Project description:BackgroundThe pathophysiology of cervical dystonia is still unclear. Recent evidence points toward a network disorder affecting several brain areas. The objective of this study was to assess the saccadic inhibition as a marker of corticostriatal function in cervical dystonia.MethodsWe recruited 31 cervical dystonia patients and 17 matched healthy controls. Subjects performed an overlap prosaccade, an antisaccade, and a countermanding task on an eye tracker to assess automatic visual response and response inhibition.ResultsCervical dystonia patients made more premature saccades (P = 0.041) in the overlap prosaccade task and more directional errors in the antisaccade task (P = 0.011) and had a higher rate of failed inhibition in the countermanding task (P = 0.001).ConclusionsThe results suggest altered saccadic inhibition in cervical dystonia, possibly as a consequence of dysfunctional corticostriatal networks. Further studies are warranted to confirm whether these abnormalities are affected by the available therapies and whether this type of impairment is found in other focal dystonias. © 2021 The Authors. Project description:Visual guidance of gait is an important skill for everyday mobility. While this has often been studied using eye-tracking techniques, recent studies have shown that visual exploration involves more than just the eye; head movement and potentially the whole body is involved for successful visual exploration. This study aimed to assess coordinative patterns associated with head movement and it was hypothesized that these patterns would span across the body, rather than being localized. Twenty-one (after exclusions) healthy young adult volunteers followed a treadmill walking protocol designed to elicit different types of head movements (no stimuli compared to stimuli requiring horizontal, vertical, and mixed gaze shifts). Principal Component Analysis was used to establish whole-body correlated patterns of marker movement (Principal Movements; PMs) related to the activity of the head. In total 37 higher order PMs were found to be associated with head movement, two of these showed significant differences between trials associated with strong head rotations in the horizontal and sagittal plane. Both of these were associated with a whole-body pattern of activity. An analysis of the higher order components revealed that exploratory head movements are associated with distinct movement patterns, which span across the body. This shows that visual exploration can produce whole-body movement patterns that have a potentially destabilizing influence. These findings shed new light on established results in visual search research and hold relevance for fall and injury prevention.

Project description:Tapping or clapping to an auditory beat, an easy task for most individuals, reveals precise temporal synchronization with auditory patterns such as music, even in the presence of temporal fluctuations. Most models of beat-tracking rely on the theoretical concept of pulse: a perceived regular beat generated by an internal oscillation that forms the foundation of entrainment abilities. Although tapping to the beat is a natural sensorimotor activity for most individuals, not everyone can track an auditory beat. Recently, the case of Mathieu was documented (Phillips-Silver et al. 2011 Neuropsychologia 49, 961-969. (doi:10.1016/j.neuropsychologia.2011.02.002)). Mathieu presented himself as having difficulty following a beat and exhibited synchronization failures. We examined beat-tracking in normal control participants, Mathieu, and a second beat-deaf individual, who tapped with an auditory metronome in which unpredictable perturbations were introduced to disrupt entrainment. Both beat-deaf cases exhibited failures in error correction in response to the perturbation task while exhibiting normal spontaneous motor tempi (in the absence of an auditory stimulus), supporting a deficit specific to perception-action coupling. A damped harmonic oscillator model was applied to the temporal adaptation responses; the model's parameters of relaxation time and endogenous frequency accounted for differences between the beat-deaf cases as well as the control group individuals.

Project description:BackgroundChildhood dystonia is a disorder that involves inappropriate muscle activation during attempts at voluntary movement. Few studies have investigated the muscle activity associated with dystonia in children, and none have done so in the hands.MethodsIn this study, we measured surface electromyographic activity in four intrinsic hand muscles while participants attempted to perform an isometric tracking task using one of the muscles.ResultsChildren with dystonia had greater tracking error with the task-related muscle and greater overflow to non-task muscles. Both tracking error and overflow correlated with the Barry-Albright Dystonia scale of the respective upper limb. Overflow also decreased when participants received visual feedback of non-task muscle activity.DiscussionWe conclude that two of the motor deficits in childhood dystonia--motor overflow and difficulties in actively controlling muscles--can be seen in the surface electromyographic activity of individual muscles during an isometric task. As expected from results in adults, overflow is an important feature of childhood dystonia. However, overflow may be at least partially dependent on an individual's level of awareness of their muscle activity. Most importantly, poor single-muscle tracking shows that children with dystonia have deficits of individual muscle control in addition to overflow or co-contraction. These results provide the first quantitative measures of the muscle activity associated with hand dystonia in children, and they suggest possible directions for control of dystonic symptoms.

