOVA indicated that forest type was the significant factor for bark-inhabiting myxomycetes. Diderma effusum was primarily obtained from mixed forests, while Clastoderma debaryanum and Colloderma oculatum were more common in coniferous forests. The RDA analysis indicated that the vegetation, pH, water retention, and elevation were the primary factors that affected the bark-inhabiting myxomycetes.

Checklist of aquatic Diptera (Insecta) of Plitvice Lakes National Park, Croatia, a UNESCO world heritage site.

Project description:Studies on aquatic Diptera in the Plitvice Lakes National Park (Croatia) conducted in the last 50 years have produced 157 species and 7 taxa of aquatic Diptera placed in 13 families. Samples were collected at 25 sampling sites representing the four main types of karst aquatic habitats: spring, stream, tufa barriers and lakes. All records of all the aquatic families of Diptera in Plitvice Lakes NP are summarized, including previously unpublished data. Twelve species new for Plitvice Lakes NP are recorded for the first time, belonging to the families: Chironomidae - Labrundinia longipalpis (Goetghebuer, 1921), Nilothauma brayi (Goetghebuer, 1921), Potthastia longimanus Kieffer, 1922, Polypedilum (Polypedilum) nubeculosum (Meigen, 1804), Tanytarsus brundini Lindeberg, 1963; Dixidae - Dixella autumnalis (Meigen, 1838); Scathophagidae - Acanthocnema latipennis Becker, 1894 and Stratiomyidae - Oxycera pardalina Meigen, 1822, Oxycera limbata Loew, 1862, Oxycera turcica Ustuner & Hasbenli, 2004, Nemotelus pantherinus (Linnaeus, 1758), Oplodontha viridula (Fabricius, 1775). The most species-rich family was the Chironomidae with 62 species (and an additional seven taxa), followed by the Empididae with 22 species and Limoniidae with 19 species. The highest number of species was recorded in springs. The relatively low number of species in certain families and the complete absence of some aquatic families shows that further research into the aquatic Diptera in Plitvice Lakes NP is needed.

Legionella species diversity in an acidic biofilm community in Yellowstone National Park.

Project description:Legionella species are frequently detected in aquatic environments, but their occurrence in extreme, acidic, geothermal habitats has not been explored with cultivation-independent methods. We investigated a predominately eukaryotic algal mat community in a pH 2.7 geothermal stream in Yellowstone National Park for the presence of Legionella and potential host amoebae. Our analyses, using PCR amplification with Legionella-specific primers targeting 16S rRNA genes, detected four known Legionella species, as well as Legionella sequences from species that are not represented in sequence databases, in mat samples and cultivated isolates. The nonrandom occurrence of sequences detected at lower (30 degrees C) and higher (35 to 38 degrees C) temperatures suggests that natural thermal gradients in the stream influence Legionella species distributions in this mat community. We detected only one sequence, Legionella micdadei, from cultivated isolates. We cultured and sequenced partial 18S rRNA gene regions from two potential hosts, Acanthamoeba and Euglena species.

Discovering Hidden Diversity of Characins (Teleostei: Characiformes) in Ecuador's Yasuni National Park.

Project description:BackgroundManagement and conservation of biodiversity requires adequate species inventories. The Yasuní National Park is one of the most diverse regions on Earth and recent studies of terrestrial vertebrates, based on genetic evidence, have shown high levels of cryptic and undescribed diversity. Few genetic studies have been carried out in freshwater fishes from western Amazonia. Thus, in contrast with terrestrial vertebrates, their content of cryptic diversity remains unknown. In this study, we carried out genetic and morphological analyses on characin fishes at Yasuní National Park, in eastern Ecuador. Our goal was to identify cryptic diversity among one of the most speciose fish families in the Amazon region. This is the first time that genetic evidence has been used to assess the species content of the Napo Basin, one of the richest regions in vertebrate diversity.ResultsPhylogenetic analyses of partial mitochondrial 16S ribosomal RNA gene (∼600 pb) DNA sequences from 232 specimens of the family Characidae and its closest groups revealed eight candidate new species among 33 species sampled, representing a 24% increase in species number. Analyses of external morphology allowed us to confirm the species status of six of the candidate species.ConclusionsOur results show high levels of cryptic diversity in Amazonian characins. If this group is representative of other Amazonian fish, our results would imply that the species richness of the Amazonian ichthyofauna is highly underestimated. Molecular methods are a necessary tool to obtain more realistic inventories of Neotropical freshwater fishes.

Genetic diversity and origin of the feral horses in Theodore Roosevelt National Park.

Project description:Feral horses in Theodore Roosevelt National Park (TRNP) represent an iconic era of the North Dakota Badlands. Their uncertain history raises management questions regarding origins, genetic diversity, and long-term genetic viability. Hair samples with follicles were collected from 196 horses in the Park and used to sequence the control region of mitochondrial DNA (mtDNA) and to profile 12 autosomal short tandem repeat (STR) markers. Three mtDNA haplotypes found in the TRNP horses belonged to haplogroups L and B. The control region variation was low with haplotype diversity of 0.5271, nucleotide diversity of 0.0077 and mean pairwise difference of 2.93. We sequenced one mitochondrial genome from each haplotype determined by the control region. Two complete mtDNA sequences of haplogroup L were closely related to the mtDNA of American Paint horse. The TRNP haplotype B did not have close matches in GenBank. The phylogenetic test placed this sequence in a group consisting of two horses from China, one from Yakutia, and one from Italy raising a possibility of historical transportation of horses from Siberia and East Asia to North America. Autosomal STR loci were polymorphic and indicated that the TRNP horses were distinctly different from 48 major horse breeds. Heterozygosity, mean number of alleles, and other measures of diversity indicated that TRNP herd diversity was below that observed for most other feral herds and domestic breeds. Both mtDNA and STRs demonstrated that the existing genetic data sets of horses are insufficient to determine the exact origins of the TRNP horses. However, measures of nuclear and mitochondrial diversity have elucidated management needs. It is recommended that new genetic stock be introduced and that adaptive management principles are employed to ensure that unique mitochondrial lineages are preserved and genetic diversity is increased and maintained over time.
