Decoding intentions from movement kinematics.

ABSTRACT: How do we understand the intentions of other people? There has been a longstanding controversy over whether it is possible to understand others' intentions by simply observing their movements. Here, we show that indeed movement kinematics can form the basis for intention detection. By combining kinematics and psychophysical methods with classification and regression tree (CART) modeling, we found that observers utilized a subset of discriminant kinematic features over the total kinematic pattern in order to detect intention from observation of simple motor acts. Intention discriminability covaried with movement kinematics on a trial-by-trial basis, and was directly related to the expression of discriminative features in the observed movements. These findings demonstrate a definable and measurable relationship between the specific features of observed movements and the ability to discriminate intention, providing quantitative evidence of the significance of movement kinematics for anticipating others' intentional actions.

Single-trial decoding of movement intentions using functional ultrasound neuroimaging.

Project description:New technologies are key to understanding the dynamic activity of neural circuits and systems in the brain. Here, we show that a minimally invasive approach based on ultrasound can be used to detect the neural correlates of movement planning, including directions and effectors. While non-human primates (NHPs) performed memory-guided movements, we used functional ultrasound (fUS) neuroimaging to record changes in cerebral blood volume with 100 μm resolution. We recorded from outside the dura above the posterior parietal cortex, a brain area important for spatial perception, multisensory integration, and movement planning. We then used fUS signals from the delay period before movement to decode the animals' intended direction and effector. Single-trial decoding is a prerequisite to brain-machine interfaces, a key application that could benefit from this technology. These results are a critical step in the development of neuro-recording and brain interface tools that are less invasive, high resolution, and scalable.

Decoding social intentions in human prehensile actions: Insights from a combined kinematics-fMRI study.

Project description:Consistent evidence suggests that the way we reach and grasp an object is modulated not only by object properties (e.g., size, shape, texture, fragility and weight), but also by the types of intention driving the action, among which the intention to interact with another agent (i.e., social intention). Action observation studies ascribe the neural substrate of this 'intentional' component to the putative mirror neuron (pMNS) and the mentalizing (MS) systems. How social intentions are translated into executed actions, however, has yet to be addressed. We conducted a kinematic and a functional Magnetic Resonance Imaging (fMRI) study considering a reach-to-grasp movement performed towards the same object positioned at the same location but with different intentions: passing it to another person (social condition) or putting it on a concave base (individual condition). Kinematics showed that individual and social intentions are characterized by different profiles, with a slower movement at the level of both the reaching (i.e., arm movement) and the grasping (i.e., hand aperture) components. fMRI results showed that: (i) distinct voxel pattern activity for the social and the individual condition are present within the pMNS and the MS during action execution; (ii) decoding accuracies of regions belonging to the pMNS and the MS are correlated, suggesting that these two systems could interact for the generation of appropriate motor commands. Results are discussed in terms of motor simulation and inferential processes as part of a hierarchical generative model for action intention understanding and generation of appropriate motor commands.

Adults Do Not Distinguish Action Intentions Based on Movement Kinematics Presented in Naturalistic Settings.

Project description:Predicting others' actions is an essential part of acting in the social world. Action kinematics have been proposed to be a cue about others' intentions. It is still an open question as to whether adults can use kinematic information in naturalistic settings when presented as a part of a richer visual scene than previously examined. We investigated adults' intention perceptions from kinematics using naturalistic stimuli in two experiments. In experiment 1, thirty participants watched grasp-to-drink and grasp-to-place movements and identified the movement intention (to drink or to place), whilst their mouth-opening muscle activity was measured with electromyography (EMG) to examine participants' motor simulation of the observed actions. We found anecdotal evidence that participants could correctly identify the intentions from the action kinematics, although we found no evidence for increased activation of their mylohyoid muscle during the observation of grasp-to-drink compared to grasp-to-place actions. In pre-registered experiment 2, fifty participants completed the same task online. With the increased statistical power, we found strong evidence that participants were not able to discriminate intentions based on movement kinematics. Together, our findings suggest that the role of action kinematics in intention perception is more complex than previously assumed. Although previous research indicates that under certain circumstances observers can perceive and act upon intention-specific kinematic information, perceptual differences in everyday scenes or the observers' ability to use kinematic information in more naturalistic scenes seems limited.

