Project description:Previous published evidence for the occurrence of an exotic armadillo tick, Amblyomma auricularium (Conil), in Florida is scant, but we found it is fully established and integrated into the state's tick fauna. We collected 11,192 specimens of this tick from naturalized nine-banded armadillos, Dasypus novemcinctus L., and 14 other species of wild native mammals and birds in Florida, while sampling statewide during 2004 through 2007. In all, we document its specific presence only in 14 contiguous South Florida counties. Moreover, we report the first collections of A. auricularium from the Virginia opossum (Didelphis virginiana Kerr), common raccoon [Procyon lotor (L.)], cotton deermouse [Peromyscus gossypinus (Le Conte)], gray fox [Urocyon cinereoargenteus (Schreber)], eastern spotted skunk [Spilogale putorius (L.)], and white-tailed deer [Odocoileus virginianus (Zimmerman)]. For the first time on birds, we report the collection of this tick from the broad-winged hawk [Buteo platypterus (Vieillot)], northern cardinal [Cardinalis cardinalis (L.)], Carolina wren [Thryothorus ludovicianus (Latham)], gray catbird [Dumetella carolinensis (L.)], and yellow-rumped warbler [Setophaga coronata (L.)]. In addition, we report unattached A. auricularium collected from humans for the first time, and additional new collections from domestic dogs, Canis lupus familiaris L.

Project description:New records of 14 stygobiont crustacean species pertaining to six Malacostraca orders from 32 cenotes are presented, with their associated caves of the state of Yucatan, Mexico, together with an individual account for each species. Species composition of most of the investigated cenotes is examined for the first time. A thermosbaenacean and two amphipod species were not formally recorded to the cenote ecosystems of the state of Yucatan prior to our research. Distribution data of a cirolanid isopod previously known only from its type locality is also provided. Barcodes of mitochondrial cytochrome c oxidase subunit I for the reported peracarid species previously lacking this information have been included in present study as tools for species identification and a baseline of further molecular genetic analyses.

Project description:Over 4,950 asymptomatic weed samples from more than 20 weed species that are host plants for curtoviruses were collected from ten chile pepper fields in southern New Mexico (NM) during 2003, 2004 and 2005 to identify whether they were infected with curtoviruses and to determine which curtoviruses were distributed in the weed population. Polymerase chain reaction using primers designed to detect a portion of the coat protein (cp) gene were used to detect curtoviruses, and infected plants were further tested for specific curtoviruses using primers designed to detect to a portion of the replication-associated protein (rep) gene. Amplification of the cp gene was successful from 3.7, 1.17, and 1.9% of the weed samples in 2003, 2004, and 2005, respectively. Seventy-three amplicons from those samples were sequenced and compared to well-characterized curtoviruses. Analysis of the rep nucleotide sequences showed that approximately 32.9% of the weed isolates tested were closely related to beet mild curly top virus (BMCTV). Approximately 12.4% were closely related to beet severe curly top virus (BSCTV). The rest of the weed isolates (54.7%), which shared a very high level of nucleotide sequence identity to each other, represent a new curtovirus species. Using eight primers designed for PCR, complete genomes of three curtoviruses isolated from chile pepper samples representing the three groups of curtoviruses in southern New Mexico were sequenced. Comparisons of whole sequences of the genomes revealed that the DG2SW171601 isolate (2,929 nucleotides) was nearly identical to BMCTV-W4 (approximately 98% nucleotide sequence identity). The LRME27601 isolate (2,927 nucleotides) was most closely related to BSCTV (approximately 92% nucleotide sequence identity). The LJN17601 isolate (2,959 nucleotides) shared only from 49.9 to 88.8% nucleotide sequence identity with other well-characterized curtoviruses. Based on the accepted cut-off of 89%, we propose that the LJN17601 isolate is a member of a new curtovirus species. Chile peppers infected with this virus in the field express chlorotic stunting symptoms, so we propose the name pepper yellow dwarf virus (PeYDV). This new curtovirus species may be the result of mutations in the genome and recombination between BMCTV-W4 and BSCTV.

A key to the Iranian species of Nenteria is also presented, and Trachycilliba abantica (Bal & Őzkan, 2007) is reported for the first time in Iran.