Project description:The first steps in numerical cognition are usually done in conjunction with fingers. Following the assumption that abstract concepts stay associated with the sensory-motor information that was present during their acquisition and consolidation, mental number representations should always be associated with the respective finger counting components. We tested whether finger movements that imply finger counting actually prime the corresponding number concepts in adults. All participants counted number 1 with their thumb and incremented sequentially to number 5 with their pinky. In the experiment, participants sequentially and repeatedly pressed five buttons from thumb to pinky. Each button press triggered the visual presentation of a random number between 1 and 5 that had to be named aloud, resulting in 20% counting-congruent and 80% counting-incongruent finger-number mappings. Average naming latencies were significantly shorter for congruent than incongruent finger-number combinations. Furthermore, there was a distance effect where primes partly co-activated numerically close target numbers and with decreasing priming for more distant prime-target pairs. Overall, these results provide further evidence that number representations are strongly associated with finger counting experience, making fingers an effective tool for number comprehension.

Project description:We examined how people organize redundant kinematic control variables (finger joint configurations) while learning to make goal-directed movements of a virtual object (a cursor) within a low-dimensional task space (a computer screen). Subjects participated in three experiments performed on separate days. Learning progressed rapidly on day 1, resulting in reduced target capture error and increased cursor trajectory linearity. On days 2 and 3, one group of subjects adapted to a rotation of the nominal map, imposed either stepwise or randomly over trials. Another group experienced a scaling distortion. We report two findings. First, adaptation rates and memory-dependent motor command updating depended on distortion type. Stepwise application and removal of the rotation induced a marked increase in finger motion variability but scaling did not, suggesting that the rotation initiated a more exhaustive search through the space of viable finger motions to resolve the target capture task than did scaling. Indeed, subjects formed new coordination patterns in compensating the rotation but relied on patterns established during baseline practice to compensate the scaling. These findings support the idea that the brain compensates direction and extent errors separately and in computationally distinct ways, but are inconsistent with the idea that once a task is learned, command updating is limited to those degrees of freedom contributing to performance (thereby minimizing energetic or similar costs of control). Second, we report that subjects who learned a scaling while moving to just one target generalized more narrowly across directions than those who learned a rotation. This contrasts with results from whole-arm reaching studies, where a learned scaling generalizes more broadly across direction than rotation. Based on inverse- and forward-dynamics analyses of reaching with the arm, we propose the difference in results derives from extensive exposure in reaching with familiar arm dynamics versus the novelty of the manual task.

Project description:Movements of the upper limb are controlled mostly through the contralateral hemisphere. Although overall activity changes in the ipsilateral motor cortex have been reported, their functional significance remains unclear. Using human functional imaging, we analyzed neural finger representations by studying differences in fine-grained activation patterns for single isometric finger presses. We demonstrate that cortical motor areas encode ipsilateral movements in 2 fundamentally different ways. During unimanual ipsilateral finger presses, primary sensory and motor cortices show, underneath global suppression, finger-specific activity patterns that are nearly identical to those elicited by contralateral mirror-symmetric action. This component vanishes when both motor cortices are functionally engaged during bimanual actions. We suggest that the ipsilateral representation present during unimanual presses arises because otherwise functionally idle circuits are driven by input from the opposite hemisphere. A second type of representation becomes evident in caudal premotor and anterior parietal cortices during bimanual actions. In these regions, ipsilateral actions are represented as nonlinear modulation of activity patterns related to contralateral actions, an encoding scheme that may provide the neural substrate for coordinating bimanual movements. We conclude that ipsilateral cortical representations change their informational content and functional role, depending on the behavioral context.