Decoding moral judgments from neural representations of intentions.

Project description:Intentional harms are typically judged to be morally worse than accidental harms. Distinguishing between intentional harms and accidents depends on the capacity for mental state reasoning (i.e., reasoning about beliefs and intentions), which is supported by a group of brain regions including the right temporo-parietal junction (RTPJ). Prior research has found that interfering with activity in RTPJ can impair mental state reasoning for moral judgment and that high-functioning individuals with autism spectrum disorders make moral judgments based less on intent information than neurotypical participants. Three experiments, using multivoxel pattern analysis, find that (i) in neurotypical adults, the RTPJ shows reliable and distinct spatial patterns of responses across voxels for intentional vs. accidental harms, and (ii) individual differences in this neural pattern predict differences in participants' moral judgments. These effects are specific to RTPJ. By contrast, (iii) this distinction was absent in adults with autism spectrum disorders. We conclude that multivoxel pattern analysis can detect features of mental state representations (e.g., intent), and that the corresponding neural patterns are behaviorally and clinically relevant.

The kinematics of cyclic human movement.

Project description:Literature mentions two types of models describing cyclic movement-theory and data driven. Theory driven models include anatomical and physiological aspects. They are principally suitable for answering questions about the reasons for movement characteristics, but they are complicated and substantial simplifications do not allow generally valid results. Data driven models allow answering specific questions, but lack the understanding of the general movement characteristic. With this paper we try a compromise without having to rely on anatomy, neurology and muscle function. We hypothesize a general kinematic description of cyclic human motion is possible without having to specify the movement generating processes, and still get the kinematics right. The model proposed consists of a superposition of six contributions-subject's attractor, morphing, short time fluctuation, transient effect, control mechanism and sensor noise, while characterizing numbers and random contributions. We test the model with data from treadmill running and stationary biking. Applying the model in a simulation results in good agreement between measured data and simulation values. We find in all our cases the similarity analysis between measurement and simulation is best for the same subjects-[Formula: see text] and [Formula: see text]. All comparisons between different subjects are [Formula: see text] and [Formula: see text]. This uniquely allows for the identification of each measurement for the associated simulation. However, even different subject comparisons show good agreement between measurement and simulation results of differences ?run = 6.7±4.7% and ?bike = 5.1±4.5%.

Atypical basic movement kinematics in autism spectrum conditions.

Project description:Individuals with autism spectrum conditions have difficulties in understanding and responding appropriately to others. Additionally, they demonstrate impaired perception of biological motion and problems with motor control. Here we investigated whether individuals with autism move with an atypical kinematic profile, which might help to explain perceptual and motor impairments, and in principle may contribute to some of their higher level social problems. We recorded trajectory, velocity, acceleration and jerk while adult participants with autism and a matched control group conducted horizontal sinusoidal arm movements. Additionally, participants with autism took part in a biological motion perception task in which they classified observed movements as 'natural' or 'unnatural'. Results show that individuals with autism moved with atypical kinematics; they did not minimize jerk to the same extent as the matched typical control group, and moved with greater acceleration and velocity. The degree to which kinematics were atypical was correlated with a bias towards perceiving biological motion as 'unnatural' and with the severity of autism symptoms as measured by the Autism Diagnostic Observation Schedule. We suggest that fundamental differences in movement kinematics in autism might help to explain their problems with motor control. Additionally, developmental experience of their own atypical kinematic profiles may lead to disrupted perception of others' actions.

Movement kinematics drive chain selection toward intention detection.

