Project description:The first steps in numerical cognition are usually done in conjunction with fingers. Following the assumption that abstract concepts stay associated with the sensory-motor information that was present during their acquisition and consolidation, mental number representations should always be associated with the respective finger counting components. We tested whether finger movements that imply finger counting actually prime the corresponding number concepts in adults. All participants counted number 1 with their thumb and incremented sequentially to number 5 with their pinky. In the experiment, participants sequentially and repeatedly pressed five buttons from thumb to pinky. Each button press triggered the visual presentation of a random number between 1 and 5 that had to be named aloud, resulting in 20% counting-congruent and 80% counting-incongruent finger-number mappings. Average naming latencies were significantly shorter for congruent than incongruent finger-number combinations. Furthermore, there was a distance effect where primes partly co-activated numerically close target numbers and with decreasing priming for more distant prime-target pairs. Overall, these results provide further evidence that number representations are strongly associated with finger counting experience, making fingers an effective tool for number comprehension.

Project description:Thermodynamics constrains the flow of matter in a reaction network to occur through routes along which the Gibbs energy decreases, implying that viable steady-state flux patterns should be void of closed reaction cycles. Identifying and removing cycles in large reaction networks can unfortunately be a highly challenging task from a computational viewpoint. We propose here a method that accomplishes it by combining a relaxation algorithm and a Monte Carlo procedure to detect loops, with ad hoc rules (discussed in detail) to eliminate them. As test cases, we tackle (a) the problem of identifying infeasible cycles in the E. coli metabolic network and (b) the problem of correcting thermodynamic infeasibilities in the Flux-Balance-Analysis solutions for 15 human cell-type-specific metabolic networks. Results for (a) are compared with previous analyses of the same issue, while results for (b) are weighed against alternative methods to retrieve thermodynamically viable flux patterns based on minimizing specific global quantities. Our method, on the one hand, outperforms previous techniques and, on the other, corrects loopy solutions to Flux Balance Analysis. As a byproduct, it also turns out to be able to reveal possible inconsistencies in model reconstructions.

Project description:The Spatial Numerical Association of Response Code (SNARC) is the preferential association between smaller/larger magnitudes and left/right side, respectively. Some evidence suggest a link between SNARC and a left-to-right finger counting habit. We asked 268 participants to show how they use the hands to count from 1 to 10. By means of this ecological task, 80% of the sample use first the right hand (to count from 1 to 5) and the majority of them use a palm-up posture. In Experiment 2 (N = 46) right-starters were asked to categorize 1-to-5 magnitudes as even or odd, using the left and right hand. Stimuli were presented both as Arabic numbers and by means of left and right hand photographs in palm-up and palm-down posture. Results confirmed the expected SNARC effect in the Arabic condition. With hand images we found that right hand responses were better for larger than for smaller magnitudes (SNARC, mainly for left hand palm-up stimuli), showing that the SNARC can be generalized to different codes. Finally, the interactions between magnitudes and left/right hand images in palm-up and palm-down posture suggest that embodied cognition can influence numerical processing.

Project description:RNA-Seq measures gene expression by counting sequence reads belonging to unique cDNA fragments. Molecular barcodes commonly in the form of random nucleotides were recently introduced to improve gene expression measures by detecting amplification duplicates, but are susceptible to errors generated during PCR and sequencing. This results in false positive counts, leading to inaccurate transcriptome quantification especially at low input and single-cell RNA amounts where the total number of molecules present is minuscule. To address this issue, we demonstrated the systematic identification of molecular species using transposable error-correcting barcodes that are exponentially expanded to tens of billions of unique labels.We experimentally showed random-mer molecular barcodes suffer from substantial and persistent errors that are difficult to resolve. To assess our method's performance, we applied it to the analysis of known reference RNA standards. By including an inline random-mer molecular barcode, we systematically characterized the presence of sequence errors in random-mer molecular barcodes. We observed that such errors are extensive and become more dominant at low input amounts.We described the first study to use transposable molecular barcodes and its use for studying random-mer molecular barcode errors. Extensive errors found in random-mer molecular barcodes may warrant the use of error correcting barcodes for transcriptome analysis as input amounts decrease.

Project description:Human non-hairy (glabrous) skin of the fingers, palms and soles wrinkles after prolonged exposure to water. Wrinkling is a sympathetic nervous system-dependent process but little is known about the physiology and potential functions of water-induced skin wrinkling. Here we investigated the idea that wrinkling might improve handling of wet objects by measuring the performance of a large cohort of human subjects (n?=?40) in a manual dexterity task. We also tested the idea that skin wrinkling has an impact on tactile acuity or vibrotactile sensation using two independent sensory tasks. We found that skin wrinkling did not improve dexterity in handling wet objects nor did it affect any aspect of touch sensitivity measured. Thus water-induced wrinkling appears to have no significant impact on tactile driven performance or dexterity in handling wet or dry objects.

Project description:The embodied cognition framework suggests that neural systems for perception and action are engaged during higher cognitive processes. In an event-related fMRI study, we tested this claim for the abstract domain of numerical symbol processing: is the human cortical motor system part of the representation of numbers, and is organization of numerical knowledge influenced by individual finger counting habits? Developmental studies suggest a link between numerals and finger counting habits due to the acquisition of numerical skills through finger counting in childhood. In the present study, digits 1 to 9 and the corresponding number words were presented visually to adults with different finger counting habits, i.e. left- and right-starters who reported that they usually start counting small numbers with their left and right hand, respectively. Despite the absence of overt hand movements, the hemisphere contralateral to the hand used for counting small numbers was activated when small numbers were presented. The correspondence between finger counting habits and hemispheric motor activation is consistent with an intrinsic functional link between finger counting and number processing.

Project description:Although many three-dimensional pointing gesture recognition methods have been proposed, the problem of self-occlusion has not been considered. Furthermore, because almost all pointing gesture recognition methods use a wide-angle camera, additional sensors or cameras are required to concurrently perform finger gesture recognition. In this paper, we propose a method for performing both pointing gesture and finger gesture recognition for large display environments, using a single Kinect device and a skeleton tracking model. By considering self-occlusion, a compensation technique can be performed on the user's detected shoulder position when a hand occludes the shoulder. In addition, we propose a technique to facilitate finger counting gesture recognition, based on the depth image of the hand position. In this technique, the depth image is extracted from the end of the pointing vector. By using exception handling for self-occlusions, experimental results indicate that the pointing accuracy of a specific reference position was significantly improved. The average root mean square error was approximately 13 pixels for a 1920 × 1080 pixels screen resolution. Moreover, the finger counting gesture recognition accuracy was 98.3%.
