The unexpected finding of Parapholidoptera castaneoviridis in south-eastern Romania (Insecta, Orthoptera, Tettigoniidae).

ABSTRACT: The Balkano-Anatolian genus Parapholidoptera comprises 21 species and the westernmost one, Parapholidoptera castaneoviridis, previously recognized to occur in western Turkey, north-eastern Greece and south-eastern Bulgaria is recorded for the first time from south-eastern Romania, almost 300 km away from the closest known locality. Illustrations and measurements of morphological characters are given and the male calling song from this new, northernmost population is described.

Project description:Described from the steppe zones north of the Black Sea, Caucasus, and central Asia, Gryllotalpa stepposa Zhantiev was recently recorded from a few localities in Greece, R. Macedonia, and Bulgaria. In May 2015, several specimens were collected from Ivrinezu Mare in Romania, which suggested a continuous distribution area of the species, stretching from the central Balkans to central Asia. Thus, to reveal its actual range of occurrence, a survey of several Orthoptera collections became mandatory and, as expected, a large number of misidentified specimens of Gryllotalpa stepposa were discovered, providing new data on the species distribution in south-eastern Europe, including also the first records of this mole cricket in Serbia and Hungary. Here a full locality list is presented of this species west of Ukraine and Moldova and the current geographic distribution of the genus Gryllotalpa in the Balkans is revised. A key for distinguishing the mole crickets in south-eastern Europe and a distribution map for this region are presented.

Wolf diet and prey selection in the South-Eastern Carpathian Mountains, Romania.

Project description:The Romanian wolf population, one of the largest in Europe, occupies a total home-range of 154500 km2 and is spread across a variety of landscapes-from anthropized hills and plateaus to remote, densely forested mountains. However, this population is markedly understudied, and even basic knowledge of the species' feeding habits is deficient. Wolf diet was assessed based on 236 scat samples collected between November 2013 and October 2014, by following pre-established transects (total length = 774 km). The study area (600 km2) is a multi-prey ecosystem in the southern sector of the Eastern Romanian Carpathians. Our results emphasize that more than 80% of the wolf diet is based on wild ungulates. The wild boar is clearly selected (D = 0.74) and is the most common species in the diet (Bio = 72%), while roe deer (Bio = 10%) and red deer (Bio = 5%) have a smaller contribution. Domestic species represented the second-largest prey category in both seasons. Among them, dog is a particularly important source of food (Bio 3.5-10.9%). Other domestic species (goat, sheep, horse) have marginal importance in the wolf diet and seasonal occurrence. Standardized niche breadths are low in both seasons (BAw = 0.07, BAs = 0.12), and a high degree of overlap in the resources used has been observed (Ôws = 0.99). Our study represents the first step towards understanding the wolf foraging behaviour in the Romanian Carpathians and is valuable to address the complex issues of wolf and wild ungulate population management and conservation.

Genes expressed in the pleuropodia from embryos of the locust Schistocerca gregaria (Insecta, Orthoptera).

Project description:RNA-seq was used to study the genes expressed in peculiar glandular organs that transiently appear in insect embryos and are called the pleuropodia. The locust Schistocerca gregaria (Orthoptera) was used as a model. The purpose of the study is to identify the function of these organs. Our results suport the hypothesis (Slifer, 1937) that the pleuropodia secrete enzymes that digest the serosal cuticle before hatching. Additionally, we found that the pleuropodia may also have other functions, such as in embryonic immunity. The pleuropodia are peculiarly modified limbs. We show that in their early stages both legs and pleuropodia share a similar genetic landscape, but as the appendages become more morphologically diverse they became also more diverse genetically.

The Evolutionary Patterns of Genome Size in Ensifera (Insecta: Orthoptera).

