Identification of new cytotypes of Valeriana jatamansi Jones, 1970 (Valerianaceae) from North-Western Himalayan region of India.

ABSTRACT: Valeriana jatamansi, a medicinally important species of the family Valerianaceae, has been cytologically studied in different geographical areas of North-Western Himalayan region of India. The tetraploid cytotype with chromosome numbers 2n=32 is in conformity with the earlier reports of the species from different parts of the world. An octoploid cytotype (2n=64) makes a new addition for the species on a worldwide basis, whereas the diploid cytotype (2n=16) is new to India have been reported for the first time in India. These cytotypes (2n=16, 32, 64) show significant variations with respect to morphology as well as geographical distribution in the Western Indian Himalayas. Further, anomalous populations have been marked with meiotic abnormalities in the form of cytomixis, chromosomal stickiness, unoriented bivalents, formation of laggards and bridges resulting in abnormal microsporogenesis, and production of heterogeneous-sized fertile pollen grains along with reduced pollen fertility.

Amanitatullossiana, a new species, and two new records of AmanitasectionLepidella from north-western Himalaya, India.

Project description:Amanitatullossiana, a new species of Amanita [subgenus Lepidella] section Lepidella from India is described. The species is characterised by its ash grey to brownish-grey pileus covered with dark grey to greyish-black universal veil remnants, the upper part of its rooting stipe base covered by several rows of recurved scales, broadly ellipsoid to ellipsoid basidiospores, absence of basidial clamp connections and pileal remnants of universal veil comprising abundant, disordered inflated cells intermixed with scattered filamentous hyphae. Molecular phylogenetic analysis and morphology both support the association of A.tullossiana with species of Bas' stirps Cinereoconia - A.cinereoconia and A.griseoverrucosa. Two species, A.griseoverrucosa and A.virgineoides are reported here as new records for India.

Gliophorusglutinosus sp. nov. (Hygrophoraceae, Agaricales) from Eastern Himalayan region of India.

Project description:An interesting species of the genus Gliophorus (sect.Glutinosae), collected from Sikkim Himalaya in India, is described here as G.glutinosus sp. nov. after thorough morphological examination and phylogenetic analysis. The species is mainly characterised by its strongly glutinous basidiomata throughout, particularly on the twisted stipe, decurrent lamellae with glutinous edge, gelatinised cheilocystidia, presence of pleuropseudocystidia and absence of clamps in hyphae of the pileipellis. This communication includes detailed morphological description, illustrations, comparison with the allied taxa, nrITS based phylogeny of this novel taxon and a key to the species under Gliophorussect.Glutinosae.

Assessment of bias in morphological identification of carnivore scats confirmed with molecular scatology in north-eastern Himalayan region of Pakistan.

Project description:Scats are often used to study ecological parameters of carnivore species. However, field identification of carnivore scats, based on their morphological characteristics, becomes difficult if many carnivore species are distributed in the same area. We assessed error rates in morphological identification of five sympatric carnivores' scats in north-eastern Himalayan region of Pakistan during 2013-2017. A sample of 149 scats were subjected to molecular identification using fecal DNA. We used a confusion matrix to assess different types of errors associated with carnivore scat identification. We were able to amplify DNA from 96.6% (n = 144) of scats. Based on field identification of carnivore scats, we had predicted that out of 144 scats: 11 (7.6%) scats were from common leopard, 38 (26.4%) from red fox, 29 (20.1%) from Asiatic jackal, 37 (25.7%) from yellow throated martin, 14 (9.7%) from Asian palm civet and 15 (10.4%) from small Indian civet. However, molecular identification revealed and confirmed nine were scats (6.24%) from common leopard, 40 (27.8 %) from red fox, 21 (14.6%) from Asiatic jackal, 45 (31.25%) from Asian palm civet, 12 (8.3%) scats from small Indian civet, while 11 scats (7.6%) were found from Canis lupus Spp., three (2%) from dog, one (0.7 %) scat sample from porcupine, and two (1.4%) from rhesus monkey. Misidentification rate was highest for Asian palm civet (25.7%), followed by red fox (11.1%) and Asiatic jackal (9.7%) but least for common leopard scats (4.2%). The results specific to our study area concur with previous studies that have recommended that carnivore monitoring programs utilize molecular identification of predator scats. Using only morphological identification of scats can be misleading and may result in wrong management decisions.

Detection and molecular characterization of chicken infectious anaemia virus in young chicks in Punjab region of north-western India.

Project description:Chicken infectious anaemia (CIA) is an important viral disease of chicken causing significant immunosuppression and severe anaemia worldwide. Occurrence of severe disease and mortality is noticed in young chicks (2-3 weeks). Vertical mode of transmission increases chance of infection and persistence of virus among the infected flocks. The current study was conducted in Punjab state for confirmation and genetic characterization of CAV among chicken flocks of various poultry farms. DNA was extracted from the tissue samples and subjected to polymerase chain reaction (PCR) of VP1 gene and whole genome. PCR products were further sequenced for confirmation of chicken infectious anaemia virus (CIAV) genome in the clinical samples. PCR amplification of DNA from the tissue samples yielded expected product size of 1350 bases of VP1 gene and 2.3 kb of whole genome. Out of 16 commercial poultry farms, 11 were confirmed with presence of CIAV, and out of 65 birds, 39 were found positive (60%) for CIAV genes. Among the various organs, the presence of viral gene was detected at highest level in thymus when compared with other organs. It is concluded that chicken infectious anaemia virus detected from Punjab state is closely related to other Indian isolates and neighbouring countries which necessitates need of more intensive studies with a greater number of samples for implementing effective control measures.