Project description:The ability to understand intentions based on another's movements is crucial for human interaction. This ability has been ascribed to the so-called motor chaining mechanism: anytime a motor chain is activated (e.g., grasp-to-drink), the observer attributes to the agent the corresponding intention (i.e., to drink) from the first motor act (i.e., the grasp). However, the mechanisms by which a specific chain is selected in the observer remain poorly understood. In the current study, we investigate the possibility that in the absence of discriminative contextual cues, slight kinematic variations in the observed grasp inform mapping to the most probable chain. Chaining of motor acts predicts that, in a sequential grasping task (e.g., grasp-to-drink), electromyographic (EMG) components that are required for the final act [e.g., the mouth-opening mylohyoid (MH) muscle] show anticipatory activation. To test this prediction, we used MH EMG, transcranial magnetic stimulation (TMS; MH motor-evoked potentials), and predictive models of movement kinematics to measure the level and timing of MH activation during the execution (Experiment 1) and the observation (Experiment 2) of reach-to-grasp actions. We found that MH-related corticobulbar excitability during grasping observation varied as a function of the goal (to drink or to pour) and the kinematics of the observed grasp. These results show that subtle changes in movement kinematics drive the selection of the most probable motor chain, allowing the observer to link an observed act to the agent's intention.

A cryptography-based approach for movement decoding.

Project description:Brain decoders use neural recordings to infer the activity or intent of a user. To train a decoder, one generally needs to infer the measured variables of interest (covariates) from simultaneously measured neural activity. However, there are cases for which obtaining supervised data is difficult or impossible. Here, we describe an approach for movement decoding that does not require access to simultaneously measured neural activity and motor outputs. We use the statistics of movement-much like cryptographers use the statistics of language-to find a mapping between neural activity and motor variables, and then align the distribution of decoder outputs with the typical distribution of motor outputs by minimizing their Kullback-Leibler divergence. By using datasets collected from the motor cortex of three non-human primates performing either a reaching task or an isometric force-production task, we show that the performance of such a distribution-alignment decoding algorithm is comparable to the performance of supervised approaches. Distribution-alignment decoding promises to broaden the set of potential applications of brain decoding.

Purkinje cell microzones mediate distinct kinematics of a single movement.

Project description:The classification of neuronal subpopulations has significantly advanced, yet its relevance for behavior remains unclear. The highly organized flocculus of the cerebellum, known to fine-tune multi-axial eye movements, is an ideal substrate for the study of potential functions of neuronal subpopulations. Here, we demonstrate that its recently identified subpopulations of 9+ and 9- Purkinje cells exhibit an intermediate Aldolase C expression and electrophysiological profile, providing evidence for a graded continuum of intrinsic properties among PC subpopulations. By identifying and utilizing two Cre-lines that genetically target these floccular domains, we show with high spatial specificity that these subpopulations of Purkinje cells participate in separate micromodules with topographically organized connections. Finally, optogenetic excitation of the respective subpopulations results in movements around the same axis in space, yet with distinct kinematic profiles. These results indicate that Purkinje cell subpopulations integrate in discrete circuits and mediate particular parameters of single movements.

Simulating movement of tRNA into the ribosome during decoding.

Project description:Decoding is the key step during protein synthesis that enables information transfer from RNA to protein, a process critical for the survival of all organisms. We have used large-scale (2.64 x 10(6) atoms) all-atom simulations of the entire ribosome to understand a critical step of decoding. Although the decoding problem has been studied for more than four decades, the rate-limiting step of cognate tRNA selection has only recently been identified. This step, known as accommodation, involves the movement inside the ribosome of the aminoacyl-tRNA from the partially bound "A/T" state to the fully bound "A/A" state. Here, we show that a corridor of 20 universally conserved ribosomal RNA bases interacts with the tRNA during the accommodation movement. Surprisingly, the tRNA is impeded by the A-loop (23S helix 92), instead of enjoying a smooth transition to the A/A state. In particular, universally conserved 23S ribosomal RNA bases U2492, C2556, and C2573 act as a 3D gate, causing the acceptor stem to pause before allowing entrance into the peptidyl transferase center. Our simulations demonstrate that the flexibility of the acceptor stem of the tRNA, in addition to flexibility of the anticodon arm, is essential for tRNA selection. This study serves as a template for simulating conformational changes in large (>10(6) atoms) biological and artificial molecular machines.
