Project description:Chironomids are one of the most abundant aquatic insects and are widely distributed in various biological communities. However, the lack of high-quality genomes has hindered our ability to study the evolution and ecology of this group. Here, we used Nanopore long reads and Hi-C data to produce two chromosome-level genomes from mixed genomic data. The genomes of Smittia aterrima (SateA) and Smittia pratorum (SateB) were assembled into three chromosomes, with sizes of 78.45 Mb and 71.56 Mb, scaffold N50 lengths of 25.73 and 23.53 Mb, and BUSCO completeness of 98.5% and 97.8% (n = 1,367), 5.68 Mb (7.24%) and 1.94 Mb (2.72%) of repetitive elements, and predicted 12,330 (97.70% BUSCO completeness) and 11,250 (97.40%) protein-coding genes, respectively. These high-quality genomes will serve as valuable resources for comprehending the evolution and environmental adaptation of chironomids.

Project description:The karyotype features and gene COI sequence of Chironomusheteropilicornis Wülker, 1996 from the Gydan Peninsula are presented for the first time. Nine banding sequences were determined, eight of them hpiA2, hpiB1, hpiC1, hpiC2, hpiD1, hpiE1, hpiF3 and hpiG1 were previously known from European, Georgian (South Caucasus) and Siberian populations. One new banding sequence for Ch.heteropilicornis, hpiB2, was found. The hpiA2 banding sequence was found in all individuals, and this is its second finding after the Georgian population (Karmokov 2019). The hpiF3 banding sequence was found only in the homozygous state. Additional B-chromosomes are absent. The genetic distances (K2P) between Ch.heteropilicornis COI gene sequence from Gydan Peninsula and Norway are 1.1--1.3%, and Georgia - 1.8%, much lower than the commonly accepted threshold of 3% for species of genus Chironomus Meigen, 1803. The phylogenetic tree for COI gene sequences estimated by Bayesian inference showed geographically determined clusters of Norway and Gydan and a separate lineage of the Georgian population of Ch.heteropilicornis. The analysis of karyotype and COI gene sequences shows that the population of Ch.heteropilicornis from the Gydan Peninsula has an intermediate position within the Ch.pilicornis group between Georgian, Yakutian and Norwegian populations. The position of Ch.pilicornis Fabricius, 1787 from Canada and Greenland on the phylogenetic tree is discussed.

Project description:Members of the genus Parachironomus Lenz known from China and Japan are revised, and a key to their male adults is given. Parachironomuspoyangensis sp. n. is described in this life stage. Parachironomusfrequens (Johannsen) and Parachironomusmonochromus (van der Wulp) are recorded from China for the first time, thus are redescribed from Chinese specimens. Parachironomuskamaabeus Sasa & Tanaka and Parachironomustoneabeus Sasa & Tanaka are new junior synonyms of Parachironomusfrequens. Three Chinese or Japanese species formerly placed in Parachironomus are transferred to other genera, resulting in the new combinations Cryptochironomusinafegeus (Sasa, Kitami & Suzuki), Demicryptochironomus (Irmakia) lobus (Yan, Sæther, Jin & Wang), and Microchironomuslacteipennis (Kieffer). Chironomussauteri Kieffer, Parachironomuskisobilobalis Sasa & Kondo and Parachironomuskuramaexpandus Sasa are removed from Parachironomus; the last of these three denotes a valid species of uncertain generic placement, the first two are nomina dubia.

Project description:Non-biting midges of the fly family Chironomidae are extremely abundant and diverse in Arctic regions and are essential components of Arctic ecosystems. Modern identification tools based on documented records of Arctic chironomid species are therefore important for ecological research and environmental monitoring in the region. Here, we provide an updated review of the chironomid fauna of the Svalbard archipelago and the island of Jan Mayen, Norway. Our results show that a total of 73 species distributed across 24 genera in four subfamilies are known from these areas. Our review treats 109 taxa, including nomina dubia and misidentifications. It includes morphological identification keys to all known species as well as photographs of most taxa and DNA barcodes of 66 species. Taxonomic remarks are given for selected taxa, including previous misidentifications and erroneous records. Chironomus islandicus, Tvetenia bavarica, Limnophyes schnelli, Metriocnemus brusti and Metriocnemus fuscipes as well as the genera Allocladius, Corynoneura and Bryophaenocladius are reported from Svalbard for the first time, while Procladius (Holotanypus) frigidus, Stictochironomus psilopterus, Chaetocladius incertus, Orthocladius (Orthocladius) mixtus and Smittia longicosta, previously considered as junior synonyms or nomina dubia, are revived as valid species based on examination of type material or literature. Twenty species within eleven genera are introduced with interim names. Metriocnemus similis is regarded as a junior synonym of Metriocnemus ursinus, and Smittia incerta, Smittia flexinervis and Smittia spitzbergensis are regarded as nomina dubia. Valid taxa no longer considered as part of the Svalbard fauna are Parochlus kiefferi, Arctopelopia barbitarsis, Procladius (Holotanypus) crassinervis, Diamesa lindrothi, Diamesa incallida, Diamesa lundstromi, Chironomus hyperboreus, Sergentia coracina, Camptocladius stercorarius, Chaetocladius dissipatus, Chaetocladius dentiforceps, Chaetocladius laminatus, Chaetocladius perennis, Cricotopus (Cricotopus) humeralis, Cricotopus (Cricotopus) polaris, Hydrosmittia ruttneri, Limnophyes edwardsi, Metriocnemus picipes, Metriocnemus tristellus, Orthocladius (Eudactylocladius) gelidus, Orthocladius (Euorthocladius) thienemanni, Orthocladius (Orthocladius) obumbratus, Orthocladius (Orthocladius) rhyacobius, Paralimnophyes, Paraphaenocladius impensus, Psectrocladius (Monopsectrocladius) calcaratus, Psectrocladius (Psectrocladius) psilopterus, Psectrocladius (Psectrocladius) ventricosus, Smittia lasiophthalma, Smittia lasiops and Zalutschia tatrica.