Project description:The ability to imagine the future (prospection) relies on many of the same brain regions that support memory for the past. To date, scientific research has primarily focused on the neural substrates of episodic forms of prospection (mental simulation of spatiotemporally specific future events); however, little is known about the neural substrates of semantic prospection (mental simulation of future nonpersonal facts). Of particular interest is the role of the medial temporal lobes (MTLs), and specifically the hippocampus. Although the hippocampus has been proposed to play a key role in episodic prospection, recent evidence suggests that it may not play a similar role in semantic prospection. To examine this possibility, amnesic patients with MTL lesions were asked to imagine future issues occurring in the public domain. The results showed that patients could list general semantic facts about the future, but when probed to elaborate, patients produced impoverished descriptions that lacked semantic detail. This impairment occurred despite intact performance on standard neuropsychological tests of semantic processing and did not simply reflect deficits in narrative construction. The performance of a patient with damage limited to the hippocampus was similar to that of the remaining patients with MTL lesions and amnesic patients' impaired elaboration of the semantic future correlated with their impaired elaboration of the semantic past. Together, these results provide novel evidence from MTL amnesia that memory and prospection are linked in the semantic domain and reveal that the MTLs play a critical role in the construction of detailed, multi-element semantic simulations.

Project description:When performing a goal-directed action with a tool, it is generally assumed that the point of control of the action system is displaced from the hand to the tool, implying that body and tool function as one system. Studies of how actions with tools are performed have been limited to studying either end-effector kinematics or joint-angle coordination patterns. Because joint-angle coordination patterns affect end-effector kinematics, the current study examined them together, with the aim of revealing how body and tool function as one system. Seated participants made point-to-point movements with their index finger, and with rods of 10, 20, and 30 cm attached to their index finger. Start point and target were presented on a table in front of them, and in half of the conditions a participant displacement compensated for rod length. Results revealed that the kinematics of the rod's tip showed higher peak velocity, longer deceleration time, and more curvature with longer rods. End-effector movements were more curved in the horizontal plane when participants were not displaced. Joint-angle trajectories were similar across rod lengths when participants were displaced, whereas more extreme joint-angles were used with longer rods when participants were not displaced. Furthermore, in every condition the end-effector was stabilized to a similar extent; both variability in joint-angle coordination patterns that affected end-effector position and variability that did not affect end-effector position increased in a similar way vis-à-vis rod length. Moreover, the increase was higher in those conditions, in which participants were not displaced. This suggests that during tool use, body and tool are united in a single system so as to stabilize the end-effector kinematics in a similar way that is independent of tool length. In addition, the properties of the actual trajectory of the end-effector, as well as the actual joint-angles used, depend on the length of the tool and the specifics of the task.

Project description:Finger-tapping experiments were conducted to examine whether the dynamics of intrapersonal and interpersonal coordination systems can be described equally by the Haken-Kelso-Bunz model, which describes inter-limb coordination dynamics. This article reports the results of finger-tapping experiments conducted in both systems. Two within-subject factors were investigated: the phase mode and the number of fingers. In the intrapersonal experiment (Experiment 1), the participants were asked to tap, paced by a gradually hastening auditory metronome, looking at their fingers moving, using the index finger in the two finger condition, or the index and middle finger in the four-finger condition. In the interpersonal experiment (Experiment 2), pairs of participants performed the task while each participant used the outside hand, tapping with the index finger in the two finger condition, or the index and middle finger in the four-finger condition. Some results did not agree with the HKB model predictions. First, from Experiment 1, no significant difference was observed in the movement stability between the in-phase and anti-phase modes in the two finger condition. Second, from Experiment 2, no significant difference was found in the movement stability between the in-phase and anti-phase mode in the four-finger condition. From these findings, different coordination dynamics were inferred between intrapersonal and interpersonal coordination systems against prediction from the previous studies. Results were discussed according to differences between intrapersonal and interpersonal coordination systems in the availability of perceptual information and the complexity in the interaction between limbs derived from a nested structure.