Project description:Genomic size variation has long been a focus for biologists. However, due to the lack of genome size data, the mechanisms behind this variation and the biological significance of insect genome size are rarely studied systematically. The detailed taxonomy and phylogeny of the Ensifera, as well as the extensive documentation concerning their morphological, ecological, behavioral, and distributional characteristics, make them a strong model for studying the important scientific problem of genome size variation. However, data on the genome size of Ensifera are rather sparse. In our study, we used flow cytometry to determine the genome size of 32 species of Ensifera, the smallest one being only 1C = 0.952 pg with the largest species up to 1C = 19.135 pg, representing a 20-fold range. This provides a broader blueprint for the genome size variation of Orthoptera than was previously available. We also completed the assembly of nine mitochondrial genomes and combined mitochondrial genome data from public databases to construct phylogenetic trees containing 32 species of Ensifera and three outgroups. Based on these inferred phylogenetic trees, we detected the phylogenetic signal of genome size variation in Ensifera and found that it was strong in both males and females. Phylogenetic comparative analyses revealed that there were no correlations between genome size and body size or flight ability in Tettigoniidae. Reconstruction of ancestral genome size revealed that the genome size of Ensifera evolved in a complex pattern, in which the genome size of the grylloid clade tended to decrease while that of the non-grylloid clade expanded significantly albeit with fluctuations. However, the evolutionary mechanisms underlying variation of genome size in Ensifera are still unknown.

Rampant nuclear insertion of mtDNA across diverse lineages within Orthoptera (Insecta).

Project description:Nuclear mitochondrial pseudogenes (numts) are non-functional fragments of mtDNA inserted into the nuclear genome. Numts are prevalent across eukaryotes and a positive correlation is known to exist between the number of numts and the genome size. Most numt surveys have relied on model organisms with fully sequenced nuclear genomes, but such analyses have limited utilities for making a generalization about the patterns of numt accumulation for any given clade. Among insects, the order Orthoptera is known to have the largest nuclear genome and it is also reported to include several species with a large number of numts. In this study, we use Orthoptera as a case study to document the diversity and abundance of numts by generating numts of three mitochondrial loci across 28 orthopteran families, representing the phylogenetic diversity of the order. We discover that numts are rampant in all lineages, but there is no discernable and consistent pattern of numt accumulation among different lineages. Likewise, we do not find any evidence that a certain mitochondrial gene is more prone to nuclear insertion than others. We also find that numt insertion must have occurred continuously and frequently throughout the diversification of Orthoptera. Although most numts are the result of recent nuclear insertion, we find evidence of very ancient numt insertion shared by highly divergent families dating back to the Jurassic period. Finally, we discuss several factors contributing to the extreme prevalence of numts in Orthoptera and highlight the importance of exploring the utility of numts in evolutionary studies.

Trade-off between flight capability and reproduction in Acridoidea (Insecta: Orthoptera).

Project description:In many insect taxa, there is a well-established trade-off between flight capability and reproduction. The wing types of Acridoidea exhibit extremely variability from full length to complete loss in many groups, thus, provide a good model for studying the trade-off between flight and reproduction. In this study, we completed the sampling of 63 Acridoidea species, measured the body length, wing length, body weight, flight muscle weight, testis and ovary weight, and the relative wing length (RWL), relative flight muscle weight (RFW), and gonadosomatic index (GSI) of different species were statistically analyzed. The results showed that there were significant differences in RWL, RFW, and GSI among Acridoidea species with different wing types. RFW of long-winged species was significantly higher than that of short-winged and wingless species (p < .01), while GSI of wingless species was higher than that of long-winged and short-winged species. The RWL and RFW had a strong positive correlation in species with different wing types (correlation coefficient r = .8344 for male and .7269 for female, and p < .05), while RFW was strong negatively correlated with GSI (r = -.2649 for male and -.5024 for female, and p < .05). For Acridoidea species with wing dimorphism, males with relatively long wings had higher RFW than that of females with relatively short wings, while females had higher GSI. Phylogenetic comparative analysis showed that RWL, RFW, and GSI all had phylogenetic signals and phylogenetic dependence. These results revealed that long-winged individuals are flight capable at the expense of reproduction, while short-winged and wingless individuals cannot fly, but has greater reproductive output. The results support the trade-off between flight and reproduction in Acridoidea.

Re-visiting phylogenetic and taxonomic relationships in the genus Saga (Insecta: Orthoptera).