Project description:The genus Cladosporium is restricted to dematiaceous hyphomycetes with a coronate scar type, and Davidiella teleomorphs. In the present study numerous cladosporium-like taxa are treated, and allocated to different genera based on their morphology and DNA phylogeny derived from the LSU nrRNA gene. Several species are introduced in new genera such as Hyalodendriella, Ochrocladosporium, Rachicladosporium, Rhizocladosporium, Toxicocladosporium and Verrucocladosporium. A further new taxon is described in Devriesia (Teratosphaeriaceae). Furthermore, Cladosporium castellanii, the etiological agent of tinea nigra in humans, is confirmed as synonym of Stenella araguata, while the type species of Stenella is shown to be linked to the Teratosphaeriaceae (Capnodiales), and not the Mycosphaerellaceae as formerly presumed.

Project description:The karyotypes of six African Chironomus species (Chironomus alluaudi Kieffer, 1913, Chironomus transvaalensis Kieffer, 1923, Chironomus sp. Nakuru, Chironomus formosipennis Kieffer, 1908, Chironomus prope pulcher Wiedemann, 1830, Chironomus sp. Kisumu) were investigated; four of these karyotypes were described for the first time (Chironomus sp. Nakuru, Chironomus formosipennis, Chironomus prope pulcher, Chironomus sp. Kisumu). Of the six Chironomus karyotypes, three had "pseudothummi" cytocomplex chromosome arms combinations AE CD BF G (Chironomus alluaudi, Chironomus transvaalensis, Chironomus sp. Nakuru), two had "thummi"cytocomplex arms combinations AB CD EF G (Chironomus formosipennis, Chironomus prope pulcher), and one had "parathummi"armcombinations AC BF DE G (Chironomus sp. Kisumu). Thus, three of the ten main cytocomplexes known were detected in Africa. Detailed photomaps of all chromosome arms, with the exception of arms B and G, were prepared for the karyotypes of Chironomus alluaudi, Chironomus transvaalensis, Chironomus sp. Nakuru, Chironomus prope pulcher; the karyotypes of Chironomus formosipennis, Chironomus sp. Kisumucould only be fragmentarily mapped. Endemic African banding sequences were characteristic for most of the chromosomal arms in all species studied. However, basic sequences, which can be present in different Chironomus species on different continents (Wülker, 1980; Kiknadze et al. 2008), were also detected also in several African species (Chironomus alluaudi, Chironomus sp. Nakuru, and Chironomus formosipennis). The banding sequences of African species studied allow discussion of the derivation of modern banding patterns from hypothetical species, living before separation of cytocomplexes and continents.

Project description:Chironomids show a wide distribution and can occupy several habitats due to their high adaptive capacity in different freshwater environments. The genus Polypedilum is found along a wide elevational and environmental gradient in the neotropics, and its genetic variability could help to elucidate factors determining its distribution and tolerance to the environmental changes of different species or populations. This study examines the genetic variability of Polypedilum in an important biogeographic area that acts as a geographical barrier of biodiversity at the border of the Choco and Tumbes biomes. We identified five Polypedilum morphotypes using classic taxonomic methods. We examined 68 Polypedilum individuals from eight sampling sites in El Oro Province, Ecuador, analyzing the putative molecular species using the cytochrome c oxidase subunit 1 (CO1) mitochondrial gene fragment. Then, we calculated molecular diversity indices, Haplotype diversity (Hd), and θs and θπ estimators. Seven Polypedilum OTUs were determined from which a high molecular diversity was registered. A CCA was conducted to understand the population composition in relation to environmental characteristics. Results indicated that dissolved oxygen and temperature are the main environmental factors affecting Polypedilum distribution across elevational gradients and between basins.

Project description:A new species of Denopelopia from Brazil is described based on adult male and pupa. The male of the new species can be distinguished from all other species of the genus by the genitalia and fore-tibial spur morphology. The pupa is very similar to those of Denopelopia atria, but it can be distinguished by the absence of distinct constrictions in the respiratory atrium of the thoracic horn. Generic diagnosis to male and pupa of Denopelopia is emended and keys to male and pupae of known species are provided.

Project description:The species Chironomus sp. "Florida" has several qualities that make it a potential aquatic laboratory model to be used in Puerto Rico. Its use as such, however, requires a rearing protocol and life cycle description not previously reported. The present study addresses this lack of information by first describing a rearing method obtained through three years of observations. Next we describe and discuss the life cycle and the effects of temperature and feeding on development. The species has a short life cycle (typically 11 days) and larval stages easily identified using body measurements. Temperature affects the duration of the life cycle, with warm temperatures producing faster development than cold temperatures. The effects of different food concentrations vary: in large water volumes, concentrations of 2 mg/larva/day produce faster developmental times, but at low water volumes, small food concentrations of 0.5 mg/larva/day produce faster developmental times. The rearing protocol and life cycle parameters presented in this study are intended to promote the use of this species as a laboratory model. The fast development of Chironomus sp. "Florida" makes it ideal for toxicological studies.