A review of the genus Lordiphosa Basden in India, with descriptions of four new species from the Himalayan region (Diptera, Drosophilidae).

Project description:All Indian species of the genus Lordiphosa Basden are reviewed, with descriptions of four new species, L. curva Fartyal & Toda, sp. n. of the denticeps species group and L. ayarpathaensis Kandpal & Singh, sp. n., L. makaibarensis Pradhan & Chatterjee, sp. n. and L. srinagarensis Sati & Fartyal, sp. n. of the nigricolor species group. Two of the new species, L. ayarpathaensis and L. makaibarensis, were found visiting flowers of Hedychium spicatum and Datura suaveolens, respectively. This is the first record of flower visitation in Lordiphosa flies. In addition, L. parantillaria (Kumar & Gupta, 1990), syn. n. is synonymized with L. antillaria (Okada, 1984). Supplementary and revised descriptions for L. antillaria and L. neokurokawai (Singh & Gupta, 1981) and a key to all Indian species of Lordiphosa are provided.

Comparative litter decomposability traits of selected native and exotic woody species from an urban environment of north-western Siwalik region, India.

Project description:Exotic plants can potentially modify ecosystem functions like cycling of nutrients by adjusting their decomposition rates. However, these effects are largely unknown for urban ecosystems, though they act as reservoirs of exotic plants. The present study evaluated the decomposition rates of five native and five exotic (three invasive and two non-invasive) species by conducting the litter bag experiment. Our study, however, did not find any significant differences in overall decomposition rates of native and exotic species but decomposition rates were strongly correlated with initial chemical quality of the litter. Further, litter carbon, lignin to nitrogen ratio and carbon to nitrogen ratio seemed to be good predictors for decomposition rates in this study. Interestingly, invasive exotic species had higher decomposition rate while non-invasive exotic species showed a slower rate as compared to the native species. In conclusion, our study indicates that invasive exotic plants try to maintain a higher chemical quality of litter than native and non-invasive exotic species which promotes their rapid decomposition. Thus, the better chemical quality of litter may facilitate the naturalisation and invasion of exotic plants irrespective of their origin.

Bio-prospecting Bacterial Diversity of Hot Springs in Northern Himalayan Region of India for Laccases.

Project description:Bacterial diversity of hot springs of northern Himalayan region of India was studied and explored for laccases, the multicopper enzymes applicable in a large number of industries due to their ability to utilize a wide range of substrates. 220 operational taxonomic units (OTUs) out of 5551 sequence reads for bacterial diversity and 3 OTUs out of 19 sequence reads for Laccase like multicopper oxidases (LMCOs) diversity were generated. Bacteroidetes (74.28%) was the most abundant phylum including genus Paludibacter (66.96%), followed by phylum Proteobacteria (24.53%) including genera Chitinilyticum (7.55%) and Cellvibrio (6.14%). In case of laccase diversity, three LMCO sequences showed affiliation with proteobacteria and one with two domain laccase from uncultivable bacteroidetes. LMCO sequences belonged to H and N families.

North-Western Himalayan Common Beans: Population Structure and Mapping of Quantitative Anthracnose Resistance Through Genome Wide Association Study.

Project description:Common bean (Phaseolus vulgaris L.) is an important legume crop of north-western (NW) Himalayan region and the major disease that causes catastrophic loss to the crop is anthracnose, which is caused by Colletotrichum lindemuthianum. The pathogen is highly diverse and most of the commercial cultivars are susceptible to different races prevalent in the region. The lack of information on the genomic regions associated with anthracnose resistance in NW Himalayan common bean population prompted us to dissect Quantitative Resistance Loci (QRLs) against major anthracnose races. In this study, 188 common bean landraces collected from NW region were screened against five important anthracnose races and 113 bean genotypes showed resistance to one or multiple races. Genotyping by sequencing (GBS) was performed on a panel of 192 bean lines (4 controls plus 188 Indian beans) and 22,589 SNPs were obtained that are evenly distributed. Population structure analysis of 192 bean genotypes categorized 188 Indian beans into two major clusters representing Andean and Mesoamerican gene pools with obvious admixtures. Many QRLs associated with anthracnose resistance to Indian C. lindemuthianum virulences (race 3, 87, and 503) are located at Pv04 within the gene models that encode typical resistance gene signatures. The QRLs associated with race 73 are located on Pv08 and overlaps with Co-4 anthracnose resistance gene. A SNP located at distal end of Pv11 in a gene model Phvul.011G202300 which encodes a LRR with a typical NB-ARC domain showed association with race 73 resistance. Common bean genomic regions located at Pv03, Pv09, and Pv11 showed association with resistance to anthracnose race 2047. The present study showed presence of many novel bean genomic regions associated with anthracnose resistance. The presence of Co-4 and Co-2 genes in our material is encouraging for breeding durable anthracnose resistant cultivars for the region.

Project description:A new species of Vigna Savi, subgenus Ceratotropis (Piper) Verdc., Vignayadavii S.P. Gaikwad, R.D. Gore, S.D. Randive & K.U. Garad, sp. nov. is described and illustrated here. It is morphologically close to Vignadalzelliana (Kuntze) Verdc. but differs in its underground obligate cleistogamous flowers on positively geotropic branches, hairy calyx, small corolla, linear style beak and dimorphic seeds with shiny seed coat.

Project description:The present study employed 16S rRNA amplicon sequencing to survey the prokaryotic microbiota on Farpuk Cave, revealing a diverse bacterial community with 4,021 operational taxonomical units (OTUs), mainly dominated by the genus Rhodococcus. Moreover, 18.17% of the OTUs were unclassified at the phylum level, suggesting the existence of novel bacterial species.