Project description:This work includes taxonomic data on four species of oribatid mites of the family Otocepheidae from Yunnan, China. Two new species of the genera Basiceramerus and Eurostocepheus are described, respectively. Basiceramerusovatus sp. nov. differs from B.bangladeshensis Corpuz-Raros & Gruèzo, 2008 by the wavy marginalis passing the base of the adanal setae, prodorsal condyles well separated from median ones, a ventral ridge present, and the anal plate foveolate; it differs from B.igorotus Corpuz-Raros & Gruèzo, 2011 from Vietnam by the wavy marginalis passing the base of the adanal setae, a connected tutorium and lamelliform expansion, a ventral ridge present, a smooth genital plate, and seta an 2 located close to the median margin of the anal opening; it differs from B.igorotus from the Philippines by the lamellar setae inserted behind the tip of the lamella, separated prodorsal condyles, lyrifissure im posterior to gla, genital plate smooth, anal plate foveolate, and the wavy marginalis passing the base of the adanal setae. Eurostocepheus (Eurostocepheus) sinutussp. nov. differs from other known species of this genus by having a ventral groove between the genital aperture and the ventral ridge, eight pairs of notogastral setae, and distinctly shorter and thinner notogastral setae p 1, p 2, p 3, h 3. Eurostocepheus (Eurostocepheus) aquilinus Aoki, 1965 and E. (E.) mahunkai Mondal & Kundu, 1999 are reported for the first time from China.

Project description:Amblyomma maculatum Koch sensu lato (s.l.) ticks are the vector of Rickettsia parkeri in Arizona, where nine cases of R. parkeri rickettsiosis have been identified since the initial case in 2014. The current study sought to better define the geographic ranges of the vector and pathogen and to assess the potential public health risk posed by R. parkeri in this region of the southwestern United States. A total of 275 A. maculatum s.l. ticks were collected from 34 locations in four counties in Arizona and one county in New Mexico and screened for DNA of Rickettsia species. Rickettsia parkeri was detected in 20.4% of the ticks, including one specimen collected from New Mexico, the first report of R. parkeri in A. maculatum s.l. from this state. This work demonstrates a broader distribution of A. maculatum s.l. ticks and R. parkeri in the southwestern United States than appreciated previously to suggest that R. parkeri rickettsiosis is underrecognized in this region.

Project description:Dermacentor (Indocentor) auratus Supino, 1897 is a prominent ixodid vector of numerous pathogens of public health and veterinary importance. Using long-range PCR of two overlapping regions sequenced on an Illumina MiSeq machine, the complete mitochondrial genome of D. auratus is reported here. The resulting contigs were able to be assembled into a complete and circularised genome which had the general organisation of the mitochondrial genomes of the Metastriates. It had a total length of 14,766 bp and contained 37 genes, including 13 protein-coding genes, 22 transfer RNA genes, and 2 ribosomal RNA genes, as well as 2 non-coding control regions and 3 tick-boxes. The phylogenetic analysis on the whole mitogenome confirmed the position of D. auratus within the Dermacentor clade.

Project description:During a study to identify zoonotic pathogens in northwestern Mexico, we detected the presence of a rickettsial agent in Dermacentor parumapertus ticks from black-tailed jackrabbits (Lepus californicus). Comparison of 4 gene sequences (gltA, htrA, ompA, and ompB) of this agent showed 99%-100% identity with sequences of Rickettsia parkeri.

Project description:The American dog tick, Dermacentor variabilis, is a veterinary- and medically- significant tick species that is known to transmit several diseases to animal and human hosts. The spatial distribution of this species in North America is not well understood, however; and knowledge of likely changes to its future geographic distribution owing to ongoing climate change is needed for proper public health planning and messaging. Two recent studies have evaluated these topics for D. variabilis; however, less-rigorous modeling approaches in those studies may have led to erroneous predictions. We evaluated the present and future distribution of this species using a correlative maximum entropy approach, using publicly available occurrence information. Future potential distributions were predicted under two representative concentration pathway (RCP) scenarios; RCP 4.5 for low-emissions and RCP 8.5 for high-emissions. Our results indicated a broader current distribution of this species in all directions relative to its currently known extent, and dramatic potential for westward and northward expansion of suitable areas under both climate change scenarios. Implications for disease ecology and public health are discussed.