Project description:Twelve of the 13 bushcricket species of the Saga genus are bisexuals and diploids, except the parthenogenetic and tetraploid bush cricket, Saga pedo. Despite a continuous research effort stretching through the 1900s, the taxonomic relationships of the Saga species are still disputed. In this study, our primary aim was to reveal natural relationships of the European Saga species and three of their Asian relatives, with special attention to the problematic taxonomy of two subspecies: S. campbelli campbelli and S. c. gracilis. Following a phylogenetic analysis of eight species, a comprehensive study was carried out on the above three taxa by using acoustic and morphometric approaches in parallel. Our phylogenetic data showed that European Saga species evolved from a monophyletic lineage. The geographical transitional species S. cappadocica was positioned between European and Asian lineages supporting the idea that the European Saga lineage originated phylogeographically from the Asian clade. The above results showed better agreement with the morphological data than with earlier ones based either on karyology or acoustic information only. After reviewing our data, we concluded that Saga pedo has most likely evolved from S. c. gracilis and not from S. rammei or S. ephippigera, as proposed by earlier studies. S. c. gracilis shares the same ITS2 haplotype with S. pedo, indicating that the latter could have evolved from populations of the former, probably through whole genome duplication. Based on acoustic and morphometric differences, we propose to elevate the two subspecies, S. campbelli campbelli and S. c. gracilis, to species level status, as Saga gracilis Kis 1962, and Saga campbelli Uvarov 1921. The present work sets the stage for future genetic and experimental investigations of Saginae and highlights the need for additional comprehensive analysis involving more Asian Saga species.

Narrow Genetic Diversity of Wolbachia Symbionts in Acrididae Grasshopper Hosts (Insecta, Orthoptera).

Project description:Bacteria of the Wolbachia genus are maternally inherited symbionts of Nematoda and numerous Arthropoda hosts. There are approximately 20 lineages of Wolbachia, which are called supergroups, and they are designated alphabetically. Wolbachia strains of the supergroups A and B are predominant in arthropods, especially in insects, and supergroup F seems to rank third. Host taxa have been studied very unevenly for Wolbachia symbionts, and here, we turn to one of largely unexplored insect families: Acrididae. On the basis of five genes subject to multilocus sequence typing, we investigated the incidence and genetic diversity of Wolbachia in 41 species belonging three subfamilies (Gomphocerinae, Oedipodinae, and Podisminae) collected in Turkey, Kazakhstan, Tajikistan, Russia, and Japan, making 501 specimens in total. Our results revealed a high incidence and very narrow genetic diversity of Wolbachia. Although only the strains belonging to supergroups A and B are commonly present in present, the Acrididae hosts here proved to be infected with supergroups B and F without A-supergroup variants. The only trace of an A-supergroup lineage was noted in one case of an inter-supergroup recombinant haplotype, where the ftsZ gene came from supergroup A, and the others from supergroup B. Variation in the Wolbachia haplotypes in Acrididae hosts within supergroups B and F was extremely low. A comprehensive genetic analysis of Wolbachia diversity confirmed specific features of the Wolbachia allelic set in Acrididae hosts. This result can help to elucidate the crucial issue of Wolbachia biology: the route(s) and mechanism(s) of Wolbachia horizontal transmission.

Sand fly fauna of South-Eastern Romania, with the description of Phlebotomus (Transphlebotomus) simonahalepae n. sp. (Diptera: Psychodidae).

Project description:BackgroundAn entomological study was conducted in the Canaraua Fetii Special Protection Area in the Dobrogea region, South-Eastern Romania. Four sand fly species were recorded at this location between 1968 and 1970: Phlebotomus neglectus, Ph. balcanicus, Ph. sergenti and Sergentomyia minuta. The most abundant sand fly species recorded at that time were Ph. balcanicus and Se. minuta. In the context of a countrywide study to update the sand fly species diversity, we surveyed the same area, recording also a previously unknown Ph. (Transphlebotomus) sp., for which we provide a formal description here.MethodsSand flies were collected between July and August in 2018 and 2019 in three sites from Canaraua Fetii, Dobrogea region, Romania. The general aspect of the landscape is of a canyon (vertical, narrow walls and deep valleys). Species identification was done using both morphological and molecular analyses.ResultsOut of 645 collected sand flies, 644 (99.8%) were morphologically identified as Ph. neglectus, while one female specimen (0.2%) was assigned to a previously unknown species, belonging to the subgenus Transphlebotomus. The morphological and molecular examination of this specimen showed that it is a previously unknown species which we formally describe here as Phlebotomus (Transphlebotomus) simonahalepae n. sp. Also, Ph. balcanicus, Ph. sergenti, and Se. minuta (previously recorded in this location) were not present.ConclusionsThe study revealed for the first time the presence of sand flies of the subgenus Transphlebotomus in Romania. Moreover, a new species, Ph. simonahalepae n. sp., was described based on a female specimen, raising the number of species in this subgenus to six. In the investigated natural habitat, the predominant species was Ph. neglectus instead of Ph. balcanicus and Se. minuta (recorded as the predominant species in 1968-1970).
